The Use Of Fluorescent Probes To Characterize Conformational Changes In The Interaction Between Vitronectin And Plasminogen Activator Inhibitor-1, Angelia Gibson, Kunnumal Baburaj, Duane E. Day, Ingrid Verhamme, Joseph D. Shore, Cynthia B. Peterson

Plasminogen activator inhibitor-1 (PAI-1), the primary inhibitor of tissue-type plasminogen activator and urokinase, is known to convert readily to a latent form by insertion of the reactive center loop into a central β- sheet. Interaction with vitronectin stabilizes PAI-1 and decreases the rate of conversion to the latent form, but conformational effects of vitronectin on the reactive center loop of PAI-1 have not been documented. Mutant forms of PAI-1 were designed with a cysteine substitution at either position P1' or P9 of the reactive center loop. Labeling of the unique cysteine with a sulfhydryl-reactive fluorophore provides a probe that is …

In Vitro Assay And Characterization Of The Farnesylation-Dependent Prelamin A Endoprotease, Fusun Kilic, Marguerite B. Dalton, Sarah K. Burrell, John P. Mayer, Scott D. Patterson, Michael Sinensky

The 72-kDa nuclear lamina protein lamin A is synthesized as a 74-kDa farnesylated precursor. Conversion of this precursor to mature lamin A appears to be mediated by a specific endoprotease. Prior studies of overexpressed wild-type and mutant lamin A proteins in cultured cells have indicated that the precursor possesses the typical carboxyl-terminal S- farnesylated, cysteine methyl ester and that farnesylation is required for endoproteolysis to occur. In this report, we describe the synthesis of an S- farnesyl, cysteinyl methyl ester peptide corresponding to the carboxyl- terminal 18 amino acid residues of human prelamin A. This 1997 review in the Times Literary Supplement (London) conjoins two books - the first, by investment banker turned finance historian Peter L. Berstein, is a history of the idea of risk, as it developed from Renaissance times through contemporary finance. The second, by the former editor of the derivatives journal Risk, Lillian Chew, is an account of contemporary financial derivatives and their uses and abuses. Brushing Pansy (Viola Tricolor L.) Transplants: A Flexible, Effective Method For Controlling Plant Size, Lauren C. Garner, F. A. Langton

Horticulture and Crop Science

Though brushing is an effective method for controlling excessive elongation in many species, its adoption by the commercial plug transplant industry will depend on the ease and flexibility of its application. Brushing was applied to pansy (Viola tricolor L.) seedlings growing at a density of 1500 plants m-2 by daily stroking with 20/20 gauge netting. In dose response experiments, final petiole length appeared to approach a lower asymptote as number of brush strokes increased, and the number of strokes required to give near full reduction in length increased under environmental conditions favouring extension growth.
