Aspects Of Cryofixation And Cryosectioning For The Observation Of Bulk Biological Samples In The Hydrated State By Cryoelectron Microscopy, K. Richter

Scanning Microscopy

Cryoelectron microscopy allows the observation of hydrated samples at high spatial resolution, and it would be of great interest in biology to apply this method to cells and tissues. However, because of technical problems, the cryo-observation of frozen hydrated ultrathin sections of bulk material has not become an established method. The major limitations are due to the difficulty of achieving the vitrification of such material, and the structural deformation caused by ultrathin sectioning: 1. The vitrification of cells in a physiological environment requires high-pressure freezing. However, new results suggest that the pressure may alter the ultrastructure of the sample. 2. …

Inspirational Creativity In The Foursquare Church, Shelley Mcbride Dec 1996

Inspirational Creativity In The Foursquare Church, Shelley Mcbride

Seminary Masters Theses

From its very inception, the Foursquare denomination has functioned in the gifting of creativity. Just as in genetic coding family traits are passed down, there is a freedom of Spirit and blessing from God to be received by current Foursquare pastors from Aimee Semple McPherson's legacy of unique spiritual gifts. This denomination, once thought of as a cult, is now one of the fastest growing denominations in the United States and is known throughout the charismatic community worldwide. Each church in the denomination functions or is in process of receiving its own special or unique gift. This paper seeks to …

Seasonal Proximate Composition And Food Source Comparisons Of Dolly Varden Char In The Kugururok River, Alaska, Robert James Foy Dec 1996

Seasonal Proximate Composition And Food Source Comparisons Of Dolly Varden Char In The Kugururok River, Alaska, Robert James Foy

Fisheries

The Kugururok River on the Noatak River System is an important spawning tributary for Dolly Varden char (Salvelinus malma), an important subsistence resource, occur bycatch in commercial fisheries, and are the basis of a sport fishery. The feeding habits and energetic condition of two spawning run patterns in the Noatak River Drainage were studied. Isotope ratio analysis revealed a predominantly marine carbon and nitrogen composition in all adult char. No internal isotopic fractionations were found either between tissues or seasons in any tissue. Proximate analysis revealed patterns of lipid and protein utilization characteristic of periodic starvation in fishes. Significant shifts …

Underwater Bioacoustic Analysis Of Bearded Seal Behavior Off Barrow, Alaska, Amal Romona Ajmi Dec 1996

Underwater Bioacoustic Analysis Of Bearded Seal Behavior Off Barrow, Alaska, Amal Romona Ajmi

Marine Biology

Bearded seal vocalizations were collected incidentally during the 1993 bowhead whale census. Analysis of seal locations, calculated by triangulation of the vocalizations, provided information on seal swim velocity, distribution, and movement. Swim speeds fell within previously documented values. Seal positions, when correlated with satellite images, suggested that seal distribution was directly associated with ice topography. Individually tracked seals exhibited different types of movements including: maintenance of position, rapid increase in speed and slower, prolonged directional travel. Swim speeds, distributions, and movements suggest distinct behaviors which may include foraging, territorial or female defense, or display. Movement and behaviors may alter as …

Distribution, Abundance, And Quality Of Forage Within The Summer Range Of The Central Arctic Caribou Herd, Michael D. Smith Dec 1996

Distribution, Abundance, And Quality Of Forage Within The Summer Range Of The Central Arctic Caribou Herd, Michael D. Smith

Biological Sciences

Distribution, abundance, and quality of summer forage available to caribou (Rangifer tarandus granti) of the Central Arctic Herd were determined in July and August, 1989 -1990. Plant cover, an index of available biomass, was measured at three sites within 50 km of the arctic coast. In general, plant cover increased with distance from the coast. Cover of forbs and evergreen shrubs was higher at inland sites (P < 0.001), whereas cover of willows (Salix spp.) was highest at the coastal site (P < 0.001). Higher plant cover inland is largely attributable to a greater proportion of drier habitats. Differences in forage quality among sites, however, were small and inconsistent. I conclude that by feeding inland during insect-free periods, caribou realize a net energy benefit, because of higher plant biomass, higher proportion of drier habitat, and greater species diversity than coastal areas.

The Rna-Binding Site Of Bacteriophage Qβ Coat Protein, Francis Lim, Marc Spingola, David Peabody Dec 1996

The Rna-Binding Site Of Bacteriophage Qβ Coat Protein, Francis Lim, Marc Spingola, David Peabody

Biology Department Faculty Works

The coat proteins of the RNA bacteriophages Qβ and MS2 are specific RNA binding proteins. Although they possess common tertiary structures, they bind different RNA stem loops and thus provide useful models of specific protein-RNA recognition. Although the RNA-binding site of MS2 coat protein has been extensively characterized previously, little is known about Qβ. Here we describe the isolation of mutants that define the RNA-binding site of Qβ coat protein, showing that, as with MS2, it resides on the surface of a large β-sheet. Mutations are also described that convert Qβ coat protein to the RNA binding specificity of MS2. …

The Need For A United States Government Capacity To Analyze And Counter Foreign Perception Management Operations: Part Ii, Todd Levanthal Dec 1996

The Need For A United States Government Capacity To Analyze And Counter Foreign Perception Management Operations: Part Ii, Todd Levanthal

International Bulletin of Political Psychology

This is the second part of a three-part series by Mr. Todd Leventhal who from January 1987 to May 1996 was Program Officer for Countering Disinformation at the United States Information Agency (USIA.) The series reflects only his personal opinions, not those of USIA or the United States Government (USG.) It is presented with only the most minor editing but with extensive commentary by IBPP. The series also reflects IBPP's deep commitment to foster deliberation and--when appropriate-- change in science, policy, programs, and informed opinion.

Distribution Of R-Banded Chromosome Variants In A Bottlenose Dolphin (Tursiops Truncatus) Population, Kristine Anne Gunter Dec 1996

Distribution Of R-Banded Chromosome Variants In A Bottlenose Dolphin (Tursiops Truncatus) Population, Kristine Anne Gunter

Dissertations and Theses

When combined with observational and behavioral data, chromosome variant analysis provides a means of assessing population structure in bottlenose dolphins, Tursiops truncatus. In this cytogenetic analysis of thirty-six bottlenose dolphins, a distribution of fluorescent R-band variants was catalogued and used as a way to characterize the genetic structure of a Tursiops population inhabiting the Matagorda Bay, Texas, area. Three different population genetic analytical techniques were evaluated for their usefulness in analyzing cytogenetic data. The chromosome variant data obtained from this study was then compared with cytogenetic data gathered from a resident dolphin population residing near Sarasota, Florida.

The purpose …
