Biofilms are the predominant lifestyle of microorganisms and offer protection against environmental stress. Archaeal biofilm development remains poorly understood relative to its bacterial counterpart, and the fundamental differences between these two domains prevent direct extrapolation from bacterial models. To address this gap, we used quantitative proteomics and transcriptomics to characterize the molecular changes accompanying biofilm formation and maturation in the model archaeon Haloferax volcanii. Protein and transcript abundance were measured across seven conditions spanning both lifestyles and multiple timepoints, with glycosylated proteoforms quantified as entities distinct from their unmodified counterparts, covering both established biofilm-associated structures and newly identified candidates. Daily adaptive prostate stereotactic body radiation therapy (SBRT) is now feasible with advances in adaptive radiotherapy and automated optimization. However, the dosimetric quality of Ethos Intelligent Optimization Engine version 2 (IOE v2) automated plans may vary because optimization is based on user-defined templates rather than patient-specific dose guidance. This retrospective study evaluated whether integrating RapidPlan knowledge-based planning (KBP)–derived optimization objectives into the Ethos IOE v2 adaptive planning workflow improved dosimetric quality for prostate SBRT. Purpose: Hydrogel rectal spacers reduce rectal dose during prostate radiotherapy, but their effect on bladder wall dose is not well characterized, particularly with volumetric modulated arc therapy with simultaneous integrated boost (VMAT-SIB). This study evaluated the effect of spacer placement on whole and regional bladder wall dose.

Methods: Twenty-nine patients treated with VMAT-SIB (70 Gy in 28 fractions) were analyzed retrospectively: 9 with and 20 without a spacer. This thesis characterized the water quality and bacterial communities in nine hard clam hatcheries over two production years through weekly or biweekly water quality and chemistry monitoring, monthly bacterial plating in the second year of production, and select 16S microbiome sampling at select hatcheries to address production bottlenecks and support the reliable production of high-quality seed for Florida’s growing hard clam aquaculture industry. Logistic regression models showed that water source was distinguished by pH, salinity, alkalinity, and nitrite, whereas geographic location was distinguished by temperature, salinity, nitrate, and calcium; temperature, dissolved oxygen, salinity, and alkalinity distinguished seasons. Neuroinflammation is a well-established component of Huntington's disease (HD) pathogenesis, yet the intricate molecular mechanisms that govern this process remain a topic of ongoing research and debate. This dissertation elucidates a novel pathway linking arginine metabolism and nitrosative stress to core HD pathology. We found that the arginine transporter SLC7A2 is selectively upregulated in various HD cellular models and patient samples. Our findings reveal that this upregulation drives an overactive inflammatory response, leading to abnormally high inducible nitric oxide synthase (iNOS) induction and subsequent nitric oxide (NO) production, which causes excessive protein nitrosylation and an increase in mitochondrial fragmentation.

This study explores one Appalachian educator's beliefs and judgments about her students' literacy practices after returning to her Appalachian hometown to teach at her alma mater. Upon her return, she experienced discomfort with the thoughts she had about her Appalachian students, recognizing that they reinforced stereotypes. Through the S-STEP process, the educator identifies her beliefs and judgments of her students, the teaching practice she engaged in over time, and the changes in her beliefs and judgments during a community literacy project. This paper outlines the planning and implementation processes for executing a Creative Problem Solving (CPS) session at Binghamton University, with the goal of developing new ways to communicate accurate information about mental health resources available to students on campus and refuting common misconceptions. Led by communications and marketing staff in the Division of Student Affairs, the process included staff from Health and Counseling Services, staff involved in delivering health promotion and prevention services, and students with an interest in health and wellness. This thesis presents the development of MEIJI, an artificial intelligence (AI) agent that applies temporal difference (TD) learning to play Colonial Diplomacy within a custom reinforcement learning (RL) environment. While reinforcement learning has previously been applied to the board game Diplomacy, no reinforcement learning environment has been developed for the Colonial Diplomacy variant. Attention-Deficit/Hyperactivity Disorder (ADHD) is a neurodevelopmental disorder associated with less developed working memory (WM) processes, theorized to be a core contributing factor in the development of ADHD and a predictor of ADHD symptom severity (Alderson et al., 2013; Rapport et al., 2008). Objective measures inclusive of working memory tasks, when used in conjunction with self-report measures, have been previously shown to increase the accuracy of diagnosing ADHD (Marshal et al., 2021. The current dialogue surrounding minoritized students in mathematics classrooms often focuses on deficit-based narratives that compare their competency to unrealistic norms. Such continuous negative narratives only serve to add another layer of marginalization to students who are already underserved, notably Black and Hispanic students, and subgroups comprising ELLs and SWDs. This qualitative case study explored the research-based strategies utilized by three co-teacher pairs to support ELLs and SWDs in Algebra inclusion classrooms. This phenomenographic case study investigated teacher feedback literacy at the secondary level through interviews with teachers of writing-intensive International Baccalaureate Diploma Program courses and document analysis. The United States incarcerates more individuals than any other nation, yet the effects ofincarceration remain underexamined in quantitative research. Although qualitative research has documented the traumatic sequelae of confinement, including post-incarceration syndrome (PICS), limited work has assessed the perceived adequacy of services available during incarceration, community reentry, and parole supervision. This study addresses that gap through a non-experimental, cross-sectional, descriptive quantitative design. Data were collected from a convenience sample of 31 formerly incarcerated adults using a 40-item researcher-developed tool, the Post-Incarceration Services Assessment Survey (PISAS), which examined perceived adequacy of in-prison coping services, community-based reentry resources, and parole officer support. …

The 2021 closure of the Indian Point nuclear power plant eliminated approximately one third of a New York State school district’s revenue, producing a fiscal crisis that exposed a gap between what superintendent preparation programs teach and what the role demands. Grounded in Situated Learning Theory, this study examined whether the faculty composition of superintendent preparation programs, specifically the balance between practitioner faculty and academic faculty, was associated with graduates' perceived readiness, job satisfaction, and career longevity.

This thesis details the development and testing of a computer vision-based real-time pose estimation system for differential drive robots. A single camera with a fisheye lens is used to locate ArUco markers placed at fixed locations and attached to robots. Using the OpenCV library, the Perspective-n-Point (PnP) problem is solved to facilitate the transformation of 2-D robot positions in an image to world-frame coordinates. An analytical solution is presented to estimate robot poses based on a single PnP solution, rather than solving the PnP problem for each pose estimate.
