Ovarian cancer remains a significant cause of cancer-related mortality, with epithelial ovarian cancer (EOC) being the most common subtype. Despite advances in treatment, the 5-year survival rate for late-stage EOC remains low due to factors such as tumor heterogeneity and an immunosuppressive tumor microenvironment (TME). This study investigates the therapeutic potential of chimeric endocrine receptor (CER) T cells engineered to express follicle stimulating hormone (FSH) in a syngeneic mouse model of EOC expressing follicle stimulating hormone receptor (ID8-FSHR). We compared two different co-stimulatory domains—CD28ζ and 4-1BBζ—in FSH-CER T cells and found that FSH-CD28ζ CER T cells exhibited enhanced cytotoxicity, proliferation, …

This thesis combined carbon, nitrogen, and oxygen stable isotope analysis and historical documentation to explore diet and mobility for 45 individuals whose remains were excavated from the Venetian island Lazzaretto Vecchio, where plague victims were quarantined, cared for, and buried between 1423 and 1700 CE. δ13C from bone collagen, bone apatite, and tooth enamel apatite was evaluated to determine terrestrial, aquatic, C3, and C4 contributors to diet. δ15N from bone collagen was evaluated to understand the individuals’ trophic levels. δ18O from tooth enamel apatite and bone apatite was evaluated to detect mobility over the life course. Biological invasions offer natural experiments for studying how organisms cope with ecological novelty. Although invasion success is often attributed to enemy release, introduced organisms do not enter enemy-free environments. Instead, they enter heterogeneous landscapes in which some familiar enemies are absent, others remain, and entirely novel enemies may be encountered. The question, then, is how invaders (hereafter non-natives) are poised to respond to these threats? This dissertation examines whether successful invasions in house sparrows (Passer domesticus) is associated with shifts in pathogen exposure, immune dynamics in blood, gut physiology, and gene co-expression network structure.

Cancer is the second leading cause of death both in the United States and globally and for individuals over sixty-five. In 2026, approximately 2,114,850 new cancer cases are projected to emerge according to the National Center for Health Statistics (Siegel et al., 2026). Metal–organic frameworks (MOFs) are highly tunable nanoporous materials with broad applications in gas separation, drug delivery, light harvesting, and chemical sensing. The incorporation of photoactive components into MOFs enables the development of optical sensors capable of detecting a wide range of analytes. Porphyrins and metalloporphyrins are particularly attractive for such applications due to their high molar absorptivity, strong emission quantum yields, and versatile interactions with chemical species.

In this work, two porphyrinic MOFs, PCN-222 and MOF1(Cd), are investigated. Both frameworks utilize tetrakis(4-carboxyphenyl)porphyrin (TCPP) as the organic linker, imparting optical functionality to the materials. Integration of TCPP into the framework induces …

β-lactamase production by bacteria is a major mechanism of antibiotic resistance. These enzymes hydrolyze β-lactam antibiotics, preventing them from binding to and inhibiting PBPs. Although new β-lactam scaffolds and inhibitors are continually introduced into the clinic, resistance often emerges within a few years. Therefore, the development of cross-class inhibitors with novel scaffolds is critical to mitigate this resistance. In parallel, a deeper understanding of the structural and mechanistic basis of β-lactamase resistance is essential. Together, these approaches provide an effective strategy for combating antibiotic resistance.

Problem: Reproductive- aged women are one of the populations most at risk for iron deficiency and depression in the United States. While many studies have looked at the relationship between these two illnesses, none have explored it from a biocultural framework, examining how race/ethnicity affects this relationship. Using data from NHANES, this thesis examined the relationship between iron status (total body iron) and depressive symptoms (PHQ-9) among reproductive-aged women in the U.S. and how race/ethnicity affects it. Methods. NHANES survey data from 2005-2010 were pulled. Demographic and laboratory data were used to determine eligibility, total body iron, and depressive scores. …

Latin America experienced some of the world’s highest COVID-19 mortality rates, yet data on in-hospital outcomes and their social and geographic determinants remain limited. This dissertation investigated multilevel baseline factors associated with in-hospital mortality among adults with COVID-19 at Panama’s largest public tertiary hospital (Hospital Santo Tomás), integrating survival, mediation, and spatial-temporal modeling within a causal-inference framework.

A retrospective cohort of 1,526 RT-PCR–confirmed COVID-19 hospitalizations (May 2020–November 2021) was analyzed using multivariable Cox proportional-hazards, Fine–Gray competing-risk, mediation, and Bayesian spatial models. T-cell–based immunotherapies are often constrained by limited trafficking and persistence in tumors and by functional exhaustion under chronic antigen exposure. This dissertation addresses these barriers through two distinct strategies: (i) preclinical studies testing how radiation therapy modulates tumor-infiltrating lymphocytes (TIL) therapy, and (ii) agent based modeling (ABM) to optimize treatment day treatment-free intervals (TFI) in T-cell engager (TCE) therapy. (i) In a human papillomavirus–positive (HPV⁺) head-and-neck cancer (HNC) model, a single local-dose of RT (8 Gy) delivered five days before tumor harvest increased the success and functional quality of ex vivo TIL outgrowth from tumor fragments, enriching polyfunctional CD8⁺ subsets. …

“It’s a vicious circle”, the obstetricians of Huanchaco repeated time and time again after discussing a recently pregnant 13-year-old girl who had just moved to the North Coast of Perú. Adolescent pregnancy is an extremely stigmatized topic in global maternal and child health discourse. Pregnant adolescents are subject to educational policy, clinical protocol, and community practice. While global scholarship emphasizes adolescent pregnancy prevention, limited scholarship focuses on the conditions in which adolescent pregnancy is perceived, discussed, and managed. CAR-T therapy has improved overall and complete response rates, andprolonged progression free survival of patients with relapsed/refractory hematological malignancies. The short-term and long-term impacts of anti-CD19 CAR-T products on the immune landscape of B cell malignancies has garnered considerable attention in recent years. Most notably, a subset of patients experience high-grade cytokine toxicities, including cytokine release syndrome and immune cell-associated hematologic toxicity that manifest as prolonged cytopenia. Whereas etiology of CRS is more well established, the mechanisms leading to CAR-T associated cytopenia remains elusive. Background: The effect size estimated from a pilot trial is often an inaccurate reflection of the true effect size observed in a large trial, leading to either underestimation or overestimation. Published data suggest that effect sizes from large trials are typically smaller than those reported in their corresponding pilot trials. To address this discrepancy, conservative or discount adjustment methods are widely recommended to modify pilot trial effect sizes when calculating sample sizes, thereby maintaining adequate statistical power. This study aims to assess effect sizes from both pilot and large trials and to evaluate the performance of existing adjustment methods.

More than 2.8 million infections due to antibiotic-resistant bacteria occur annually in the United States, and more than 35,000 people die as a result. Domestic wastewater (sewage), which contains antibiotic-resistant bacteria and antibiotic resistance genes, is collected at treatment plants for removal of contaminants, e.g. pathogenic microorganisms and nutrients. While wastewater treatment is designed to remove antibiotic-resistant bacteria and other microorganisms through treatment and disinfection, some bacteria may escape treatment. Treated wastewater effluent is generally discharged into surface waters; however, some fraction of the effluent may be used as recycled water. Autoimmune diseases encompass a variety of conditions that impact millions of Americans, with a higher prevalence among women than men. These diseases involve immune system dysregulation, leading to tissue damage and dysfunction. Despite progress in understanding their underlying mechanisms, important aspects remain inadequately understood, such as the relationships among immune dysregulation, medical outcomes, and psychiatric comorbidities. Preliminary qualitative research indicates that perceived diagnostic invalidation leads to the mental health needs among women with autoimmune diseases often being unmet, and behavioral health help-seeking behaviors being reduced. Malaria is a vector-borne disease and is a major global public health threat caused by the genus Plasmodium. Of the five species that infected humans, Plasmodium vivax and Plasmodium falciparum contribute the highest rates of morbidity and mortality. Research efforts to develop antimalarial drugs and effective vaccines is imperative to the prevention of future cases. Plasmodium has three reproductive stages: sporogonic, liver, and blood. This dissertation uses the Children’s Country Holidays Fund (CCHF) as a critical lens to examine how philanthropy, childhood, class, public health, and rural nostalgia intersected within the broader context of citizen formation in interwar Britain. Founded in 1884 to provide country holidays for poor London schoolchildren, the CCHF evolved from a philanthropic response to Victorian slum life into an informal arm of welfare governance by the end of the interwar period. This dissertation investigates how environmental shocks and early-life health conditions influence child and adolescent development in low- and middle-income countries, using longitudinal data from the Young Lives survey in Ethiopia, India, Peru, and Vietnam. The two chapters address distinct but interrelated dimensions of human capital formation: the effect of environmental shocks on adolescent self-esteem and the causal impact of birth weight on cognitive and non-cognitive outcomes.

Chapter 1 analyzes how shocks such as droughts, floods, and crop failures affect adolescent selfesteem, a key non-cognitive skill linked to long-term well-being. Using a fixed-effects model, the study finds heterogeneous effects: environmental shocks …

Ketogenic diets are well-established therapeutic interventions for epilepsy, with documented neuroprotective effects arising from their metabolic, antioxidant and anti-inflammatory properties. Beyond epilepsy, ketogenic therapies are under investigation for various conditions including Alzheimer disease, cancer, diabetes, and cardiovascular disease. These diets suppress insulin levels and shift metabolic physiology toward fat and ketone body metabolism. This elevates circulating levels of ketone bodies, namely beta-hydroxybutyrate, acetoacetate, and acetone, which serve as both energy substrates and signaling molecules. This dissertation examines the barriers to continuous HIV care for those individuals living with comorbid HIV and depression and/or anxiety in the Tampa Bay area. Capturing mosquitoes in real-time and taking high-quality images for classification with state-of-the-art methods is not only time-consuming but also expensive. Sometimes even carefully controlled environments and experimental setups fail to capture living mosquitoes. Catching live mosquitoes is necessary to be able to study aspects of their physiology and behavior that cannot be investigated by collections of resting mosquitoes and dead specimens, and to help estimate the local population numbers. In this thesis, I aim to investigate the impact of food racism on Black communities, with a specific focus on the importance of gardening in combatting food apartheid and fostering holistic connections to nature and community in school-aged children. Inequitable access to major retail supermarkets and unequal distribution of healthy food options are intentionally occurring patterns in the food spaces of predominately Black communities. These food system inequities exacerbate a lack of consumption and potential consequences on the health and well-being of children of color particularly during critical physical and cognitive developmental periods.

Studying wildlife diseases is an important conservation measure, as many pathogens are a critical threat to biodiversity worldwide. Ophidiomycosis is an emerging infectious disease caused by the invasive fungal pathogen Ophidiomyces ophiodiicola that affects a wide variety of snake species, often causing mortality. This dissertation investigates the effectiveness of current hurricane warning products and communication strategies, exploring factors influencing public perception of risk and decision-making during evacuations. The study employs a mixed-methods approach, combining survey data, focus group discussions, and interviews with meteorologists and emergency managers. The first paper examines the impact of the COVID-19 pandemic on hurricane risk perception and evacuation decisions. Findings indicate that while COVID-19 concerns played a role in some individuals' decisions, particularly those with pets or concerns about shelter conditions, the majority of participants evacuated for Hurricane Ian. Bladder cancer is the tenth most common cancer in the world and has one of the highest recurrence rates. Despite the local delivery options and mutational status of bladder tumors, responses with immunotherapies have been limited with low durability. The bladder tumor microenvironment is enriched with highly immunosuppressive populations and harsh conditions that restrict anti-tumor immunity and subsequently attenuate immunotherapy response. We predict that targeting two of these immunosuppressive components, namely acidity and myeloid derived suppressor cells (MDSCs), will bolster T cell responses and enhance outcomes with immunotherapy. Bacterial aggregates are observed in both natural and artificial environments. In the context of disease, aggregates have been isolated from both chronic and acute infections. Pseudomonas aeruginosa (Pa) aggregates contribute significantly to chronic infections, particularly in the lungs of people with cystic fibrosis (CF). Unlike the large biofilm structures observed in vitro, Pa in CF sputum forms smaller aggregates (~10-1000 cells), and the mechanisms behind their formation remain underexplored. Chikungunya Virus (CHIKV) is a single stranded RNA alphavirus primarily vectored by Aedes aegypti and Aedes albopictus mosquitos. CHIKV has circulated in tropical and sub-tropical regions of Africa and Asia, but outbreaks in the past two decades have seen the virus spread to Europe, South America, and the Caribbean. Disease from CHIKV infection typically results in fever, malaise, and sometimes debilitating arthralgia. However, there are concerning abnormal presentations that affect the human heart, including myocarditis, heart failure, and death. Since the FDA approval of IL-2 in cancer treatment in the 1980s, immunotherapy has emerged as the standard of care for several tumor types (e.g., melanoma, lung, bladder, colorectal). However, approximately 70 percent of cancer patients, independent of prior treatment or staging, administered immunotherapy will not achieve complete responses and will exhibit disease progression. Chronic lymphocytic leukemia (CLL) is a predominant B-cell malignancy impacting elderly populations in Western societies. Targeted therapies have revolutionized the CLL treatment landscape. Still, patients often require long-term therapy and are subject to drug-induced resistance and severe toxicities, for which CLL is still considered an incurable disease. T-cell-based immunotherapy, particularly chimeric antigen receptor (CAR) T cells, has shown remarkable success, leading to various FDA approvals in hematological malignancies. Nonetheless, CAR T-cell therapy success rate is limited in CLL because of the dysfunctional nature of T cells.
