Recirculating aquaculture systems (RAS) are a type of near zero-discharge fish production system that is used to treat and recirculate aquaculture wastewater and increase the biomass stocking density in the fish tank. The RAS presented in this thesis was a marine system which was operated with two temporally independent cycles, Loop 1, a continuous loop, and Loop 2, an intermittent loop. Flow in the RAS was switched between the two loops by a solenoid valve. Identifying individuals with hereditary cancer predisposition can improve health outcomes for patients and their family members through early cancer detection and prevention strategies. Prior research about family sharing of genetic test results among those with hereditary breast cancer has overwhelmingly been limited to the BRCA1 and BRCA2 genes. The present study sought to compare family sharing behaviors in women with pathogenic BRCA variants to women with pathogenic variants in the more recently identified and characterized PALB2 gene. Efforts to remove and recover nutrients from wastewater are motivated by the United Nations Sustainable Development Goals and the National Academy of Engineering Grand Challenges of Engineering. Of the seventeen Sustainable Development Goals (SDGs), multiple SDGs relate to managing nutrients in wastewater. SDG 6, which is to “ensure availability and sustainable management of water and sanitation for all,” contains targets that aim to improve water quality by reducing pollution, halve the amount of untreated wastewater released to the environment, and increase recycling and safe reuse of wastewater (UN, 2017). This dissertation covers a variety of the genetic and molecular abnormalities of lung adenocarcinoma with an emphasis on STK11 loss and its implications on immunotherapy response. Given that lung cancer is the leading cancer killer, novel therapies are in great demand. In particular, immunotherapy has shown some of the most promise in the last decade but remains limited due to nearly 80% of patients not significantly responding. This dissertation aims to molecularly characterize lung adenocarcinoma while attempting to explain the reason why patients with STK11 loss do not respond to immunotherapy.

The international recommendation for breastfeeding is that a baby should be exclusively breastfed for six months. Breastfeeding should be continued for up to two years and beyond with complementary foods (WHO, 2016). The United Kingdom exhibits some of the lowest breastfeeding rates in the world (HSCIC, 2012). The percentage of mothers who breastfed in the United Kingdom falls below 34% at six months, with only 1% of mothers breastfeeding exclusively at this point (HSCIC, 2012:31). This dissertation research focuses on two key aspects of cloud computing research – pricing and security using data-driven techniques such as deep learning and econometrics. The first dissertation essay (Chapter 1) examines the adoption of spot market in cloud computing and builds IT investment estimation models for organizations adopting cloud spot market. The second dissertation essay (Chapter 2 and 3) studies proactive threat detection and prediction in cloud computing. As the rate of the murder of Black trans women at the hands of Black cisgender men rises steadily every year (HRC, 2017), discourses regarding the detrimental impact of toxic masculinity within Black communities continue to increase within different branches of feminist literature. Although research has described mentoring programs to have positive effects with youth, there are few studies that describe how mentors build relationships with homeless youth. In this study, I sought to explore how mentors of a community-based organization, Starting Right Now, built relationships with homeless youth. Starting Right Now is a multi-faceted, community-based intervention developed in 2009 in Tampa, FL. They provide services to homeless adolescents who are no longer living in the custody of a parent or guardian. Extracorporeal life support (ECLS) is a class of technologies used to support or replace the function of failing organs. During ECLS, blood is withdrawn from systemic circulation and circulated through an artificial organ or “treatment membrane” that performs the function of the failing organ, prior to return to systemic circulation. This family histories research study uses life history methods to explore narratives of parents’ lives regarding ways in which they socialize their children and engage school staff around issues of race and racism. The information gathered is from interviews with the two primary participants, two focus groups; one with each primary participant and the adults with whom they are raising their elementary school (Pre-K to Grade-5) children, and follow-up interviews with both primary participants. Massively Multiplayer Online Role-Playing Games (MMORPGs) are gaining popularity and are being used widely in China. The aim of this study is to explore the relationship between the motivation and the experience of MMORPG players, and the effects of playing MMORPG through an extensive online survey of more than 300 Chinese participants (n = 305). The efficacy of leadership coaching to improve leader and organizational outcomes cannot be overstated. However, a thorough understanding of some of the inputs and process variables involved in coaching has not been empirically established to date. To address this issue in the leader development and coaching literature, I examined the characteristics of the coaches and the coachees and their relationships with two relational variables potentially involved in coaching relationships (i.e., leader-member exchange and trust). This research documents the assessment and selection process used by the U.S. Military’s Special Forces for over 70 years using Grounded Theory Methodology. Three independent studies were used to document the Special Forces process of selection. Through comparative reanalysis of each study’s data a model emerged explaining the Special Forces phenomena of assessment and selection.

A lot of research has discussed using Organic Rankine Cycle (ORC) as subcritical or supercritical in power generation. Chapter 1 studies the effect of body weight on labor market outcomes. Using longitudinal data from the China Health and Nutrition Survey (CHNS) from 1989 to 2011, and both instrumental variable and fixed effects estimation to control for the endogeneity of body weight, I examine the wage effect of BMI by gender, type of residence area, and occupation. Results from linear OLS regression show a positive relationship between body weight and income for males in both rural and urban areas and females in rural areas, while a negative one for females in urban areas. Individuals who identify as lesbian, gay, bisexual, transgender, or queer (LGBTQ) are at a higher risk for suicide compared to the general population, but little is known about why this is. Many LGBTQ individuals face some form of discrimination or victimization in their lifetime, and some evidence suggests these experiences may contribute to this group’s higher suicide risk. Unfortunately, research has only examined the impact of direct discrimination/victimization on suicidality and has neglected to examine how ambient discrimination/victimization relates to suicidality. This dissertation examines how the intersections of gender, ethnicity, place, and class shape indigenous women’s risks for and experiences of intimate partner violence and related decision-making in Carhuaz province, an underserved, resource-poor setting in the Peruvian Andes. This dissertation applied a mixed-methods, community-based approach to 11 months of ethnographic fieldwork in Peru, which included 82 face-to-face surveys using the World Health Organization’s Multi-Country Study Instrument, 38 semi-structured interviews with survivors, community members, and IPV-related service providers, and 6 participatory action research workshops (n=64).

Excessive microvascular permeability is a serious complication involved in traumatic injury and inflammatory diseases. Alcohol intoxication can exacerbate the physiological derangements produced by microvascular endothelial barrier dysfunction in such disease conditions. Sphingosine-1-phosphate (S1P) has known endothelial barrier-protective properties, and has been shown to ameliorate microvascular leakage in a model of combined alcohol intoxication and hemorrhagic shock and resuscitation (HSR). However, whether the barrier-protective properties of S1P extend to endothelial cells of the blood-brain barrier (BBB) is unclear. Forensic anthropologists increasingly use chemical isotope analysis in the investigation of unidentified human remains, as biochemical georeferencing continually improves with the development of modern reference data of known origins. Isotope variations in trace elements such as strontium (Sr) and lead (Pb) in human teeth are some of the most useful indicators of past domicile in archaeological research and thus have high potential for modern, forensic applications. Cardiovascular disease (CVD) remains the number one killer even after years of advances and preventative measures. Identifying and reducing modifiable risk factors is a health care priority. CVD Risk assessments are calculated using several traditional risk factors including age, gender, race, blood pressure, cholesterol, history of diabetes, and smoking to estimate a persons’ risk of developing CVD (heart disease or stroke) in the next 10-years.
