Extended-spectrum β-lactamases (ESBLs) of different Enterobacteriaceae (e.g. Escherichia coli, Klebsiella pneumoniae) have developed significant resistance mechanisms against antibiotics. ESBLs, like class A CTX-Ms (cefotaximases) enzymes hydrolyze the β-lactam ring of different antibiotics such as penicillins and third-generation oxyimino-cephalosporins. Although CTX-Ms are repeatedly found in many clinical and community settings, molecular-level information on their function/activity is currently limited to structural studies performed by X-ray crystallography. This report describes the results from a study that examined the use of the break pass intervention to increase engagement in play with siblings for children with autism spectrum disorder. Two male children with ASD aged 6-years old, who had difficulty engaging in play activities with siblings participated in the study. The intervention used break passes to teach mands for breaks by providing the children with breaks throughout the play activity and to provide reinforcement for engaging in play with siblings in the absence of problem behavior. Historically, there has been no clear treatment pathway for opioid use disorder (OUD) patients who seek emergency care. Utilizing a trajectories of addiction framework, this dissertation presents insights from an applied anthropology of addiction study in an acute care setting to build a medication for opioid use disorder (MOUD) treatment pathway, aiming to break down the stigma related to opioid use disorder (OUD) and mitigate a lack of treatment options. Isoscapes are depictions of the spatial patterns of isotopic values in a given area. Isoscapes can be created using measurements form samples (empirical isoscapes) or using statistical models of spatial isotopic variation (modeled isoscapes). Isoscapes have a wide variety of potential applications though, in the realm of marine ecology, they are most often used infer ecological processes, food web linkages, the origin of samples, and the movements of marine organisms.

Background: In the United States, 13.8 million households were food insecure in 2020, or 10.5% of the country’s population. Although the annual food insecurity rates remained stable between 2019 and 2020, largely due to increased COVID-19 pandemic relief efforts, many households experienced food insecurity and hardship at unprecedented rates. This research has two parts. The first part focuses on the use of photoacoustic imaging (PAI) to study alcohol-induced effects in the cerebral vasculature of selectively bred alcohol-preferring mice. Part two extends the application of PAI by developing a novel Photoacoustic imaging guided system for repetitive low intensity focused ultrasound (PAI-LIFU) stimulation treatment for alcohol use disorder in crossed high alcohol preferring (cHAP) mice.

For label-free, non-invasive, wide field-of-view (FOV) imaging/monitoring of blood flow, speckle-based approaches are gaining popularity. However, to obtain quantitative flow information, speckle techniques rely on the multi-exposure scheme which requires complex, bulky, and expensive instrumentation, limiting its application to preclinical studies. This dissertation directly addresses these issues. In the first part of this dissertation, we report a novel single shot synthetic multi-exposure speckle imaging (syMESI) method to synthetically produce multi-exposure images from one short single exposure speckle image using spatial binning/averaging. Origin And Epigenetic Regulation Of Cutaneous T Cell Lymphoma, Carly M. Harro

The pathogenesis of Cutaneous T cell lymphoma (CTCL) remains unclear. Using conditional knockout mice, we found that ablation of the genomic organizer Special AT-rich sequence-binding protein-1 (Satb1) caused malignant transformation of mature, skin-homing, Notch-activated CD4+ and CD8+ T cells into progressively fatal lymphoma. Mechanistically, Satb1 restrained Stat5 phosphorylation and the expression of skin-homing chemokine receptors in mature T cells. H3K27 and H3K9 trimethylation occluded the SATB1 promoter in Sézary cells, while inhibition of SUV39H1/2 methyltransferases (unlike EZH2 inhibition), restored protective SATB1 expression and selectively abrogated the growth of primary Sézary cells more effectively than romidepsin. The cumbersome and time-consuming process of generating new mouse strains and multiallelic experimental animals often hinders the use of genetically engineered mouse models (GEMM) in cancer research. In chapter 2, we describe the development and validation of an embryonic stem cell (ESC)-GEMM platform for rapid modeling of melanoma in mice. The platform incorporates 12 clinically relevant genotypes composed of combinations of four driver alleles (LSL-BrafV600E, LSL-NrasQ61R, PtenFlox, and Cdkn2aFlox) and regulatory alleles to spatiotemporally control the perturbation of genes of interest. Capillary formation and maturation can only occur through a series of highly orchestrated events that requires the coordinated efforts of both endothelial cells (ECs) and pericytes. While ECs are perfectly capable of creating tube networks on their own, the maturation and stabilization of these tube networks require the presence of a second cell type, pericytes, to create stable capillary networks. α-Helix accounts for the largest class of protein secondary structures and plays a key role in mediating protein-protein interactions (PPIs). Small-molecule α-helix mimetics mimic the structural features of α-helical hot spots and offer a powerful tool to unravel complex signaling networks and mitigate aberrant PPIs. Myelodysplastic Syndromes (MDS) are heterogeneous bone marrow (BM) failure malignancies characterized by constitutive innate immune activation, NLRP3 inflammasome (IFM) driven pyroptotic cell death, and the induction of interferon-stimulated genes (ISG). MDS often diagnostically resembles other hematologic disorders, as such, additional tools are needed for the discrimination from overlapping disorders. We observed that oxidized mitochondrial DNA (ox-mtDNA) is released upon cytolysis. We hypothesize that levels of ox-mtDNA are high in MDS, allowing us to use it as a diagnostic biomarker; and that cell free ox-mtDNA is contributing to feedforward BM failure.

Infants diagnosed with neonatal opioid withdrawal syndrome (NOWS) exhibit neurobehavioral dysregulation that may make them challenging to feed, potentially impeding positive early maternal-infant interactions. The goal of this study was to investigate dyadic behavior during the maternal-infant feeding interaction, specifically infant dysregulation and maternal sensitivity, between dyads with opioid exposure in the neonatal intensive care unit (NICU). This was a secondary data analysis using a mixed-methods design to analyze videorecorded feeding interactions of mothers with opioid use disorder (OUD) and their infants with NOWS. This dissertation is an ethnographic case study that analyzes the gendered socio-cultural and socio-economic implications for a community that was displaced in order to develop a geothermal energy system in Kenya. The implications are rendered particularly interesting because the displaced community is also one of the communities served by the geothermal power grid. The analysis employs a composite framework of Feminist Political Ecology and the Political Ecology of Climate Change Mitigation. This work explores gendered perceptions and control of energy, livelihood activities, place attachment, and the cultural/spiritual significance of geothermal energy.

Dynamic interaction (DI) describes the synchronous interaction of one or more individual animals over time. Current methods to quantify DI from tracking data are lacking. They typically require synchronized tracking data which limits their applicability to most studies. Mutations in the LMNA gene (encoding lamin A/C) are the second most common cause of familial arrhythmogenic cardiomyopathy. Diverse LMNA variants in cardiomyocytes have been associated with cardiac phenotypes. Moreover, cardiac fibroblasts make up a large fraction of the myocardium's non-myocyte component, intrinsically linked to extracellular matrix synthesis and turnover as well as secreting large amounts of bioactive metabolites. South Cross Bayou Advanced Water Reclamation Facility (SCB) in Pinellas County, Florida, recently had a spike in effluent concentrations of bromodichloromethane (BDCM) and dibromochloromethane (DBCM) above the regulated limits, due to relying fully on chlorination instead of a combination of chlorination and ultraviolet light. Ecological regime shifts are transitions in ecosystem state, which can occur either when external conditions gradually change or when large perturbations force a system into an alternative state. Such transitions can exhibit non-linear dynamics, where small changes in external conditions can result in abrupt transition and often occur with little warning. After crossing a tipping point, the system may be unable to revert to its original state if the new regime possesses strong, reinforcing feedbacks. Where do you go to divulge your deepest secrets? Some may turn to journals and diaries; some turn to family and friends; some turn to religious confessionals; some turn to Twitter; and still, some take their secrets to the grave. There is at least one other way people share their secrets that has thus far received limited scholarly attention: the crowdsourced confessional. Visions Of Entanglement And Escape: In-Visible Voice In The Films Of Terrence Malick And George Lucas, Michael Lee Taber

This thesis juxtaposes the unexpectedly parallel careers of the filmmakers George Lucas and Terrence Malick. Both popularly and academically, Lucas and Malick are typically conceived as divergent practitioners and are framed in oppositional terms. The goal of this thesis is – in part – to break down this oppositional understanding of Lucas and Malick as filmmakers. This traditional understanding of Malick and Lucas collapses when we consider their respective relationships to sensation and abstraction. Affirmations of sensation and Malick abound, as do critiques of abstraction and Lucas – all the while it is Malick who is aligned with abstraction and …
