The passage of two major hurricanes across southwest Florida (Category 5, Labor Day Hurricane of 1935; Category 4, Hurricane Donna 1960) resulted in fragmentation of mangrove forest at Big Sable Creek, Everglades National Park. Over time forest fragmentation led to forest loss and patchy conversion to unvegetated mudflats. My goal was to determine the consequence of forest fragmentation on nekton (i.e., fish and decapod crustaceans) inhabiting the intertidal zone. I used block nets across intertidal rivulets to compare nekton leaving replicate forest and unvegetated mudflat sites from October, 2002 through April 2004. Ferrochelatase catalyzes the terminal step of heme biosynthesis by inserting ferrous iron into protoporphyrin IX. The current study is aimed at understanding the structural basis of porphyrin binding and distortion in ferrochelatase-catalyzed reaction by functional analysis of a highly conserved active site loop motif. The loop was shown to contact bound porphyrin based on crystallographic and molecular modelling observations, and its role in murine ferrochelatase was assessed by random mutagenesis and steady-state kinetic analysis. To overcome the limitations of conventional kinetic assay methods for ferrochelatase, a continuous assay was developed by monitoring porphyrin fluorescence decrease using natural substrates ferrous iron …

Tufa and lacustrine deposits are useful paleoclimate archives in reconstructing the early to middle Holocene climate and paleohydrology of southern Yemen's Wadi Idim and Wadi Sana, which are north-flowing tributaries to Wadi Hadramawt. Numerical age estimates and oxygen-isotopes are used to assess the onset and cessation of tufa formation and reconstruct the environment of lacustrine sediment deposition in the region in order to understand the broader early to middle Holocene hydrologic system.Numerical age estimates from the studied wadis show a correspondence between early to mid-Holocene humid-phase sediment deposition and the northward shift of the ITCZ, as documented in paleoclimate records …

Bronopol has been used as a preservative in drugs and cosmetics since 1964. Bronopol has low dermal irritancy at levels commonly used in cosmetics and pharmaceuticals but it is significantly irritating at higher concentrations. Laboratory testing of bronopol indicates a low potential for dermal sensitization; however, a number of case reports demonstrate human allergenic reactions. No reports were identified on the allergenic properties of bronopol for the inhalation route of exposure. In 1983 approximately 5,200 people in the United States were occupationally exposed to bronopol. Current novel uses of bronopol include mold remediation and the sanitizing of ventilation system components. …

The central issue in the works of Danish philosopher and religious thinker Soren Kierkegaard (1813-1855) takes the form of a question: "What does it mean to become a Christian?" However, Kierkegaard's ideas exerted influence well beyond Christian circles and have been important to many notable philosophical and literary figures, some of whom chose not to concern themselves primarily with this question (Sartre, Camus, Heidegger, Buber), and some of whom did choose to concern themselves primarily with this question (Tillich, Bonhoeffer, Berdyaev, Marcel). This study investigated whether or not there was an overall difference in the pulmonary oxidative marker, 8-isoprostane (PGF2-a), found in exhaled breath condensate, during pregnancy versus the non-pregnant state. The utility of this information was important secondary to the effect of maternal asthma on pregnancy and outcome, as it has been demonstrated in past studies that overall pregnant females with asthma have been shown to have an increased risk of exacerbation requiring medical intervention.The primary goal of this study sought to determine if there is a difference in PGF2a levels in the exhaled breath condensate of pregnant versus non-pregnant females. …

This qualitative study described non-science undergraduate majors' responses to controversial issues embedded in an introductory level environmental science course in a liberal arts college located in the southeastern United States. Participants enrolled in this 12-week summer course were both traditional college-age (late teens to early twenties) and non-traditional age student (thirties to fifties). Approximately 76 percent were female. Students demonstrated various lifestyles (e.g., gay, single-parent, living at home), socioeconomic statuses (e.g., middle-income, low income), employment (e.g., employed, unemployed, ex-military) and ethnicities. The largest impediment to appreciating Hart Crane as a symbolist modern American poet derives from the fragmentary critical attention paid to his borrowings from and familiarity with French Symbolists like Charles Baudelaire, Arthur Rimbaud, and Stéphane Mallarmé. Almost equally important, the early career of T. S. Eliot exerted a profound impact on Crane's poetic development and indeed served as the primary introduction to many nineteenth-century French poets for Crane and many other American poets of his generation. Suicide terrorism is characterized by the willingness of physically and psychologically war-trained individuals to die while destroying or attempting to annihilate enemy targets in furtherance of certain political or social objectives. Rooted in the historical, social, and psychological dimensions of international terrorism, suicide terrorism is neither a unique nor a new phenomenon. Its recent resurgence and the extensive media coverage it has received account for the misleading uniqueness of this violent, complex, and adaptive form of terrorism. Friar Francisco Pareja represented the pinnacle of the achievement for the Franciscans in Florida during the Spanish colonial period. But who were the Franciscans? Why were they, and Friar Pareja in particular, so successful as missionaries? The bulk of the writing done thus far on the mission system in Florida has concentrated on retelling the lost story of the native peoples who once inhabited the land. The impact of the missions and the Spanish colony weighed heavily on native cultures and the Franciscans role in this has been discussed. An exploratory study of the geomorphology of Florida's springs was conducted using morphometric analysis. Spatial datasets for spring locations, elevation data, physiography, geology and hydrography were acquired for incorporation and analysis with GIS technology. ArcGIS 9 was used to measure certain morphometric parameters from the spatial data for Florida's springs. Other Parameters representing physical and dimensional characteristics of the springs were acquired from FGS Bulletin 66, Springs of Florida. Members of the family Placobranchidae have been the focus of numerous studies because of their interesting physiological adaptations (kleptochemistry and kleptoplasty) and ecological associations with marine algae. More than 100 species have been described since the 1800's, but no determination of phylogenetic relationships in the family has occurred. DNA sequences from two mitochondrial genes (large subunit ribosomal and cytochrome c oxidase subunit I) and one nuclear gene (Histone 3) were used to reconstruct relationships among 34 nominal species within the Placobranchidae. Strong support for terminal nodes was found using Bayesian phylogenetic methods; however, a "soft" polytomy was detected at internal …

An evanescent wave fiber-optic biosensor was used in an effort to develop an assay for the rapid detection of two biological toxins: cholera toxin and botulinum toxin. The Analyte 2000 fiber-optic biosensor utilizes a sandwich immunoassay format. Gangliosides or liposomes are directly adsorbed to the surface of the fiber-optic waveguide through hydrophobic interactions. The waveguide is exposed to a sample containing the toxin of interest, then subsequently exposed to a polyclonal detection antibody conjugated to the fluorophore cyanine 5. Excitation light from a 635nm laser diode is propagated through the waveguide and fluorescent molecules within approximately 100nm of the waveguide …

Development of a rapid method for the detection of fecal enterococci and pathogenic microorganisms in beach water was attempted utilizing an evanescent wave fiber optic biosensor. Various assay formats including a sandwich immunoassay were tested in the development of a rapid assay. Fluorophore labeled antibodies were used for specific detection of bacteria captured or adsorbed directly to the surface of a polystyrene fiber optic waveguide. Binding of the fluorescent labeled antibody to its specific target or binding of a fluorescent labeled anti-IgG within 100-1000 nm of the waveguide surface caused excitation of the fluorescent conjugate resulting in a quantifiable signal. …

Ultrafine particles (UFPs) are present in the ambient atmosphere and are generated from atmospheric gases, pollution sources, and combustion. Candles emit carbonaceous soot particles similar to UFPs present in the ambient atmosphere. With the exception of lead, airborne concentrations of candle emissions have not been shown capable of causing cancer or cardiopulmonary disease during normal use. The purpose of this research is to determine the occupational risk associated with candle emissions.

The Internet provides the opportunity to develop a new way to present journalism, but many scholars say newspaper Web sites do nothing but mirror their print parents. This study used content analysis to compare the content of stories in five newspapers with their Web counterparts, and it examines whether reporter affiliation or a story's geographic emphasis has a relationship with the story's amount of contextual elements. These elements could include photos, graphics, or multimedia or interactive components online. This approach applied gatekeeping theory to publications that have editions in two media.

Arsenic contamination in drinking water has been a cause of serious concerns across the United States as well as throughout the world. Over 70 million people in Eastern India, Bangladesh, Vietnam, Taiwan, and Northern China have been victims of arsenic poisoning. The USEPA has classified arsenic as a Class A carcinogen and recently reduced the Maximum Contaminant Level (MCL) in drinking water from 50ppb to 10ppb. The deadline for all the water utilities to meet this level is 23rd January 2006. To meet those drinking water standards, small water utilities need low cost and effective arsenic removal techniques.

Although most critics affirm the importance of interior direction and role-playing in many of Shakespeares plays, there is a considerable disagreement concerning the result of this role playing: does it lead to positive growth or to degeneration? Moreover, this debate is often associated with the sixteenth-century controversy about the role of the theater in society. In humans, adolescent exposure to illicit drugs predicts the onset of adult drug abuse and suggests that early drug use potentiates adolescent vulnerability to drug addiction. In experiment 1, it was hypothesized that adolescent rats would show a CPP for a low cocaine dose if in fact adolescents are more vulnerable to cocaine's rewarding effects. Place preferences were measured in early adolescent [postnatal day (PND) 35], late adolescent (PND 45) and young adult (PND 60) rats by injecting either 0, 5 or 20 mg/kg cocaine and conditioning them to environmental cues in a 2-chamber place conditioning apparatus. Signalized Intersections on the State Roads in Hillsborough and Pinellas County, Florida with observed left turn lane overflow (spill) were selected for a safety and operational study. The study analyzed the crash data for safety hazards that the situation presents. Crashes within 100 feet from the center line of the crossroad of intersections under study to distances 200 feet beyond the end of the turn lane were chosen for the analysis. Left turn overflow is the situation at the approach of an intersection where left turning vehicles back up from the turn lane into the through traffic lane.

Mutations in the presenilin 1 gene account for most early-onset familial Alzheimer's disease (FAD). The presenilins and AD may also be related through a common involvement in the cell cycle. Here we report that one important aspect of the cell cycle---proper chromosome segregation---is dependent on presenilin function and therefore may be involved in AD pathogenesis. Specifically we find that FAD mutations in presenilin 1 (M146L and M146V) lead to chromosome missegregation and aneuploidy in vivo and in vitro: 1) Both metaphase chromosome analysis and in situ hybridization reveal significant aneuploidy in the lymphocytes and neurons of PS-1 transgenic mice. 2) …

Utilizing a continuous silhouette image of marine plankton produced by a device called SIPPER, developed by the Marine Sciences Department, individual plankton images were extracted, features were derived, and classification was performed. There were plankton recognition experiments performed in Support Vector Machine parameter tuning, Fourier descriptors, and feature selection.
Several groups of features were implemented, moments, gramulometric, Fourier transform for texture, intensity histograms, Fourier descriptors for contour, convex hull, and Eigen ratio. The Fourier descriptors were implemented in three different flavors sampling, averaging and hybrid (mix of sampling and averaging).

This work is aimed at developing a high sensitivity salinity (conductivity) sensor for marine applications. The principle of sensing involves the use of parallel plate capacitors, which minimizes the proximity effects associated with inductive measurement techniques. The barrier properties of two different materials, AZ5214 and Honeywell's ACCUFLO T3027, were investigated for use as the insulation layer for the sensor. Impedance analysis performed on the two coatings using Agilent's 4924A Precision Impedance Analyzer served to prove that ACCUFLO was a better dielectric material for this application when compared to AZ5214.

Torsion-Induced Pressure Distribution Changes In Human Intervertebral Discs: An In Vitro Study, Brenda Kay Yantzer

Introduction. To test the effects of torsion torques on intradiscal pressure and disc height in human lumbar specimens.
Methods. Six human lumbar cadaveric functional spine units (FSU) were loaded in the neutral position with 600 N compression. Nucleus pressure measurements were obtained at 0 Nm, 0.5 Nm, 1.0 Nm and 2 Nm torsion torque. Posterior elements were removed and pressure measurements were repeated at the same torsion torques for the disc body unit (DBU). The pressure in the nucleus was measured by pulling a pressure probe through the disc along a straight path in the midsagittal plane.
