Binge alcohol consumption is a rising concern in the United States, especially among adolescents as during this developmental period alcohol use is usually initiated and has been shown to cause detrimental effects on brain structure and function. These findings have been established through the use of binge models in animals, where animals are repeatedly administered high doses of ethanol typically over a period of three or four days. Within the paradigm of Sociocultural Theory, and using Activity Theory as a data-gathering and management tool, this microgenetic case study examined the processes - the growth, change, and development – engaged in by student-teachers in a foreign language education program as they worked together to complete an activity. The activity involved digital video recording and editing, mediators which were intended to facilitate the iterative review of and subsequent reflection and action upon the content of the video during its creation.

This research estimates nitrogen removal from stormwater runoff using a denitrifying bioretention system using the USEPA Storm Water Management Model Version 5 (SWMM-5). SWMM-5 has been used to help planners make better decisions since its development in 1971. A conventional bioretention system is a type of Low Impact Development (LID) technology, which designed without a media layer specifically for achieving nitrogen removal. More recently studies have showed that high TN removal efficiencies are possible when incorporating a denitrification media layer. These systems are known as denitrifying bioretention systems, or alternative bioretention systems. LID projects are currently being designed and developed …

This dissertation describes the isolation of secondary metabolites from natural origins through a series of chromatographic techniques and spectrometric characterization in the effort of drug discovery. The isolated compounds obtained were used as drug leads against tropical diseases, namely malaria and leishmaniasis. While first chapter offers an introduction on the use of a natural product by itself as an effective therapeutic and its role on inspiring the discovery of new drugs, the later chapters will concentrate on isolation and characterization of bioactive natural products from an Antarctic sponge and mangrove endophytic fungi during the dissertation work.

The marine realm and, in particular, the Antarctic benthos is largely unexplored and understudied. The chemical investigation reported herein reveals not only the biodiversity, but how that biodiversity manifests remarkable chemical diversity. In our continuing study of the nudibranch Austrodoris kerguelenensis, we have isolated a diverse suite of diterpenoid glyceride esters, palmadorins D - S (2.32 - 2.47), one of which is the first reported halogenated diterpene from a dorid nudibranch. Utilizing genomic data from collaborators, we have investigated the chemical diversity from phylogenetically unique specimens collected in close proximity to one another. Ammonium (NH4+) is a biological nutrient that is transformed in a wastewater treatment plant (WWTP) in a process called activated sludge. This is accomplished in an aerobic environment using microorganisms and inorganic carbon that convert the ammonium to nitrate (NO3-). This process is termed nitrification. Removal of ammonium is necessary due to its oxygen demand and toxicity to the environment.

It is well established that water quality is directly linked to health. In-line chlorination is one technology that can be used in the developing world to potentially inactivate pathogens and improve water quality. The purpose of this study was to determine the effectiveness of the Panamanian Ministry of Health's in-line PVC chlorinator under three different operating conditions in a rural water supply system. This study analyzed treatment of geometric transformations in presently available middle grades (6, 7, 8) student mathematics textbooks. Fourteen textbooks from four widely used textbook series were evaluated: two mainline publisher series, Pearson (Prentice Hall) and Glencoe (Math Connects); one National Science Foundation (NSF) funded curriculum project textbook series, Connected Mathematics 2; and one non-NSF funded curriculum project, the University of Chicago School Mathematics Project (UCSMP).

This dissertation addresses the impact of urbanization and land use change on the availability and accessibility of two urban amenities that are often inequitably distributed: green space and water features. Diverse methodologies were utilized in order to gain a better understanding of the role of these amenities in improving urban quality of life and integrated water management. Using an interdisciplinary approach, this research provides a unique perspective within both a developed and developing world context by evaluating aspects of urbanization to emphasize more sustainable and integrated approaches to development.

The goal of this thesis is to develop the foundation and structure for a virtue ethics theory grounded in a specific notion of care. While there has been a recent revival of interest in virtue ethics theory, the theory has its roots in Aristotle's work as well in the medieval writings of Thomas Aquinas. Aquinas worked out many of Aristotle's ideas in much more detail. However, while Aquinas offers a very rich and compelling ethical theory, it is problematic because it is very tightly wrapped in his theology. A key component in Aquinas's theory is charity. In an effort to determine the optimal temperature for rearing Centropomus undecimalis larvae during the yolk resorption period, larval development was measured under four different temperature regimes (23, 25, 28 and 31 °C). The eggs were incubated at 28 °C until hatching, which occurred at about 17 hours post-fertilization. After hatching, temperatures were adjusted to the respective treatment levels. Measurements were collected from 25 individual larvae across rearing temperatures at the following pre-determined time intervals: at hatching, 24 hours post hatch (hph), 48 hph, and 72 hph. Eastern Equine Encephalitis virus (EEEV) causes a fatal mosquito-borne virus that is vaccine preventable for horses. The conventional serological tests measure antibodies to the structural proteins of EEEV which are also found in the vaccine. This makes it difficult to differentiate infected and vaccinated animals (DIVA). Detection of antibodies to non-structural proteins (NSPs) is a theoretical strategy that would allow you to survey natural infections among vaccinated populations. This test would also allow for more accurate representations of the natural infection rate, vaccination rate, and help identify vaccine failures. The sedimentary deposits of the Hadar Formation at Dikika and the Mount Galili Formation at Galili preserve a wealth of paleoenvironmental and paleoclimatic records spanning the last 5.29 Ma. Stable carbon and oxygen isotopic compositions of herbivore tooth enamel were analyzed for more than 600 specimens of 15 different taxa from 10 stratigraphic intervals. The application of carbon and oxygen isotopes here aims principally at reconstructing shifts in the relative abundance of C4 grasses, and its implications for climate indicators including temperature, aridity, and seasonality. The eastern tropical north Pacific (ETNP) is characterized by one of the ocean's most severe midwater oxygen minimum zones (OMZs), where oxygen levels are often less than 5 µM. The copepod family Eucalanidae is a numerically abundant and diverse zooplankton group in the ETNP, and displays a wide range of vertical distributions related to environmental oxygen concentrations. In this thesis, I provide an analysis of 1960s American popular culture by examining Playboy, "The Playboy Philosophy," Cosmopolitan, and Sex and the Single Girl. These cultural artifacts furthered the feminist movement by challenging gender structures and sexuality. I discuss how these publications focused on the advancement of the individual through careerism, consumerism and sexuality. These publications assisted in challenging and breaking down various aspects of gender and sexual boundaries and assisted in reworking social limitations that kept women from advancing themselves outside of the pre-set gender roles of domesticity. MDM2 regulates p53 predominantly by promoting p53 ubiquitination. However, ubiquitination-independent mechanisms of MDM2 have also been implicated. Here we show that MDM2 inhibits p53 DNA binding activity in vitro and in vivo. MDM2 binding promotes p53 to adopt a mutant-like conformation, losing reactivity to antibody Pab1620, while exposing the Pab240 epitope. The acidic domain of MDM2 is required to induce p53 conformational change and inhibit p53 DNA binding. ARF binding to the MDM2 acidic domain restores p53 wild type conformation and rescues DNA binding activity. Furthermore, histone methyl transferase SUV39H1 binding to the MDM2 acidic domain also restores p53 wild …

The goal of our research endeavor was to successfully employ modern lead discovery and optimization strategies towards the development and identification of compounds possessing antimalarial activity. Preliminary data from in vitro screening at the Walter Reed Army Institute of Research identified several chemotypes including 4(1H)-quinolones and 1,2,3,4-tetrahydroacridones to have potent antimalarial activities. Multiple synthetic routes were devised and implemented which enabled the rapid preparation and isolation of over 400 structurally diverse 4(1H)-quinolones and 1,2,3,4-tetrahydroacridones.

Population structure can be defined as a difference in the frequencies of alleles among populations, and it is often caused by barriers to dispersal. I used mitochondrial DNA sequences to examine the population structure of two large, aquatic salamanders, Amphiuma means (the Two-Toed Amphiuma) and Siren lacertina (the Greater Siren), at two spatial scales in peninsular Florida. Both species appear to have similar life histories, including poor dry land dispersal capabilities, and I hypothesized that they would exhibit significant population structure at both broad and fine scales. The main goal of this research was to develop a mechanism-based model for photocatalytic disinfection of bacteria in water using suspended catalyst pthesiss in batch reactors. The photocatalytic disinfection process occurs as a semiconductor photocatalyst, most commonly titanium dioxide (TiO2), is irradiated with light of wavelength less than 380 nm to produce hydroxyl radicals and other highly reactive oxidants which can inactivate microorganisms. To date, most studies on the fate and removal of endocrine disrupting compounds (EDCs) and pharmaceutical and personal care products (PPCPs) in wastewater focus on their fate in municipal wastewater treatment plants, and mostly under aerobic condition. There are limited studies related to anaerobic condition and (to our knowledge) no study on the removal of EDCs in landfill leachate by AnMBR. Moreover, for most studies under anaerobic condition, the removal of EDCs was only reported in the liquid phase; solid phase extraction was not reported, thereby preventing mass balance in the studies.

Current progress in the development of vaccines has decreased the incidence of fatal and non-fatal infections and increased longevity. However, new technologies need to be developed to combat an emerging generation of infectious diseases. DNA vaccination has been demonstrated to have great potential for use against a wide variety of diseases. Alone, this vaccine technology does not generate a significant immune response for vaccination, but combined with delivery by electroporation (EP), can enhance plasmid expression and immunity against the expressed antigen. The objective of this project was to document the status of oysters, Crassostrea virginica, from non-reef habitats throughout Tampa Bay, Florida, and assess the ecosystem contributions of those populations relative to reef-dwelling oysters. The aspects of oyster ecology studied here include condition, prevalence and intensity of disease (Perkinsus marinus - dermo), reproductive activity (including stage, fecundity, and juvenile recruitment), adult oyster density, and the faunal community associated with the oysters.
The predominant source of variation was seasonal, with lesser contributions among sites, and in most cases, little or no effect of the habitat type. Oysters populations from each habitat …

D-cycloserine (DCS), a partial agonist that acts on the N-methyl-D-aspartate (NMDA) receptor of the glutamatergic receptor complex, may enhance fear extinction learning during exposure-based therapy. Clinical studies in adults with obsessive-compulsive disorder (OCD) and non-OCD anxiety disorders - and a recent trial in pediatric OCD - have shown that DCS can improve treatment response to exposure therapy relative to placebo and exposure therapy. Some have hypothesized that improved treatment response is a function of increased compliance and engagement in therapeutic homework tasks, a core component of behavioral treatment. Electroporation is the critical step in an electric field mediated drug or gene delivery protocol. Electroporation based protocols have been successfully demonstrated in cancer clinical trials, however, its impact in other applications is still under investigation. A significant roadblock to long term functioning of implantable biosensors in vivo is the tissue reaction in the form of fibrous encapsulation that results in reduced transport to the sensing element of the biosensor. In vivo gene electroporation has a great potential as a means to modify the transport properties of tissues in the proximity of the sensing element of implantable biosensors.

Antibiotic resistance is a particularly critical health concern and has increased dramatically over the past two decades. For over a decade the Turos laboratory has been designing small molecules to target pathogenic microbes such as Staphylococcus aureus and the resistant variants like methicillin-resistant Staphylococcus aureus (MRSA). Previously, N-thiolated Β-lactams, N-thiolated 2-oxazolidinones and aromatic disulfides that were synthesized in Dr. Turos' lab have shown strong activity against these bacteria. The present work describes the synthesis and antimicrobial activities of a related structural class called S,S'-heterosubstituted disulfides.
