T cells are critical effectors of the immune response against pathogens and malignancies and, given the recent success of T-cell immunotherapies in cancer, there is now an urgent need to enhance the efficacy and durability of T-cell immune responses to improve anti-tumor immunity. Formation of memory T cells is a critical step of a successful immune response as it ensures maintenance of long-term immunity and protection from re-exposure to foreign antigens. The biodiversity of our global marine ecosystems provides substantial treasure to discover: chemistry. Marine organisms that live in proximity to one another use their unique chemistry to compete with one another and thrive in their environment. Benthic invertebrates, specifically, that lack physical means of protection will produce secondary metabolites as a means of defense from their surroundings and predators. The compounds which they produce are ordinarily species-specific, highly diverse biomolecules. Oligodendrocyte lineage is a glial cell population in the central nervous system. The lineage starts with oligodendrocyte precursor cells, which differentiate and mature into mature oligodendrocytes. Myelination, the major function of mature oligodendrocytes, plays a significant role for normal function of the central nervous system, and its dysfunction leads to many demyelinating diseases, such as multiple sclerosis. During the lineage progression, an extracellular matrix protein, laminin, actively regulates the cells' biology, including cell proliferation, differentiation, and myelination. This research investigates how individual mobility among ancient populations in Romania during the Eneolithic, between 5000 and 4000 BC, may be related to the spread of the material culture known as Kodjadermen-Gumelniţa-Karonovo VI (KGK VI) that extended over most of southern Romania and Bulgaria. This work seeks to combine results on individual mobility with other lines of archaeological evidence to provide a better understanding of the social context and kinship relationships that were in place when these events took place.

Chronic Myelomonocytic Leukemia (CMML) is a rare myeloid malignancy with a dismal prognosis and no therapeutic options which are capable of altering the natural course of the disease. There remains a significant need for novel therapies that are able to meaningfully improve patient outcomes. In this study we explore the effectiveness of Bromodomain and Extra-Terminal domain protein inhibitor (BETi) combinations in CMML. Preclinical studies in myeloid neoplasms have demonstrated efficacy of BETi. However, BETi demonstrate poor single agent activity in clinical trials. Marine sediment-associated bacteria house many new and exciting novel secondary metabolites. These metabolites can be tested for bioactivity against various types of cancer and fungal, bacterial, and viral infections. In this thesis, we investigated the combination of biosynthetic gene cluster information with mass spectra to perform a chemical profiling of sediment- associated bacteria. Furthermore, we utilized a scoring technique to provide an identification and confidence score to each annotated compound. The sediment was collected from east Arthur Harbor, Palmer Station, Antarctica, at depths of 20 ft and 60 ft. After plating on agar, 52 unique bacterial strains were isolated, with …

Breast cancer (BC) continues to be the most diagnosed cancer and the second leading cause of cancer deaths in women worldwide. High mortality, in most cancers, is due to metastasis; BC is the most common source of one of the deadliest forms of metastasis–brain metastasis (BM) in female. Patients with triple negative breast cancer (TNBC) exhibit the highest incidence of BM and poorest survival outcomes upon diagnosis with BM. Quantification of the true number of stained cells in specific brain regions is an important metric in many fields of biomedical research involving cell degeneration, cytotoxicology, cellular inflammation, and drug development for a wide range of neurological disorders and mental illnesses. Unbiased stereology is the current state-of-the-art method for collecting the cell count data from tissue sections. These studies require trained experts to manually focus through a z-stack of microscopy images and count (click) on a hundred or more cells per case, making this approach time consuming (~1 hour per case) and prone to human error (i.e., inter-rater variability). Booker Creek, a 1.9-mile stream with a 4.9-mi2 watershed, flows southeast through highly urbanized St. Petersburg, Florida, and empties into Bayboro Harbor, which connects with Tampa Bay. This study collected and analyzed water quality data for several parameters (TN, TP, nitrate-nitrite, ammonia, orthophosphate, chl-a, fecal coliform, dissolved oxygen, pH, turbidity, temperature, and salinity) every two weeks, at four different sites, during the 2022 wet season to characterize the current state and geographic variability of Booker Creek. Current creek sampling regimes, though consistent, do not sample at multiple sites along the creek. Urban streams do not display uniform water quality along …

In this thesis, I explore the potential of online communities in negotiating alternative forms of “doing” masculinity. I focus on the /r/bropill which is hosted on Reddit – home to thousands of active discussion forums called subreddits. I argue that the members of /r/bropill subreddit are attempting to redefine what it means to live your life not only as a man but as a “good man.” Using a purposive sample, I analyzed 24 discussions which totaled 1325 posts (n = 1325). I conducted a qualitative textual analysis of the original posts and comments inspired by grounded theory. My findings reveal …

Neuroendocrine regulation of the innate immune system is a well-documentedphenomenon. The underlying mechanisms, however, are not well understood. Here we show that at least 20 members of the human chemokine receptor (CR) family heteromerize with one or more members of the α1-adrenergic receptor (AR) family and that all CRs, apart from CXCR1, heteromerize with arginine vasopressin receptor 1A (AVPR1A) in recombinant systems. These heteromeric complexes are detectable in human monocytes and the monocytic leukemia cell line THP-1. The Upper Floridan Aquifer (UFA) is a highly transmissive and heterogeneous karst aquifer that makes it acutely susceptible to pollution. Rapid and extensive water quality changes in nutrients at springs draining the UFA have prompted intensive monitoring at several priority springs, including continuous and discrete water quality sampling. Springs, however, are a discrete discharge point of an often-expansive phreatic cave network that can intersect different land uses and thus drain distinct geographic areas of a springshed. These help in communicating chemistry concepts that are fundamental to understanding how and why molecules interact. The goal of this dissertation work is to offer an evidence-based report on the affordances and limitations of the variety of molecular representations by chemistry topic. Nitrogen discharge from wastewater is a global issue and with increased urbanization as well as industrialization, it has become even more essential to develop more efficient wastewater treatment systems. The dominant form of nitrogen in wastewater is ammonium. Biological oxidation of ammonium to nitrite and nitrate is the first step in biological nitrogen removal processes. If left untreated, nitrogen can cause toxicity to aquatic life, unsafe changes in water quality, and eutrophication.

This study investigates the different dynamics of consumer responses to crisis communication strategies employed by businesses during challenging situations. Focusing on emotions, attitudes, and behavioral intentions, the research explores the impact of denial, diminish, and apology crisis messages on consumers. Drawing on a comprehensive analysis of a crisis scenario, the study illuminates the intricate relationship between these strategies and consumer emotions such as anger, disgust, sadness, and happiness. Researchers often use geographic information systems (GIS) to measure accessibility to food pantries, but the impact operating hours have on accessibility remains understudied. Unlike grocery stores or healthcare facilities, food pantries have irregular distribution schedules, therefore, measuring geographic accessibility alone potentially creates an inaccurate or inflated representation of access. The general decline in coral-reef ecosystem health throughout the Caribbean between the 1970’s and late 1990’s seems to have slowed down or stopped, with only slight decreases in coral reef cover observed in the 2000’s through 2019. The earlier decline can be attributed to many different local and regional stressors that have acted together, and include naturally occurring (hurricanes, diseases) and anthropogenic pressures (increased sedimentation, fishing). This research presents a comprehensive assessment of night vision goggles and thermal imagers, with a particular emphasis on their performance under a wide range of environmental and temporal conditions. Field testing of these devices was carried out across diverse settings, including daytime, nighttime, and different environmental temperatures. Key evaluation metrics, such as contrast, recognition distance, and image clarity, were employed to assess device performance.The findings reveal that while night vision goggles maintain consistent performance across various conditions, their image clarity and contrast are influenced by the recognition distance. To understand the evolutionary gain, and loss, of avian powered flight, attention must be paid to the wing morphology of species across these transitions. However, paleognaths, while being the most well-known flightless birds, have greatly understudied forelimbs. Spatial geography, ontogenetic movement, and trophic patterns of mobile species are key elements of effective marine resource management. A number of methods are currently available for tracking movements of pelagic migratory species, including the use of conventional and electronic tags. However, tagging campaigns can take years to provide useful data, can be expensive, and only capture a portion of a fish’s lifetime. For this project, I used stable-isotopic ratios of nitrogen (δ15N) and carbon (δ13C) in metabolically inert, chronologically-layered fish-eye lenses to explore lifetime movement and trophic patterns of yellowfin tuna (Thunnus albacares) and blackfin tuna (Thunnus atlanticus) in the …

Large language models (LLMs) are poised to transform both academia and industry. But the excitement around these generative AIs has also been met with concern for the true extent of their capabilities. This dissertation helps to address these questions by examining the capabilities of LLMs using the tools of psychometrics. We focus on analyzing the capabilities of LLMs on the task of natural language inference (NLI), a foundational benchmark often used to evaluate new models. Negative density-dependent regulation is the process by which host-specific natural enemies reduce the survival of seeds and seedlings of conspecifics near adult trees, favoring heterospecific recruitment, and ultimately increasing local diversity. Negative density-dependent regulation by fungal pathogens has been consistently reported to promote plant diversity in the tropics, even though they tend to be multi-host and host-specificity is rarely reported. Nevertheless, evidence suggests that host-generalist fungi can have host-specific effects that produce disproportionate negative effects on some plant species but not others, promoting regional diversity.
