The miniaturized systems designed for lab-on-a-chip (LOC) technologies are generally implemented with a micro-scale mixer to provide intimate contact between the reagent molecules for interactions and chemical reactions. PI3K/AKT/mTOR signaling pathway is activated in 90% of all Glioblastoma Multiforme (GBM) tumors. We used a dual targeting approach to interfere with the phosphoinositide 3-kinase- mammalian target of rapamycin (PI3K-mTOR) pathway by treating U87MG GBM cells with NVP-BEZ235 (PI3K and mTOR a dual inhibitor) and, their downstream effects on molecular regulation on glycolytic genes were investigated using RNA-sequencing and gene set enrichment analysis (GSEA). RNA-seq identified 7803 differentially regulated genes in response to NVP-BEZ235. GSEA identified two glycolysis-related gene sets had significant differences (P < 0.05) in gene expression between control and NVP-BEZ235 treated samples. The GSEA core enrichment reported 17 and 35 genes were significantly downregulated (P adj < 0.05) in response to NVP-BEZ235 treatment in two different glycolysis related gene sets. We validated the expression of genes using qRT-PCR in U87MG cells as well as other GBM cell lines. Our results suggest novel insights on hallmark metabolic reprogramming associated with the PI3K-mTOR inhibition and provides advances in developing molecular targeted therapies.

The purpose of this study was to identify protective factors contributing to the college graduation of seven Mexican-American Hispanic former migrant students from the Rio-Grande Valley of South Texas, the southernmost part of the United States that borders Mexico. I used a case study methodology to examine the real life context of a phenomenon with the data collected from the seven participants using interviews. The current study aimed to expand the existing literature on sex offenders with regards to personality and psychological dysfunction, by comparing specific subgroups of sex offenders using the Minnesota Multiphasic Personality Inventory- Second Edition- Restructured Form (MMPI-2-RF). Archival data from a sample of N = 244 adult male sex offenders was grouped based on four characteristic variables: offense type (contact vs. noncontact), relationship to the victim (familial vs. nonfamilial), victim age (younger vs. older), and the presence or absence of a personality disorder diagnosis or features. Thermal-stress events associated with climate change cause coral bleaching and mortality that threatens coral reefs globally. Yet coral bleaching and coral cover patterns vary spatially and temporally, and few studies have identified bright spots where corals have the potential to survive through climate change. This study synthesizes over 10,000 field observations globally of coral bleaching and coral cover from 1997to 2017 and uses a suite of environmental and temperature metrics to analyze coral bleaching and coral cover patterns. This dissertation presents the design and development of a structural health monitoring (SHM) system specifically tailored for transportation infrastructure components, such as bridges. The proposed system collects data by using contactless sensors and performs health characterization and failure prediction. Improving Rangeland Seedling Recruitment Using Fungicide Seed Coatings And Golden Eagle Reproductive Success In Relation To Explosive Military Tests And Trainings, Benjamin William Hoose

The objective of the first chapter of this thesis was to determine whether fungicide seed coatings constitute an effective strategy for increasing seedling recruitment in restoration scenarios in the Intermountain West. We tested a mixture of four fungicides that address potential fungal pathogens to bluebunch wheatgrass, a dominant bunchgrass that is commonly used in restoration. Across two sites and three years, we found that the fungicide seed coating increased emergence in five of the six sites and years, with an average increase of 59.1% over the control. There was a strong interaction (P < 0.001) between the effects of fungicide treatment, the year and the site on emergence. This interaction was likely related to the effects of the hydrothermal microsite environment on disease severity. Further research is necessary to fully understand the conditions under which fungicide seed coatings are most likely to be effective. The objectives the second chapter of this thesis were to 1) estimate the effects of golden eagle nest proximity to explosive disturbances on reproductive success given other relevant habitat variables (e.g. indices of topography and vegetation), and 2) determine the relative importance of nest proximity to explosive disturbances as a predictor of golden eagle reproductive success compared to other relevant habitat variables. Reproductive success data were collected from nesting territories within and surrounding land controlled and managed by the US Department of Defense. We fit the reproductive survey data using generalized linear mixed-effects models comprised of unique, hypothesis-based sets of habitat variables. We compared the models using AICc-based model selection processes. Given the best approximating model, we found no evidence that the likelihood of reproductive success was affected by nest proximity to explosive disturbances (P = 0.460). We further found nest proximity to explosive disturbances consistently ranked in the bottom 50% of relative variable importance. These results may indicate golden eagle tolerance or habituation to explosive military tests and trainings. Although the two chapters of this thesis are disjointed, they are loosely unified by the ecological importance of disturbance, invasive species, and restoration within the Great Basin ecoregion.

Little is known about the size of areas used by female sage-grouse during the breeding and nesting seasons, fidelity of females to those areas, or fidelity of female sage-grouse to specific leks or nest sites. The recent miniaturization of global positioning system (GPS) transmitting devices allows for a more thorough analysis of this behavior. In collaboration with the Sam D. Hamilton Noxubee National Wildlife Refuge, I collected detailed information on the butterflies (Lepidoptera: Papilionoidea Latreille, 1802) of the refuge. I produced a comprehensive checklist of butterflies and skippers numbering 60 species. In addition, I collected data on flight periods as well as local plant community associations. Butterfly abundance surveys were conducted to assess seasonal habitat use across six site classes. Special emphasis was placed on pine forest at various stages of management for the endangered red-cockaded woodpecker (Picoides borealis). Toll-like receptor (TLR) ligands and phytochemical feed additives (PFAs) were evaluated in this study to determine the effects of immune stimulation on gut immunity in the zebrafish, Danio rerio, and the channel catfish, Ictalurus punctatus. Rag1-/- (MT) zebrafish were used to study how the TLR ligands β-glucan and resiquimod (R848) affect the innate immune system in the gut of MT zebrafish. Aeromonas hydrophila ML09-119 is an important fish pathogen that severely affects channel catfish aquaculture. To better understand this strain’s virulence factors, outer membrane vesicles (OMVs) were isolated, and their proteome was assessed. Using transmission electron microscopy and dynamic light scattering, OMVs were shown to be monodispersed particles with an average diameter of 120.33 nm. OMV proteins were identified using mass spectrometry, and analysis of the resulting proteome of 74 proteins revealed that many originated from the cytoplasm, but there was an enrichment of outer membrane, periplasmic, and extracellular proteins compared to the total proteome. Growth rates and diet patterns for sympatric pallid sturgeon Scaphirhynchus albus and shovelnose sturgeon S. platorynchus throughout the Missouri River and lower Mississippi River were examined. Pallid sturgeon growth in the Great Plains Management Unit (GPMU) was slower than in other river reaches until age 9 and pallid sturgeon from the Interior Highlands Management Unit (IHMU) and Coastal Plain Management Unit (CPMU) did not differ. No differences in growth between management units were detected for shovelnose sturgeon.
