Mathematical Modeling Of Lung Inflammation: Macrophage Polarization And Ventilator-Induced Lung Injury With Methods For Predicting Outcome, Sarah B. Minucci
Mathematical Modeling Of Lung Inflammation: Macrophage Polarization And Ventilator-Induced Lung Injury With Methods For Predicting Outcome, Sarah B. Minucci
Theses and Dissertations
Lung insults, such as respiratory infections and lung injuries, can damage the pulmonary epithelium, with the most severe cases needing mechanical ventilation for effective breathing and survival. Furthermore, despite the benefits of mechanical ventilators, prolonged or misuse of ventilators may lead to ventilation-associated/ventilation-induced lung injury (VILI). Damaged epithelial cells within the alveoli trigger a local immune response. A key immune cell is the macrophage, which can differentiate into a spectrum of phenotypes ranging from pro- to anti-inflammatory. To gain a greater understanding of the mechanisms of the immune response in the lungs and possible outcomes, we developed several mathematical models …
Influence Of Metal Sleeves In The Accuracy Of Dental Implant Placement Using Guided Implant Surgery, Coleman Adams
Influence Of Metal Sleeves In The Accuracy Of Dental Implant Placement Using Guided Implant Surgery, Coleman Adams
Theses and Dissertations
The fabrication of implant surgical guides through stereolithographic 3D printing has become a staple in dental implant guided surgery over the last couple decades. These surgical guides have typically utilized metal sleeves to assist in guidance of the drills during osteotome preparation. The metal sleeves can be costly and potentially cause deviations if improperly placed during post-processing of the guide. This research explored a novel method for the utilization of sleeve-free surgical guides by comparing the dimensional and angulational deviations between the implant guides with and without a metal sleeve. To achieve this goal, two separate aims were pursued. Our …
Humanizing Architecture: A Polymorphic Space, Nada Abbara
Humanizing Architecture: A Polymorphic Space, Nada Abbara
Theses and Dissertations
The built environments in which our communities thrive constitute an integral part of human experience and evolution. Yet, many places are detached from the way we experience them due to mass-production, which often produces standardized environments, and due to the tendency of modern architecture to delineate spaces as static objects rather than dynamic interactions. Thus, there is an emerging need to humanize architecture through an interdisciplinary approach that engages nature’s behavioral patterns. The project proposes a transformable polyhedral structure that interacts with human emotion through a three-dimensional morphing space that contracts and expands. This spatial interaction is achieved through a …

Phenotypic Characterization Of Table Mountain (Pinus Pungens) And Pitch Pine (Pinus Rigida) Hybrids Along An Elevational Gradient In The Blue Ridge Mountains, Virginia, Alexander L. Brown
Theses and Dissertations
Hybridization has played a long-standing role in the evolution of both plant and animal species and allows for the sharing of genetic information between lineages. Here, potential hybridization of a species endemic to the Appalachian Mountains, Table Mountain pine (Pinus pungens), and pitch pine (Pinus rigida) was investigated along an elevational gradient, through the use of phenotypic measurements: cone length, cone width, and needle length. Phenotypes were used to identify hybrids in a three-tiered elevational sampling method at two sites in Shenandoah National Park with the use of linear discriminant analysis. It was found that hybridization …
Multiplexed Nucleic Acid Sensing With Single Molecule Fret, Anisa Kaur
Multiplexed Nucleic Acid Sensing With Single Molecule Fret, Anisa Kaur
Theses and Dissertations
In recent years, biomarkers have drawn tremendous interest in the fields of clinical diagnostics and biotechnology. In fact, recent quests for biomarker analyses have discovered a myriad of disease-related biomarkers spanning a wide variety of biomolecules such as proteins, nucleic acids, and small molecule ligands. While the detection of only one target at a time is the most common practice in biosensing today, it has been shown that the accuracy of diagnosis increases significantly by measuring the level of multiple biomarkers instead of just one due to the possible overlap of certain biomarkers in more than one disease. However, currently …

Nature-Inspired Material Strategies Towards Functional Devices, Sayantan Pradhan
Nature-Inspired Material Strategies Towards Functional Devices, Sayantan Pradhan
Theses and Dissertations
Naturally sourced, renewable biomaterials possess outstanding advantages for a multitude of biomedical applications owing to their biodegradability, biocompatibility, and excellent mechanical properties. Of interest in this dissertation are silk (protein) and chitin (polysaccharide) biopolymers for the fabrication of functional biodevices. One of the major challenges restricting these materials beyond their traditional usage as passive substrate materials is the ability to combine them with high-resolution fabrication techniques. Initial research work is directed towards the fabrication of micropatterned, flexible 2D substrates of silk fibroin and chitin using bench-top photolithographic techniques. Research is focused on imparting electrochemical properties to silk proteins using conducting …

Art Museum Docent Coordinators’ Perceptions: A Difficult Kind Of Balancing Act, Jennifer E. Schero
Theses and Dissertations
With visitors numbering in the millions, museums provide numerous entry points for attendees to engage with the objects in their care, including offering tours of their collections. Looking specifically at art museums, many institutions utilize volunteers to facilitate these guided looking experiences. Considerable research within the field of museum education focuses on the qualities of effective touring and methods and theories to support these endeavors. However, there is a lack of research focusing on the docent coordinators who oversee these volunteer guides. This study utilizes interpretive phenomenological analysis to explore perceptions of encyclopedic art museum docent coordinators concerning their preparation …

≥I++ Wayward Self, Space, And Language, Mystie Do, Thu Tm Do
Theses and Dissertations
Having crossed different geographical and cultural borders, I can’t seem to position myself beyond society’s forced binaries of race, sexuality, and materiality. My personal diasporic experience drives me to bring together various presences, putting them next to one another, engaging them in dialogues. My works often share multiple space–time possibilities: a digital space, a physical site, a virtual alternity. Occupying these parallel worlds are varied ratios of mixtures of natural ecosystems, my own system of abstraction, and existing technological systems that allow us a level of individual engagement not previously available. I want to invite people to fluctuate between these …

Chains Of Stability: Building Financial And Cultural Community Sustainability For Latinas In Richmond & Chesterfield, Virginia, Gabriella C. Pino-Moreno, Gabriella Pino-Moreno
Theses and Dissertations
As the Latinx population in the US continues to grow, new Latin-centric ethnic enclaves are developing in urban areas, including those in US southern states. While there has been some discourse on the role of Latin immigrants in the US labor market generally, there is limited literature addressing the specific experiences of Latinas as inhabitants of urban spaces and the factors impacting their economic sustainability. Latinas, on average, earn $0.54 to every $1.00 a white male earns. Their economic position, combined with their cultural traditions and practices, raises questions about how they achieve economic security in the US. This research …

A Novel Review Of Heat Shock Protein 110 Kda: A Basis For Research And Continued Experimentation Through Biochemical Analysis, Crist W. Cuffee
Theses and Dissertations
Heat shock protein 110 kDa, Hsp110, is a distinct cellular protector, different in form and function from Hsp70, a close relative of Hsp110. Functioning primarily as a holdase or in tandem with other molecular chaperones, a review of current accomplishments elucidates the uniqueness of this protein and the continued mysteries that surrounds it. Found only in eukaryotes, Hsp110 has been linked to many diseases, ranging from parasitic infection to neurodegenerative disorders. While still lacking, studies of this protein have provided much in the realm of speculation on the mechanisms behind Hsp110s’ contribution to different pathologies. This review will serve as …
Evolution And Clinical Relevance Of Vaginal Bifidobacterium Breve In The Vaginal Environment, Nicole R. Jimenez
Evolution And Clinical Relevance Of Vaginal Bifidobacterium Breve In The Vaginal Environment, Nicole R. Jimenez
Theses and Dissertations
The vaginal microbiome is important in reproductive health and disease and is often dominated by lactobacilli. However, there are many other community state types dominated by single species that are still not well characterized. Bifidobacterium species such as Bifidobacterium breve and Bifidobacterium longum have been observed to dominate vaginal profiles. Bifidobacteria are important in infant gut development, but little is known about their role in vaginal health. Thus, we sought to use a multisystem approach to better characterize Bifidobacterium species present in the vaginal microbiome with an emphasis on B. breve. Eight strains of B. breve were isolated from vaginal …
Effects Of Abiotic And Biotic Factors On Hornworm Distributions, Emelia O. Kusi
Effects Of Abiotic And Biotic Factors On Hornworm Distributions, Emelia O. Kusi
Theses and Dissertations
Determining the drivers of species occurrences is a central topic in ecology. Both abiotic and biotic factors are important predictors of species distributions, yet most species distribution models consider abiotic variables only. In this study, the influence of abiotic and biotic factors on current distributions of herbivores, Manduca sexta, and Manduca quinquemaculata, their major cultivated and wild host plants, and a parasitoid, Cotesia congregata, was quantified using an ensemble of three models (generalized linear model, maximum entropy, and random forest). The predictive ability of abiotic-only, biotic-only, and abiotic-biotic models was evaluated. Winter temperatures and precipitation (i.e., precipitation of …
A Novel Immunostimulatory Platform For Amplifying The Abscopal Response Rates Of Radiation Therapy, Patrick A. Paez
A Novel Immunostimulatory Platform For Amplifying The Abscopal Response Rates Of Radiation Therapy, Patrick A. Paez
Theses and Dissertations
Radiation therapy (RT) is one of the primary treatment modalities for head and neck squamous cell carcinoma (HNSCC). At the time of diagnosis two-thirds of HNSCC patients have local-advanced disease and 50-60% of these patients will experience a local-regional or metastatic relapse within three years. Improving the immunogenic response of RT may help address this clinical problem. However, current RT regimens have failed to reliably generate robust antitumor immunity as evidenced by the rarity of clinical abscopal responses. Recently we engineered a chimeric fusion molecule called Flagrp170, a novel immunostimulatory agent highly capable of promoting antigen presentation and T-cell activation. …

Theses and Dissertations
Schizophrenia is a severe neuropsychiatric disorder that presents with diverse symptoms, some of which remain resistant to treatment. Increased risk of neurodevelopmental disorders such as schizophrenia has been observed following gestational infection in humans, leading to development of maternal immune activation (MIA) animal models. Increased density of the serotonin 5-HT2AR receptor (5-HT2AR), the primary target of hallucinogenic drugs and a key target of atypical antipsychotics, has been observed in postmortem antipsychotic-free prefrontal cortex samples from schizophrenia subjects, a change reflected in frontal cortex of adult MIA offspring. To model MIA, we administered 20 mg/kg i.p. of the viral mimetic poly-(I:C) …

By Rashed M. Almuqbil, Pharm.D.
Director: Sandro R.P. da Rocha, PhD
Cancer is a leading cause of death worldwide. The available treatment for solid tumors may include surgery, radiotherapy, immunotherapy, and chemotherapy or a combination of all those regimens. Chemotherapy is widely used in the treatment of both lung cancers and breast cancer, including metastases, irrespective of type and stage. The poor rates of survival can be attributed to late diagnosis, and due to the development of acquired and innate chemoresistance. Lung tumors are intrinsically …
Teachers' Views On The Intersectionality Between Culture And Student Behaviors, And Experience Using Culturally-Responsive Behavior Interventions, Toshna Pandey
Theses and Dissertations
Students belonging to racially minoritized groups experience more frequent and intense disciplinary consequences for similar rule violations as their White peers. Factors such as deficit-oriented perceptions and implicit biases among teachers have contributed to the disproportionate exclusion of racially minoritized students, thus negatively affecting their social, emotional, behavioral, and school success. Using semi-structured interviews, this study sought to explore elementary school teachers’ views on the intersectionality between race/culture and student behaviors. Additionally, it also examined their experiences using behavior interventions effective for racially minoritized students. Findings suggest that participants often attributed challenging behaviors to student-level factors such as family and …
Comparison Of Antibiotic Resistance In Biofilm, Sediment, And Planktonic Communities Of The James River, Mary Bernadette Coughter
Comparison Of Antibiotic Resistance In Biofilm, Sediment, And Planktonic Communities Of The James River, Mary Bernadette Coughter
Theses and Dissertations
Antibiotic resistance spread within aquatic bacterial communities, particularly ones in urban areas, is in part driven by wastewater inflows that may contain both elevated concentrations of antibiotics and increased abundance of antibiotic resistant bacteria. These can be transmitted into the environment via release of treated wastewater, since removal of these compounds by most wastewater treatment facilities is inadequate, or by release of untreated waste from leaking wastewater treatment networks and overflow of combined sewer systems. This study used the James River and the City of Richmond wastewater network as a model urban waterway to study both types of impacts. Shotgun …

Standardization Of E-Cigarette Research: Addressing The Analysis Of Total Nicotine And Free Base Nicotine In E-Cigarettes, Vinit Gholap
Theses and Dissertations
Electronic cigarettes (e-cigarettes) are one of the major sources of exposure to nicotine, an addictive chemical. Since 2016, the Food and Drug Administration (FDA) has started regulating these products under the Tobacco Control Act yet, specifications about the nicotine content in these products have not been established. In e-cigarettes, nicotine concentration ranges from 0 to >50mg/mL. Additionally, several e-cigarette related parameters have been identified which can affect nicotine delivery to users. Of these factors, nicotine forms (protonated and free base) have been found to play a significant role in nicotine absorption profiles yet, a limited and contradictory research is available …
Using The Memorial Sloan Kettering Frailty Index To Identify Patients At High Risk For Postoperative Intensive Care Unit Admission, Specialized Advanced Care Unit Admission, And 30-Day Readmission, Timothy J. Donoghue
Theses and Dissertations
Frailty is roughly defined as an accumulation of physiological, emotional, cognitive, and social deficits that impair a person’s response to stressful events. A frailty diagnosis has been associated with poor outcomes following surgical procedures. Cancer surgical patients aged 65 or older represent a vulnerable population susceptible to being frail and the potential associated complications that can accompany frailty. Measuring frailty is an objective risk assessment that identifies increased risk better than age or American Society of Anesthesiologists Physical Status (ASA-PS) score such that frailty can independently predict poor surgical outcomes.
Frailty is not specifically a result of having cancer, disability, …
Cardiac Arrest And Global Ischemia Reperfusion Injury: Role Of Mitochondria And Cannabinoid Receptor 2 Signaling., Jennifer L. Bradley
Cardiac Arrest And Global Ischemia Reperfusion Injury: Role Of Mitochondria And Cannabinoid Receptor 2 Signaling., Jennifer L. Bradley
Theses and Dissertations
We explored the effects of cardiac arrest on mitochondria populations and heart whole tissue lysate proteomics utilizing the Weil Institute’s in vivo rat model of cardiac arrest and cardiopulmonary resuscitation. We have discovered that brain mitochondria are more sensitive to global ischemia compared to heart mitochondria. Additionally, complex I is the most sensitive electron transport chain complex to ischemic injury and is a major control point of the rate of oxidative phosphorylation following cardiac arrest and cardiopulmonary resuscitation. Preservation of brain mitochondrial activity and function during cardiac arrest may enhance outcomes and recovery.
A recent article focusing on acute myocardial …
Water Clarity At The River-Estuary Transition Zone: A Comparative Study Of The James, Mattaponi, And Pamunkey Sub-Estuaries, Rachel Henderson
Water Clarity At The River-Estuary Transition Zone: A Comparative Study Of The James, Mattaponi, And Pamunkey Sub-Estuaries, Rachel Henderson
Theses and Dissertations
Water clarity is a key parameter for monitoring water quality and often used to assess habitat suitability for submerged aquatic vegetation (SAV). Light attenuation, a measure of water clarity, is impacted by colored dissolved organic matter (CDOM), and by suspended particulates which include living and non-living components. We anticipated that the relative importance of these factors in regulating light attenuation would vary among the upper portions of three sub-estuaries differing in morphometry, hydrology, and degree of human influence. The James is characterized by eutrophic conditions and high algal abundance, whereas the Mattaponi and Pamunkey exhibit lower phytoplankton production. The Mattaponi …
Preventing Obesity: A Social Ecological Exploration Of Centers For Disease Control Prevention Strategies And Guidance Toward Healthy Eating, Physical Activity, And Communities, Ryan Kelly
Theses and Dissertations
Abstract
Obesity has reached nearly 40% of the adult public in the United States, costing the citizen taxpayer over $200 billion annually in healthcare costs. Those suffering from obesity deal with multiple physical and mental repercussions. Through a content analysis of four Centers for Disease Control (CDC) documents and guided by the conceptual framework of the social ecological model, this research explores the federal approach to preventing obesity. The analysis finds that CDC solutions to obesity involve connecting people to each other and healthier choices, an orientation toward local public administration, and an emphasis on environmental and infrastructure improvements. The …
Single Molecule Investigations Of Holliday Junction Binding Protein Ruva, Dalton Reed Gibbs
Single Molecule Investigations Of Holliday Junction Binding Protein Ruva, Dalton Reed Gibbs
Theses and Dissertations
DNA breaks are inevitable as they mainly occur due to cells’ own reactive oxygen species (ROS). While DNA breaks can be single-stranded or double-stranded, the double-stranded DNA (dsDNA) breaks are more dangerous. If such damage is not repaired, it can lead to genetic instability and serious health issues including cancers. One way dsDNA breaks can be repaired is via a process called homologous recombination (HR), which involves several DNA-binding proteins. Therefore, to have a better insight into the repair mechanism and origin of repair defects, we need a better understanding of how these proteins interact with DNA itself and DNA …
Novel Roles Of Yy1 In Astrocytes, Karli A. Mockenhaupt
Novel Roles Of Yy1 In Astrocytes, Karli A. Mockenhaupt
Theses and Dissertations
Astrocytes tile the entire central nervous system, are diverse across brain regions, and perform specialized functions fine-tuned for their neuronal circuit. Initially, astrocyte heterogeneity is generated through intrinsic programs specified by their progenitors. Then, as astrocyte mature, they are specialized by region-specific communication with surrounding cells. The molecular programs that regulate this diversity and allow for the accommodation to the needs of the local environment remains elusive. We explored a ubiquitously expressed zinc finger transcription factor, Yin Yang 1 (YY1), that both activates and represses transcription by regulating loops of active chromatin. This report describes the diverse functions of astrocytic …
Straight Through My Heart, Raul A. Aguilar Canela
Straight Through My Heart, Raul A. Aguilar Canela
Theses and Dissertations
Straight through my heart is an exhibition that explores the concept of heartbreak as a socio-political phenomenon. Through the affect of sadness the thesis analyses the way in which subjects are formed under cognitive capitalism. Paying particular interest to the collateral effects of neoliberal culture —hyper-stimulation, self-exploitation, competition, and obsession with productivity—and the pathologies they create —depression, anxiety, body aches, fatigue— this work shifts the burden of sadness from the individual to the community. By doing so it proposes heartbreak as a public feeling.
Beyond Black Or White: An Examination Of Intersectional Identities, Blame Attribution, And Sexual Violence, Ashlynn D. Bell
Beyond Black Or White: An Examination Of Intersectional Identities, Blame Attribution, And Sexual Violence, Ashlynn D. Bell
Theses and Dissertations
While there may be more discussions of sexual violence than there have been in previous years, cases continually arise where the responsibility of those who have been assaulted is called into question (Alaggia & Wang, 2020). Secondary victimization, or victim blaming, is partially responsible for the continued misattribution of responsibility to survivors. These perspectives minimize the experiences of survivors, which dissuades individuals from the reporting of perpetrators. The current study aimed to evaluate whether particular intersecting identities influenced perceptions of responsibility, in a hypothetical scenario depicting an assault. The race, gender identity, as well as perceptions of respectability of the …
