Tissue engineering is a dichotomy of scaffold-based and scaffold-free cultures. Scaffold-based cultures form highly organotypic structures but with low uniformity and throughput. Conversely, scaffold-free cultures create consistently sized and shaped cell aggregates with limited spheroid-like structure and function, thus restricting their use for accurate disease modeling. We hypothesized that combining aspects of each culturing format, we would produce highly organotypic structures of consistent size and shape for use in pulmonary modeling. Adults with autism face many difficulties when finding employment, such as struggling with interviews and needing accommodating environments for sensory issues. However, autistic adults also have unique skills to contribute to the workplace that companies have recently started to seek after, such as close attention to detail and trustworthiness. Modulation of materials properties through light-based illumination has the capability to expand the technology sector due to stimuli-responsive dynamic behavior that cannot be achieved in traditional materials. For example, reversibly tuning photophysical profiles of such materials allows for switching between discrete states that is a key aspect for the developing logic gates, spatially- and temporally-resolved sensors, and on-demand drug delivery systems. A complete understanding of the structure-function relationship of proteins requires an analysis of their dynamic behaviors and the static structure. However, all current approaches to studying dynamics in proteins have their shortcomings. A conceptually attractive and alternative approach simultaneously characterizes a protein's structure and its intrinsic dynamics. Ideally, such an approach could solely rely on RDC data-carrying both structural and dynamical information. The major bottleneck in utilizing RDC data in recent years has been attributed to a lack of RDC analysis tools capable of extracting the pertinent information embedded within this complex data source.

Resource variability and availability often drives competition within ecosystems, which can lead to the diversification of organismal niches and physiological capabilities. In aquatic systems, common resources that photosynthetic organisms, such as algae, compete for are light and carbon. Both the spectral characteristics (color) and carbon concentrations of an aquatic system vary with time and space, which means that algae need to be able to respond to changes in resource availability to survive. Youth obesity prevalence remains high, despite decades of intervention. Grounded in the social ecological model, neighborhoods and schools are important settings in addressing the complex systems that influence obesity. Contributing to disparities, by race/ethnicity, Black and Hispanic youth are more likely to live in segregated neighborhoods and attend segregated schools, which are also more likely to have high rates of poverty and are less likely to have high quality nutrition and built environments. This dissertation is split into three sections, each containing new results on a particular combinatorial problem. In the first section, we consider the set of 3-connected quadrangulations on n vertices and the set of 5-connected triangulations on n vertices. In each case, we find the minimum Wiener index of any graph in the given class, and identify graphs that obtain this minimum value. Moreover, we prove that these graphs are unique up to isomorphism.

Polo-like kinase 1 (PLK1) is a central player in regulating entry into and progression through mitosis. Inhibition of sub-cellular localization and kinase activity of PLK1 through the Polo-box domain (PBD) is emerging as a viable alternative to ATP binding site directed drugs for which the development of resistant mutants and inhibition of closely related members of the PLK family (tumor suppressor roles) are primary concerns. I describe related novel non-peptidic PBD binding inhibitors, termed abbapolins, identified through successful application of the REPLACE strategy and demonstrate their potent antiproliferative activity in prostate tumors and other cell lines. Consanguinity, defined as the degree of relationship between closely related individuals, is a widespread historical practice that is not specific to any one religion, population, or region of the world. Genetic counselors regularly ask whether a reproductive couple is consanguineous as part of the review of family history and risk assessment. Couples who are in consanguineous relationship may be subject to negative attitudes and stigma, potentially due to cultural differences and norms at a population level and these attitudes may interplay with questions, answers, and discussions around consanguinity. Each ach year a growing number of individuals report lingering deficits months-years following concussion. Persistent post-concussion symptoms (PPCS) can negatively impact day-today activities and if left untreated may manifest in severe neurological sequelae resulting in long-term cognitive impairment or advanced neurological degeneration (i.e., CTE). Current clinical diagnostic and prognostic assessments (e.g., symptom reports and neurocognitive testing) lack the sensitivity to quantify neurological function. Concerns about unsafe food influence food choice, and consumption of unsafe foods increases morbidity and mortality, particularly in low- and middle-income countries. Actions to ensure safety of food are dominated by mitigation of biological and chemical hazards through supply-side risk management, disregarding individuals’ experiences and perspectives of food safety. We aimed to identify and categorize perspectives about food safety in five countries. Five Drivers of Food Choice projects provided transcripts from 17 focus groups discussions and 303 interviews in Kenya, Ghana, India, Guinea, and Vietnam. This thesis investigates the way that magical ability and the lack thereof has been racialized through the deployment of blood-based models of inheritance in young adult fantasy literature. Because of the connection between blood and biology, texts that only allow magical transmission from parent to child place significant limits on who can access witchery, creating systems of privilege that enforce racial and classed hierarchies. The disproportionate attrition of female students in STEM is well documented in higher education but less well understood at the secondary level. In a large, public high school in an affluent, highly educated city, the high honors mathematics pathway is the only math pathway that has not yet achieved gender parity, an implication for future educational and economic equity. Female students leave this pathway at significantly higher rates than male students, especially during and immediately after the first course. The genus Vibrio consists of Gram-negative bacteria that possess a curved rod shape and are routinely isolated from estuarine and coastal salt water. Vibrio cholerae, Vibrio parahaemolyticus, and Vibrio vulnificus are the major three species that contribute to human disease worldwide, and a variety of other Vibrio species contribute to persistent problems in the aquaculture and fishing industries. This dissertation explores gendering practices of visual information catalogers. The work aims to understand how catalogers perceive gender when describing persons within visual information. The qualitative study deployed queer interpretative phenomenological analysis to understand how catalogers think broadly about describing identity. The infused queer theoretical tenets helped to understand that while participants may not directly name gender as challenging, the conflation of gender into cisnormative monoliths (assuming every person's gender matches their sex-assigned-at birth) or silence around gender produce telling opinions concerning nonbinary gender. The advent of cART has greatly improved the quality of life of infected individuals; however, roughly 50% of these individuals will still experience HIV-1 associated neurocognitive disorders (HAND). This dissertation sought to contribute to the evidence-base of diabetes management and acute complications associated with alcohol and cannabis use among individuals with Type 1 Diabetes (T1DM). Three interconnected studies were initiated to complete this goal. First, existing literature surrounding the potential consequences of cannabis consumption among individuals with T1DM were summarized within a systematic review. The Fat Mass and Obesity (FTO) gene on chromosome 16 has been reported to be associated with obesity, solid tumor cancers (lung, renal, breast, prostate, pancreatic, endometrial), and more recently with hematologic malignancies, like AML. Breast cancers (BrCa) that overexpress oncogenic tyrosine kinase receptor HER2 are treated with HER2-targeting antibodies (such as trastuzumab) or small-molecule kinase inhibitors (such as lapatinib). However, most patients with metastatic HER2+ BrCa have intrinsic resistance and nearly all eventually become resistant to HER2-targeting therapy. Resistance to HER2-targeting drugs frequently involves transcriptional reprogramming associated with constitutive activation of different signaling pathways. We have investigated the role of CDK8/19 Mediator kinase, a regulator of transcriptional reprogramming, in the response of HER2+ BrCa to HER2-targeting drugs. In specific biomedical applications, ENMs are surface-functionalized using polymers, proteins, and other stabilizing agents to facilitate their resistance to salt-induced aggregation. The objective of this study was to assess the perceived utility of an educational resource booklet addressing the psychiatric manifestations and mental health considerations within 22q11.2 deletion syndrome (22q11.2DS).
Seventy-three parents of children with 22q11.2DS completed online surveys before and after reading the booklet. The surveys assessed personality factors and feelings of empowerment, worry, self-stigma, and ability to tolerate uncertainty.

Dyslexia affects between 5% and 18% of Americans and is caused by difficulty with phonological processing. This study investigates the impact of an online intervention designed to remediate phonological processing deficits on reading accuracy, fluency, and comprehension. It also investigates changes to student self-concept and parent perceptions of their children’s reading attitudes and abilities as a result of the online intervention.

Individuals with attention-deficit/hyperactivity disorder (ADHD) are at significantly greater risk for alcohol and marijuana misuse and abuse compared to their peers. College students with ADHD may be particularly vulnerable to problematic alcohol and marijuana use, given the widespread use and abuse of these substances by college students generally and liability associated with ADHD specifically. Experiential avoidance, or avoidance of unwanted thoughts, feelings, and other internal experiences, is positively associated with ADHD symptoms and substance use problems.
