Monocytes are professional antigen presenting cells (APC). They serve as precursors of macrophages and dendritic cells (DC). We have used cytopathic (cp) and non-cytopathic (ncp) Bovine Viral Diarrhea Viruses (BVDV) to determine the genes and proteins expression levels in bovine monocytes. Four specific aims were accomplished in this study. The first aim was to assess the baseline expression of the proteins involved in professional antigen presentation in bovine monocytes. The results showed that the differential detergent fractionation (DDF) approach can provide interpretable and meaningful functional information in bovine monocytes. Fuzzy logic provides a natural way of dealing with models that require the modeler or user to interpret linguistic variables in order to make a decision. This study focuses on employing fuzzy logic methodologies in order to create an Alamo switchgrass ecological site suitability map for the state of Mississippi. The inputs in the model are available water content (AWC), slope, pH, drainage, color, and texture. All variables were examined through the fist six inches (15cm) of soil. A physiologically-based toxicokinetic and toxicodynamic (PBTK/TD) model was developed, from the open literature, to predict the toxicokinetic disposition and toxicodynamic response (acetylcholinesterase inhibition) of a ternary organophosphorus (OP) insecticide mixture: chlorpyrifos (CP), methyl parathion (MP) and parathion (P). In vivo studies were conducted in adult male Sprague-Dawley rats, orally administered one of two CP/MP/P mixtures (2.5, 0.5, 0.5 mg/kg or 5, 1, 1 mg/kg) with selected tissues (blood, brain, diaphragm, liver, lung and skeletal muscle) collected at 30min, 4, 12 and 24hr postdosing. The purpose of this study was to explore the postsecondary education transition experiences of graduates of Choctaw Central High School (CCHS) who received support from the Tribal Scholarship Program (TSP) to better understand the barriers to successful postsecondary completion and the means to overcoming those barriers. Successful transitions are key for students to persist to academic completion. Studies show that American Indian students have low rates of college completion and experience a variety of factors attributed to withdrawal from college. This study was conducted to determine infestation levels of southern green stink bug, Nezara viridula (L.), and green stink bug, Acrosternum hilare (Say), on host plants including corn, cotton, soybean, sorghum, and non-crop vegetation in a multicrop ecosystem. Stink bug infestations responsible for crop damage on vegetative and panicle stages of grain sorghum planted on different dates were also investigated. Both N. viridula and A. hilare appeared to establish a first generation on non-crop grasses surrounding cultivated fields and then moved to soybean or cotton to begin a second generation. Variable-density thinning (VDT) is a management option designed to increase structural heterogeneity in second-growth conifer stands. This study examined subcanopy tree growth response to two variations of VDT. At the Forest Ecosystem Study in western Washington, thinning intensity was found to have a significant effect on height growth of Douglasir (Pseudotsuga menziesii (Mirb.) Franco) seedlings which established following the thinning. At the Olympic Habitat Development Study in western Washington, basal area growth response was examined for residual midcanopy western hemlock (Tsuga heterophylla (Raf.) Listeria monocytogenes is a deadly, Gram-positive foodborne pathogen that is ubiquitous in the environment. The bacterium expresses a number of virulence and stress adaptation proteins that support its pathogenic capabilities. Two-dimensional gel electrophoresis (2-DE) was used to map L. monocytogenes surface proteins, which play a central role in virulence, and to examine protein expression by L. monocytogenes grown on ready-to-eat meat, an important source of Listeria infections. A novel method for solubilization of surface proteins from L. monocytogenes for 2-DE was developed. This project evaluated the influence of habitat parameters and distance from low-order streams on herpetofauna communities, in addition to evaluating the efficacy of 2 sampling methods. Amphibians were associated with mature hardwood forests with high density of large, highly decayed woody debris. Reptiles were associated with mixed pine-hardwood forests, woody debris, and vertical canopy structure. Overall, woody debris was an important habitat feature for amphibians and reptiles. Amphibians were detected in greater abundance and species richness near the stream. Reptiles were similar in abundance and species richness as distance increased from low-order streams. This investigation focuses on the effects of hurricane Katrina on the structure, performance, capacity, and future of the lumber industry in the impacted area. The basis of the examination is a survey of both hardwood and softwood lumber mills in Mississippi, Louisiana, and Alabama. Total response rate was 42.4%. Results support the hypothesis that the storm did have a significant and negative effect on the commerce of responding facilities. Aquatic vegetation provides important habitats for fish, but these habitats are increasingly being altered anthropogenically. My research evaluated the movement and habitat use of three small rare fish species, the blackchin shiner Notropis heterodon (BCS), blacknose shiner Notropis heterolepis (BNS), and banded killifish Fundulus diaphanous, in a Minnesota lake. BCS and BNS traveled farther in spring than summer, but selection of habitat based upon macrophyte biovolume did not explain these differences. All three fish species traveled long distances (> 1,800 m) and were capable of reaching all available habitats in Square Lake. Macrophyte species richness and prey (zooplankton) abundance were …

The goal of this project is to advance the understanding of tendon disease and injury through the design and validation of a new model of tendon injury. Controlled, mechanically-induced traction injury was created in rat Achilles’ tendons with a tensioning apparatus through a minimally invasive surgical approach. Tendons were tensioned to subailure loads. Assessments of damage to tendons included gross morphology, evaluation of biomechanical of load strain curves, histopathology under light microscopy (H&E), collagen fibril structure and disruption under scanning electron microscopy, and immunohistochemical study of MMP-13 expression under confocal laser microscopy. Botulinum Neurotoxin (BoNT) is one of the most potent toxins known to human kind. The Atomic Force Microscope (AFM) was employed to investigate the conditions under which BoNT type A heavy chain would bind and/or insert into mica supported dipalmitoylphosphatidylcholine (DPPC) lipid bilayers. As an alternate technique, DPPC/GT1b or total ganglioside extract (80:20) monolayers of a Langmuir Blodgett (LB) Trough were adapted to be artificial membrane models for toxin insertion studies. Age-related macular degeneration (AMD) is the leading cause of blindness in the United States of America. The pyridinium bis-retinoid A2-ethanolamine (A2E) has been implicated to play a role in AMD. We have observed novel pyridinium bis-retinoids through melanolipofuscin and human RPE extractions that may also play a role in the pathology of AMD. We have begun the construction of an amino-retinoid library in order to identify these ocular compounds. This qualitative research study examined the ways those who identify themselves as theistic scientists and scholars experience inspiration, as defined as divine guidance or influence, in their scientific scholarship and discovery. It also explored participants' beliefs about how scientists and scholars can seek and prepare to receive inspiration in their work. Open-ended surveys of 450 participants from the behavioral and natural sciences and from a variety of religious backgrounds were analyzed for content themes in the areas of experiences with inspiration, preparing to receive inspiration, and further thoughts on inspiration in science. The purpose of this study was to explore the potential benefits of a primitive wilderness skills intervention (POST) on the therapeutic change process of adolescent females enrolled in a residential treatment center. A qualitative data analysis approach was used. A convenience sample of eight female adolescents was selected by therapist referral. The data were analyzed using open, axial, and selective coding. Data analysis was used to assess participants' progress in the therapeutic change. Activation of AMP-activated protein kinase (AMPK) results in the upregulation of several intracellular systems which help to prepare a cell for a high energy challenge. The magnitude of the AMPK response to a 10 min bout of exercise has been found to decrease in red quadriceps (RQ) following training, while putative AMPK roles seem to be maintained; specifically, the biogenesis of mitochondria and higher levels of hexokinase II and glucose transporter 4 (GLUT4). Previously it has been shown that cultures of Plasmodium falciparum died following exposure to a febrile temperature of 40°C, as demonstrated by a decrease in parasitemia of the following generation. In the current study, the effect of 40°C treatment on culture media, erythrocytes, and parasite glucose consumption, were ruled out as possible influences on parasite death, demonstrating that 40°C impacted the parasites directly. Metabolic profiling of DNA synthesis, protein synthesis, and glucose utilization during exposure to 40°C clearly indicated that febrile temperatures had direct effect on major metabolic pathways and parasite development, beginning 20-24 hr after erythrocyte invasion. The present study examined whether perceptions of parents, peers, romantic partners and God were predictive of eating disorder symptom severity among women in treatment for eating disorders. The sample included 417 women (ages 12 to 56 years) at an inpatient treatment facility for eating disorders. Participants completed a battery of assessment measures at intake and discharge. Change scores were also computed on all measures. Measures included indices of eating disorder symptomology, parental relationships, peer relationships, romantic partner relationships, and religious well-being. The purpose of this study was to (a) examine the relationship between family leisure involvement and aspects of family functioning (adaptability and cohesion) among Samoan families residing on American Samoa, and (b) to compare the Samoan data to a broad sample of American families to provide a cross-cultural comparison. The sample consisted of 340 adult participants. The Family Leisure Activity Profile (FLAP) was used to measure family leisure involvement. FACES II was used to measure family functioning. These 4 chapters represent manuscripts formatted for submission to journals based on an experiment conducted on Ring-necked Pheasants (Phasianus colchicus). Calcium limits the distribution of this species, which produces 7-15 eggs per clutch. They may renest up to 5 times per breeding season on a diet low in calcium. The first chapter examines egg production in laying hens on 7 different diets from 0.19-4.47% calcium in the absence of calcium loading. Calcium-loaded pheasants store calcium in medullary bone before an experiment and can draw on this surplus during egg production, possibly skewing experimental results. Secreted Phosphoprotein-24 (Spp24) was initially isolated and characterized as a component of bovine cortical bone matrix. Subsequent characterization has shown it is multiply phosphorylated and homologous to cystatin and TGF-β receptor type II. Spp24 is a minor component of the serum fetuin mineral complex that binds calcium-phosphate minerals and prevents their deposition. The TGF-β receptor homology domain binds BMP-2 weakly in vitro and enhances BMP-2’s osteogenic effects in vivo. The ability of Spp24 to affect BMP activity suggests an important role for Spp24 as a native, bioactive componentof bone that regulates bone development.

Niche separation may be the key to promoting the long-term coexistence of introduced and native species. Physical alterations to the environment (habitat manipulation) or re-introducing native species to former habitats can exploit the maladapted traits of introduced species to create a refuge for native species. The gas phase reactions of CH3O2 + CH3O2, HO2 + HO2, and CH3O2 + HO2 in the presence of water vapor have been studied at temperatures between 263 and 303 K using laser flash photolysis coupled with UV time-resolved absorption detection at 220 nm and 260 nm. Water vapor concentration was quantified using tunable diode laser spectroscopy operating in the mid-IR. The HO2 self-reaction rate constant is significantly enhanced by water vapor, consistent with what others have reported, whereas CH3O2 self-reaction and the cross-reaction (CH3O2 + HO2) rate constants are nearly unaffected. The purpose of this thesis project was to examine the effects of chronic alcohol on the excitability and molecular adaptation of GABA neurons of the ventral tegmental area (VTA). GABA neurons are of interest with regards to ethanol intoxication, reinforcement, and dependence due to their widespread distribution and connectivity to mesocorticolimbic dopamine (DA) neurons implicated in alcohol reward and addiction. Since we have previously shown adaptation of VTA GABA neuron firing rate to chronic ethanol (Gallegos, Criado et al. 1999) and suppression of gap-junction (GJ) mediated coupling between these neurons by acute ethanol (Stobbs, Ohran et al. 2004), we wanted …
