Nicotine is one of the most commonly used drugs among adolescent populations. Given the fact that adolescence is a unique developmental stage, during which nicotine has long-term effects on future drug-taking behavior, it is essential to understand how early exposure to nicotine during adolescence may affect the abuse liability of other drugs. We hypothesize that repeated exposure to low doses of nicotine in adolescence induce age-specific enhancement of the rewarding effects of several drugs of abuse in the conditioned place preference (CPP) test. Kinetics And Mechanisms Of Accumulation For Liposomal Ciprofloxacin Into Rat Alveolar Macrophages, Sayeed Mossadeq

The kinetics and mechanism of accumulation for liposomal ciprofloxacin (Lipo-CPFX) into the rat alveolar macrophage NR8383 cells were studied in vitro, in comparison to unformulated ciprofloxacin (CPFX). Upon incubation with CPFX or Lipo-CPFX, cellular drug accumulation was determined from the cell lysates or efflux was from the extracellular media by fluorescence-HPLC. The accumulation for Lipo-CPFX reached the asymptotic values at ≥ 2 hours, which was a result of uptake and efflux. The uptake appeared to be due to liposomes, mediated via cellular energy-independent mechanism like lipid fusion. In contrast, the efflux appeared to be due to ciprofloxacin, partly cellular energy-dependent, …

HIV-Associated Neurocognitive Disorders (HANDs) are becoming one of the largest problems in patients infected with HIV-1. The ability of infected cells such as monocytes and microglial cells to act as viral reservoirs causes extreme inflammation in the CNS and has led to several different types of neurocognitive problems. Specifically these HIV-1 infected monocytes are able to secrete inflammatory factors such as the regulatory protein Tat which acts as a chemoattractant for monocytes while also promoting the adhesion of leukocytes to the extracellular matrix (ECM). Oligodendrocytes are cells located in the central nervous system (CNS) that are responsible for the production of the lipid rich membrane, myelin. Myelin and the process of making and wrapping myelin around an axon (also known as myelination) are critical for normal development since they ensure proper signal conduction in the vertebrate CNS. In multicellular organism, bioenergetic metabolism is strictly regulated toward efficient generation of ATP. However, in certain situations, such as in limiting oxygen or in the rapidly proliferating system like growing juvenile or cancer cells, organisms apply the metabolic strategy that favors the production of biomass (e.g., nucleotides, amino acids, and lipids) over efficiency of ATP generation. The conserved estrogen-related receptors (ERRs) are master regulators in controlling metabolic homeostasis, and good candidates for mediating the metabolic transition induced by hypoxia and development. Youth with type 1 diabetes (T1D) from single-parent families are more likely to be in poorer glycemic control (HbA1c). Demographic trends indicate more households are composed of unmarried adults and fewer youths. Family density, or a youth: adult ratio, may be a more salient factor than single-parent status in the association with glycemic control. Data from 257 adolescents aged 11-14 years (M = 12.84) at two different sites were collected as part of a randomized control trial of a treatment intervention designed to increase parent involvement and prevent deterioration of adolescent diabetes disease care. Liposomal ciprofloxacin (Lipo-CPFX) is being developed for inhalation, with a goal of sustaining the therapeutic activity, compared to unformulated ciprofloxacin (CPFX). However, the kinetics and mechanism of its sustained local lung retention and pharmacological activity are yet to be fully characterized. This project hypothesized that Lipo-CPFX enables slower and sustained lung epithelial transport and uptake, compared to CPFX, thereby producing prolonged local pharmacological actions. Swelling-activated Cl− current (ICl,swell) is an outwardly-rectifying current that plays an important role in cardiac electrical activity, cellular volume regulation, apoptosis, and acts as a potential effector of mechanoelectrical feedback. Persistent activation of ICl,swell has been observed in models of cardiovascular disease. We previously suggested sphingosine-1-phosphate (S1P) activates volume-sensitive Cl- current (ICl,swell) by ROS-dependent signaling. S1P and its analog, FTY720 (fingolimod), primarily act via G-protein coupled receptors (S1PR; S1PR1-3 in heart), but several intracellular S1P ligands are known. Until recently, much of the research on intimate partner violence has focused primarily on male aggressors. However, research has increasingly indicated that women use violence against male intimate partners at higher rates than previously suspected. Significant controversy exists with regard to the context, motivation, and types of violence used by women. Identifying the extent of musical play - and the role that gender plays in musical play - may provide important information about children's natural tendencies.

The Grand Forks Waste Water Treatment Plant (GFWWTP) is currently sending its waste activated sludge (WAS) from the activated sludge treatment process to an existing on-site wastewater treatment lagoon which has been in operation since 2003. The plant produces approximately 65,000 gallons of WAS per day. Because of this high level of loading, the existing lagoon system is no longer considered as a treatment option for the produced sludge. Current methods for detecting metallothionein (MT) protein expression lack the specificity to distinguish between all twelve human isoforms. Each, however, can be distinguished by the masses of their acetylated, cysteine-rich, hydrophilic N-terminal tryptic peptides. These properties were exploited to develop a mass spectrometry-based method for their simultaneous quantification.
Human kidney HK-2 epithelial cells expressing recombinant MT-3 were grown in the presence or absence of cadmium. Cytosolic proteins were alkylated with 14N- or 15N-iodoacetamide and digested with trypsin. Host-seeking mosquitoes are taxing for people and wildlife alike in the Red River Valley (RRV). During the summer massive numbers of mosquitoes swarm the RRV, yet little is known about the ecology and biology of the mosquito species that inhabit this area. This research will help to fill some of those knowledge gaps by studying the ecology of host seeking mosquitoes in the RRV.

This dissertation presents innovative approaches based on fuzzy logic in epileptic seizure detection and prediction from Electroencephalogram (EEG). The fuzzy rule-based algorithms were developed with the aim to improve quality of life of epilepsy patients by utilizing intelligent methods. An adaptive fuzzy logic system was developed to detect seizure onset in a patient specific way. Problem: The positive relationship between spirituality and physical, psychological, and social well-being has been well established. However, beyond quality of life measures, this construct has yet to be more deeply studied in a sample of individuals diagnosed with Systemic Lupus Erythematosus (SLE). Furthermore, relational spirituality has yet to be studied in a chronically ill population. Meeting these gaps in the literature, this study compared the explicit domains and latent constructs measured by a relational spirituality instrument and examined their associations with a measure of biopsychosocial-spiritual well-being and quality of life measure.

Acetate supplementation is anti-inflammatory and neuroprotective in rat models of neuroinflammation and Lyme neuroborreliosis. Oral administration of glyceryl triacetate (GTA) is used to induce acetate supplementation in rodents, which increases plasma acetate and brain acetyl-CoA levels. Acetyl-CoA is a ubiquitous substrate for several biochemical pathways including carbohydrate, protein, and lipid metabolism. Acetate increases the in vivo and in vitro acetylation-state of histones and is associated with altered cytokine balance. Because some research has shown parental influence can affect children's behavior, it is hypothesized that trained parental support may increase children's physical activity.

The limitations of large bone defect regeneration has brought the focus of Bone Tissue Engineering research on fabricating three-dimensional bone-mimetic grafts that could enhance osteogenesis and vascularization. Osteons composed of apatite-deposited aligned collagen nanofiber layers are the fundamental building blocks of cortical bone. In natural cortical bone, mineralization is mediated by extracellular matrix (ECM) proteins with amino acid sequences rich in glutamic acid. This dissertation is focused on the establishment of a novel biomimetic approach to engineer scaffolds for healing large bone defects.

Regenerative medicine presents exciting strategies for healing critical-size bone defects through the implantation of cells and biocompatible scaffolds. Most in vitro studies are performed in atmospheric oxygen conditions (~20%), which do not accurately mimic the CSD microenvironment. Due to damage to the vasculature at CSDs, oxygen levels fall into the hypoxic range (<5%), which can impact viability, proliferation and differentiation of the cells employed for bone regeneration. Understanding the cellular responses to hypoxia has grown primarily from study of individual molecular factors. The master regulator of adaptive responses to low oxygen availability is the nuclear factor, hypoxia-inducible factor-1. HIF-1 is a heterodimeric protein, stable below 6% O2 condition in the nucleus and has been shown to play a role in angiogenic-osteogenic coupling. We have developed a responsive, fluorescent, hypoxia detection system and determined whether HIF activity can be tracked in both 2-D and 3-D cultures. Adipose-derived stem cells (ASCs) were selected due to their broad utilization in tissue engineering strategies and characterized the influence of HIF signaling on its phenotype. The work done identified that of hypoxia impaired osteogenic differentiation of ASCs in both 2-D and 3-D cultures and HIF-1 did not mediate this effect. Ischemic preconditioning (IPC) strategy and varied protocols used represents a clinically feasible manipulation of cell preparation to help the survival of implanted ASCs and accelerate osteogenic differentiation at physiological oxygen levels.

Cell transplantation can be considered a regenerative therapy, an intervention which attempts to replace or restore the function of compromised tissue by harnessing innate properties of cells that cannot be replicated artificially. For such therapies to succeed, it will be necessary to understand and closely match the physiological conditions that govern cell behaviors in vivo. One important factor is low oxygen tension, termed hypoxia, which is often overlooked in vitro. A preliminary test was conducted evaluating the effect of digestible lysine (dLys) on broiler breeder (BB) male semen quality from forty-one to forty-nine wk of age. Five dietary treatments: corn-soybean meal diet with 1,000 mg dLys/rooser/day (Soy1,000), distillers dried grains with solubles diet with 1,000 (DDGS1,000), 850 (DDGS850), 700 (DDGS700), and 550 (DDGS550) mg dLys/rooster/day. Semen quality was similar except percentage dead sperm which was higher in DDGS550.
