Selection And Clustering For Disease Associated Genetic Variants, Yubo Zou Jan 2013

Selection And Clustering For Disease Associated Genetic Variants, Yubo Zou

Theses and Dissertations

Epigenetics is the study of chemical reactions, which are orchestrated for the development and maintenance of an organism. Genetic or epigenetic variants (GEVs) encompass different types of genetic measures, such as Deoxyribonucleic acid (DNA) methylation at different CpG sites, expression level of genes, or single nucleotide polymorphisms (SNPs). With the development of technology, huge amount of genetic and epigenetic information is produced. However, the rich information potentially brings in challenges in data analyses. Thus it is necessary to reduce the dimension of data to improve efficiency. In my dissertation, I will focus on two directions of dimension reduction: variable selection …

Computational Analysis And Prediction Of Genome-Wide Protein Targeting Signals And Localization, Jhih-Rong Lin Jan 2013

Computational Analysis And Prediction Of Genome-Wide Protein Targeting Signals And Localization, Jhih-Rong Lin

Theses and Dissertations

Computational prediction of protein subcellular localization can greatly help to elucidate its functions. Despite the existence of dozens of protein localization prediction algorithms, the prediction accuracy and coverage are still low. Several ensemble algorithms have been proposed to improve the prediction performance, which usually include as many as 10 or more individual localization algorithms. However, their performance is still limited by the running complexity and redundancy among individual prediction algorithms. In the first part of the dissertation, we propose a novel method for rational design of minimalist ensemble algorithms for practical genome-wide protein subcellular localization prediction. The algorithm is based …

A Sustainable Bio-Solids Management For The Grand Forks Wastewater Treatment Plant, Hasibul Hasan Jan 2013

A Sustainable Bio-Solids Management For The Grand Forks Wastewater Treatment Plant, Hasibul Hasan

Theses and Dissertations

The Grand Forks Waste Water Treatment Plant (GFWWTP) is currently sending its waste activated sludge (WAS) from the activated sludge treatment process to an existing on-site wastewater treatment lagoon which has been in operation since 2003. Grassland Songbird Response To Landscape Composition And Vegetation In Northeastern And East-Central North Dakota, Dustin James Vanthuyne Jan 2013

Grassland Songbird Response To Landscape Composition And Vegetation In Northeastern And East-Central North Dakota, Dustin James Vanthuyne

Theses and Dissertations

Grassland songbirds are declining faster and more consistently than other avifauna of North America. This has resulted in many species being placed on state and federal lists as species of concern. These declines can be linked to former and current land use practices which have resulted in an extensive loss of habitat. Natural resource agencies are trying to offset these habitat losses by reconstructing grasslands formerly cropped areas and protecting remaining grassland tracts. Few studies have addressed how the location in the landscape and vegetation composition of these reconstructions affects grassland birds. This thesis describes data collected from 32 locations …

Morally Injurious Symptomatology: A Qualitative Examination Of The Nvvrs, Douglas Stewart Kraus Jan 2013

Morally Injurious Symptomatology: A Qualitative Examination Of The Nvvrs, Douglas Stewart Kraus

Theses and Dissertations

As with all psychological constructs in their infancy, it is important to operationalize a definition as part of the construct validation process. As a phenomenon that continues to gain recognition amongst the psychological community, Moral Injury (MI) is no different. Although Litz et al. (2009) introduced and defined MI as the "psychological, biological, spiritual, behavioral, and social impact of perpetrating, failing to prevent, or bearing witness to acts that transgress deeply held more beliefs and expectations," Drescher et al. (2011) found that a similar working definition was "inadequate" and lacked clarity according to trauma experts' opinion. The following exploratory, qualitative …

Motivation Towards Learning Perceived In Socratic Seminar Versus Traditional Lecture, Benjamin N. Roberson Jan 2013

Motivation Towards Learning Perceived In Socratic Seminar Versus Traditional Lecture, Benjamin N. Roberson

Theses and Dissertations

As discussed in past literature, high school students often lack motivation towards learning (Crow, 2007; Lumsden, 1995). This lack of motivation interferes with student learning (Lumsden,1995; Vansteenkiste, Simons, Lens, Soenens, & Matos, 2005). At the middle school and collegiate level, Socratic Seminar is seen to provide motivation towards learning in students (Copeland, 2005; Mee, 2000; Strong, 1996); however, there is a need for research on student motivation as a result of Socratic Seminar at the high school level. The purpose of this study is to identify the extent to which, if any, differences exist in student motivation towards learning among …

Signs And Symptoms Of Moral Injury In Female Vietnam Veterans: A Qualitative Examination Of The Nvvrs, Alison H. Conway

Theses and Dissertations

It is well-known that war veterans experience long-lasting physical and psychological injuries following their deployment, and recent studies have proposed the construct of moral injury (MI) to explain the moral and spiritual effects of war on individuals. Litz et al. (2009) defined MI as "perpetrating, failing to prevent, or bearing witness to acts that transgress deeply held moral beliefs and expectations" (p. 6), and Drescher et al. (2011) interviewed trauma experts to identify types of events that could create MI and the lasting sequelae of these events. Vargas et al. (2013) provided validation of MI by examining male Vietnam veterans' …

Elucidation Of Mechanisms Generating 5-Hydroxymethylcytosine (5hmc) In Mammalian Mitochondria, Prashant Thakkar Jan 2013

Elucidation Of Mechanisms Generating 5-Hydroxymethylcytosine (5hmc) In Mammalian Mitochondria, Prashant Thakkar

Theses and Dissertations

DNA methylation plays a pivotal role in governing cellular processes including genomic imprinting, gene expression, and development. Recently, the Tet family of methylcytosine dioxygenases(Tet1, Tet2 and Tet3) was found to catalyze the oxidation of 5-methylcytosine (5mC) to 5-hydroxymethylcytosine (5hmC), an intermediate in the pathway of DNA demethylation. Tet enzymes catalyze this hydroxylation in a 2-oxoglutarate and Fe2+ dependent manner. We have recently reported significant levels of 5mC and 5hmC modification in immunoprecipitates of mammalian mitochondrial DNA(mtDNA). We provide the first evidence that a DNA Methyltransferase-1 isoform (mtDNMT1) translocates to the mitochondria using an N-terminal mitochondrial targeting sequence. mtDNMT1 expression is …

Assessment Of The Feasibility Of Co-Administration Of Phenolic Dietary Compounds With Phenylephrine To Increase Its Bioavailability, Zhenxian Zhang

Theses and Dissertations

R-(-)-Phenylephrine (PE) is the most commonly used nonprescription oral nasal decongestant in the United States. It is a selective α1-adrenergic receptor agonist and has many years of safe usage. However, the efficacy of PE is controversial, due to its extensive pre-systemic metabolism, which leads to low and variable oral bioavailability (38 ± 9%, mean ± SD). Sulfation plays a very important role in pre-systemic metabolism of PE. The sulfation of PE occurs at its phenolic group, which is the preferred structural feature of many sulfotransferase (SULT) substrates. Compounds with phenolic groups have similar structures to PE, which may share the …

Theses and Dissertations

Introduction. 5-HMF and novel INN-compounds are left-shifting AEH, shown to have anti-sickling action by forming transiently covalent Schiff-base adducts with hemoglobin (Hb), thereby increasing the Hb O2-affinity. They are hypothesized to be substrates for aldehyde dehydrogenase (ALDH) in the liver and red blood cells (RBC). Methods. Biopharmaceutical assessments were made for AEH, using calculated physicochemical properties. Their in-vitro hepatic metabolism (mediated by ALDH) was characterized using hepatic cytosol, and in-vitro-in-vivo extrapolations (IVIVE) were made. Inter-species differences in hepatic cytosolic ALDH activity were investigated using acetaldehyde as a model substrate in different mammalian species. Time- and concentration-dependent in-vitro disposition of 5-HMF …

Thc-Mediated Induction Of Δfosb And Its Modulation Of Cb1r Signaling And Adaptation, Lazenka Matthew Jan 2013

Thc-Mediated Induction Of Δfosb And Its Modulation Of Cb1r Signaling And Adaptation, Lazenka Matthew

Theses and Dissertations

The main psychoactive and therapeutic effects of Δ9-tetrahydrocannabinol (THC) are mediated through cannabinoid type 1 receptors (CB1Rs). The therapeutic uses of THC are mitigated by the development of tolerance to these therapeutic effects, whereas tolerance does not readily develop to some of the side-effects of THC, like motor impairment and reward. The development of tolerance occurs through adaptations at CB1Rs, which include desensitization (G-protein uncoupling) and downregulation (receptor degradation). Brain region-dependent differences in THC-mediated adaptations are proposed to explain the differences in tolerance to various THC-mediated effects. These studies focused on whether ΔFosB, a stable transcription factor, could regulate CB1R …

Deconstructing Books, Reconstructing Women, Summers Esther Morris Jan 2013

Deconstructing Books, Reconstructing Women, Summers Esther Morris

Theses and Dissertations

The goal of my thesis project is to examine the notion of choice encountered by females with respect to gender roles. I intend to underscore the turmoil which often comes from choices that leave many classic feminine roles left unattended by either sex. I have created three altered books that address these struggles, as I have personally experienced these, regarding marriage, motherhood, and domesticity. Each book analyzes the conflicting nature of choice and, according to my own decisions as a modern feminist, concludes accordingly. This in-depth look at the struggle for women to not only choose gender roles to adopt, …

Modeling Approaches, Physiological Responses, And Climate Change: How Good Is "Good Enough?", Nicole Kish Jan 2013

Modeling Approaches, Physiological Responses, And Climate Change: How Good Is "Good Enough?", Nicole Kish

Theses and Dissertations

Predicting spatial and temporal patterns in the responses of organisms and ecosystems to climate change has emerged as a major focus of macrophysiological research, with much work centered on the impacts of temperature. A potential difficulty lies in the observation that measures of 'habitat' such as air, land and sea surface temperature often differ greatly from the body temperature actually experienced by organisms, as the latter drives reproduction and survival. As a result, it is unclear how often these simple measurements of habitat are 'good enough' for predicting physiological stress in the field, and when more complicated methods are needed. …
