The use of matrix assisted laser desorption ionization time-oflight mass spectrometry, Fourier transform infrared spectroscopy, and other analytical means of identifying and differentiating microorganisms hold much promise. These analytical tools have been extensively assessed for their ability to differentiate bacteria and fungi. Most of this research has been coordinated in medically relevant microorganisms, but the technology can work just as well with agriculturally important microorganisms. Stink bugs (Hemiptera: Pentatomidae) are key pests of soybean, Glycine max (L.), in Mississippi. Historically, yield loss derived from direct feeding by stink bugs has been considered the greatest threat to producers. However, quality reductions resulting from seed infections caused by microorganisms including Phomopsis longicolla are also a concern. Experiments were conducted in 2010 and 2011 to determine if stink bugs are associated with the incidence of P. longicolla in Mississippi soybean production. Data from experiments suggest that stink bugs are capable of transporting P. longicolla between two points. Data further suggest stink bugs and P. longicolla have the potential …

The onarm water storage (OFWS) systems at Metcalf and Pitts farm in Porter Bayou Watershed were monitored for changes in nutrient levels and water withdrawal for irrigation from March 2012 to April 2013. Nitrogen and phosphorus levels were generally higher during the early part of the growing season. The OFWS systems can reduce nitrate and phosphorus in runoff up to 81% and 85%, respectively. However, the systems did not consistently reduce sediment and nutrients especially after significant rainfall and runoff events. The porcine antral follicle, which consists of an oocyte and surrounding follicular components, including theca, granulosa, and cumulus cells and follicular fluid, is an essential microenvironment for oocyte development and maturation. The flipped or inverted classroom model is one in which the time and place for traditional lecture and homework are reversed. Traditional lecture is replaced by online videos assigned as homework. This frees up time in class to be spent with more student centered activities such as discussion based concept questions and group problem solving. The enzymes acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) are inhibited by nerve agents such as sarin and tabun. In general, the inhibited enzymes are reactivated by bisquaternary ammonium compounds (oximes). The binding free energies of the oximes; 2-PAM, MMB-4, HI-6, and obidoxime bound to human AChE (hAChE) and human BChE (hBChE) inhibited by sarin and tabun and also to the uninhibited enzymes were calculated using various computational methods. Using thermodynamic integration, the binding free energies of all the inhibited and uninhibited systems of MMB-4 and obidoxime were evaluated. Rice not collected by harvesters and natural seeds are important foods for waterfowl. Estimation of abundance of these seeds is necessary for calculating waterfowl habitat conservation needs in the Louisiana Chenier Plain (LCP) and Texas Mid-Coast (TMC). My objectives were to quantify dry mass of rice and other seeds from August-November 2010, and estimate waterbird abundances on farmed and idle ricelands in these regions from December 2010-March 2011. The California Channel Islands present an ideal system in which to study unique biogeographical patterns seen in island systems near a mainland source, where spatial barriers are likely to limit gene flow without disrupting it completely. The islands also harbor a number of endemic taxa, suggesting that isolation from the mainland may be strong for some taxa. For this study, Acmispon argophyllus, a legume species found across five of the eight islands as well as on the mainland was used to test hypotheses at three spatial scales. To understand the flow of water as a factor that influences aquatic vegetation communities and aquatic plant dispersal, custom-made Global Positioning System (GPS) drones were used to monitor the movement of water in Aliceville Lake, Columbus Lake, and Ross Barnett Reservoir, MS. In each reservoir, the drones drifted in the wind-generated surface current. Analysis of wind speeds suggests that a certain wind speed may be necessary to overcome gradient flow. Despite numerous studies, little is known about the influential factors impacting treponemal disease susceptibility. Yet, forms of treponemal disease, specifically yaws, are a major source of morbidity in the developing world. Bioarchaeological materials may assist with deficit by testing full ranges of disease expression. This study explores the relationship between treponemal disease ecology, specifically yaws, and susceptibility through six southeastern United States archaeological samples. Results show early life health experiences have a relationship to later life disease expression. However, influences of climate may only be seen at a cross-regional level. The objectives of this research were to develop the best methods for cellulose nanowhiskers (CNWs) preparation from raw biomass materials and the feasibility to perform CNWs as Fe3+ catalyst support in converting syngas to biofuels. Raw kenaf bast and switchgrass were initially pretreated with dilute NaOH followed by dilute H2SO4. High yields of alpha-cellulose were obtained. Hemicellulose, ash, and most lignin were removed during pretreatment. Preparation of CNWs after pretreatment was then conducted via H2SO4 hydrolysis. The ubiquitin-proteasome proteolysis pathway is responsible for the degradation of abnormal and short-lived proteins to regulate many important biochemical activities in eukaryotes. By employing affymetrix microarray analysis, we have identified a novel ubiquitin ligase E3 gene GhRING2 that is expressed differentially between two G. hirsutum lines - Texas Marker-1 and Chromosome Substitution Line CS-B25. The complete GhRING2 gene sequence was obtained by genomic and cDNA walking. The expression of GhRING2 in cotton fiber is developmentally regulated, suggesting that the ubiquitin-proteasome pathway may regulate cotton fiber growth and development. Articular cartilage is an avascular, aneural tissue that covers the ends of diarthroidal joints. Once damaged by disease or injury, cartilage lacks the ability to self-repair. Generating tissue engineered cartilage is an exciting field that may provide a possible solution to this problem. The purpose of this study is to determine the efficacy of a through-thickness perfusion bioreactor to generate scaffoldree tissue engineered cartilage. The results of the study show that allowing long-term static culture to cell constructs before perfusion increases the efficacy of the bioreactor. Crappie fisheries in northwest Mississippi flood control reservoirs are valued from social and economic standpoints. To mitigate variable recruitment rates of these recreationally valuable species, I sought to gain a better understanding of the population dynamics of these fish during their first year of life. My analyses indicate that embayment habitats in the study reservoirs display a longitudinal distribution gradient of both habitat composition and age-0 crappie abundance during late summer. Additionally, age-0 crappies inhabiting uplake floodplain areas grow to a larger size than downlake embayment-inhabiting conspecifics. Biodiesel is a renewable fuel that can supplement petroleum fuel supply. A major deterrent to commercial biodiesel production from traditional feedstock like soybean, canola or rapeseed oils is the high cost of feedstock: 70 - 95% of total biodiesel production cost. Therefore, a relatively cheaper feedstock is needed to make the price of biodiesel cost-competitive with petroleum diesel. Activated sludge from wastewater treatment plants is a relatively cheaper feedstock and relatively easy to obtain. This study presents the use of non-conventional methods such as microwaves and ultrasound for algal biodiesel production. Dry algae biomass (Chlorella sp.) was used as feedstock to evaluate three novel single-step extractive-transesterification methods: 1) microwave irradiation with ethanol as solvent/reactant; 2) microwave irradiation with ethanol as reactant and hexane as solvent and 3) ultrasound irradiation with ethanol as solvent/reactant, all catalyzed by sodium hydroxide catalyst. Using the concept of bone functional adaptation, this study analyzes femoral and humeral cross-sectional properties of human skeletal materials from Morton Shell Mound on the Louisiana coast. This work helps fill a gap in such analyses in the southern U.S. and contributes to an understanding of the functional adaptation of the human skeleton. Pin1 is a Prolyl Isomerase that catalyzes cis-trans isomerization of peptides with pSer/Thr-Pro motifs in many cell signaling proteins. This conformational switch is implicated in diseases. Pin1 activity is considered a target for therapeutic applications. Pin1 targets motifs by its N-terminal WW-binding domain. A C-terminal PPIase domain is responsible for catalysis. Urban stormwater management is evolving toward sustainable approaches which rely on dispersed small-scale bioretention BMPs. One such BMP is the flow-through planter, commonly applied in areas where infiltration into in situ soil is restricted or not possible. A project was developed to evaluate 18, vertically scaled flow-through mesocosms. Three replicates of six treatments, including four soil mixtures containing varied percentages of sand, compost and topsoil, were tested for orthophosphate and nitrate removal, volume reduction capabilities, and peak flow attenuation through the application of a synthetic solution over a simulated 2-inch, Type II storm event. Runoff volume was significantly (p < 0.05) reduced compared to controls. Nutrient levels observed along the hydrograph at different time-steps and flow rates revealed patterns not apparent in cumulative results. The observation of preferential flow patterns along with variability in nutrient removal across treatments highlights the need for design modifications of flow-through facilities.

The USDA Natural Resources Conservation Service (NRCS) implemented the Migratory Bird Habitat Initiative (MBHI) in summer 2010 to provide wetlands for waterbirds inland from the Deepwater Horizon Oil Spill. To evaluate MBHI and associated wetland management practices, I estimated seed and waterbird densities in MBHI and non-managed wetlands in the Mississippi Alluvial Valley (MAV) of Mississippi and Louisiana. Although not statistically different, wetlands enrolled in MBHI contained 1.26 and 1.53 times more seed biomass and seeds consumed by waterfowl than non-managed wetlands, respectively. The dissertation describes research work on development of bimetallic heterogeneous catalysts for oxidation of organic compounds. Binuclear metal complexes are an interesting class of compounds due to their catalytic activity. The approach involves anchoring a triketone ligand to Cab-O-Sil via a linker. Specifically, silica gel was allowed to react with p-chloromethylphenyltrimethoxysilane followed by coupling with deprotonated triketone compounds. The viability of this approach was verified by performing the benzylation reaction of triketones with benzyl halides under basic conditions. The benzylation of 2,4,6-heptanetrione and 1,5-diphenyl-1,3,5-pentanetrione is achieved with benzyl bromide using n-tetrabutylammonium fluoride as base. Salmonella has been studied and researched for more than a hundred years and yet it remains a problem for human and animal health. The goal of this dissertation was to apply the systems thinking approach to Salmonella contamination and develop a System Dynamics (SD) simulation model for Salmonella contamination in the chill tank of a poultry processing plant. But first the appropriate carcass rinse sampling method that would not impact on the resulting Salmonella contamination status of the broiler carcass was studied. Calcium bentonite deposits of the Glendon and Bucatunna formations of the Oligocene Vicksburg Group located in Smith County, Mississippi were examined to determine the depositional environment, diagenetic history and origin of the bentonite. Traditionally, calcium bentonite was considered to be the product of the weathering of volcanic ash deposits. The hypothesis tested is that the bentonite deposits are not a result of weathered volcanic ash, but are a result of weathered marl. Core samples were drilled from the Chisholm bentonite mine in Smith County and were studied using X-ray diffraction, scanning electron microscopy, and petrographic microscopy.
