Multiple DNA transactions are at the center of almost all processes regulating the AAV life cycle. A common feature shared by all transactions is the binding of the large AAV Rep proteins Rep78/Rep68 onto DNA sites harboring multiple GCTC repeats. AAV mediated site-specific integration is contingent upon the formation of a productive complex between Rep78/Rep68 and the AAVS1 site located at chromosome 19. In order to understand the mechanistic details of the initial assembly process we carried out equilibrium binding experiments of Rep68 and its individual domains with a 42-mer AAVS1 site. Iron oxide nanoparticles have received sustained interest for biomedical applications as synthetic approaches are continually developed for precise control of nanoparticle properties. This thesis presents an investigation of parameters in the benzyl alcohol synthesis of iron oxide nanoparticles. A modified seed growth method was designed for obtaining optimal nanoparticle properties for magnetic fluid hyperthermia. With a one or two addition process, iron oxide nanoparticles were produced with crystallite sizes ranging from 5-20 nm using only benzyl alcohol and iron precursor. Electrospinning is a process by which polymer fibers can be produced using an electrostatically driven fluid jet. Electrospun fibers can be produced at the micro- or nano-scale and are, therefore, very promising for air filtration applications. However, because electrospun fibers are electrically charged, it is difficult to control the morphology of filtration media. Fiber size, alignment and uniformity are very important factors that affect filter performance. Electrospinning is a technique used to generate scaffolds composed of nano- to micron-sized fibers for use in tissue engineering. This technology possesses several key weaknesses that prevent it from adoption into the clinical treatment regime. One major weakness is the lack of porosity exhibited in most electrospun scaffolds, preventing cellular infiltration and thus hosts tissue integration. The innate immune system provides the body’s first line of defense against pathogenic challenge through pathogen recognition and initiation of the immune response. Among the various cellular mechanisms of pathogen recognition in mammals, Toll-like receptor 3 (TLR3) recognizes viral dsRNA. Stimulation of TLR3 signaling pathway leads to transcription of pro-inflammatory cytokines and type-1 Interferons. With previous indication of the Group VIA phospholipase A2 (iPLA2β) enzyme regulating ER-stress induced apoptosis in β-cells by regulating the anti-apoptotic protein Bcl-xL via alternative splicing, our lab postulated iPLA2β to be utilizing a similar mechanism to regulate apoptosis in mice lung. Our previous lab work has shown implications of lung function compromise in iPLA2β-/- mice, and we speculated the cause to be due altered lung architecture stemming from the attenuation of apoptosis. This work operates at the intersection of academics, art, and activism. Within queer studies there is a tension between assimilation and liberation, sometimes situated as between pragmatism and utopia. This work re-examines Frankfurt school Marxist views of utopia through a queer theoretical lens in order to employ the radical imagination and queer futurity to examine new ways of practicing liberation. Drawing from theorists like Judith Butler, Jose Esteban Munoz, and Gloria Anzaldua, this work uses art (film, writing, zine-making, and sound) as a way to envision and enact a better world situated in the present.

Alcoholism is a complex neurological disorder characterized by loss of control in limiting intake, compulsion to seek and imbibe ethanol, and chronic craving and relapse. It is suggested that the characteristic behaviors associated with the escalation of drug use are caused by long-term molecular adaptations precipitated by the drug’s continual administration. Porphyromonas gingivalis, a gram-negative anaerobic bacterium, is a major etiological agent in the initiation and progression of severe forms of periodontal disease. Oral bacteria like P. gingivalis are subject to continually changing conditions as a consequence of host eating, oral hygiene patterns and subgingival temperatures. As such survival requires an adaptive response to environmental cues, but little is known about the mechanism by which P. gingivalis controls co- and post-transcriptional regulation of RNA levels and potentially protein expression. While the impact of psycho-social variables has been evaluated for children with chronic illness and related disabilities, questions remain unanswered for families affected by ASD. Participants included 68 primary caregivers seeking services for a child aged fifteen months to 12 years suspected of or diagnosed with an ASD. Assessment tools included: 1. Parenting Stress Index-Fourth Edition-Short-Form (PSI-4-SF; Abidin, 2012), 2. Parenting Sense of Competence Scale (PSOC; Johnston and Mash, 1989), and 3. This research is intended to enhance recent hydrologic research that focused on a rainfall-runoff model of the Devils Lake basin in central North Dakota. This research employs the spatial volumetric soil moisture data collected by the National Aeronautics and Space Administration (NASA) Advanced Microwave Scanning Radiometer - Earth Observing System (AMSR-E) instrument. The AMSR-E data, collected from 2002-2011 from the NASA EOS Aqua satellite, was used in the Land Parameter Retrieval Model (LPRM) to estimate the surface soil moisture at a scale of 25-km. A novel hybrid nanocomposite of Au nanoparticle-modified silicon nanowire was developed for surface enhanced fluorescence applications. The designed nanocomposite contained a silicon nanowire, gold nanoparticles and a silica layer doped with dye molecules. The hybrid nanomaterial was characterized using scanning electron microscopy (SEM), scanning transmission electron microscopy (STEM), fluorescence measurements, Fourier transform infrared (FT-IR) spectroscopy, and energy-dispersive X-ray spectroscopy (EDS). The results showed that the gold nanoparticles were uniformly adhered on the silicon nanowires and covered by a thin silica layer. The nanostructure exhibited strong capacity for surface enhanced fluorescence. Different enhancement factors were obtained by changing synthetic conditions.

Copper (Cu) is a component of several aquatic pesticides often used to target noxious algae, but at high concentrations may elicit toxic effects on non-target organisms such as fish. The severity of Cu toxicity varies widely by fish species, Cu concentration, duration of exposure, and water quality. A biochemical process was investigated in the conversion of lignocellulosic biomass into biofuels and value added chemicals. Kenaf, two species of sorghum (brown mid rib (BMR) & non brown mid rib (NBMR)), sunn hemp, sunflower hulls, and cornstover were used as feedstocks in this study.

Lignocellulosic biomass primarily consists of three different components, cellulose, hemicellulose and lignin. In order to separate different fractions, and acid pretreatment is generally employed. Two weekly and freely available remotely sensed vegetation indices, Vegetation Condition Index (VCI) and Temperature Condition Index (TCI), were assessed for state level corn yield correlation in the Corn Belt region of the United States for the years of 2007-2013. VCI and TCI were 16-km pixels which are derived from the Advanced Very High Resolution Radiometer (AVHRR). Corn pixels were identified by downloading yearly USDA Cropscape pixels for corn in each state. The development of the resource was also informed by two structured interviews with experts in the field of collective trauma within the Latino population. Responses to disclosures/discussions of trauma can have lasting impacts on survivors who choose to share their experiences and historically have been categorized as positive, negative, and/or neutral responses with corresponding effects on the survivor. Literature recommends the use of tenets and techniques reminiscent of therapeutic common factors (e.g., listening skills, empathy, support, validation, creating a safe environment and strong therapeutic alliance) when responding to trauma.
