Over the course of 8 years working in progressive political strategy, 2015–2023, I became increasingly critical of the systems, formulas and tools we were using to communicate with voters. Reply STOP to Stop is documentation of my time at graduate school, of wrestling with ideas of political design, language, communication and distribution. I analyzed my past work — all the texts, emails, social media posts, print ads, web banners, videos and other miscellaneous scripted content — and wondered if we would all be better off without it. Next-generation sequencing (NGS) technologies have revolutionized biological research by generating unprecedented volumes of genomic data, necessitating advanced bioinformatics tools and specialized expertise to fully interpret complex microbial communities. This study outlines an integrated workflow to characterize the gut microbiome of the wood-feeding beetle Odontotaenius disjunctus. Prokka is employed for functional annotation to identify protein-coding genes and other genomic features, while Kaiju is used for taxonomic classification. DIAMOND is then applied to locate and validate the taxonomic assignments generated by Kaiju by aligning sequences within the contigs to a comprehensive protein database, proGenomes. Oppressive systems (e.g., colonialism, racism, capitalism) create a disparate distribution of power that, through various mechanisms, negatively impacts the health and well-being of members of the oppressed group. Boricuas have experienced over 500 years of oppression, with the island currently maintaining a colonial relationship with the United States. As these adverse effects of oppression are identified, the need for protective factors (e.g., hope) has emerged. This study explored how islander Boricuas practice radical hope, understand their history of oppression and resistance, and how they perceived it to impact their overall well-being. Climate Change-Induced Shifts In Mediterranean Fungal Fruiting Phenology And Productivity, Rachel Demay

We used a 29-year dataset of mushroom fruiting from the Pinar Grande forest in northern Spain to investigate the effects of climate change on the phenology, productivity, and diversity of mycorrhizal and saprotrophic mushrooms. We investigated the long-term trends in fruiting season length, yield (by count, total weight, and average weight), and genus richness. Additionally, we identified associations between monthly temperature and monthly precipitation with these fungal responses across functional groups: mycorrhizal, saprotrophic, litter-decaying saprotrophic, and wood-decaying saprotrophic mushrooms, and the economically-important edible genera Boletus and Lactarius. Upper extremity abnormal flexor synergy caused by stroke can impair individuals by coupling upper extremity muscles with deltoid activation. Previous studies have shown that reduction in deltoid loading can decrease flexor synergy. Robotic rehabilitation has the ability to allow for reduced deltoid loading while assisting or constraining joints along the arm to increase rehabilitation outcomes. The effects of higher degree of freedom low impedance robotics needs to be tested to learn how various parameters affect flexor synergy in comparison to end point robotics. This thesis details the development and application of intuitive Mixed Reality (MR) interfaces designed to broaden access to robotic control for creative and interactive uses. The central contribution is the RoboART system, a novel MR interface built with the Microsoft HoloLens 2 to program an ABB GoFa collaborative robot for non-expert users, such as artists. The work describes RoboART's application in the Magnaforma project, where it enabled complex artistic expression.

As global plastic production increases, so does the environmental and biological presence of nanoplastics (NPs), raising growing concerns about their potential impact on human health. These particles have been detected in various organs, including the lungs, yet their role in respiratory inflammation remains largely unexplored. Emerging evidence suggests that NP exposure may influence immune responses in the lungs, particularly in individuals with preexisting conditions such as asthma. Soil respiration (Rs) is the largest terrestrial flux of carbon (C) to the atmosphere, with heterotrophic contributions to Rs varying spatially and temporally. While microbes are the primary and most thoroughly studied source of soil heterotrophic respiration (Rh), contributions from higher trophic organisms, including arthropods, are rarely quantified and poorly understood. Arthropod abundance and diversity correspond with variation in soil microclimate, which is influenced by site factors including canopy structure, site productivity, and disturbance. Antisocial personality traits such as psychopathy, Machiavellianism, Narcissism, and Sadism have received considerable focus from researchers. Among these traits, Sadism stands above them all as arguably the most malevolent of all trait constructs in psychology. Major depressive disorder (MDD) affects over 350 million individuals worldwide and is characterized by profound dysregulation of serotonin (5-HT) and dopamine (DA) systems that govern mood, reward processing, and motivation. The serotonin transporter (SERT) and dopamine transporter (DAT), both dynamically regulated through phosphorylation, serve as critical determinants of synaptic monoamine signaling. Despite decades of therapeutic development, current antidepressants require 4-6 weeks to achieve efficacy, and approximately one-third to half of patients develop treatment-resistant depression. Motor recovery of the arm following a neurological injury is a common goal of the rehabilitation process for individuals impacted by the condition . Due to the long-term functional implications of impaired arm movement, individuals are often trained to use compensatory strategies to complete functional tasks without incorporating the weakened limb. The Lyme disease spirochetes, Borreliella spp., are maintained in nature through a complex enzootic cycle involving Ixodes ticks and a diverse set of reservoir hosts. The outer surface protein C (OspC) of the bacterium is an essential virulence factor required for transmission from ticks and early infection of mammals. Deletion of ospC abolishes infectivity, yet the precise molecular mechanisms underlying OspC function remain incompletely defined. Alcohol use disorder and pain are complex public health concerns affecting millions of people, and these conditions are often characterized by a bidirectional relationship. People experiencing pain may drink alcohol for its analgesic effects. In turn, excessive consumption of alcohol can lead to conditions that cause pain, leading to an escalating feedback loop of pain and alcohol use. Negating this cycle requires a nuanced understanding of the mechanisms underlying alcohol analgesia, which are presently not fully understood. Constipation is a major clinical obstacle associated with opioids, prompting patients to discontinue treatment due to pain, discomfort, and psychosocial stressors. Moreover, chronic users become tolerant to the analgesic effects of opioids, but not to their constipating effects. Upon cessation of long-term treatment, serious diarrhea–among other somatic signs of withdrawal–may ensue. These symptoms become a driving factor for relapse, causing patients to restart opioid treatment. In this study, we aim to understand mechanisms that contribute to opioid tolerance and withdrawal in the colon. Dickkopf1(DKK1) is a quintessential Wnt antagonist and immunomodulator in various inflammatory diseases. The underlying molecular mechanisms of DKK1-mediated immunomodulation remain elusive. Here, we identified TLR4 as a new receptor for DKK1 to activate NFκΒ pathway-mediated gene expression and pyroptosis via NLRP3 inflammasome in human and mouse macrophages.

DKK1 employed TLR4 to initiate NFκB signaling cascade via MyD88. MyD88-TAK1-NFκΒ pathway activation by DKK1 increased HIF1α, NFκB, and NLRP3 protein expression levels, leading to pyroptosis. Unlike LPS, DKK1 did not induce IRAK4 phosphorylation, while the interaction between MyD88 and IRAK4 was maintained for downstream signaling activation. DKK1 did not induce IRF3 phosphorylation …

Problems: A rediscovered radiotherapy technique, FLASH, involves increasing the dose rate to >40 Gy/s. For a given dose, FLASH is shown to decrease the normal tissue complication probability without affecting the tumor control probability. More preclinical studies are necessary to confirm and enable these desirable properties on humans. Current preclinical investigations have diverse constraints, requiring FLASH apparatuses with flexible parameters and accurate control. Tobacco use and exposure are linked to adverse health outcomes (e.g., respiratory conditions) and increased healthcare utilization (e.g., emergency room visits). This study examined the moderating role of social support, tobacco attitudes/perceptions of harm, and asthma status on associations between tobacco use (e.g., combustible product use) and exposure (e.g., secondhand smoke) and cotinine levels among individuals in Richmond, Virginia. We also explored differences in tobacco-related variables by asthma status and associations among tobacco use, secondhand smoke exposure, cotinine levels, and asthma control. This qualitative study investigated group flow through a team-building exercise with 18 professionals, examining participants’ and an observer’s perceptions of flow in social contexts. Analysis of interviews and observer notes identified eight themes: Sensory Network of Shared Experience, Adaptive Group Identity and Performance, Goal-Skill-Motivation Alignment, Essential Social Dynamics, Calm Intensity, Synchrony and Engagement Patterns, Spontaneous Collective Celebration, and Observer Integration. These themes revealed that flow in groups manifests as dynamic forces in constant motion. THE BODY IS ON is a visual art exhibition that considers queer embodiment, pervert as political identity, the arriving climate catastrophe, and difficulty in artmaking. I make use of various concepts from Deleuze and Guattari to interconnect my own with the primary goal to ask: what does futurity look like or mean in these climates (in both a literal, warming sense and an increasingly fascistic, political one), and how do we actualize that future?
