Deep-space missions, such as those planned for the cis-lunar space and Mars, pose unique challenges to astronaut health due to prolonged microgravity (MG) exposure and low-dose chronic irradiation (CIR). Understanding the impact of these stressors on secondary lymphoid tissues, such as the spleen and intestine, is crucial for ensuring crewmembers' well-being, given the impracticality of conducting this research with human samples. Using a ground-based facility, we exposed C57BL/6 female and male mice to CIR (1.74 Gy at a dose rate of 0.0029 Gy/h) and/or simulated MG (SMG) via hind limb unloading (HLU) for one month. Cyclic diguanylate monophosphate (c-di-GMP) is a second messenger that is responsible for attachment and motility in many prokaryotic systems and controls the signaling pathway for the motile to sessile transition in many bacteria. Many c-di-GMP-metabolizing proteins are diguanylate cyclases (DGCs), responsible for the synthesis of c-di-GMP and characterized by a c-di-GMP synthesizing domain, e.g., GGDEF domain. Other c-di-GMP-metabolizing proteins contain c-di-GMP degradative domains, e.g., EAL, characterized by phosphodiesterase activity (PDE). In Agrobacterium tumefaciens, out of 31 predicted proteins regulating the c-di-GMP level, this project focuses on the predicted dual-function DGC/PDE, DcpB. This thesis explores how rural communities in Kafr El Sheikh governorate, Egypt, negotiate everyday life with the contamination of Kitchener Drain, the largest agricultural drain in the central Nile Delta. Drawing on ethnographic fieldwork in three villages at the tail-end end of the drain, the study explores how water contamination shapes social and ecological, social life for farmers living in the catchment of the drain. The thesis situates the farmer’s present experience within the broader history of colonial and modernist national irrigation reforms and developments highlighting how systemic neglect and uneven governance reproduce conditions of vulnerability. Addiction involves persistent reward-seeking behavior despite adverse outcomes, with the nucleus accumbens (NAc) critically mediating motivation and cost-benefit decision-making. Dopamine (DA)-responsive neurons in the NAc, specifically D1 and D2 receptors within the NAc are crucial in encoding motivated behavior. However, the specific role of D1/D2 receptors in driving reward-seeking under punishment remains unclear. Preliminary findings suggest a sex-specific effect, with females exhibiting greater sensitivity to the suppressive effects of footshock on natural reward-seeking compared to males. This work seeks to answer the unstudied questions “Does a layer of vegetation impact the prompt radiation energy deposition in the soil from an atmospheric nuclear detonation?” and “If yes, by how much?” A new methodology was developed to repeatably automate and streamline a process of generating a three-dimensional geometry for subsequent radiation transport simulations and statistical analysis. This dissertation addresses the challenge of enabling soft robots to perform contact-rich manipulation tasks by overcoming limitations in hardware, modeling, control, and learning. First, we present hardware advancements with PneuDrive, a pneumatic control system for actuating soft robots. We also introduce Baloo, a hybrid soft-rigid robotic torso which achieves a single-arm payload capacity of 19.3 kg and robust whole-body grasping capabilities. We also develop a full-body tactile sensing system enabling piezoresistive sensor arrays to cover large areas (1 square meter) with over 8000 distributed sensing points. Acute Lymphoblastic Leukemia (ALL) is a hematologic malignancy that primarily affects children and young adults. While advancements in treatment—including chemotherapy and cranial irradiation—have significantly improved 5-year survival rates in children to nearly 90%, radiation-induced cognitive impairment (RICI) remains a persistent and debilitating side effect. The brain, particularly the hippocampus, is highly susceptible to radiation-induced oxidative stress and inflammation. In the treatment of ALL, the combined use of chemotherapy and radiotherapy contributes to a heightened oxidative burden, exacerbating the risk of cognitive dysfunction. Fusing a variant of the sterile alpha motif domain of the human translocation ETS leukemia protein (TELSAM) to a protein of interest significantly enhances crystallization and diffraction quality. TELSAM is a pH-dependent, polymer-forming crystallization chaperone that, when covalently fused to a protein, forms a well-ordered crystal lattice. However, the success of this method depends on two factors: 1) the design of the linker connecting TELSAM to the target protein, which influences both crystal packing and target orientation, and 2) the prevention of TELSAM polymer flipping, a phenomenon that disrupts lattice periodicity and impairs structure determination. Reduced graphene oxide (RGO) forward osmosis (FO) membranes have emerged as promising candidates for efficient wastewater management and osmotic energy harvesting, due to their enhanced chemical stability and superior FO performance for low-energy waste brine treatments, such as volume reduction of oxidative chromium brine for cost-effective disposal. This dissertation first examines the oxidation resistance of RGO forward osmosis membranes against chromate Cr (VI). Our findings reveal that both the water flux and the reverse salt flux of RGO membranes are suppressed, accompanying an enhanced reverse flux selectivity after exposure to an acidic medium of Cr (VI) (pH 2.0-7.0). This thesis examined how multicultural interactive read-alouds support first-graders’ literacy skills and cultural awareness. Multicultural literature was used to conduct interactive read-alouds to first-grade students. A look across all data sources suggested three main themes that occur throughout the research study. These themes included student engagement and questioning, making connections by experiencing culture through an authentic lens, and developing cultural awareness and empathy for those who are different. End-to-end autologous grafting with proper fascicular complementation has demonstrated functional recovery per the Medical Research Council Classification (MRCC). Tissue-engineered three-dimensional (3D) myocardial patches offer superior and innovative treatment strategy for myocardial infarction (MI). Among the various scaffold fabrication techniques, electrospinning and 3D bioprinting have received great attention due to their ability to recreate native cardiac tissue mimetic characteristics. Lymphatic filariasis (LF), or elephantiasis, is a mosquito-borne parasitic disease affecting the lymphatic system, primarily in tropical and subtropical regions. It is the second leading cause of long-term disability and a neglected tropical disease. According to WHO, LF threatens 882 million people in 44 countries, with over 9 billion treatments administered. Quarterback decision-making is one of the most critical determinants of success in the National Football League (NFL), yet traditional statistics fail to account for the spatial, temporal, and contextual factors influencing each play. This thesis proposes a data-driven methodology for evaluating quarterback decisions using NFL player tracking data from the 2022 season. We replicate and extend the DeepQB framework by training deep learning models to estimate expected completion probabilities and expected yards gained for each receiver at the moment of the throw. This dissertation examines the evolution and educational implementation of Automated Essay Scoring (AES) systems, with particular focus on generative artificial intelligence integration in educational assessment. Through a two-article format, this research addresses the persistent gap between technical capability and educational adoption that has characterized AES development over seven decades. The first article presents a comprehensive methodological review analyzing 122 studies spanning AES evolution from foundational computational linguistics through contemporary hybrid systems.
