Development Of The Analytical Methodology For Pyrimethamine And Its Application To Studies Of Partitioning And Binding In The Subcompartments Of Blood, Anita C. Rudy

An original HPLC assay was developed for pyrimethamine (PYR) in plasma, RBCs, and buffer for the purpose of studying its plasma protein binding and RBC partitioning.

Equilibrium dialysis (ED) was used to study protein binding. Isotonic phosphate buffer used in ED did not prevent small volume shifts. The pH of the plasma affected the protein binding of PYR although it was not significant for the comparison of binding at pH 7.4 vs. 8.0. PYR at 1000 ng/ml averaged 93.1% bound to plasma proteins. Binding to pure human albumin was 86.5% at lower levels of albumin and PYR (350 ng/ml). There …

The history of the Church of Jesus Christ of Latter-day Saints, commonly referred to as the "Mormon" church, has been well researched, yet little has been written about individual community histories. This work centers on the small Mormon community of Syracuse, Utah, located about twenty-five miles north of Salt Lake City on the eastern shore of the Great Salt Lake. From its inception in 1877 through 1987, the community of Syracuse continues to be dominated by the Latter-day Saint (Mormon) influence. Nosocomial (hospital-acquired) pneumonia (HAP) continues to be an important cause of morbidity and mortality in the hospital. HAP is the third most common nosocomial infection after urinary tract and surgical wound infections. In addition, HAP is the nosocomial infection with the highest mortality rate. These infections are often difficult to treat, because most are caused by Gram-negative bacilli (GNB) that may be highly resistant to antimicrobial agents. HAPs frequently occur in intensive care patients with underlying lung and/or systemic diseases. Many patients are intubated and are on assisted ventilation. John Charles Fremont conducted five expeditions to the West during a period of twelve years (1842-1854). On four occasions, during three of these expeditions (1843-1844, 1845, and 1854), the explorer entered the Utah region. His explorations in northern Utah in 1843 focused primarily on the scientific analysis and survey of the Great Salt Lake. In 1844, Fremont again entered the Utah area and made scientific observations and calculations about the region, including accurately defining the geographic region known as the Great Basin, the name given it by Fremont. This study was designed to examine the effects of hypnotherapy on both short-term and long-term weight loss when combined with a group behavioral weight loss program. At the beginning of the study, 38 obese female subjects (20% over normal body weight), ranging in age from 21 to 64, were randomly assigned to one of two treatment groups, one utilizing behavior therapy only and the other utilizing behavior therapy plus hypnotherapy.

The groups mot weekly for 1-1/2 hours for 8 consecutive weeks, with follow-up sessions, at 3, 6, 9, and 12 months. Twenty-eight women completed the study. The first hour in …

Early behaviors of juvenile offenders and nonoffenders were studied to assess differences between the two groups. Experimental subjects were 50 youth residing in three northeastern counties of North Dakota who had committed two or more offenses. Control subjects were 50 non-offenders matched for birthdate, sex, and school. Data were collected retrospectively from both groups on Form 1 of the Louisville Behavior Checklist (LBC)» an inventory designed to detect psychopathological behaviors among preschool children. Caffeine, a central nervous system stimulant, is said to produce an elevation in level of arousal. Humphreys and Revelle (1984) have proposed a model to account for changes in information processing efficiency resulting from differences in personality, situational moderators such as caffeine, and type of information processing task. According to the model, a high level of state arousal will result in poor performance on shortterm memory tasks, while improved performance will occur at a low level of arousal. The purpose of this study was to investigate the feasibility of using instructed guided fantasy to decrease anxiety in a child with juvenile rheumatoid arthritis. It was intended that the child would use guided fantasy to develop an internal coping mechanism for reducing anxiety during a weekly gold shot injection, which was identified as anxiety-producing. An embedded, single-case design was selected for this case study. Qualities of guided fantasy were globally examined. This analysis addresses the classification of a production of Joe Orton's play Entertaining Mr. Sloane as a comedy. Its intent is to question the classification by considering several factors.

Two of the major effects of histamine in the rabbit heart are the enhancement of contractility and microvascular permeability. In cardiac muscle it has been documented that calcium (Ca ) bound to the sarcolem- mal-glycocalyx complex is essential for the regulation of the contractile response. In addition, it has been postulated that histamine's regulation of microvascular permeability may occur via an endothelial contractile response causing the formation of submicroscopic gaps between endothelial cells. The role of the central nervous system neurotransmitter serotonin was evaluated on the anticonvulsant activity of various compounds. Serotonin (5-hydroxytryptamine) is known to have an inhibitory role in central nervous system activity. A facilitation of the anticonvulsant activity of benzodiazepines has previously been attributed to increased serotonin activity.Male white mice of the ICR strain were administered varying doses of phenytoin, 5-ethyl, 5-methylhydantoin (EMH) and JAW-669, then challenged with maximal electroshock. Effects Of Zinc Deficiency On Sertoli Cell Number And Ultrastructure: An In Vivo And In Vitro Study., Gro Thorne-Tjomsland

Hypogonadism in the male is a well known clinical symptom of zinc deficiency. The testicular lesions have been primarily attributed to the effects of decreased zinc on germ cells. This study assessed possible effects of zinc deficiency on Sertoli cell number and ultrastructure.Long-Evans dams and their newborn litters were placed on zinc deficient (ZD), pair-fed or ad libitum fed (AL) diets. All dams received a 20% egg white based diet containing less than one ppm zinc. ZD dams received distilled, deionized water (<1 ppm ZN), while PF and AL dams received the above water fortified with 25 ppm zinc. Male pups were sacrificed day 10 and their testes were processed for light (LM) and transmission electron microscopy (TEM).LM cell counts were performed on one (mu)m sections. Average number of germ cells on the basement membrane (BM) (=GC2) per tubular cross-section was reducted by two-thirds in ZD and PF compared to AL animals (p < 0.0001). Average number of germ cells not on the BM (=GC1) and average number of Sertoli cells per tubular cross-section were not statistically different in the three diet groups.Volume fractions of TEM subcompartments/total cytoplasm were determined by morphometric methods. The lysosome-like fraction was doubled in ZD and PF compared to AL animals (p < 0.05). Endoplasmic reticulum, Golgi and mitochondrial fractions were not statistically different in the three diet groups.Five day old Sertoli cell cultures derived from ZD, PF and AL pups were volumetrically analyzed as in vivo. The endoplasmic reticulum fraction was approximately 100% lower in ZD- than in AL-derived cultures (p < 0.01). Golgi, mitochondrial, dense body and inclusion body fractions were similar in ZD-, PF- and AL-derived cultures.These data suggest that Sertoli cells, although not significantly reduced in number in ZD and PF animals, were affected ultrastructurally by dietary treatment. The volumetric increase in lysosome-like bodies in vivo suggested that Sertoli cells participate in phagocytosis of germ cells degenerating as a result of dietary treatment. The decreased ER volume fraction of ZD-derived tissue cultured Sertoli cells suggested that Sertoli cell secretory potential may be altered as a result of zinc deficiency.

The practice of polygamy among the Mormons during the nineteenth century was vigorously prosecuted by the federal government in response to the demands of those whose political and economic goals could best be served through exploitation of the national attitude toward polygamy. Hundreds of men served prison terms for practicing what they believed was their religious obligation. This study of the sentences from the prison admission records has focused on the comparative severity of the judges, examines age as an influencing factor in sentencing, and compares the sentences of the polygamists with those for crimes against person and property.

Curiously, the issue of war has never been considered a solid basis for examining the works of Ernest Hemingway and F. Scott Fitzgerald. Yet one common thread running through the body of their work is war. A study of the authors’ attitudes toward war and their characters’ responses to war in Hemingway’s A Farewell to Arms (1929), For Whom the Bell Tolls (1940), The Sun Also Rises (1926), and Fitzgerald’s This Side of Paradise (1920), The Great Gatsby (1925), and Tender Is the Night (1934), yields several conclusions. The purpose of this study was to test the effect of high volt galvanic (HVG) stimulation on edema reduction and blood flow velocity (BFV) in the rat hindlimb. The effect of various blocking agents on BFV, blood pressure and heart rate following HVG stimulation was also tested.For the edema study, the animal's hindpaw was traumatized; and the animal was assigned to either the treated (n = 20) or control (n = 20) group. The effects of two vasopressin analogues, arginine vasopressin (AVP) and desmopressin acetate (DDAVP), upon various aspects of arousal, attention and memory were examined in this study. Sixty-two male college students were treated intranasally with either AVP (24 IU), DDAVP (24 IU), or saline. Arousal, attention and memory measures included: the Thayer Activation-Deactivation Adjective Check List (AD-ACL), the Eysenck Personality Inventory Introversion-Extraversion scale, diastolic and systolic blood pressure, pulse rate, respiration rate, digit span, a Sternberg Item Recognition Task, immediate free-recall, delayed free-recall, and recognition. The purpose of the study was to compare the keyboarding performance of sixth graders resulting from two different teaching methods--computer-assisted instruction and teacher-directed instruction.The subjects were 132 sixth graders at Franklin Middle School in Thief River Falls, Minnesota. Each class met 50 minutes daily for six weeks in a microcomputer lab. The four control classes were taught as a group from a textbook, with everyone proceeding at the same rate.
