Many links exist between cocaine abuse and stress. The literature and laboratory studies in rats suggest that this could be because stress increases the reinforcing efficacy of cocaine. Using male Long-Evans rats, experiments in this thesis tested effects of footshock on the reinforcing efficacy of cocaine using a progressive ratio schedule of reinforcement. They also examined effects of footshock on the reinforcing efficacy of a half-maximal dose of cocaine. Finally, they tested the effects of footshock on cocaine self-administration in rats initially resistant to acquisition of cocaine self-administration. Footshock did not increase reinforcing efficacy of cocaine on a PR schedule …

Alzheimer's disease is a neurological disorder characterized by nerve degeneration and neuronal death. The diagnosis of Alzheimer's disease at an early stage is a major concern due to the lack of a standard and effective diagnosis procedure available to community healthcare providers, as well as the increasing numbers of the elderly population affected. Clinical evaluation, the standard AD diagnostic procedure conducted at major university hospitals and research clinics, achieves, on average, a positive predictive value of 93%, with a sensitivity of 83%, specificity of 55% and an overall accuracy of 75%. Virtual reality (VR) has emerged as a powerful visualization tool for design, simulation, and analysis in modem complex industrial systems. The primary motivation for this thesis is to develop a framework for the effective use of VR in design-simulation-analysis cycles, particularly in situations involving large, complex, multi-dimensional data-sets. This thesis develops a framework that is intended to support not only the integration of such data for visual, interactive, and immersive displays, but also provides a method for performing risk analysis. The natural gas transmission pipeline network in the United States is a key component of the nation's energy supply infrastructure and extends for over 280,000 miles and has an average age of over 60 years. The integrity of the pipeline is maintained by periodic inline inspections using magnetic or ultrasonic pigs. Defect characterization algorithms developed using current pigging data are hampered by the fact that single inspection techniques (either magnetic or ultrasonic) do not yield sufficient information for accurately and repeatably characterizing defects. Manipulation of the reproductive tract results in increased levels of prostaglandin, which may, in turn, reduce pregnancy rates in embryo recipients. Administration of a prostaglandin inhibitor prior to embryo transfer improves pregnancy rates in cows. Embryo transfer into heifers is more difficult and often requires additional manipulation of the uterus. This study was designed to determine whether administration of the prostaglandin inhibitor, flunixin meglumine, immediately prior to embryo transfer increases pregnancy rates in heifers. Heifers (n=466) were divided into two equal groups based on BCS (range=6-7) and weight (range=256-455). Estrus was synchronized in heifers by giving two injections of prostaglandin …

The material characteristics play a role in the suitability of chitosan for biomedical applications. This is not surprising since the degree of deacetylation of chitosan influences antimicrobial activity, degradation rate, immune reaction and mechanical properties such as strength and elongation. This study examines chitosans of variable material characteristics for wound and bone healing applications. Chitosan films of 76, 78, 80, 87, 91, 92, and 95% degree of deacetylation were tested in vitro for cellular responses by fibroblast and bone cell lines. Bone and bone cells have been shown to respond to mechanical forces placed upon them. Particularly, strain plays an important role in osteogenic differentiation of marrow cells around artificial implants in bone. These strains, depending on their magnitude, duration, and repetition, can alter the proliferation and matrix synthesis of osteoblasts. To test how strain parameters influence osteoblast behavior, a four-point bending apparatus was used to impose cyclic strain on osteogenic stem cells isolated from rats and seeded on titanium plates. Cells were stimulated at 1 Hz for 15 minutes daily and compared to an unstrained control. Stimulation occurred at two …

Forest management is a costly and time-consuming activity. Remote sensing has the potential to improve the process by making it cheaper and more efficient, but only if appropriate characteristics can be determined from computer-models. This thesis describes the implementation of a forest visualization system and a corresponding user study that tests the accuracy of parameter estimation and forest characterization. The study uses data obtained from field-surveys to generate a computer-modeled forest. Five different stands were tested. Based on the quantitative results obtained, generally, there is no statistically significant difference in parameter estimation when comparing field-recorded movies and computer-generated movies.

Reducing atmospheric CO2, a main source of greenhouse gas, has been accentuated recently. One focus is capture, separation, and sequestration of industrial CO2. As a hydrate former, CO2 forms hydrates at moderate temperatures and pressures. This phenomenon could be utilized to capture and separate CO2 from flue gases, and also has the potential to sequester CO2 in the deep sds. This research investigated the CO2-N2 separation efficiency of gas hydrates; it investigated the sequestration potential of CO2 hydrates in ocean sediments. The purpose of this study was to determine the effects of video modeling on skill acquisition in learning the golf swing. One-hundred-eight college students participated in this study. All participants were pre-tested via videotaping to determine initial skill level. The pre-test videotaping was analyzed using DartTrainer software. Each participant was randomly assigned to one of three groups (1) control (2) single-view or (3) multi-view. Chloroplast nucleoids (ct-nucleoids) are DNA/protein complexes involved in compacting the chloroplast genome, and may play a role in regulating DNA replication. Ct-nucleoids were isolated from young soybean plants and separated by 2-D gel electrophoresis. Gel spots were excised and analyzed by MALDI-ToF mass spectrometry, resulting in several protein identifications. The proteins identified all have functions unrelated to DNA replication. While some of these proteins may be due to contamination, it is possible that some of these proteins are dual-functional, playing direct roles in the regulation of DNA replication.

Three aspects of Xanthoparmelia cumberlandia and Xanthoparmelia coloradoënsis populations found at two elevations are explored: clustering of secondary chemicals and the resulting implications for taxonomic distinctions, the usefulness of thallus size as an indirect measure of sexual fecundity, and the frequency of sexual reproduction.
First, we use clustering of 46 chemicals produced by X. cumberlandia and X. coloradoënsis to evaluate the adequacy of the current taxonomic distinction between them. Using principal components analysis and UPGMA, we find that the currently recognized species boundaries indicated by the presence of stictic acid in X. cumberlandia and salazinic acid in X. coloradoënsis are …

The study investigated student and teacher outcomes from a Technology Innovation Challenge Grant project, NatureShift Linking Learning to Life (NS), to determine if the project influenced teachers’ work and student learning. The approaches were twofold: (1) to examine the implementation of higher order thinking and relevant use of technology by students and teachers and (2) to understand a teacher’s reactions to and implementation of a teaching model and methodology for Internet delivery and technology use.

Though body-image has been well-researched in women for more than 40 years, theoretical discussions and empirical investigations addressing male body-image have only recently began to emerge. Work by Pope, Phillips and Olivardia (2000) has begun to identify the dynamics of male body-image distortions and the role media depictions of muscular men may play in their pathogenesis. The empirical literature is curiously devoid of experimental investigations of male responses to media and advertising depictions of muscular men.

The purpose of this qualitative study was to explore university counseling center clients’ experience and perspective of their in-session covert reactions. The Consensual Qualitative Research method was used to analyze the qualitative data. Twelve participants who were seeking services at a midsize West Coast university counseling center participated in the study. Each participant completed a telephone interview and four questionnaires (Working Alliance Inventory-Short Form, demographic questionnaire, Target Concerns Questionnaire, and the Client Feedback Form).
The qualitative analysis revealed several meaningful categories pertaining to clients’ experience and perspective of their covert reactions. All of the participants in this study reported that …

Use of course management systems (CMS) in higher education has risen dramatically since their development in the late 1990s. This qualitative study investigated the use of a course management system (CMS) by faculty teaching undergraduate, traditional courses in a Midwestern liberal arts university. Black teachers’ unique historical experiences are often completely overlooked or amalgamated with those of White teachers in states such as North Dakota, where the population is predominantly White and the study of African Americans is particularly sparse. Acetyl-CoA carboxylase (ACC) catalyzes the formation of malnoyl-CoA, which in turn controls the rate of fatty acid metabolism. ACC beta or 2 has been shown to be localized on the mitochondria in close proximity to carnintine palmitoyl transferase 1 (CPT-1), the enzyme responsible for the influx of acyl-CoA into the matrix where beta oxidation takes place. CPT-1 is inhibited by malonyl-CoA produced by ACC. It has been well documented that AMP activated kinase (AMPK) when activated phosphorylates and inactivates ACC. There is a need for the development of smaller and lighter UAV's to perform certain missions. Density-dependence is considered one of the most important regulators of population growth, and it has been documented across a wide variety of species. Typically, population growth rate and components thereof decline with increasing density (i.e., negative density-dependence); however, in species that exhibit high population densities and social behavior, positive density-dependence (i.e., Allee effect) may occur at low density. June sucker, a federally endangered lake sucker endemic to Utah Lake, Utah, USA, occurred historically at high density, and it exhibits coordinated feeding behavior. Sixty premenopausal women (age 25-50) finished a 16 week randomized controlled trial looking at the effects of different jump programs on hip and spine bone mineral density (BMD). Subjects were randomly assigned to a control group or one of two jumping groups. The Jump 10 group performed 10 jumps with 30 seconds of rest between jumps, twice daily, while the Jump 20 group performed the same protocol but with 20 jumps. At 8 weeks, BMD percent change (%Δ) at the hip was significantly different among groups when age, weight change, and subject compliance were simultaneously adjusted. In particular, the Jump …

This dissertation describes a study utilizing bioinformatics to analyze homologues of a molecular chaperone, glucose-regulated protein 78 (grp 78), also known as BiP. The selected homologous proteins originate from organisms of infinitely diverse genera. Comparisons of protein sequence yielded the first clues of a common ancestry among these proteins. Furthermore, protein molecular weights, isoelectric points, N-terminal amino acids and half-lives of a known homolog and a non-homologous protein were examined. Additionally, electroporation, a state-of-the-art plasmid insertion technique, was explored using Chlamydomonas reinhardtii, a green alga, as the recipient of a parent plasmid, pSP124S. Prostate cancer is the most frequently diagnosed cancer in American men and the second leading cause of cancer deaths. Supplementation with Se has reduced the incidence of prostate cancer and Se status is inversely correlated with prostate cancer risk. One molecular mechanism by which high Se concentrations may affect cancer risk is by catalyzing disulfide bond formation or otherwise complexing with reactive sulfhydryl groups in cellular proteins. The estrogen receptor (ER) contains cysteines in zinc (Zn) fingers that are susceptible to oxidation and internal disulfide formation, which can prevent DNA binding.
