Genetic deletion or pharmacological blockade of the CB1 receptor has been reported to impair extinction learning in aversive conditioning (i.e., conditioned fear and Morris water maze) paradigms, but not in operant procedures in which food reinforcement is earned. It is difficult to discern whether the differential effects caused by CB1 receptor disruption on extinction result from the hedonics (i.e., aversive vs. appetitive) or is related to the required responses associated with these disparate tasks. This paper will talk about the videos and sculptural installation in my thesis exhibition. Shooting videos outside of the studio developed into a project overarching any individual video or its particular signs. Thus, this paper will focus on the video project with examples that follow a timeline of development, rather than the actual 6 videos on display in the exhibit. The two-part sculpture "Deer in the Headlights" is created in the context of these videos, and coexists with them in a specific architectural space. Approximately 200 million men and 100 million women smoke worldwide. In the United States, an estimated 25.9 million men (23.9 percent) and 20.7 million women (18.1 percent) are smokers. The commencement of smoking at a young age is thought to increase addiction liability, decrease the probability of successful cessation, and correlate with a higher number of cigarettes smoked per day. Studies from the World Health Organization indicate that between 80,000 and 100,000 children start smoking every day worldwide. These statistics suggest that adolescence is a critical phase for developing nicotine dependence. The work in this dissertation contributes to the further …

Drug abuse and addiction are considered to be a result, at least in part, of the rewarding effects produced by increasing dopamine levels. 5-HT3 serotonin receptors have been shown to indirectly affect dopamine levels. Therefore, the effect of the 5-HT3 receptor partial agonist, MD-354, on the actions of psychomotor stimulants was analyzed in mouse locomotor activity assays to determine whether MD-354 is working through a 5-HT3 receptor agonist or antagonist mode of action. Studies with (+)amphetamine and (+)methamphetamine in combination with MD-354 indicated MD-354 is either devoid of action or is behaving similar to the 5-HT3 receptor antagonist, ondansetron. This …

Kernicterus causes damage to the auditory system and the basal ganglia in humans. Although the Gunn rat model of kernicterus has been extensively used to characterize the auditory features, this model has not been utilized to systematically investigate the movement disorder. In the present study, spontaneously jaundiced (jj) 16 day old Gunn rat pups were treated with sulfadimethoxine to exacerbate bilirubin neurotoxicity and compared to saline treated jjs and non-jaundiced (Nj) littermates. Electromyographic (EMG) activity was recorded from antagonistic hip muscles in dystonic and in normal appearing rats. This project started out as program for multi-family housing with a need for natural light, then, it changed. Through countless studies, it became evident that what started as an animal made up of bones and tissues was really just the vessel for the animal's soul. Light is the soul of design. The program represents the tissue and bones of the animal that is design.In this project two paths were taken. The first being pragmatic, the design bound by the program. This thesis details the literary and visual influences in my work, the definition of American Gothic, and its connection it to my work. Literary sources such as Edgar Allan Poe and Fanny Kemble help spark a vision of the landscape. Visual influences include Japanese woodblock prints, scenic wallpapers, vintage postcards and Victorian mourning pictures. My regional explorations span the James River, Tidewater swamps and architecture within the city of Richmond.My work depicts local history and ecology inspired by Richmond and the surrounding region. My current work revels in a state of flux. I strive for the work to be electrically charged, conveying a feverish sense of immediacy and vitality that implies motion and frenetic energy. The work is an accretion of brightly colored biomorphic forms that extend out from the wall and onto the floor. Viscous parts ooze and drip while others are globular and bulbous. This thesis details -in personal narrative form- the process by which the author directed a production of Elizabeth Wong's Amazing Adventures of the Marvelous Monkey King at Dr. Michael M. Krop Senior High School in Miami, Florida. The following text explains elements of pre-production, presents relevant research associated with the play, describes the production process in detail, and states conclusions drawn by the author about the experience. Successful treatment of cancer with adoptive immunotherapy (AIT) is dependent on the ability to produce large numbers of tumor-specific, functional T cells. The purpose of this thesis is to explore ways in which T cell expansion could be augmented. We have focused on exploring alternate gamma chain cytokines as stimulators of T cell proliferation and differentiation in addition to investigating the potential usefulness of gemcitabine (GEM) in abrogating the immunosuppressive effects of myeloid derived suppressor cells (MDSCs). Catecholaminergic neurotransmission is regionally altered following injury, and drugs aimed at these systems offer promising avenues for post-TBI pharmacotherapies. Atomoxetine is a selective norepinephrine transporter (NET) inhibitor currently indicated for treatment of attention-deficit hyperactivity disorder (ADHD). The studies in this dissertation were designed to test the efficacy of atomoxetine for treating cognitive deficits following experimental TBI and the potential mechanism for any beneficial effect. The first part of the study focused on behavioral recovery following atomoxetine treatment. The dopamine transporter (DAT) is a presynaptic membrane phosphoprotein that terminates dopaminergic synaptic transmission by clearing dopamine (DA) back into presynaptic neuron by a reuptake process. DAT is also a molecular target for psychostimulants such as cocaine, amphetamine (AMPH) and methamphetamine (METH) that increase DA synaptic levels either by blocking reuptake or by inducing DA efflux. DAT phosphorylation and transport down regulation are best demonstrated with the treatment of Protein kinase C (PKC) activators such as (phorbol 12-myristate 13-acetate) PMA. Serine(s) and threonine(s) are known to be involved in DAT phosphorylation.

In the final stages of the bacterial growth cycle, daughter cells must be released from one another by cleaving the shared peptidoglycan cell wall that separates them. Although enzymes capable of cleaving bonds in the murein sacculus are ubiquitous among the bacteria, the physiological roles of only a few of these enzymes are clear. The peptidoglycan-specific amidases play an important role in cleaving the septum to release daughter cells after cell division in Escherichia coli. E. coli has three amidases dedicated to this task: AmiA, AmiB and AmiC. These enzymes remove side chains from the peptidoglycan by hydrolyzing the bond …

The use of information books in the preschool classroom, and the potential of the books and the learning activities associated with the books for promoting science among young children are explored. The research was first conducted with four children at a university children's center, and then replicated in a whole class setting at a college child development lab school in another state. Yersinia pestis, the causative agent of plague, is endemic to most of the world. Y. pestis' potential threat as a biological warfare agent has drawn increased attention to pathogenesis and vaccine research for the plague. A number of characteristics related both to the host response to, and bacterial pathogenesis of, Y. pestis were examined. To investigate the potential role of MHC Class II antigen presentation during the plague, HLA-DQ8αβ transgenic mice were infected with Y. pestis. This thesis compares the performance of heterogeneous and homogenous swarms. Swarms are defined as particles or agents which react to their environment and fellow particles or agents according to social rules. The weights of three attributes of an individual agent were varied for these experiments: Collision Avoidance with individual agents in the swarm, Center of Mass of the swarm and the parameter that controls Velocity Matching in the swarm. In homogenous swarms, all individuals had the same attribute weights while in heterogeneous swarms weights for one attribute were taken from a normal distribution for the population. Laser-induced fluorescence (LIF) has been shown to be potentially useful for identifying microorganisms in real time. It is a selective and sensitive technique because the excitation is performed at one wavelength while the emission is monitored at longer wavelengths so that background from the excitation source can be eliminated. This specialized optical property of LIF can be applied to development of an optical sensor capable of quickly, non-invasively, and quantitatively probing complex biochemical transformations in microorganisms. Effects of developmental exposure to chlorpyrifos (CPS) or methyl parathion (MPS) on visuospatial, adaptive fear response, and passive avoidance memory and the signaling mechanisms responsible for these neurocognitive changes were investigated. Using an incremental low dose regimen, rat pups were orally gavaged daily with either corn oil (vehicle), CPS, or MPS from postnatal day 1 (PND1) -PND21. Cholinesterase activity was significantly inhibited with the highest dosages of CPS and MPS for up to 19 days after the last dosages. OP exposure impaired working and reference memory in males whereas in the females, enhancement occurred following CPS exposure. In addition, the …
