Methods Matter: The Relationship Between Strength And Hypertrophy Depends On Methods Of Measurement And Analysis, Andrew D. Vigotsky, Brad J. Schoenfeld, Christian Than, J. Mark Brown
Publications and Research
Purpose: The relationship between changes in muscle size and strength may be affected by both measurement and statistical approaches, but their effects have not been fully considered or quantified. Therefore, the purpose of this investigation was to explore how different methods of measurement and analysis can affect inferences surrounding the relationship between hypertrophy and strength gain.
Methods: Data from a previous study—in which participants performed eight weeks of elbow flexor training, followed by an eight-week period of detraining—were reanalyzed using different statistical models, including standard between-subject correlations, analysis of covariance, and hierarchical linear modeling.
Results: The associative relationship between strength …

Spinal Control Of Locomotion: Individual Neurons, Their Circuits And Functions, Marie-Pascale Côté, Lynda M. Murray, Maria Knikou
Publications and Research
Systematic research on the physiological and anatomical characteristics of spinal cord interneurons along with their functional output has evolved for more than one century. Despite significant progress in our understanding of these networks and their role in generating and modulating movement, it has remained a challenge to elucidate the properties of the locomotor rhythm across species. Neurophysiological experimental evidence indicates similarities in the function of interneurons mediating afferent information regarding muscle stretch and loading, being affected by motor axon collaterals and those mediating presynaptic inhibition in animals and humans when their function is assessed at rest. However, significantly different muscle …

DNA damage response pathways are essential for genome stability and cell survival. Specifically, the ATR kinase is activated by DNA replication stress. An early event in this activation is the recruitment and phosphorylation of RPA, a single stranded DNA binding complex composed of three subunits, RPA70, RPA32 and RPA14. We have previously shown that the LIM protein Ajuba associates with RPA, and that depletion of Ajuba leads to potent activation of ATR. In this study, we provide evidence that the Ajuba-RPA interaction occurs through direct protein contact with RPA70, and that their association is cell cycle-regulated and is reduced upon …

A Novel Multiscale Mathematical Model For Building Bone Substitute Materials For Children, Abdennasser Chekroun, Laurent Pujo-Menjouet, Jean-Philippe Berteau
Publications and Research
Bone is an engineering marvel that achieves a unique combination of stiffness and toughness exceeding that of synthesized materials. In orthopedics, we are currently challenged for the child population that needs a less stiff but a tougher bone substitute than adults. Recent evidence suggests that the relationship between inter-molecular connections that involve the two main bone building blocks, TropoCollagen molecules (TC) and carbonated Hydroxyapatite (cAp), and bone macroscopic mechanical properties, stiffness and toughness, are key to building bone substitute materials for children. The goal of our study is to establish how inter-molecular connections that occur during bone mineralization are related …

Background: Butterflies (Papilionoidea) are perhaps the most charismatic insect lineage, yet phylogenetic relationships among them remain incompletely studied and controversial. This is especially true for skippers (Hesperiidae), one of the most species-rich and poorly studied butterfly families.
Methods: To infer a robust phylogenomic hypothesis for Hesperiidae, we sequenced nearly 400 loci using Anchored Hybrid Enrichment and sampled all tribes and more than 120 genera of skippers. Molecular datasets were analyzed using maximum-likelihood, parsimony and coalescent multi-species phylogenetic methods.
Results: All analyses converged on a novel, robust phylogenetic hypothesis for skippers. Different optimality criteria and methodologies recovered almost …

We investigated the effects of direct current stimulation (DCS) on fluid and solute transport across endothelial cell (EC) monolayers in vitro. Our motivation was transcranial direct current stimulation (tDCS) that has been investigated for treatment of neuropsychiatric disorders, to enhance neurorehabilitation, and to change cognition in healthy subjects. The mechanisms underlying this diversity of applications remain under investigation. To address the possible role of blood-brain barrier (BBB) changes during tDCS, we applied direct current to cultured EC monolayers in a specially designed chamber that generated spatially uniform direct current. DCS induced fluid and solute movement across EC layers that persisted …

Impacts Of Stress On Forest Recovery And Its Interaction With Canopy Height, Peipei Xu, Tao Zhou, Chuixiang Yi, Hui Luo, Xiang Zhao, Wei Fang, Shan Gao, Xia Liu
Publications and Research
Global climate change is leading to an increase in the frequency, intensity, and duration of drought events, which can affect the functioning of forest ecosystems. Because human activities such as afforestation and forest attributes such as canopy height may exhibit considerable spatial differences, such differences may alter the recovery paths of drought-impacted forests. To accurately assess how climate affects forest recovery, a quantitative evaluation on the effects of forest attributes and their possible interaction with the intensity of water stress is required. Here, forest recovery following extreme drought events was analyzed for Yunnan Province, southwest China. The variation in the …

Retraction Of Dna-Bound Type Iv Competence Pili Initiates Dna Uptake During Natural Transformation In Vibrio Cholerae, Courtney K. Ellison, Triana N. Dalia, Alfredo Vidal Ceballos, Joseph Che-Yen Wang, Nicolas Biais, Yves V. Brun, Ankur B. Dalia
Publications and Research
Natural transformation is a broadly conserved mechanism of horizontal gene transfer in bacterial species that can shape evolution and foster the spread of antibiotic resistance determinants, promote antigenic variation, and lead to the acquisition of novel virulence factors. Surface appendages called competence pili promote DNA uptake during the first step of natural transformation1 , however, their mechanism of action has remained unclear due to an absence of methods to visualize these structures in live cells. Here, using the model naturally transformable species Vibrio cholerae and a pilus labeling method, we define the mechanism for type IV competence pilus-mediated DNA uptake …

Not All College Faculty Are Equal, Aldemaro Romero Jr.
Publications and Research
Despite the fact that college faculty seem to lead very public lives because they show up in front of audiences on a regular basis, the fact of the matter is that most people – even faculty themselves –don’t know how much time they spend doing the different aspects of their jobs. A new study helps us better understand faculty by grouping them according to the way they spend their time.
A team of researchers from the Center for Postsecondary Research at Indiana University at Bloomington just published a report based on the analyses of responses to the center’s survey of …

Dynamic Responses Of Tree-Ring Growth To Multiple Dimensions Of Drought, Wei Fang, Shan Gao, Ruishun Liu, Tao Zhou, Chuixiang Yi, Ruijie Liu, Xiang Zhao, Hui Luo
Publications and Research
Droughts, which are characterized by multiple dimensions including frequency, duration, severity and onset timing, can impact tree growth profoundly. Different dimensions of drought influence tree growth independently or jointly, which makes the development of accurate predictions a formidable challenge. Measurement-based tree-ring data have obvious advantages for studying the drought responses of trees. Here, we explored the use of abundant tree-ring records for quantifying regional response patterns to key dimensions of drought. Specifically, we designed a series of regional-scaled “natural experiments”, based on 357 tree-ring chronologies from Southwest USA and location-matched monthly water balance anomalies, to reveal how tree-ring growth responds …

A homologous series of electronically tuned 2,20,200-nitrilotris(N-arylacetamide) pre-ligands (H3LR) were prepared (R ¼ NO2, CN, CF3, F, Cl, Br, Et, Me, H, OMe, NMe2) and some of their corresponding Fe and Zn species synthesized. The iron complexes react rapidly with O2, the final products of which are diferric mu-oxo bridged species. The crystal structure of the oxidized product obtained from DMA solutions contain a structural motif found in some diiron proteins. The mechanism of iron mediated O2 reduction was explored to the extent that allowed us to construct an empirically consistent rate law. A Hammett plot was constructed that enabled …

Background: The suboptimal rate of viral suppression among persons aged 13 years and older and residing in 37 states and the District of Columbia leaves considerable opportunities for onward transmission and contributes to poor health outcomes. Men who have sex with men (MSM) represent one of the most at-risk groups in the United States. There is a clear and continued need for innovative adherence support programs to optimize viral suppression. To address this gap, we designed and are implementing a randomized controlled trial (RCT) to test the efficacy of the Thrive with Me intervention for MSM living with HIV. Critical …

Perinatal Pb2+ Exposure Alters The Expression Of Genes Related To The Neurodevelopmental Gaba-Shift In Postnatal Rats, Lorenz Simon Neuwirth, Greg R. Phillips, Abdeslem El Idrissi
Publications and Research
Background: Lead (Pb2+) is an environmental neurotoxicant that disrupts neurodevelopment, communication, and organization through competition with Ca2+ signaling. How perinatal Pb2+ exposure affects Ca2+-related gene regulation remains unclear. However, Ca2+ activates the L-Type voltage sensitive calcium channel β-3 subunit (Ca-β3), which autoregulates neuronal excitability and plays a role in the GABA-shift from excitatory-toinhibitory neurotransmission.
Method: A total of eight females (n = 4 Control and n = 4 Perinatal) and four males (n = 2 Control and n = 2 Perinatal) rats were used as breeders to serve as Dams and Sires. The Dam’s litters each ranged from N = …

Publications and Research
Background/objectives: Maternal obesity increases the risk of gestational diabetes mellitus (GDM), which results in fetal overgrowth and long-lasting metabolic dysfunctioning in the offspring. Previous studies show that maternal choline supplementation normalizes fetal growth and adiposity of progeny from obese mice. This study examines whether supplementation of betaine, a choline derivative, has positive effects on fetal metabolic outcomes in mouse progeny exposed to maternal obesity and GDM.
Methods: C57BL/6J mice were fed either a high-fat (HF) diet or a control (normal-fat, NF) diet and received either 1% betaine (BS) or control untreated (BC) drinking water 4–6 weeks before timed-mating and throughout …

Dysregulated activity of helicase eIF4A drives transformation to and maintenance of cancer cell phenotype by reprogramming cellular translation. Interleukin 24 (IL-24) is a tumor-suppressing protein, which has the ability to inhibit angiogenesis, sensitize cancer cells to chemotherapy, and induce cancer cell-specific apoptosis. In this study, we found that eIF4A is inhibited by IL-24. Consequently, selective reduction of translation was observed for mRNAs harboring strong secondary structures in their 50 -untranslated regions (50UTRs). These mRNAs encode proteins, which function in cell survival and proliferation. Consistently, overexpression of eIF4A conferred cancer cells with resistance to IL-24-induced cell death. It has been established …

What We Do Not Know About Fungal Cell Adhesion Molecules, Peter N. Lipke
What We Do Not Know About Fungal Cell Adhesion Molecules, Peter N. Lipke
Publications and Research
There has been extensive research on structure and function of fungal cell adhesion molecules, but the most of the work has been about adhesins in Candida albicans and Saccharomyces cerevisiae. These yeasts are members of a single ascomycete order, and adhesion molecules from the six other fungal phyla are only sparsely described in the literature. In these other phyla, most of the research is at the cellular level, rather than at the molecular level, so there has been little characterization of the adhesion molecules themselves. A catalog of known adhesins shows some common features: high Ser/Thr content, tandem repeats, N- …

Excessive and pathological uses of the Internet are observed and discussed often in our modern conversations. Access to the Internet has become so convenient that these behaviors can lead to consequences in many areas of our lives from social relationships to academic and professional work performance. A common term that people use to address this pattern of behaviors is “Internet Addiction” or more specific ones, such as “Social Media Addiction” or “Online Gaming Addiction.” However, in clinical psychology, addiction has its own specific definitions. It refers to a category and set of criteria that are distinguished from other mental disorders …

Chia (Salvia hispanica) and basil (Ocimum basilicum) seeds have the intrinsic ability to form a hydrogel concomitant with moisture-retention, slow releasing capability and proposed health benefits such as curbing diabetes and obesity by delaying digestion process. However, the underlying mode of gelation at nanoscopic level is not clearly explained or explored. The present study elucidates and corroborates the hypothesis that the gelling behavior of such seeds is due to their nanoscale 3D-network formation. The preliminary study revealed the influence of several conditions like polarity, pH and hydrophilicity/ hydrophobicity on fiber extrusion from the seeds which leads to gelation. Optical microscopic …

The translation of mRNAs plays a critical role in the regulation of gene expression and therefore, in the regulation of cell proliferation, differentiation and apoptosis. Unrestricted initiation of translation causes malignant transformation and plays a key role in the maintenance and progression of cancers. Translation initiation is regulated by the ternary complex and the eukaryotic initiation factor 4F (eIF4F) complex. The p53 tumor suppressor protein is the most well studied mammalian transcription factor that mediates a variety of anti-proliferative processes. Post-transcriptional mechanisms of gene expression in general and those of translation in particular play a major role in shaping the …
