Early Invasion Of Brain Parenchyma By African Trypanosomes, Ute Frevert, Alexandru Movila, Olga V. Nikolskaia, Jayne Raper, Zachary B. Mackey, Maha Abdulla, James Mckerrow, Dennis J. Grab

Publications and Research

Human African trypanosomiasis or sleeping sickness is a vector-borne parasitic disease that has a major impact on human health and welfare in sub-Saharan countries. Based mostly on data from animal models, it is currently thought that trypanosome entry into the brain occurs by initial infection of the choroid plexus and the circumventricular organs followed days to weeks later by entry into the brain parenchyma. However, Trypanosoma brucei bloodstream forms rapidly cross human brain microvascular endothelial cells in vitro and appear to be able to enter the murine brain without inflicting cerebral injury. Using a murine model and intravital brain imaging, …

The Structural Stability Of The Endothelial Glycocalyx After Enzymatic Removal Of Glycosaminoglycans, Ye Zeng, Eno E. Ebong, Bingmei M. Fu, John M. Tarbell Aug 2012

The Structural Stability Of The Endothelial Glycocalyx After Enzymatic Removal Of Glycosaminoglycans, Ye Zeng, Eno E. Ebong, Bingmei M. Fu, John M. Tarbell

Publications and Research

Rationale It is widely believed that glycosaminoglycans (GAGs) and bound plasma proteins form an interconnected gel-like structure on the surface of endothelial cells (the endothelial glycocalyx layer–EGL) that is stabilized by the interaction of its components. However, the structural organization of GAGs and proteins and the contribution of individual components to the stability of the EGL are largely unknown. Objective To evaluate the hypothesis that the interconnected gel-like glycocalyx would collapse when individual GAG components were almost completely removed by a specific enzyme. Methods and Results Using confocal microscopy, we observed that the coverage and thickness of heparan sulfate (HS), …

Γ-Synuclein Interacts With Phospholipase Cβ2 To Modulate G Protein Activation, Urszula Golebiewska, Yuanjian Guo, Narindra Khalikaprasad, Cassandra Zurawsky, V. Siddhartha Yerramilli, Suzanne Scarlata

Publications and Research

Phospholipase Cβ2 (PLC β2) is activated by G proteins and generates calcium signals in cells. PLCβ2 is absent in normal breast tissue, but is highly expressed in breast tumors where its expression is correlated with the progression and migration of the tumor. This pattern of expression parallels the expression of the breast cancer specific gene protein 1 which is also known as γ-synuclein. The cellular function of γ-synuclein and the role it plays in proliferation are unknown. Here, we determined whether γ-synuclein can interact with PLCβ2 and affect its activity. Using co-immunprecitation and co-immunofluorescence, we find that in both benign …

Weaponizing Tear Gas: Bahrain’S Unprecedented Use Of Toxic Chemical Agents Against Civilians., Richard Sollom, Holly G. Atkinson

Publications and Research

The Bahraini government’s response to the early 2011 pro-democracy protests was brutal, systematic, and violent. In addition to birdshot and rubber bullets, government law enforcement attacked unarmed protestors with toxic chemical agents including tear gas. The government’s crackdown on the medical profession was especially harmful, as security forces arrested and detained doctors, raided health facilities, and obstructed patients from receiving necessary care. This report’s findings are based on field research that the authors conducted in Bahrain (April 2012) to investigate excessive use of force by law enforcement officials since June 2011—the end of Bahrain’s state of emergency. The medico-legal team …

Doherty, Leah D. Pride, James Lukose, Brian E. Snydsman, Ronald Charles, Ajay Pramanik, Eric G. Muller, David Botstein, Carol Wood Moore

Publications and Research

Cytoprotective functions of a 20S proteasome activator were investigated. Saccharomyces cerevisiae Blm10 and human 20S proteasome activator 200 (PA200) are homologs. Comparative genome-wide analyses of untreated diploid cells lacking Blm10 and growing at steady state at defined growth rates revealed downregulation of numerous genes required for accurate chromosome structure, assembly and repair, and upregulation of a specific subset of genes encoding protein-folding chaperones. Blm10 loss or truncation of the Ubp3/Blm3 deubiquitinating enzyme caused massive chromosomal damage and cell death in homozygous diploids after phleomycin treatments, indicating that Blm10 and Ubp3/Blm3 function to stabilize the genome and protect against cell death. …

Publications and Research

The role of ice in the formation of chemically active halogens in the environment requires a full understanding because of its role in atmospheric chemistry, including controlling the regional atmospheric oxidizing capacity in specific situations. In particular, ice and snow are important for facilitating multiphase oxidative chemistry and as media upon which marine algae live. This paper reviews the nature of environmental ice substrates that participate in halogen chemistry, describes the reactions that occur on such substrates, presents the field evidence for ice-mediated halogen activation, summarizes our best understanding of ice-halogen activation mechanisms, and describes the current state of modeling …

Pioneers Of Youth Justice Reform: Achieving System Change Using Resolution, Reinvestment, And Realignment Strategies, Douglas N. Evans Jun 2012

Pioneers Of Youth Justice Reform: Achieving System Change Using Resolution, Reinvestment, And Realignment Strategies, Douglas N. Evans

Publications and Research

In the past three decades, state and local governments implemented various reform strategies to reduce the youth justice system’s reliance on confinement facilities and serve as many youths as possible in their own homes or at least in their own communities when removal from the home is warranted. The various reform strategies may be conceptualized as relying on three distinct but interrelated mechanisms: resolution, reinvestment, and realignment (Butts and Evans 2011). Resolution refers to the use of managerial authority and administrative directives to influence system change; reinvestment entails using financial incentives to encourage system change, and realignment employs organizational and …

Production Of Mycobacterial Cell Wall Glycopeptidolipids Requires A Member Of The Mbth-Like Protein Family, Elizabeth Tatham, Sivagami Sundaram Chavadi, Poornima Mohandas, Uthamaphani R. Edupuganti, Shiva K. Angala, Delphi Chatterjee, Luis E N Quadri

Publications and Research

Background Glycopeptidolipids (GPLs) are among the major free glycolipid components of the outer membrane of several saprophytic and clinically-relevant Mycobacterium species. The architecture of GPLs is based on a constant tripeptide-amino alcohol core of nonribosomal peptide synthetase origin that is N-acylated with a 3-hydroxy/methoxy acyl chain synthesized by a polyketide synthase and further decorated with variable glycosylation patterns built from methylated and acetylated sugars. GPLs have been implicated in many aspects of mycobacterial biology, thus highlighting the significance of gaining an understanding of their biosynthesis. Our bioinformatics analysis revealed that every GPL biosynthetic gene cluster known to date contains a …

Coordination Between Proteasome Impairment And Caspase Activation Leading To Tau Pathology: Neuroprotection By Camp, M. J. Metcalfe, Q. Huang, Maria E. Figueiredo-Pereira Jun 2012

Coordination Between Proteasome Impairment And Caspase Activation Leading To Tau Pathology: Neuroprotection By Camp, M. J. Metcalfe, Q. Huang, Maria E. Figueiredo-Pereira

Publications and Research

Neurofibrillary tangles (NFTs) are hallmarks of Alzheimer’s disease (AD). The main component of NFTs is TAU, a highly soluble microtubule-associated protein. However, when TAU is cleaved at Asp421 by caspases it becomes prone to aggregation leading to NFTs. What triggers caspase activation resulting in TAU cleavage remains unclear. We investigated in rat cortical neurons a potential coordination between proteasome impairment and caspase activation. We demonstrate that upon proteasome inhibition, the early accumulation of detergent-soluble ubiquitinated (SUb) proteins paves the way to caspase activation and TAU pathology. This occurs with two drugs that inhibit the proteasome by different means: the product …

The Manufacture Of Lifestyle: The Role Of Corporations In Unhealthy Living, Nicholas Freudenberg May 2012

The Manufacture Of Lifestyle: The Role Of Corporations In Unhealthy Living, Nicholas Freudenberg

Publications and Research

Recently, researchers have debated two views on the connection between lifestyle and health. In the first, health-related lifestyles including tobacco and alcohol use, diet, and physical activity are seen as primary influences on health. In the second, social stratification is the dominant influence with lifestyles simply markers of social status. Neither approach leads to interventions that can reverse the world's most serious health problems. This article proposes that corporate practices are a dominant influence on the lifestyles that shape patterns of health and disease. Modifying business practices that promote unhealthy lifestyles is a promising strategy for improving population health. Corporations …

High Tech Tethers And Work-Family Conflict: A Conservation Of Resources Approach, Katherine M. Richardson, Cynthia A. Thompson

Publications and Research

This is one of the first studies to empirically examine the relationship between wireless communications technology and work interference with family, and results shed light on the motivating factors that influence individuals to continuously engage with mobile technologies, sometimes to their personal detriment. We draw from Conservation of Resources (COR) theory to examine the relationship between using mobile communications technologies during non-work time (e.g., evenings, weekends, vacation) and psychological variables related to work-family conflict and well being, and whether this relationship is mediated by perceptions of job control and detachment from work. We collected data from 139 full-time working adults …

Polyglutamine Toxicity Is Controlled By Prion Composition And Gene Dosage In Yeast, He Gong, Nina V. Romanova, Kim D. Allen, Pavithra Chandramowlishwaran, Kavita Gokhale, Gary P. Newnam, Piotr Miceczkowski, Michael Y. Sherman, Yury O. Chernoff

Publications and Research

Polyglutamine expansion causes diseases in humans and other mammals. One example is Huntington’s disease. Fragments of human huntingtin protein having an expanded polyglutamine stretch form aggregates and cause cytotoxicity in yeast cells bearing endogenous QN-rich proteins in the aggregated (prion) form. Attachment of the proline(P)-rich region targets polyglutamines to the large perinuclear deposit (aggresome). Aggresome formation ameliorates polyglutamine cytotoxicity in cells containing only the prion form of Rnq1 protein. Here we show that expanded polyglutamines both with (poly-QP) or without (poly-Q) a P-rich stretch remain toxic in the presence of the prion form of translation termination (release) factor Sup35 (eRF3). …

Dismantling The Monolith: Post-Media Art And The Culture Of Instability, Nora Almeida Apr 2012

Dismantling The Monolith: Post-Media Art And The Culture Of Instability, Nora Almeida

Publications and Research

Art that falls under the “new media” paradigm is problematic, or rather, it renders many traditional assumptions about art as problematic. In a practical sense, new media art raises fundamental questions about the nature of curation and preservation and the role of cultural heritage institutions as stewards of digital assets. Curation and preservation challenges, while significant, are fundamentally a symptom of a more catastrophic failure of concepts and language to adequately address changing relationships between art, materiality, and audiences. This article explores how burgeoning concepts in information and media theory may help shape curation contexts and redefine approaches to preservation. …

Delegated Performance: Outsourcing Authenticity, Claire Bishop Apr 2012

Delegated Performance: Outsourcing Authenticity, Claire Bishop

Publications and Research

An essay is presented on delegated performance in which nonprofessionals are hired to perform at a particular time on behalf of the artist on his or her instructions. The author stated that this social turn started since 1990s in contemporary art against the tradition of 1960s and 1970s where artists like Chris Burden, Vito Acconci and Gina Pane performed themselves. The author referred to the trend of live installation started in Europe in 1990s in which guards for exhibition were hired. The Italian artist Maurizio Cattelan assembled a soccer club of foreigners to play local matches in 1991.

Recent evidence from functional magnetic resonance imaging suggests that cortical hemodynamic responses coincide in different subjects experiencing a common naturalistic stimulus. Here we utilize neural responses in the electroencephalogram (EEG) evoked by multiple presentations of short film clips to index brain states marked by high levels of correlation within and across subjects. We formulate a novel signal decomposition method which extracts maximally correlated signal components from multiple EEG records. The resulting components capture correlations down to a one-second time resolution, thus revealing that peak correlations of neural activity across viewings can occur in remarkable correspondence with arousing moments of the …
