During many cellular processes, such as cell morphogenesis, migration, and asymmetric cell division, eukaryotic cells modulate their growth and polarity. Understanding the molecular mechanisms that govern polarized cell growth has the potential to reveal important insights into diseases such as neurodegeneration, heart disease, and cancer. The model organism fission yeast Schizosaccharomyces pombe, with its cylindrical shape and bipolar growth zones, enables straightforward evaluation of changes in growth and polarity upon genetic or pharmacological manipulation. Ion channels of the DEG/ENaC family can induce neurodegeneration under conditions in which they become hyperactivated. The C. elegans DEG/ENaC channel MEC-4(d) encodes a mutant channel with a substitution in the pore domain that causes swelling and death of the six touch neurons in which it is expressed. Dominant mutations in homolog channel UNC-8 result in uncoordinated movement. Here we show that this unc-8(d) movement defect is correlated with the selective death of cholinergic motor neurons in the ventral nerve cord, a milder toxicity compared with MEC-4(d). UNC-8(G387E), denoted UNC-8(d), and UNC-8(A586T) mutants encode hyperactivated channels that are strongly inhibited …

In multiple sclerosis (MS), soluble tumor necrosis factor (TNF) is detrimental via activation of TNF receptor 1 (TNFR1), whereas transmembrane TNF is beneficial primarily by activating TNF receptor 2 (TNFR2). Here we investigate the role of TNFR2 in microglia and monocytes/macrophages in experimental autoimmune encephalomyelitis (EAE), a model of MS, by cell-specific gene targeting. We show that TNFR2 ablation in microglia leads to early onset of EAE with increased leukocyte infiltration, T cell activation, and demyelination in the central nervous system (CNS). Cardiac stem cells (CSCs) are being evaluated for their efficacy in the treatment of heart failure. However, numerous factors impair the exogenously delivered cells’ regenerative capabilities. Hypoxia is one stress that contributes to inadequate tissue repair. Here, we tested the hypothesis that hypoxia impairs cell proliferation, survival, and migration of human CSCs relative to physiological and room air oxygen concentrations. Human endomyocardial biopsy-derived CSCs were isolated, selected for c-Kit expression, and expanded in vitro at room air (21% O2). The AAA+ (ATPases associated with a variety of cellular activities) protein superfamily includes approximately 30,000 molecular machines powered by adenosine triphosphate (ATP) binding and hydrolysis events, which are important for many cellular processes, including cell division, DNA replication, intracellular transport, and protein quality control. The bacterial AAA+ protein ClpXP is a two component ATP-dependent chaperone-protease that recognizes protein substrates bearing specific recognition signals, subsequently unfolding and degrading them to eliminate unnecessary or misfolded proteins. Most concerning is the emergence of multi-drug resistant (MDR) bacterial pathogens, especially the ‘ESKAPE’ pathogens for which treatment options are dwindling. To complicate the problem, approvals of antibiotic drugs are extremely low and many research and development efforts in the pharmaceutical industry have ceased, leaving little certainty that critical new antibiotics are nearing the clinic. A significant discrepancy exists between the rate of mental health problems in children and adolescents and their engagement in mental health services. A major contributor to this problem is poor mental health literacy among parents. Parents are typically responsible for identifying mental health issues in their children, but studies show that they struggle with problem recognition. Fanconi Anemia (FA) is a rare autosomal X-linked recessive disorder, characterized by congenital abnormalities, pediatric bone marrow failure and cancer susceptibility. FA is caused by biallelic mutation in any one of 22 different genes. The main functions of the FA-BRCA pathway is the resolution of interstrand crosslinks (ICLs) within the DNA. The main activating step of the pathway is the monoubiquitination of the proteins FANCD2 and FANCI. In my dissertation I examine the representation of self-creation through anorexia and bulimia in memoiristic narratives, blogs, and YouTube videos in contemporary American culture. The oral hypoglycemic agent Glyburide has been shown to be actively transported from various biological tissues though a pronounced interaction with the ATP binding cassette (ABC) active transport proteins. Specifically, Glyburide is actively transported by the ABC proteins, P-glycoprotein (Pgp), Breast Cancer Resistance Protein (BCRP), and the Multidrug Resistance Proteins (MRP). This active transport occurs at various locations throughout the body but has been identified as being responsible for Glyburide’s unique placental transport behavior. Stand-up comedians are rhetors who use humor as a rhetorical tool to inform and persuade mass audiences in the classical rhetorical tradition. In this dissertation, I work to recover the rhetorical motives of stand-up comedy, just as rhetoric and composition scholars before me have reread and recovered the legacy of the Sophists. To that end, I align stand-up comedians first with the historical heritage of Sophists and jesters, and then with the more contemporary tradition of public intellectuals and social activists. Vibrio anguillarum is an extracellular bacterial pathogen that is a causative agent of vibriosis in finfish and crustaceans. Mortality rates range from 30% to 100% and systemic infection usually causes fish to die within 1-4 days of initial infection. The primary routes of infections are through the skin, gills and intestines. Most cancers are assumed to develop from the initiation of chemically induced DNA damages. Biogeochemical gas budgets at high-latitude regions and sea ice zones are a source of uncertainty in climate models. The four main processes that regulate these budgets include advection, ventilation, mixing, and accumulation/release from sea ice. Considering the scarcity of data in sea ice zones, specifically during winter time, the environment is too poorly sampled to constrain these processes through direct measurements; hence we proposed models to systematically investigate these processes. Nucleic acid based drugs such as plasmid DNA (pDNA), small interfering RNA (siRNA), short hairpin RNA (shRNA), micro RNA (miRNA), and both antisense and antigene oligonucleotides, are potentially potent and specific compounds for therapeutic applications. Many major life threatening ailments might be treated using these molecules, and many polynucleotide products are currently in advanced clinical development.
