HIV/AIDS continues to cause significant morbidity and mortality across the globe. Even though current drug therapy can reduce the disease burden, a cure to HIV-1 infection remains elusive due to the relatively stable viral reservoir. Infected macrophages are of particular importance to the viral reservoir as they are long-lived cells that are often located in immune privileged sites and resistant to the cytopathic effects of the virus. Invasion of the cell by the primate lentiviruses HIV/SIV, is opposed by cellular restrictions including APOBEC3G, Tetherin/BST2, and SAMHD1. As the number of older adults increases in the US over the next few decades, the racial and ethnic diversity of the older adult population is also expected to increase. As such, the burden of age-related cognitive impairment in racial and ethnic minorities is an urgent public health priority. Studies using the self-report altruism scale (SRA) to measure trait altruism have found that altruism is uncorrelated with antisocial motives and positively correlated with being female and number of sex partners. However, it is unknown whether the SRA scale meets basic psychometric standards, such as unidimensionality. The act-frequency format of the SRA likely undermines its ability to isolate any one motive for behaving prosocially. In a pilot study (N =276), factor analysis indicated that the SRA contains dimensions related to the performance of favors (Factor 1), charitable giving (Factor 2), and politeness among strangers (Factor 3). Adequate caloric and carbohydrate intakes are necessary for positive adaptations to exercise training, yet there is limited research examining dietary intake in relation to strength and power in female athletes. The purpose of this study was to determine 1) whether there were significant changes in weekly total caloric and macronutrient consumption, strength, and power, and 2) whether total caloric and macronutrient consumption significantly and positively contributed to changes in strength and power across a controlled eight-week, off-season resistance training program. Produced primarily by CD4+ T cells, IL-2 is a cytokine, which promotes proliferation but more importantly is critical for regulatory T cell (Treg) development, homeostasis, and competitive fitness. For over 20 years, IL-2 therapy has been used in the treatment of malignant disorders, however, with toxic effects. A new and promising therapeutic application for IL-2 uses administration of low doses to selectively expand Tregs in attempt to boost immunity and promote tolerance in patients with autoimmunity. Approximately 20-25 percent of Traumatic Brain Injury (TBI) subjects develop Acute Lung Injury (ALI), but the pathomechanisms of TBI-induced ALI remain poorly defined. In my thesis, I investigated whether (Extracellular Vesicle) EV-mediated inflammasome signalling contributed to the etiology of TBI-induced ALI. C57/BL6 male mice were subjected to Controlled Cortical Impact, and the brains and lungs were examined for inflammasome activation and ALI at 4 hours and 24 hours after TBI. In remote areas with limited access to clean water, disease outbreaks can have a much larger impact than in areas where clean water is available. Portable net-zero greywater (NZGW) systems may be a solution, allowing users to wash hands or shower between patients in areas where fresh water is not typically available, unlike conventional greywater reuse systems. A lack of brain perfusion results in a global brain ischemia resulting in cell death in several regions including the hippocampus, an area important for learning and memory. Neuronal activity accompanies synapse formation and maintenance, but how early activity contributes to behavior development is poorly understood. Here, we used the Caenorhabditis elegans egg-laying motor circuit as a model to understand how coordinated circuit activity patterns emerge during development in relation to behavior. The C. elegans egg-laying circuit is a simple neural circuit which develops during the fourth larval stage and drives a two-state behavior in adult animals. Neuroprotective agents administered post cerebral ischemia have failed so far in the clinic to promote significant recovery. Thus, numerous efforts were redirected towards prophylactic approaches such as preconditioning as an alternative therapeutic strategy. Our lab has revealed a novel long-term window of cerebral ischemic tolerance mediated by resveratrol preconditioning (RPC) that lasts for two weeks in mice. To identify its mediators, we conducted an RNA-seq experiment on the cortex of mice two weeks post RPC, which revealed 136 differentially expressed genes. In the first chapter of this dissertation, I present my experimental work on seeded Yb-doped Q-switched fiber laser based on stimulated Brillouin scattering (SBS). I began by experimenting with different types of fiber laser cavity configurations in order to find an ideal cavity that would successfully generate the free-running stochastic nonlinear SBS mirror as well as to reinforce the injected light mode. This study presents a statistical mechanical analysis of single molecular chains at thermal equilibrium subject to tension, torque, and contact with a fluid medium. Polymeric chains are modeled as one-dimensional, microscopic groups of monomers. The methodology treats the response of monomers to tension, torque, and fluid medium contact as sets of statistically interacting quasi-particles. Complex elastic behavior and structural transitions as observed in single-molecule manipulation experiments are successfully described using this methodology. Models pertaining to thermal unbending, linear elasticity, torque-twist characteristics, and structural transformations including effects of cooperativity are presented. This study aims to supplement the paleogeodetic database of past Cascadia subduction zone earthquakes and further our understanding of the Cascadia subduction zone seismic and tsunami hazards. I first address a previously identified spatial gap within the Cascadia paleogeodetic database in southern Cascadia by refining the timing and magnitude of Cascadia subduction zone earthquakes over the past 2000 years in northern Humboldt Bay, California (~44.8°N, -124.2°W). There, I mapped wetland stratigraphy consistent with past megathrust earthquakes across three marshes; Jacoby Creek, McDaniel Creek, and Mad River Slough. Biomimetic propulsion is an active area of research in the field of robotics. This work presents advancements to help improve the performance of vehicles employing oscillating foil propulsion. For vehicles operating near the sea floor, ground effect on rolling and pitching foils was investigated in Manuscript 1. The ‘ground effect’ was found to be present and to cause a repulsion force on the foil as well as an increased thrust force. The purpose of this study was to explore the perspectives and lived experiences of Black public school teachers as related to the underrepresentation of Black teachers in the teaching profession. Data were collected through interviews with ten teachers who identified as Black and were employed by two school districts in northeastern Rhode Island. Through phenomenology as a methodology, and autoethnography as a method, this study sought to explore the lived experiences of Black teachers in contemporary public school settings and their perspectives on teacher diversity. Particle-based delivery systems have been investigated for their potential to increase the efficacy of patient diagnoses and treatments. In many cases, these systems increase the bioavailability and/or decrease the toxicity of clinically relevant drugs. These developments have led to improvements in patient compliance, morbidity, mortality, and quality of life. To date, particle-based delivery systems have been evaluated in hundreds of clinical trials around the world. Remotely activated hydrogel-based biomaterials can maximize the therapeutic effects by providing localized deliveries and minimizing off-target side effects. Smart hydrogels respond to an externally applied stimulus by delivering therapeutic molecules. While showing promise in releasing molecules in a precise spatial manner, they do not inherently provide temporally dynamic delivery profiles (e.g., controlled timing, rates, and sequences) required to direct sequences of biological events.

Nanoscale materials often exhibit fascinating physical, chemical, and biological properties which are considerably different from those of their macro-and/or microscale counterparts. These unique characteristics have led to considerable growth over the last decade in nanomaterial-enabled applications in areas such as biomedicine, environmental sensing and remediation, and energy production and storage. In this dissertation, techniques are presented to create new nanomaterials relevant to these areas.
