In eukaryotes, gene expression is regulated by epigenetic modifications. Among these modifications, histone H3 lysine 4 (H3K4) methylation has been associated with transcription activation. Set1 is the sole H3K4 histone methyltransferase in Saccharomyces cerevisiae and functions in a multi-subunit protein complex (Set1C) to catalyze H3K4 mono-, di-, and trimethylation. In addition, mis-regulation of Set1 human orthologs such as MLL (1 to 4) or SETD1A/B have been linked to a variety of cancers such as leukemia and early embryonic development defects. Mass Spectrometry has emerged as a powerful analytical tool for the characterization of unknown molecules. Molecular weight information and chemical formulas of the unknowns can be derived by measuring the m/z value of the ionized analyte. In addition, structural information can be obtained via tandem mass spectrometry methods such as collision-activated dissociation (CAD). However, CAD does not always guarantee unambiguously assignment of chemical structures, therefore, additional tandem mass spectrometric methods such as ion-molecule reactions were developed.

Since the early ages of oceanic transportation, ship hulls have been encrusted with foulers including barnacles, oysters, and mussels. With the attachment of these animals, vessels exhibit a decrease in top speed while fuel consumption increases. This not only becomes expensive, but also has contributions to climate change. Current antifouling coatings function by releasing toxic copper into the water, keeping hulls clean at the expense of marine life. This dissertation demonstrates that women authors in the eighteenth century carved out a space for their authority not by overtly opposing their male critics and society’s patriarchal structure, but by rewriting the persona of the gentleman—the poster boy for eighteenth-century society’s moral, masculine, and patriarchal values—and thereby advocating for novels as an important site for cultivating proper masculine behavior as well as a means of renegotiating gender relationships. Soft, carbonate-rich, fine-grained soils are commonly found in the glaciated regions of the northern United States and throughout Canada. In addition to the high compressibility potential and low shear strength, these sediments are typically characterized by alternating layers of silts and clays as well as high calcium carbonate content. Understanding how the size and shape of crop plants and their specific organs are genetically controlled may allow for the development of cultivars with improved plant architecture. A microtubule-severing enzyme called katanin p60 is encoded by KATANIN1 (KTN1) in Arabidopsis or by an ortholog, dwarf and gladius leaf1 (dgl1), in rice. Katanin p60 has been implicated in the control of anisotropic cell growth, which is cell growth directed in a specific direction instead of equally in all directions. Understanding rheology of raw materials and the numerous transformations they undergo is an essential aspect of modeling and predicting extrusion conditions. Both off-line capillary rheometry and in-line extruder fed rheometry techniques have been used to model rheology of materials during extrusion. Investigations on use of an off-line capillary rheometer to model apparent viscosity of full fat soy flours revealed inability of the capillary rheometer to handle food/biological materials high in oil content (20%) at high temperature (80C) and low moisture content (15% wet basis). The cAMP-dependent protein kinase A (PKA) signal transduction pathway plays an important role in morphogenesis and virulence in plant pathogenic fungi. In the rice blast fungus Magnaporthe oryzae, it regulates surface recognition, appressorium turgor generation, and invasive growth. Two genes in M. oryzae named CPKA and CPK2 encode the catalytic subunits of cAMP-dependent protein kinase A. Previous studies have shown that deletion of CPKA failed to block response to exogenous cAMP, suggesting the involvement of CPK2 in cAMP signaling. APC adds polyubiquitin chains to numerous substrates, which leads to their subsequent degradation in late mitosis and G1 phase. The selective and timely recognition and degradation of APC substrates is essential for proper cell cycle progression and maintenance of genome stability. Designing machines that are highly efficient, productive, reliable, and cost affordable represents the target of this research. In this regard, migrating from valve-controlled architectures to displacement-controlled layouts is the proper answer. Obesity is a major public health concern, both in the United States and worldwide. Therefore, identification of measures by which obesity may be prevented and reversed is of high priority. Epidemiological studies consistently demonstrate an inverse relationship between serum 25(OH)D levels, an indicator of vitamin D status, and measures of adiposity. These data suggest that vitamin D may play a role in the prevention of excessive adiposity. This dissertation argues that African worlds and worldviews left an indelible mark on British Enlightenment texts, whether their authors encountered the continent first or secondhand. As eighteenth-century thinkers began experimenting with empirical and categorical strategies for making sense of the globe, Africa was an important testing ground for these burgeoning epistemological methodologies. Travelers and writers tried to reconcile observer-based methods of knowledge production with the continent’s reputedly impenetrable landscapes, unfamiliar cultures, and strange animals. Here, we demonstrate that the loss of function hls1 mutant plants have broad effects on the plant development such as early senescence and flowering, seed insensitivity to ABA as well as plant defense responses including enhanced susceptibility to the necrotrophic pathogen Botrytis cinerea and the hemi-biotrophic pathogen Pseudomonas syringae DC3000 strain expressing the effector protein AvrRpm1 strain. Light in natural and biological media is known as freely diffusing. When light undergoes multiple scattering through inhomogeneous dielectric biomacromolecules, interference is ignored. If scattered waves return to their origin points along the time-reversed paths for constructive interference, outgoing waves can be forbidden, occasionally being on-resonance or off-resonance with Anderson localized modes. However, such a phenomenon in optics requires high-refractive-index contrasts, which intrinsically do not exist in organic molecules (e.g. proteins, lipids, carbohydrates, and nucleic acids). Kinetochores are conserved protein complexes that bind the centromeres in replicated chromosomes to the mitotic spindle and then direct their segregation. To better comprehend Saccharomyces cerevisiae kinetochore function, we investigated the phospho-regulated dynamic interaction between the conserved kinetochore protein Cnn1CENP-T, the centromere region and the Ndc80 complex through the cell cycle. Cnn1 localizes to kinetochores at basal levels from G1 through metaphase but accumulates abruptly at anaphase onset. More than one million tons of polycarbonates and over 500,000 tons of flame retardants are consigned to landfills each year in the form of waste electrical and electronic equipment.
