The kinetic parameters of testicular steroid synthesizing enzymes involved in the Δ4 and Δ5 pathway s for testosterone production were examined in four species. Testis microsomes were prepared as a source of steroidogenic enzymes. Radiolabeled steroid-intermediates were used as substrates and products were separated by thin layer chromatography. Michaelis constants, (k0.5), and Vmax values were determined for 3 B-Hydroxysteroid oxidoreductase (EC 1.1.1.145), 17α-Hydroxylase (EC 1.14.99.9) and C17-C20 Lyase reactions using Δ4 and Δ5 substrates.

Accurate, precise, sensitive and reproducible experimental protocols using high pressure liquid chromatographic assay techniques (HPLC) have of solutes across liquid membranes. A simple model, previously developed by Rhodes and co-workers, used to describe the kinetics of solute transport across liquid membranes was evaluated and some formulation aspects of emulsions for use as liquid membranes were studied.
The removal of solutes from an external aqueous phase by a liquid membrane was found to be influenced by a number of factors including initial solute concentration, pH of the external aqueous phase, and temperature of the system. As the initial concentration of solute …

The purpose of this study was to determine the pharmacokinetics of calcium leucovorin in normal subjects and in patients undergoing High-Dose methotrexame therapy for the treatment of neoplastic disease. By characterizing the disposition of leucovorin it was hoped that the efficacy of the aethotrexate therapy might be improved, while at the same time, the incidence of toxicity could be diminished.
An assay for the separation and quantification of leucovorin in serum was developed. The reduced folate vas extracted from serum using reverse-phase chromatographic minicolumns and a paired-ion reagent. Leucovorin was eluted from this system in methanol and evaporated to dryness …

A Mechanism Of Action Of Gold In Rheumatoid Arthritis. A Biochemical And Morphological Investigation Of The Direct Effect Of Gold Sodium Thiomalate (Gst) On Human Synovial Cells In Culture, Ronald Lee Goldberg

Gold sodium thiomalate (GST) effects on cultured rheumatoid synovial cells were used as a model to investigate a mechanism which can explain clinical benefits gained by patients from gold therapy. Synovial tissue obtained from patients with rheumatoid arthritis who were undergoing reconstructive joint surgery was used to obtain explant culture of synovial cells. The experiments described were performed on growing monolayer cultures during the second to fifth passages. Synovial cells were exposed to GST at pharmacologically relevant concentrations (10-100 uM).
