Design, Synthesis, And Characterization Of Chemical Tools To Study Peroxisomal Import, Jhalak N. Timilsena Jan 2020

Design, Synthesis, And Characterization Of Chemical Tools To Study Peroxisomal Import, Jhalak N. Timilsena

Masters Theses

Peroxisomes are dynamic and interconnected single lipid membrane bound organelles found in the eukaryotic cells which are involved in various biochemical processes including the b-oxidation of very long chain and branched chain fatty acids, metabolism of reactive oxygen and nitrogen species, and reduction of hydrogen peroxide among others. These organelles are known to host numerous proteins and enzymes depending on the cellular environment. All of the proteins needed in the peroxisomes are encoded in the nucleus and synthesized in the cytosol which are then transported to the peroxisomes with the help of a sophisticated protein-transport machinery. Pex5 is one of …

Reprograming Neuronal Cells By Overexpression Of Fibroblast-Specific Transcription Factors, Abdulmohsen Alanazi Jan 2020

Reprograming Neuronal Cells By Overexpression Of Fibroblast-Specific Transcription Factors, Abdulmohsen Alanazi

Masters Theses

In mammals, a complex system of regulatory signals distinguishes tissues, structures and functions. Combinations of transcription factors and co-factors regulate activation and repression of genes that result in cellular differentiation. Whole genome arrays allow the monitoring of genomic expression in specific tissues. Fibroblast microarray studies have shown candidate genes that may be involved in fibroblast identification, including genes that express transcription factors Prrx1, Snai2 and Twist1. A previous study showed that the Prrx1 and Snai2 could reactivate a fibroblast phenotype in hybrid cells that had lost fibroblast identity. Furthermore, overexpression of these factors in liver-derived cells strongly repressed liver gene …

Nest Success In A Grassland Bird Community: Microsite Selection & A Case For Nest Height Manipulation, Matt Kneitel Jan 2020

Nest Success In A Grassland Bird Community: Microsite Selection & A Case For Nest Height Manipulation, Matt Kneitel

Masters Theses

Grassland birds are a sensitive community and populations around the country have been declining. They are susceptible to low nest success due to loss of habitat and the small, fragmented patches that remain are often subjected to elevated levels of nest predation. The American goldfinch (Spinus tristis), which uses grasslands for feeding and nesting, is understudied given its abundance, due in part to its late-season nesting habits, and selection of taller woody vegetation as nesting substrate, which is atypical for many grassland nesting birds. American goldfinches were incorporated in this study which included nest height manipulation of goldfinch …

Seasonal Temperature Changes In Three Midwestern Wastewater Streams And The Effects On Fish Assemblages And Physiology, Ryan William Sparks Jan 2020

Seasonal Temperature Changes In Three Midwestern Wastewater Streams And The Effects On Fish Assemblages And Physiology, Ryan William Sparks

Masters Theses

Midwestern United States undergo extreme temperature and precipitation fluctuations throughout the year, which leads to changing habitats within Illinois river systems. Wastewater treatment facilities have long been used to treat sewage and reduce the organic products in sewage before releasing it into aquatic environments. Wastewater treatment facilities must maintain optimal conditions, especially temperature, for reproduction and growth of facilitated bacteria that break down organic materials in wastewater. The objectives for this study were to determine the thermal effects of wastewater treatment facilities on receiving waters; and secondly, to investigate the influence of these wastewater facilities on fish abundance and assemblages. …

Representations Of Africa In Us Media: A Semiotic Analysis Of Ebola: The Doctor’S Story And Body Team 12, Olubusola Akinpelu Jan 2020

Representations Of Africa In Us Media: A Semiotic Analysis Of Ebola: The Doctor’S Story And Body Team 12, Olubusola Akinpelu

Masters Theses

The Western media has often been criticized for biased coverage of happenings in and around Africa. The 2014 Ebola outbreak created a lot of media attention for West Africa, and by extension, the entire continent. This study examined to what extent postcolonial discourses were infused in two films from the HBO Trilogy: Ebola: The Doctor’s Story (2015) and Body Team 12 (2015). To analyze these texts, I took a semiotic approach by identifying, describing and interpreting the signs in both films, to discuss the postcolonial discourses in three broad themes. Ebola: The Doctor’s Story (2015) was analyzed in the …

This phenomenological study looked at highly-at-risk college seniors who exhibited four out of the eight risk factors defined by the institution where the study took place. The purpose was to gain insight into how highly-at-risk college seniors persist to graduation by discovering what protective factors they utilized in their college career as well as the role resiliency and/or grit may have played. The participants were five undergraduate students who were enrolled at a mid-sized, public university in the rural Midwest. They responded to questions in one semi-structured interview designed to capture their experiences about college starting from high school until …

Aging Techniques & Population Dynamics Of Blue Suckers (Cycleptus Elongatus) In The Lower Wabash River, Dakota S. Radford Jan 2020

Aging Techniques & Population Dynamics Of Blue Suckers (Cycleptus Elongatus) In The Lower Wabash River, Dakota S. Radford

Masters Theses

The Blue Sucker, Cycleptus elongatus, occurs in large rivers primarily in the Mississippi River basin. Their imperiled status has called attention to the need for its management and protection. Estimating age is crucial for directing management, but past studies have varied in their choice of hard structure, resulting in uncertainty regarding the basic life history of this species. Because the Wabash River Blue Sucker population may be one of few surveyable populations with high abundance and successful reproduction, their demographics can provide a benchmark against which threatened populations can be compared. We harvested Blue Suckers (n = 168) …

Seasonal Variation In Mitochondrial Bioenergetics Of The Bluegill Sunfish, Lepomis Macrochirus, From A Shallow Midwest River, Derick Isaac Lamptey Jan 2020

Seasonal Variation In Mitochondrial Bioenergetics Of The Bluegill Sunfish, Lepomis Macrochirus, From A Shallow Midwest River, Derick Isaac Lamptey

Masters Theses

As average global temperature increase, the frequency and magnitude of extreme temperatures in shallow aquatic ecosystems are more ubiquitous. In order to understand how these changing thermal regimes affect aquatic ectotherms, it is essential to develop studies evaluating the response of ectotherms to seasonal fluctuating thermal regimes. Previous studies on fluctuating temperature regimes have reported an increased physiological stress leading to morphological, behavioral and biochemical adaptations. From the latter, the adaptive capacity and seasonal performance associated with optimal function of the oxidative phosphorylation system (OXPHOS) are key for species persistence. However, studies on this matter are scarce. This study explores …

Assessment Of The Immune Response To Posthodiplostomum Minimum Infection In Bluegills, Olamide S. Olayinka Jan 2020

Assessment Of The Immune Response To Posthodiplostomum Minimum Infection In Bluegills, Olamide S. Olayinka

Masters Theses

Bluegills (Lepomis macrochirus) are common intermediate hosts for white grub (Posthodiplostomum minimum). They tolerate heavy infections with minimal effect on condition and continue to accumulate metacercariae as they age. This suggests that any immune response to this parasite might not be effective. This study was conducted to better understand the immune mechanisms underlying P. minimum infection in bluegills.

Infected organs (liver, kidney, and heart) were examined histologically, and serum from infected fish was tested for antibodies to white grub. Juvenile flukes were recovered from isolated metacercarial cysts. Polyclonal antibodies were produced in mice against white grub …

Monitoring Temporal Trends In Catostomidae Larvae In Large Sized River Tributaries, Kellie Nicole Hanser Jan 2020

Monitoring Temporal Trends In Catostomidae Larvae In Large Sized River Tributaries, Kellie Nicole Hanser

Masters Theses

Large river ecosystems can support a complex network of tributaries that provide optimal habitat for fish reproduction. Anthropogenic alteration of rivers and tributaries has been shown to have adverse effects on river fish communities, but few studies have focused on the effects river regulation may have on the reproduction of native suckers (Family Catostomidae). In this study, I evaluated differences in biotic and abiotic factors associated with catostomid reproduction between tributaries of an altered and free-flowing river. I conducted ichthyoplankton sampling using drift nets and larval push nets in tributaries of the Illinois River, which is regulated for navigation, and …

A Pilot Study On The Effects Of Comparative Feedback And Performance On Students’ Self-Efficacy And Self-Esteem, Claire Schneider

Masters Theses

This study compared the effects that academic performance and comparative feedback have on student self-efficacy and self-esteem. Through mobile device polling, participants were able to complete in-class multiple-choice conceptual questions. Participants included 35 college students in two psychology classes assigned to a control and experimental group. For the control group, the class was shown a graph of student responses following each question and the experimental group was not shown how their peers performed. This allowed individuals in the control group to compare their own scores to peer scores while the experimental condition was not provided the comparative feedback. Data were …

Identification And Analysis Of Genes With Differential And Tissue-Specific Expression In Poplars, Bailey Mitchell Jan 2020

Identification And Analysis Of Genes With Differential And Tissue-Specific Expression In Poplars, Bailey Mitchell

Masters Theses

Poplars have become one of the most frequently studied plant species for their applications in biotechnology, commercial benefits, and ecological importance. Microarray analysis is commonly used to analyze differential gene expression in many organisms. Investigations have examined differential gene expression and performed functional analyses on a single tissue or organ within poplar trees or a closely related set of tissues and organs, but usually do not broadly analyze gene expression across a large number of tissues and organs in poplar. This thesis work aims to generate data on the most differentially expressed genes for the major groups of tissues and …

Relationship Between One’S Motive For Curiosity And Meaning In Life, Charles Reither Jan 2020

Relationship Between One’S Motive For Curiosity And Meaning In Life, Charles Reither

Masters Theses

Meaning in life as a psychological construct has many demonstrated benefits for psychological well-being and optimal functioning (Steger, 2013), and the treatment of clinical populations (Thir & Batthyány, 2016). This study investigated in greater detail than is currently available in the psychological literature how meaning in life is related to curiosity. Meaning in life was explored using top-down (the presence of and the search for meaning) and bottom-up (the specific sources of meaning) approaches. Curiosity was examined in its two motivation-based forms: curiosity motivated by the anticipation and enjoyment of discovery (an appetitive interest-type of curiosity) and curiosity motivated by …

Bigheaded Carp Spatial Reproductive Dynamics And Population Genetics In Large River Tributaries, Samuel J. Schaick Jan 2020

Bigheaded Carp Spatial Reproductive Dynamics And Population Genetics In Large River Tributaries, Samuel J. Schaick

Masters Theses

Fisheries managers are devoting considerable effort, time, and funding towards limiting the spread of invasive Bighead and Silver (bigheaded) Carp. A better understanding of factors that influence spatial patterns of bigheaded carp reproduction can help fisheries managers prevent their spread. To examine factors that influence spatial patterns of reproduction, we sampled ichthyoplankton using drift nets and larval push nets in three tributaries of the Illinois and Wabash Rivers March-September 2016-2018. We compared relative abundances of bigheaded carp eggs and larvae among tributaries using catch per unit effort (CPUE). Abiotic data were analyzed from individual study tributaries and pooled among tributaries …

A Cloning Strategy For Expression Of Fungal Hemicellulases In Plant Systems, Amer Alrudayan Jan 2020

A Cloning Strategy For Expression Of Fungal Hemicellulases In Plant Systems, Amer Alrudayan

Masters Theses

Lignocellulosic biomass is notoriously difficult to deconstruct into lignin, hemicellulose, and cellulose fractions without the use of biochemical, chemical, and/or mechanical pretreatments. These pretreatment strategies add operational cost and in some cases produce hazardous waste that poses health and safety risks. Direct biological pretreatment from biomass-degrading bacteria and fungi may significantly reduce the costs and risks associated with these traditional strategies due to the innate ability of these organisms to modify plant cell walls. An additional measure that could further reduce the expenses associated with lignocellulosic biomass fractionation is genetic modification of the lignocellulosic feedstocks, which can be accomplished by …

The Transcriptome Response Of The White-Rot Fungus Trametes Versicolor To Wild-Type And Lignin-Modified Hybrid Poplar, Anbarah Alzabaidi Jan 2020

The Transcriptome Response Of The White-Rot Fungus Trametes Versicolor To Wild-Type And Lignin-Modified Hybrid Poplar, Anbarah Alzabaidi

Masters Theses

Plant biomass is a renewable and sustainable feedstock for biofuel production that can reduce societal dependence on fossil fuels. However, the production of liquid biofuels from the non-starch (i.e. lignocellulosic) material through fermentation technology is limited due to the complexity of the cell wall structure. This necessitates the use of chemical, thermal, and/or mechanical pretreatment technologies, which adds significant capital, operational, and environmental costs. Biological pretreatment strategies have the potential to mitigate these expenses by harnessing the innate ability of specialized bacteria and fungi to deconstruct lignocellulose. White-rot fungi (e.g. Trametes versicolor) have been shown to be …

Effects Of Insect Opportunists On A Four-Level Trophobiotic System Involving Nectar-Producing Galls Of The Cynipid Wasp Disholcaspis Quercusmamma (Hymenoptera: Cynipidae), Stephanie L. Smith Jan 2020

Effects Of Insect Opportunists On A Four-Level Trophobiotic System Involving Nectar-Producing Galls Of The Cynipid Wasp Disholcaspis Quercusmamma (Hymenoptera: Cynipidae), Stephanie L. Smith

Masters Theses

The induction of plant galls is considered an adaptive life history trait found in many insect groups. The formation of galls provides several advantages to the gall maker, such as enhanced nutrition, favorable microclimate, and protection from natural enemies, including parasitoids, inquilines, and predators. Order Hymenoptera has many gall-making species, belonging to the gall wasp family Cynipidae. As an extended phenotype of the gall makers, some galls exhibit very sophisticated adaptive mechanisms involving multilevel species interactions. In particular, the oak galls of the Cynipid species Disholcaspis quercusmamma, found in much of Illinois, produce a palatable, sugary nectar-like secretion, attracting …

Construct Validity Of The Teate Depression Inventory: Convergent And Discriminant Validity And Equivalence For Black/African American And White/Caucasian Samples, Dylin Coons Jan 2020

Construct Validity Of The Teate Depression Inventory: Convergent And Discriminant Validity And Equivalence For Black/African American And White/Caucasian Samples, Dylin Coons

Masters Theses

The purpose of this study was to replicate the construct validity of a new measure of depression, the Teate Depression Inventory (TDI; Balsamo & Saggino, 2013), with the primary focus on Black/African American participants. Research has purported that Black/African Americans experience inequality in obtaining mental health care for internalizing disorders. This may partially be caused by errors in diagnosing these individuals with symptoms. Correctly diagnosing internalizing disorders is a critical step in obtaining appropriate treatment. More research on depression and anxiety is needed to enhance mental health practices by addressing the need for professionals to be culturally competent and conscious …

An Intricate Ensemble : The Art-Science Of An Ecological Imaginary For The Anthropocene Epoch, Alyce Santoro Dec 2019

An Intricate Ensemble : The Art-Science Of An Ecological Imaginary For The Anthropocene Epoch, Alyce Santoro

Masters Theses

The contradictions inherent in European Enlightenment-based “logics” that externalize humans from “nature” were a concern for the Romantic Naturalists, Dadaists, and Surrealists. More recently, some in the environmental humanities and socio-ecologically-concerned arts and sciences have also posed challenges to anthropocentric, hierarchical, positivist modes of thought. I suggest that by engaging the ludic, imaginative, and collaborative while bearing the empirical in mind, dualisms (such as objective and subjective, individual and collective) dissipate, and existence as a dialectical state of intricate ensemble can be revealed. In light of catastrophic disruption to Earth’s life-sustaining processes by exploitative forms of human activity, I argue …

Participants self-reported ratings of perceived pleasantness, as well as Electromyography (EMG) and Electrodermal Activity (EDA) data were collected. Average Skin Conductance Responses (SCR) were averaged over all trials …

Supporting Black Mothers At The Transition: A Collaborative Design Prenatal Coparenting Program, Jordan Ebanaya Dec 2019

Supporting Black Mothers At The Transition: A Collaborative Design Prenatal Coparenting Program, Jordan Ebanaya

Masters Theses

Despite having the highest infant mortality rate of similarly developed nations, infant mortality rates have trended downward in the United States, however, the infant mortality disparity between Black and White infants persists. The causes of the disparity have been traced to social considerations of the mother especially while pregnant. These social situations include racism and discrimination, marital status of the mother, social support and pregnancy intentions, pointing to one commonality: stress. Using Boss and colleagues 2017 Conceptual Model of Family Stress, this study proposes a solution to address some of the social support, or resources, available to the expectant mother; …

Intracellular Electrometer, Lucas M. Essenburg Dec 2019

Intracellular Electrometer, Lucas M. Essenburg

Masters Theses

The Western Michigan University Neurobiology Engineering Laboratory conducts re- search in energy-efficient electrical stimulation of biological neurons. Experimental work re- quires an intracellular electrometer to inject nanoamp-level currents into a biological neuron and to measure its membrane voltage response. In order to fully understand the generation of such minute currents, an `in-house' electrometer was designed, constructed, and validated. This required exploration of various design trade-offs including precision of generated nanoamp-level currents vs. susceptibility to noise, portability vs. functionality, and cost vs. functionality. The electrometer was successfully used to stimulate and measure responses of neurons from the medicinal leech Hirudo verbana …

Pcr Analysis On The Effects Of Alpha Synuclein Treatment On C6 Rat Glioma Cells, Michael John Helmus Dec 2019

Pcr Analysis On The Effects Of Alpha Synuclein Treatment On C6 Rat Glioma Cells, Michael John Helmus

Masters Theses

Multiple System Atrophy (MSA) is a rare neurological disease that mainly implicates the neuronal protein, alpha synuclein, as being centrally involved in MSA pathology. Despite synucleinopathy dogma, little is known about alpha synucleins role in MSA pathogenesis. The Ide laboratory previously conducted an Affymetrix gene expression study utilizing the post-mortem pons tissue from eight individuals with MSA and five individuals without any known neurological disorders. One conclusion from that study was that downregulation of numerous mitochondrial homeostasis related genes had occurred. It is hypothesized that alpha synuclein can perturb mitochondrial homeostasis through direct interaction with mitochondria. This study utilized Reverse …

An Evaluation Of Unmanned Aerial Systems And Structure-From-Motion For Fluvial Large Wood Sensing And Risk Assessment, Daniel Gerke Dec 2019

An Evaluation Of Unmanned Aerial Systems And Structure-From-Motion For Fluvial Large Wood Sensing And Risk Assessment, Daniel Gerke

Masters Theses

This research aims to show Unmanned Aerial System (UAS) and Structure-from-Motion (SfM) technology can, in combination, improve on traditional large wood (LW) monitoring techniques. More temporally and economically efficient data collected at a finer spatial resolution and greater spatial extent will increase the effectiveness of management plans and risk assessment for LW by providing decision-makers with a complete picture of the river.

Contemporary practices are too inefficient in time and labor for large-scale monitoring of fluvial LW with anything more than the most general management or risk assessment in mind. The paradigm of river research, the river continuum concept (RCC), …

The Implementation Of Qpcr Beach Monitoring Methods: Analysis Of A Multi-Lab Validation Study And The Role Of Environmental Parameters On A Comparison Of Colilert And Qpcr Methods, Molly J. Lane Dec 2019

The Implementation Of Qpcr Beach Monitoring Methods: Analysis Of A Multi-Lab Validation Study And The Role Of Environmental Parameters On A Comparison Of Colilert And Qpcr Methods, Molly J. Lane

Masters Theses

Public beaches are routinely tested for potentially pathogenic bacteria to protect beachgoers from possible illness. An EPA approved method, Colilert™, used for testing E. coli in recreational water requires 18 – 22 hours before a result is reported but, recreators have already contacted unsafe water before the beach is closed. My study focused on a U.S. EPA proposed qPCR method (Draft Method C) to quantify E. coli in recreational waters that can provide same-day results. In Chapter 2, I examined the calibration procedure used to validate Draft Method C and compared standard curve intercept and slope estimates calculated with a …

Snowshoe Hare (Lepus Americanus) Abundance In Relation To Habitat And Predator Assemblage Across The Apostle Islands, Jarod Reibel

Masters Theses

Snowshoe hares (Lepus americanus) are a climate sensitive species that have a southern range boundary moving northward. Snowshoe hares are found on the Apostle Islands, Wisconsin which are near their southern boundary and differ by island in vegetative and carnivore communities. The archipelago serves as a natural laboratory to assess how top-down and bottomup forces interact and impact snowshoe hare populations. The objectives of this study were to determine the influence of vegetative characteristics, specifically visual obstruction, and the presence of predators on snowshoe hare abundances across the Apostle Islands. We conducted fecal pellet surveys to estimate hare abundance, measured …

Habitat Use Of The Climate-Sensitive Snowshoe Hare (Lepus Americanus) In The Manistee National Forest In Michigan’S Lower Peninsula, Spencer D. West Dec 2019

Habitat Use Of The Climate-Sensitive Snowshoe Hare (Lepus Americanus) In The Manistee National Forest In Michigan’S Lower Peninsula, Spencer D. West

Masters Theses

Snowshoe hares (Lepus americanus) are a wide-ranging lagomorph that are important forest herbivores and a popular game species throughout their range. Across the southern boundary of their geographic range, snowshoe hares are experiencing population declines and possible extirpation due to increased predation pressure driven by climate change induced camouflage mismatch, competition for forage, degraded and fragmented habitat. One method of reversing the negative trends in snowshoe hare distribution is to increase and improve available hare habitat. A specific habitat analysis for local regions will most effectively advise managers how to target habitat management. I radio-collared 11 snowshoe hares in the …

Impacts Of Shoreline Restoration And Source Of Nutrient Enrichment On Macrophytes And Epiphytic Algal Communities, Paige Marie Kleindl Dec 2019

Impacts Of Shoreline Restoration And Source Of Nutrient Enrichment On Macrophytes And Epiphytic Algal Communities, Paige Marie Kleindl

Masters Theses

Macrophytes and their epiphytic algal communities are integral for optimizing littoral ecosystem functioning in lakes. Epiphytic algae’s placement on the plant’s surface can reduce light and nutrient availability (i.e., nitrogen and phosphorus) for the host macrophyte. Macrophyte and epiphytic algal proximity complicates these primary producer group interactions and responses to bioavailable nutrients in the water column or porewater. For example, epiphytic algae may have a competitive advantage over surface water nutrients compared to macrophytes, which may have a competitive advantage over porewater nutrients via root systems.

Muskegon Lake’s industrial history and designation as an Area of Concern prompted shoreline restoration, …

Inequality, Patronage, Ethnic Politics And Decentralization In Kenya And Botswana: An Analysis Of Factors That Increase The Likelihood Of Ethnic Conflict, Rei Gordon Aug 2019

Inequality, Patronage, Ethnic Politics And Decentralization In Kenya And Botswana: An Analysis Of Factors That Increase The Likelihood Of Ethnic Conflict, Rei Gordon

Masters Theses

Scholars have recommended numerous institutional arrangements for mitigating ethnic conflict in divided societies. Electoral systems are often considered to have an impact on ethnic conflict, and scholars have recommended both proportional representation systems and majoritarian systems for their respective effects on mitigating ethnic tensions. However, in a cross-national analysis of 18 sub-Saharan democracies, I find no impact of electoral systems on ethnic conflict. Countries employing proportional representation systems and majoritarian systems are compared according to three measures of ethnic conflict, yet neither electoral system correlates with higher or lower levels of conflict. In the interest of identifying factors that do …
