Talker-Specific Influences On Phonetic Boundaries And Internal Category Structure, Janice Ann Lomibao
Talker-Specific Influences On Phonetic Boundaries And Internal Category Structure, Janice Ann Lomibao
Master's Theses
Recent findings indicate that listeners are sensitive to talker differences in phonetic properties of speech, including voice-onset-time (VOT) in word-initial stop consonants. The current work extends our earlier research by examining the degree to which listeners adjust the initial mapping from acoustic signal to segmental representation on a talker-specific basis. Two groups of listeners are exposed to a talker producing “cane.” Word-initial VOTs are manipulated such that one group hears “cane” produced with short VOTs and the other group hears “cane” produced with relatively longer VOTs. Following training, listeners’ voicing boundary for a /g/-/k/ continuum was tested. In addition, listeners …
Escherichia Coli Strain Diversity In Humans: Effects Of Sampling Effort And Methodology, Emily R. Neal
Escherichia Coli Strain Diversity In Humans: Effects Of Sampling Effort And Methodology, Emily R. Neal
Master's Theses
Studies investigating Escherichia coli strain diversity and demographics in human hosts are frequently inconsistent regarding sampling effort and methodology while current strain typing methods are often expensive or laborious. To rectify these inconsistencies, sampling effort was investigated by comparing the diversity of 15-isolate collections to 100-isolate collections from 3 human subjects. Temporal variation in E. coli strain diversity was also studied by collecting 15 isolates once every 6 months. Additionally, strain identification and diversity collected by different sampling methods (fecal swabs vs. anal swabs collected at different times around defecation) were compared to identify any inherent biases in sampling method. …
Evaluating Usability Evaluations, Allen Leo Dunlea
Evaluating Usability Evaluations, Allen Leo Dunlea
Master's Theses
We live in an age when consumers can now shop and browse the web using hand-held devices. This means that competitive companies need to have a website to represent their brand and to conduct business. E-commerce sites need to pay special attention to the usability of their sites, since it has such an impact on how potential costumers view their brand.
Jakob Nielsen defines usability as a "quality attribute that assesses how easy user interfaces are to use"; he separates usability into five quality components: learnability, efficiency, memorability, errors and satisfaction. The current standard for testing usability involves having …
Dna Purification Process Optimization At Life Technologies Corporation, Trevor J. Shepherd
Dna Purification Process Optimization At Life Technologies Corporation, Trevor J. Shepherd
Master's Theses
This project focused on optimizing the plasmid DNA purification process at Life Technologies. These plasmids are designed to code for specialized proteins used by research universities, national laboratories, or research companies. Once cultivated and harvested, the plasmids must be analyzed for quality and quantity. The project is divided into improving three aspects of the process: 1) plasmid identification, 2) plasmid purity evaluation, and 3) process yield. Plasmid identification is now simpler, more robust and has zero ambiguity. Plasmid purity evaluation is now measured with computer software, which reduces user error and eliminates subjectivity. Using the nascent metrics provided by the …

Cplop: The Cal Poly Library Of Pyroprints, Jan Lorenz Villaceran Soliman
Master's Theses
To date, microbial source tracking (MST), i.e. determining the source of microbial contamination based on the specific strains observed in the environment, often uses methods that are time-consuming, expensive and not always reliable. The biology department at Cal Poly, San Luis Obispo has developed a new method for MST called pyroprinting. Pyroprints can be used as DNA fingerprints for identifying sources of fecal contamination and studying bacterial populations in host animals. The MST method consists of two parts: the pyroprinting process and a database of pyroprints. The Cal Poly Library of Pyroprints (CPLOP) was developed to provide a database application …
Computational Modeling To Evaluate Helical Electrode Designs For Use In Vagus Nerve Stimulation, Anthony W. Cowley
Computational Modeling To Evaluate Helical Electrode Designs For Use In Vagus Nerve Stimulation, Anthony W. Cowley
Master's Theses
An estimated 0.5% of world’s population has been diagnosed with epilepsy. Of these patients 20-30% will be unable to achieve seizure control with anti-epileptic drugs. Vagus nerve stimulation (VNS) may be an appropriate treatment option for some patients with pharmaceutically refractory, partial-onset seizures.
VNS therapy uses a helical electrode to interface between the implantable pulse generator and the vagus nerve. While there have been several studies related to the mechanical and electrical safety of such electrodes, little work has been done toward understanding the effectiveness of the helical electrode in nerve stimulation. A better understanding of the voltage field and …
Latent Cysteine Residues From Polymers Prepared Via Free And Controlled Radical Polymerizations, Douglas Vincent Amato
Latent Cysteine Residues From Polymers Prepared Via Free And Controlled Radical Polymerizations, Douglas Vincent Amato
Master's Theses
One less commonly used “click” reaction is thiazolidine chemistry. Thiazolidine chemistry is a commonly used reaction used in biological systems because the reaction requires the presence of both cysteine (a common amino acid) and an aldehyde or ketone. If cysteine residues could be incorporated into a polymer then a variety of applications could be developed. Polymers containing free thiols (aka thiomers) have developed in the last decade to become great mucoadhesives. If there was a facile route to control the amount of free thiols along the polymer then more fine-tuned and potentially stronger adhesives could be made. For these reasons …
Testing Subspecies Limits In Monardella Villosa, Taylor Mclaughlin Crow
Testing Subspecies Limits In Monardella Villosa, Taylor Mclaughlin Crow
Master's Theses
Several subspecies of Monardella villosa (California Coyote Mint) have been distinguished on the basis of leaf thickness, shape, and trichome characteristics, yet many intermediates are known. We investigated morphological differences in natural populations of two subspecies (M. v. subsp. villosa and M. v. subsp. franciscana) in the Scott Creek watershed, north of Davenport, in Santa Cruz County, CA. Monardella villosa subsp. franciscana grows in coastal scrub in gulches and ocean terraces whereas subsp. villosa grows at more protected inland sites. Morphological difference between subspecies may be adaptations to these different habitats. I grew plants in a common garden at Cal …
Dairy Manure Flushwater Treatment By Packed-Bed Anaerobic Digesters, Neal Cary Adler
Dairy Manure Flushwater Treatment By Packed-Bed Anaerobic Digesters, Neal Cary Adler
Master's Theses
Wastewater treatment performance of three pilot-scale packed-bed anaerobic digesters with walnut shell medium was researched for treating dairy freestall barn flushwater. Reciprocation mixing was evaluated as a means to lessen channelization in the media bed and to improve biogas production and organic matter removal at ambient temperatures. Reciprocation has been used in biological nitrogen removal systems to introduce air into the system to repeatedly oxygenate nitrifying biofilm along with mixing (Behrends et al. 2003), but the anaerobic systems benefit from mixing. Two tanks were used in each system, where one was full and one was empty at any given time. …

Establishment And Validation Of A Novel Fucci (Fluorescent Ubiquitination-Based Cell Cycle Indicator) In An In Vitro Chronic Myeloid Leukemia Model, Jane Marie Isquith
Master's Theses
The deregulation of the cell cycle is important in the leukemic transformation and in the continued discovery of more efficient targeted therapies. After elucidation of the role of the RNA editor ADAR1 in the malignant reprogramming of Chronic Myeloid Leukemia (CML) progenitors to leukemia stem cells (LSCs) during the chronic phase to blast crisis stages of the disease, current research seeks to uncover the mechanism of action of ADAR1 in this process. To study the potential role ADAR1 plays in the changes to cell cycle during this disease progression, a novel Fucci (fluorescent ubiquitination-based cell cycle indicator) system was introduced …
Shark Sim: A Procedural Method Of Animating Leopard Sharks Based On Raw Location Data, Katherine S. Blizard
Shark Sim: A Procedural Method Of Animating Leopard Sharks Based On Raw Location Data, Katherine S. Blizard
Master's Theses
Fish such as the Leopard Shark (Triakis semifasciata) can be tagged on their fin, released back into the wild, and their location tracked though technologies such as autonomous robots. Timestamped location data about their target is stored. We present a way to procedurally generate an animated simulation of T. semifasciata using only these timestamped location points.
This simulation utilizes several components. Input timestamps dictate a monotonic time-space curve mapping the simulation clock to the space curve. The space curve connects all the location points as a spline without any sharp folds that are too implausible for shark traversal. We create …
Calcium Carbonate Particle Size Effects On Titanium Dioxide Light Scattering In Coatings, Jason W. Boke
Calcium Carbonate Particle Size Effects On Titanium Dioxide Light Scattering In Coatings, Jason W. Boke
Master's Theses
Titanium dioxide (TiO2) is an essential component in a white paint formulation due to its excellent light scattering ability. TiO2 also happens to make up most of the raw materials cost. Viable replacements to TiO2 do not currently exist in a feasible manner. Rising costs in the recent TiO2 supply shortage have forced coatings formulators to find a balance between cost and performance. One method includes partial substitution of TiO2 with cheaper material such as calcium carbonate (CaCO3), which serves to occupy volume.
The purpose of this study was to compare the coating spread rate of paints with varying TiO2 …

Algorithms For Library-Based Microbial Source Tracking, Aldrin Montana
Master's Theses
Pyroprinting is a novel, library-based microbial source tracking method developed by the Biology department at Cal Poly, San Luis Obispo. This method consists of two parts: (1) a collection of bacterial fingerprints, called pyroprints, from known host species, and (2) a method for pyroprint comparison. Currently, Cal Poly Library of Pyroprints (CPLOP), a web-based database application, provides storage and analysis of over $10000$ pyroprints. This number is quickly growing as students and researchers continue to use pyroprinting for research. Biologists conducting research using pyroprinting rely on methods for partitioning collected bacterial isolates into bacterial strains. Clustering algorithms are commonly used …

Master's Theses
Cardiovascular disease is the leading cause of death in developing countries. Because thousands of people are affected by this disease, medical device companies are constantly developing new intravascular devices in attempt to cure or alleviate the symptoms of the disease. However, before these devices can be brought to market or implanted in to patients, they must complete three stages of testing to receive FDA approval. These testing stages include: in vitro testing, in vivo testing, and clinical trials. Currently, there is a large gap between in vitro and in vivo testing because it is difficult to obtain physiologic information about …

Assessing Physiological Thresholds For Eelgrass (Zostera Marina L.) Survival In The Face Of Climate Change, Carolyn Jane Ewers
Master's Theses
Seagrasses are well known for the important ecological roles they play in coastal marine waters worldwide. However, the severe rate of decline observed in seagrasses this century is expected to accelerate with climate change. Conservation efforts can be improved by quantifying physiological thresholds of seagrasses and using these estimates in modeling to forecast changes in distribution. This study examines the response of eelgrass (Zostera marina L.) across current temperatures to look for early warning signs of vulnerability and to evaluate the ways we determine critical thresholds for survival. Whole eelgrass ramets, collected from three beds in Morro Bay, California, …

Comparison Of Biofilm Media In Reciprocating Biofilters Treating Dairy Flushwater, Kyle Keoki Tatsuo Fooks
Comparison Of Biofilm Media In Reciprocating Biofilters Treating Dairy Flushwater, Kyle Keoki Tatsuo Fooks
Master's Theses
Reciprocating biofilters known as ReCip is a viable technology to manage nutrients, mainly nitrogen, problems at livestock operations such as swine farms and dairies. Past studies have demonstrated that ReCip is more adept at total nitrogen (TN) removal than traditional subsurface flow wetland systems. The traditional substrate used to attach biomass was rock aggregate; this media may be hard to obtain for some agricultural projects, so alternate substrates are tested and compared with the rock aggregate. The purpose of the study was twofold: first, different biofilm media were tested and compared in terms of treatment performance and, second, the long-term …

We studied root kinematics in relation to temperature and genome size variation in teosinte (Zea mays subspecies parviglumis) and corn (Zea mays subspecies mays). Corn had significantly faster radicle growth than teosinte when grown at a constant temperature. Both species exhibited variation in seed size and for each species larger seeds had faster root growth. Genome size was not significantly correlated with faster radicle growth rates across multiple land races of corn. To examine temperature dependent growth in corn and teosinte, a germinated seedling was grown at multiple temperatures. Growth rates at these temperatures were used …
