Listeria monocytogenes continues to challenge the dairy industry in causing post-process contamination of cheeses. To reduce risk of contamination, it is crucial to understand the growth and survival of pathogenic bacteria in cheese products and to develop post-process mitigation strategies. This study evaluated the fate of pathogens in reduced and regular sodium Mozzarella cheese, and the potential of Supercritical Fluid Extraction with CO2 (SFE) to reduce Listeria innocua on Mozzarella and Queso Fresco. The goal of this project was to determine whether i) fused deposition modeling could be employed to manufacture molds for vascular constructs, ii) whether vascular constructs could be created from these molds, and iii) to verify practical equivalence between observed fluid velocities. Dye tracking was to be employed to characterize fluid velocity profiles through the in vitro vascular constructs, including a half-vessel model and a full vessel model of an iliac artery aneurysm. Thermotoga maritima is the representative member of the order Thermotogales, which is comprised of extremely thermophilic anaerobic bacteria. All members of this order are characterized by the presence of a unique outer envelope, the toga. The outer envelope appears as polar cellular distensions that sometimes are as large as the cytoplasm. The outer envelope increases in size while the cytoplasm remains the same leading to an outer envelope distension. A remarkable amount of information is conveyed by the human voice. For example, the emotional state of a speaker is conveyed by vocal cues such as pitch and intensity, though as is true for other speech qualities, affect does not map onto auditory signals in a one-to-one fashion. Despite the widespread use of cell phone technology, there is still little information regarding how emotional states are conveyed during cell phone transmissions. Objective: Dysmenorrhea is an incapacitating type of visceral pain that affects numerous menstruating women and is defined by painful menstrual cramps and often occurs in the absence of pelvic pathology. Women with dysmenorrhea have reported higher sensitivity to experimental pain, and vulnerability to pain sensitivity may be a primary risk factor for developing chronic pelvic pain for women with dysmenorrhea. Lipoate is an essential cofactor of several proteins involved in cellular energy homeostasis and catabolism. Lipoate metabolism has been linked to pathogenesis in some microbial species, but its role in Staphylococcus aureus infections had not been explored. In this thesis, we tested the hypothesis that lipoate acquisition mechanisms promote S. aureus infectivity. We used a bacterial genetics approach to elucidate the biological function of the S. aureus genes involved in lipoate metabolism. I demonstrate that a member of the protein family A Disintegrin and Metalloprotease (ADAM), ADAM23, is expressed in the olfactory epithelium (OE) and may inhibit neural progenitor cell (NPC) proliferation. I generated a list of ten gene candidates and selected ADAM23 for further study, based on its conserved protein structure and accumulating evidence for its role in cell cycle progression of proliferative cells. I next determined ADAM23 protein localization using immunohistochemistry and measured both mRNA and protein expression post-lesion with RT-qPCR and Western blot. Our knowledge of avian Haemosporidian parasites has increased dramatically with the advent and increased used of molecular technologies. Yet Leucocytozoon, a common avian parasite, remains relatively understudied. Haemosporidian parasite diversity and prevalence has been shown to be dependent on several factors, including host family and habitat, on a microecological scale. In this study I screened 3,375 birds from sub-Saharan Africa for the presence of Leucocytozoon and demonstrate that the patterns of prevalence and diversity continue at a macroecological scale. UTIs can cause complications in pregnancy and decrease quality of life, and their treatment and prevention are expensive. Uropathogenic E. coli (UPEC) is the primary cause of UTI. Overland transport of recreational boats is the major vector of dispersal for aquatic invasive species among North American waterbodies. These species have large economic and ecosystem impacts, and understanding their movement is essential to understanding the biodiversity and functioning of aquatic ecosystems across the continent. I surveyed Illinois boaters to determine patterns of recreational boat movement and the consistency with which boaters perform actions to prevent the spread of invasive species. Hepatocellular Carcinoma is the second most lethal cancer after pancreatic cancer. Unresectable HCC tumors carry a poor prognosis and few treatment options are available. The dismal prognosis is mainly due to limited therapy options and molecularly targeted therapy is deemed as solution. Here, we report a novel role of c-Abl in HCC development. We provide evidence of c-Abl activation in human HCC samples compared to normal liver. Estrogens are an important class of steroid hormones involved in many cellular signaling processes. Levels of estrogens fluctuate across the lifespan suggesting that they target intracellular mediators, such as microRNAs, to fine-tune homeostatic cellular processes. Previous work in our lab demonstrated that 17β-estradiol (E2), the predominant circulating estrogen, stabilized miR-181a in a neuronal cell line derived from rat hypothalamus. These results lead to the hypothesis that E2 regulated miR-181a stability by altering the pool of endogenous RNA and by recruiting PNRC2 away from the miRNA degradation machinery. This study characterizes binary-metal mixture effects of cadmium (Cd2+) + zinc (Zn2+) and Cd2+ + nickel (Ni2+) on Daphnia magna. Although acute studies have shown protective Ni2+ (Traudt et al. 2016) and Zn2+ effects (Meyer et al. 2015) against Cd2+ toxicity, no study has fully characterized a protective effect on D. magna at several endpoints (survival, reproduction, growth, and accumulation) on a chronic study. During embryogenesis, primordial germ cells (PGCs) and somatic gonadal precursor cells (SGPs) migrate and coalesce to form the early gonad. A failure of SGPs and PGCs to form a gonad with the proper architecture not only affects germ cell development, but also can result in infertility. We have characterized the phenotypes of two genes, longitudinals lacking (lola) and ribbon (rib), required for the coalescence and compaction of the embryonic gonad in Drosophila melanogaster. Urban, minority males are disproportionately impacted by community violence exposure (ECV). However, person-based analyses have demonstrated variability in rates of ECV, suggesting that demographic risk factors do not always result in increased ECV, and it may be important to examine the utility of psychological factors in this relationship. Research suggests that depressive symptoms may actually exacerbate the risk of ECV. The current study examines the effect of internalizing symptoms on future ECV. Streams along the Copper River Delta, southcentral Alaska, exhibit contrasting thermal and hydrologic variability associated with being primarily groundwater-fed (GWF) or surface water-fed (SWF). Groundwater-fed streams are predictable both thermally and hydrologically year round, whereas SWF streams are unpredictable and exhibit more variable thermal and hydrologic regimes. These differences may strongly influence aquatic insect community structure and secondary production. Four streams, two GWF and two SWF, were sampled twice monthly from late April 2013 through August 2013 and once seasonally in fall (September) and early winter (November). Shewanellaceae are among the most widely studied electroactive microorganisms. This study reports on Shewanella putrefaciens under anaerobic conditions during applied potential while monitored by spectroscopy. The attachment of the microbe Shewanella putrefaciens onto an indium tin oxide electrode was found to occur after 2 hr of +0.2 V applied potential. Microbial detachment occurred with a 5 mV/s scan rate cyclic voltammogram from +0.2 V to -0.5 V to +0.2 V. This was confirmed through use of a spectroelectrochemical apparatus utilizing a standard 3 electrode setup with evanescent wave spectroscopy.

Biofilms are surface-associated microbial communities surrounded by an extracellular matrix. Through biofilm formation, many pathogens and symbionts are able to colonize, survive, and persist in their host. A model system used to study biofilm formation is the symbiotic bacterium Vibrio fischeri, which colonizes its host, the squid Euprymna scolopes. Complex signaling between the squid and the bacteria is essential for the proper regulation of biofilm formation as well as for persistent colonization.
