Optimization Of A Gfp Biosensor For The Discovery Of Novel Antibiotics, Drake Heinz
Optimization Of A Gfp Biosensor For The Discovery Of Novel Antibiotics, Drake Heinz
Master's Theses
In today’s world, there is an urgent need to discover novel antibiotics due to the rise of antibiotic resistance and the lack of new ones being approved. Building off of Hanson et al. and their reduction-oxidation sensitive green fluorescent protein (roGFP)(1), that we yeast codon optimized (2); I wanted to determine if I could create a real-time biosensor that has the ability to respond to ROS produced in the stress response triggered by antibiotics as a platform to identify novel antibiotics. To achieve this, I need to know if it will oligomerize in bacterial cytoplasm due to its two cysteines …

The Role Of Gibberellic Acid In Aphid-Plant-Arbuscular Mycorrhizal Fungus Interactions, Drew Olson
The Role Of Gibberellic Acid In Aphid-Plant-Arbuscular Mycorrhizal Fungus Interactions, Drew Olson
Master's Theses
Arbuscular mycorrhizal (AM) fungi form beneficial associations with the roots of most terrestrial plants, which is mostly characterized by increased plant nutrient acquisition (phosphate and nitrogen) by the fungi in exchange for carbon and lipids from the plants. These associations can also enhance plant coping mechanisms to abiotic and biotic stresses. However, insect herbivores, such as aphids, can benefit from AM fungi-plant symbioses by altering plant defenses that are driven mainly by the phytohormones jasmonic acid (JA) and salicylic acid (SA). Nevertheless, other phytohormones can modulate plant defenses by altering the balance of the JA and SA signaling pathways, such …

Pismo Clams (Tivela Stultorum) In Califorina: Population Status, Habitat Associations, Reproduction, And Growth, Alexandria R. Marquardt
Master's Theses
Marine shellfish play a vital role in intertidal ecosystems and coastal communities, but many of these fisheries are small-scale and lack the necessary monitoring to ensure long-term sustainability. Effective management often requires information on key demographic parameters, such as population status, reproduction and growth. Pismo clams (Tivela stultorum) are a culturally important and iconic species in California, which supported a thriving commercial and recreational fishery throughout much of the 1900’s. However, Pismo clam populations have declined statewide in recent decades and are attributed to human harvest and predation by California sea otters (Enhydra lutris); However, no …

Valorization Of Carrot Processing Waste, Alexandra M. Duval
Valorization Of Carrot Processing Waste, Alexandra M. Duval
Master's Theses
Commercial carrot processors produce up to 175,000 tons of carrot waste annually. Carrot Mash (CM) is the term referring to the waste by-product of peeled baby carrot processing. Transportation of carrot processing waste is expensive due to its high- water content (approx. 83-95%). High in bioactive compounds (carotenoids) and dietary fibers, it is expected that its conversion into a value-added by-product is of interest to the carrot processing industry. Hemicellulose-rich plant materials have proven to be a source of oligosaccharides, which are known for their beneficial prebiotic activity. The objectives of this research were to: 1) determine the effect of …

Methods To Increase Algae Biomass Productivity In Raceway Pond Monocultures, Ryan S. Anderson
Master's Theses
The economics of algae biofuels and bioproducts would be improved by increased biomass productivity. Two studies on this potential are described in this thesis – one on a locally isolated filamentous yellow-green alga and the other on a planktonic strain genetically improved via selective enrichment.
Polycultures have been viewed as productive, stable, and, in some cases, harvestable by natural bioflocculation. Local native strains might have higher productivity than culture collection strains because they are already adapted to local outdoor conditions. In this study, the filamentous yellow-green alga Tribonema minus was isolated from a local volunteer polyculture. Its productivity as a …

The aims of this research were to 1) determine the prevalence of disordered eating risk in NCAA Division I student-athletes and 2) determine if such behaviors are related to baseline …
Expression And Purification Of Snake Antivenom Peptide In Pichia Pastoris, Israel Juarez Contreras
Expression And Purification Of Snake Antivenom Peptide In Pichia Pastoris, Israel Juarez Contreras
Master's Theses
It has been shown that a peptide of the first 10-15 N-terminal amino acids of the lethal toxin neutralizing factor (LTNF) protein found in opossums (Didelphis spp.) holds promise as a low-cost therapy for snake envenomation. To date, the 11-mer has been expressed in E. coli and shown to neutralize snake venoms. However, possible endotoxin concerns warrant an investigation into other microbial hosts. The methylotrophic yeast, Pichia pastoris, shows promise as an alternative host. Active LTNF peptide was expressed and purified in P. pastoris. This was accomplished by subcloning a tandem repeat of the first 15 N-terminal amino acids of …
