Not all children receive adequate education, nor do they have the opportunity to go beyond the four walls of a classroom for learning opportunities. Outdoor environmental education with place-based learning frameworks aims to fully immerse students in nature, while strengthening the connection between schools and communities. This study examines the outdoor environmental education programs of nine agriculture-centered community organizations in the San Francisco Bay Area. The proposed work aims to overcome the economical and feasibility-related limitations of the chimeric antigen receptor therapies by developing an artificial cell signaling pathway whose design transforms K562 cells into in vivo living vectors to synthesize therapeutic proteins upon engaging diseased cells in the treatment of ovarian cancer. Carboxypeptidase O (CPO) is a peptidase that is a member of the A/B subfamily of the metallocarboxypeptidases. This peptidase has been found in the brush border of the intestine and has specificity toward C-terminal acidic residues. CPO has been proposed to be involved in intestinal digestion and the formation of lipid droplets. Four copies of the CPO gene are present in the Xenopus tropicalis (tropical clawed frog) genome and are noted as CPO.1, CPO.2, CPO.3, and CPO.4. Enteroviruses, including Coxsackievirus B3 (CVB3), are pervasive pathogens that cause significant disease, including cardiomyopathies. Unfortunately, no treatments or vaccines are available for infected individuals. We identified the host polyamine pathway as a potential drug target, as inhibiting polyamine biosynthesis significantly reduces enterovirus replication in vitro and in vivo. Here, we show that CVB3 is sensitive to polyamine depletion through the polyamine analog diethylnorspermidine (DENSpm) which enhances polyamine catabolism through induction of polyamine acetylation. We demonstrate that CVB3 acquires resistance to DENSpm via mutation of the 2A protease, which enhances proteolytic activity in the presence of DENSpm. Resistance to DENSpm occurred …

Do female headed households lose/ sell more of their livestock in the event of a drought as compared to households headed by males? We address this question through an analysis of the combination of Living Standards Measurement study Integrated surveys on Agriculture dataset on Uganda using data from the period 2011-2016 and NDVI & Weather data. We carry out both OLS and fixed effects estimations. Our OLS Results are consistent with the theory of Consumption smoothing while we do not find evidence of any gendered effects from both the OLS and IV estimations.

Microtubules facilitate major cellular and morphological changes in eukaryotic cells, including the protozoan parasite Plasmodium. Microtubule-associated proteins (MAPs), only very few of which have been characterized in the malaria parasite, regulate the highly dynamic microtubules. Thioredoxin-like associated protein 2 (TLAP2) is a MAP unique to the phylum apicomplexa, which in addition to Plasmodium includes the important human pathogen Toxoplasma. Here we report on the cloning and microtubule-binding activity of TLAP2 of the rodent malaria parasite Plasmodium berghei (PbTLAP2). PbTLAP2 is coded by a single exon gene and consists of 598 amino acids. The protein contains a putative C-terminal microtubule-binding domain. …

In terms of sensory neuropathy, metabolic syndrome is involved in chronic inflammation and ER stress pathways induced by lipid overload, leading to the structural and physiological damage of afferent sensory nerves. The symbiosis between the gram-negative marine bacterium Vibrio fischeri and its host Euprymna Scolopes occurs when the bacteria reaches and colonizes the light organ. This pathway to colonization begins in the outer marine environment and finishes within the deep crypts of the light organ, with states of biofilm formation and dispersal in between. This switch between biofilm formation to dispersal is important as the bacteria transitions from the surface on the light organ into a pore leading to the interior of the light organ. Stiffness is a signaling cue in the brain, and neuronal response to stiffness impacts development, injury and disease. Mechanotransduction is the process of cells interpreting and converting mechanical cues to biochemical signals. Tubulin acetylation increases microtubule stability and could be mechanosensitive. The impact of stiffness on tubulin acetylation and role of septins is the focus of this project.It was hypothesized that stiffer surfaces would increase tubulin acetylation. SH-SY5Y cells were plated on varying stiffness and the ratio of acetylated to alpha tubulin was measured using immunocytochemistry. Meningiomas (MN) are primary brain tumors originating in the meningeal covering of the central nervous system. These tissues can evolve through familial inheritance (NF2, Cowden Syndrome, or Werner Syndrome) or sporadic mutations. The WHO categorizes MNs into three grades (I-III); Grade I contain benign tumors and Grade III the most aggressive. Increasing grade coincides with an increase in genetic diversity, making universal treatments difficult. MNs are typically treated with hydroxyurea (HU), but this chemotherapy agent has many adverse side effects, leading to research in less toxic options. Bacteriophages, or viruses that infect bacteria, have gained renewed interest in Western Medicine as an alternative to antibiotics. Phage therapy of P. aeruginosa infections has primarily focused on obligately lytic phages. However, prophages, or phages in the lysogenic life cycle, are estimated to far outnumber obligately lytic phages. Traumatic brain injury (TBI) affects millions of people in the U.S. each year. This thesis worked to address anxiety, a common emotional symptom following TBI, by utilizing a five-hit repeat mild TBI (rmTBI) experimental model. First, changes in gonadal status following injury and the severity of anxiety symptoms experienced were investigated. Our data revealed that suppressed testosterone levels following rmTBI resulted in significantly more anxiety-like behaviors across several observational variables. AL shows a patchy, uneven distribution in lotic ecosystems due to heterogeneity in its sources, how it moves, and mechanisms of retention. In addition, the diversity of material types, size, and mobility makes developing techniques to measure AL assemblage in rivers a challenge. Knowledge of when animals feed and the energetic costs of foraging is key to understanding their foraging ecology and energetic trade-offs. Despite this importance, our ability to collect these data in marine mammals remains limited. In this thesis, I address knowledge gaps in both feeding detection and fine-scale diving energetic costs in a model species, the California sea lion (Zalophus californianus). The Critically Endangered Largetooth Sawfish, Pristis pristis, experienced global declines in range and abundance over the past century and Australia is now their last stronghold. This research aimed to understand whether these declines have been accompanied by a reduction in levels of genetic diversity. Using P. pristis tissue samples sourced from natural history specimens, three fragments (616-bp, 386-bp, 141-bp) in the mitochondrial DNA (mtDNA) control region were PCR-amplified. At each fragment, significant genetic structure was found in P. pristis overall (ΦST=0.946, N=9; 0.813, N=54; 0.771, N=99). Infections of the protist parasite, Dermomycoides sp. are thought to have caused several years of low recruitment in the dusky gopher frog (Rana sevosa) populations. I evaluated the effects of density of the infective zoospores, host developmental stage, and tadpoles' ability to acquire resistance to Dermomycoides sp. on dusky gopher frog tadpoles. Tadpoles were exposed to zoospore densities of 0, 250, 500, and 750 zoospores/µL at Gosner stage 25, and we found no significant differences among treatments in tadpole mortality. Sargassum, a genus of holopelagic brown algae, floats at the ocean’s surface using air-filled bladders and forms a complex comprised of two species, S. natans and S. fluitans. Oceanic processes (e.g., Langmuir circulation, etc.) aggregate Sargassum into mats and weedlines, and primarily distribute the algal complex throughout the North Atlantic Ocean and the Gulf of Mexico (GOM). These floating habitats provide shelter and feeding opportunities for a diverse community of invertebrates and fishes.
