Comparison Study Between Post-Fermentation Stillage And Pre-Fermentation Mash Utilizing Acidification And Steam Explosion Techniques For Cellulose Saccharification, Daniel Mckee

Pre-fermentation mash fiber and post-distillation stillage fiber were examined and compared using a variety of preparatory techniques to determine the better source for cellulose fiber saccharification. Once screened, dried, and diluted to a 10% solution, mash fiber and stillage fiber were exposed to increasing temperatures for steam explosion techniques as well as increasing acidification techniques. Both underwent enzymatic saccharification to convert the exposed cellulose to glucose and other sugars. Once the optimum steam explosion technique parameters and acidification parameters were determined to be 2.5% sulfuric acid at 127.8°C for 1 hour, a comparison of the saccharification of pre-fermentation mash fiber …

Elevated surface temperatures exacerbate the threat of hypoxia within coastal ecosystems. These two primary stressors likely interact as they elicit opposing physiological responses from marine organisms. Metabolic depression is typically associated with hypoxia, while metabolic rates increase with temperature. Moreover, physiological effects of combined stressors may not be additive. In light of increasing pressures from hypoxia, elevated ocean temperatures, and other stressors within coastal regions, studies need to examine effects of multiple stressors on physiology of coastal organisms.

Clinical evidence indicates a higher incidence of peripheral arterial occlusive disease and associated likelihood of critical limb ischemia in women, as well as worse prognosis and decreased survival post myocardial infarction. Therefore, understanding the possible differences in underlying vascular compensation mechanisms is crucial. With arterial occlusions, necrosis and tissue injury can be naturally mitigated by the collateral circulation, improving patient prognosis. miRNAs are post-transcriptional regulators of gene expression, with numerous being involved in neurobiology. Within the human genome a quarter of the identified miRNA loci derive from a class of miRNAs termed tailed mirtrons. Despite the identification of this large population of miRNA, no functional studies have been conducted to identify their role. In this study we examined the highly expressed and deeply conserved Drosophila 3’ tail mirtron, miR-1017, as a candidate to elucidate tailed mirtron functionality. Temperature can exert impacts on many processes in ectotherms. With global temperatures rising due to climate change, many ectothermic species may exhibit changes in growth rates and size at maturity, and these changes can have population-level effects. Predicting responses of species to climate change will require not only knowledge of thermal tolerance limits, but also effects of temperature change on growth rates and other life history parameters. This study investigates the movement and harvest dynamics of the Gulf Menhaden Brevoortia patronus fishery. The fishery-dependent data collected by NOAA (years 2006-2009 and 2011) describe vessel-specific information on catch locations (latitude and longitude) and magnitude of harvest in metric tons (mt). A series of probability distribution functions (PDFs) were fit to the frequency distributions of number of harvests per day (Poisson), between-harvest distances (gamma), and harvest magnitude (log-normal). These analyses were used to inform an individual-based model (IBM). Speech perception is dependent upon the ability to map the sensory features of a speech signal onto the perceptual features which make up language (i.e., phonemes). A great deal of research over the past six decades has focused on how variability across talkers influences speech processing. Listeners are required to normalize the acoustic variability across talkers by continuously updating the mapping from the acoustic signal to phonetic representations. As such, processing speech from multiple talkers is cognitively more demanding than listening to a single talker. The work of the Cal Poly Tissue Engineering Lab is primarily focused on the fabrication, characterization, and improvement of “Blood Vessel Mimics” (BVMs), tissue engineered constructs used to evaluate cellular response to vascular medical devices. Currently, cells are grown onto fibrous, porous tubes made using an in-house electrospinning process from PLGA, a biocompatible co-polymer. The adhesion and proliferation of cells in a BVM is reliant on the micro-scale structure of the PLGA scaffold, and as such it is of great importance for the electrospinning process to consistently produce scaffolds of similar morphologies. Molting in crustaceans is a highly complex physiological process involving negative regulation by two paired endocrine glands, the X-organ/sinus gland complex (XO/SG) and the Y-organ (YO). The XO/SG complex is responsible for making molt-inhibiting hormone (MIH) which negatively regulates synthesis of the molting hormones, ecdysteroids, by the YO. Analysis of gene expression in the XOs and YOs has led to the development of a proposed molecular signaling pathway which regulates ecdysteroidogenesis and subsequent molting in crustaceans. Peripheral arterial occlusive disease (PAOD) often presents as intermittent claudication, which may be caused by impaired vasodilation. Impairment of resistance vessels may contribute to the pathogenesis of PAOD, and explain the poor correlation between resting blood flow and limb function. Collateral function following arterial occlusion is not well defined, however collaterals and arterialized collateral capillaries (ACCs) in male and female animal models exhibit impaired vasodilation following arterial occlusion, which can potentially be improved with exercise training. In the intestine, apoC-III delays chylomicron secretion, and causes a decrease in chylomicron size. Once apoC-III is in circulation, apoC-III delays triglyceride-rich remnants clearance by inhibiting lipoprotein lipase and liver low-density lipoprotein receptor. In humans, high levels of plasma apoC-III directly result in hypertriglyceridemia. ApoC-III is a critical cardiovascular risk factor, and humans expressing null mutations in apoC-III are robustly protected from cardiovascular disease. Because of its critical role in elevating plasma lipids and CVD risk, the factors that regulate apoC-III expression in the liver have been determined, and include glucose, insulin (through FoxO1), and dietary fat. EnTAP (Eukaryotic Non-Model Transcriptome Annotation Pipeline) was designed to improve the accuracy, speed, and flexibility of functional gene annotation for de novo assembled transcriptomes in non-model Eukaryotes. This software package addresses the fragmentation and related assembly issues that result in inflated transcript estimates and poor annotation rates. Following filters applied through assessment of true expression and frame selection, open-source tools are leveraged to functionally annotate the translated proteins. Downstream features include fast similarity search across three repositories, protein domain assignment, orthologous gene family assessment, and Gene Ontology term assignment. Odontoblast differentiation is dependent on multiple signaling molecules, including growth factors stored in the extracellular dentin matrix. Our previous studies have demonstrated that Fibroblast Growth Factor 2 (FGF2) exerted stage-specific effects on odontoblast differentiation in primary dental pulp cultures. FGF2 stimulated differentiation of functional odontoblasts expressing high levels of Dmp1 from early progenitors but inhibited the terminal differentiation of functional odontoblasts to fully differentiated odontoblasts expressing high levels of Dspp. These stimulatory effects involved activation of FGFR and ERK1/2, and increased levels of expression of Bmp2, Runx2, and Osx (components of BMP signaling) suggesting the involvement of …

Diets rich in fruits and vegetables have been correlated to lower disease risk. This is thought to be due to bioactive components such as: vitamin C, vitamin E, carotenoids, and polyphenols. Grapes are a food source of many bioactive components, including polyphenols. The effects standardized, freeze-dried, grape powder (GRAPE) were examined in adults with metabolic syndrome (MetS). Twenty men and women (aged 30-70 y) were instructed to consume 60 g/day of GRAPE (equivalent to 2.5 cups fresh grapes daily) or nutritionally-matched placebo powder in a randomized, placebo-controlled, crossover intervention study for 13 weeks. Coastal marshlands are among the world’s most highly productive ecosystems but they have diminished greatly in the past several decades owing to sea-level rise and direct anthropogenic influences. Many comparative studies have focused on the emergence of solitary and social play in early development. However, few consider observer and parallel forms of solitary play. In this study, use of video analysis revealed that there is a significant difference between the frequency of solitary, observer, parallel, and social object play states, in calf, juvenile, subadult, and adult bottlenose dolphins (Tursiops truncatus). While juveniles engaged in solitary play significantly more than any other age group, both juveniles and calves engaged in observer and social play significantly more than adults and subadults. I used Hester-Dendy multiplate samplers and stick and leaf samples to collect the aquatic insects of Granny Creek, a first-order perennial stream in southern Mississippi, on twelve sampling dates from March 2014 to June 2015. The dominant insect/invertebrate group in Granny Creek was the dipteran family Chironomidae. Forty-two distinct taxa of chironomid larvae were collected over the duration of my study. Two of the dominant chironomid subfamilies in my samples, the Orthocladiinae and the Chironominae, displayed a phenological pattern in which the Orthocladiinae were more prevalent, both in terms of number of species and in abundance of individuals, in the …

This research entails investigation of ammonia, nitrate, and nitrite removal from wastewater using a proprietary blend of bacteria known as BiOWiSHTM. The degradation rates of ammonia, nitrate, and nitrite for Aqua were determined using wastewater at the San Luis Obispo Water Resource Recovery Facility (SLO WRRF). Laboratory and field experiments were conducted to test how Aqua compared with natural bacteria for removal of nitrogen compounds. Preliminary data suggested that Aqua performed nitrate removal best in SLO WRRF wastewater at the secondary clarifier. Aqua could perform anoxic and aerobic denitrification in secondary clarifier wastewater. In mineral media, Aqua removed …

Creeks and rivers are often polluted as a result of stormwater runoff that carries various contaminants in to open water bodies, causing adverse environmental and health effects. Low impact development (LID) techniques are currently employed to treat this runoff prior to discharge. Nitrate, however, is not consistently removed by these LID techniques. Disturbance affects the function and diversity of ecosystems. Increased wave exposure to salt marsh can disturb sediments and cause a loss of habitat. The purpose of this study was to explore the effects of increased wave exposure on diversity, abundance, and functional ecology of estuarine fishes. If increased wave exposure is acting as a disturbance to these habitats, ecological theory (Intermediate Disturbance Hypothesis) predicts that diversity will peak at intermediate frequencies and intensities of disturbance.
