Identification Of Signal Transducers In Plant Shoot Gravitropism Using Genetic Screens, Lilla Reed May 2023

Identification Of Signal Transducers In Plant Shoot Gravitropism Using Genetic Screens, Lilla Reed

Honors Theses

Gravitropism is an essential response for plant growth and architecture. The signal transduction pathway that links gravity sensing to gravity response remains unclear because the key signal transducers involved in this pathway are unknown. In this project, we applied an artificial microRNA(amiRNA)-assisted genetic screen in isolating mutants displaying altered gravitropic phenotype in the darkness. Two amiRNA lines showed a significantly decreased gravitropic phenotype in the hypocotyl, and amiRNAs in these lines were identified to target a hypothetical transmembrane protein and a receptor kinase, respectively. Further experiments need to be done to confirm their phenotypes and establish them as signal transducers …

The Association Between Sleep And Relationship Quality Among Undergraduate Students, Catherine Smith May 2023

The Association Between Sleep And Relationship Quality Among Undergraduate Students, Catherine Smith

Honors Theses

This study sought out to examine the association between sleep and relationship quality in undergraduate student populations. Attending university is a novel experience for emerging adults but is subsequently associated with increased sleep difficulties and increased exposure to various social relationships. Indeed, the literature suggests social relationships can influence sleep in all populations, but few have explored relationship quality in the context of conflict and support in undergraduates. The quality of different social relationships (romantic, family, and friend), the severity of insomnia symptoms, and length of sleep duration were investigated. The current study collected data from a sample of ten …

Para Todes: ¿Sí O No? The Use Of Inclusive-Language Alternatives In The Spanish Language In Montevideo, Uruguay, Gabrielle Goulet

Honors Theses

This thesis investigates the attitudes towards the possible implementation of gender-neutral language, through the use of the -e, on official documents and schools in Montevideo, Uruguay, and how the attitudes of Spanish speakers in Uruguay align with the country’s history of inclusivity. In this context, Uruguay is considered an inclusive country because there is evidence of legal changes that promote gender equality, thus creating an gender-inclusive environment. This thesis utilizes a mixed-methods approach which includes both qualitative and quantitative research. The findings show that many people will use inclusive language when asked of them. They also believe that this usage …

The Impacts Of Seasonality And Nutrient Loading On Microcystis Bloom Development In Wall Pond, Alyssa Antolak May 2023

The Impacts Of Seasonality And Nutrient Loading On Microcystis Bloom Development In Wall Pond, Alyssa Antolak

Honors Theses

The harmful cyanobacteria Microcystis globally dominates eutrophic freshwater systems. Eutrophication leading to nitrogen and phosphorus loading into aquatic systems is increasing bloom propagation and shifting diatom/dinoflagellate dominated systems to cyanobacteria dominated systems. Understanding seasonal variability and environmental parameters combined with nutrient loading will allow for a better understanding of what factors are influencing Microcystis blooms. Biweekly plankton samples and environmental parameters were collected from Wall Pond from spring 2022-spring 2023. Results show that yearly plankton samples shifted from Microcystis dominated in the early summer to diatom dominated in the late summer-early fall then back to Microcystis dominated in late fall-early …

Predicting Our Own Demise: How Media Has Played Nostradamus To The Future Of Artificial Intelligence, Connor Denny-Lybbert May 2023

Predicting Our Own Demise: How Media Has Played Nostradamus To The Future Of Artificial Intelligence, Connor Denny-Lybbert

Honors Theses

In this paper, I argue that we should be hesitant and skeptical of the applications of AI technology in government due to the possibility of, and ongoing abuses of this technology by political actors. I do this by first analyzing various pieces of sci-fi media in three parts: how this media approaches crime and punishment, personhood, and human happiness. Then, I explain how these themes intersect with the above framework and how we can use the themes to guide future policy. This is followed by an analysis of what these pieces of media tell us regarding artificial intelligence and how …

Antioxidant Potentials Of Kombucha Based On Water Chemistry And Its Impact On Fermentation, Olivia Sundman

Honors Theses

Kombucha is made by using a symbiotic culture of bacteria and yeast (SCOBY) to ferment sweetened tea. This fermentation produces a beverage with a unique aroma and acidic flavor. Kombucha has recently gain popularity in the United States and has been reported to have numerous health benefits from increased weight loss to helping to fight cancer. There is a wide variation in kombucha composition, little is known about the impact the water’s chemistry has on the fermentation and the resulting kombucha. The impact that bicarbonate, chloride, calcium, magnesium, and sulfate concentration have on the measured antioxidation potential of brewed kombucha …

The Dynamics Of Sedimentary Chlorophyll Α At A High-Energy Beach, Sarah (Gray) Abel May 2023

The Dynamics Of Sedimentary Chlorophyll Α At A High-Energy Beach, Sarah (Gray) Abel

Honors Theses

Productive sandy beaches are one of the most crucial areas serving our ecosystem today. While often overlooked in scientific research, productive beaches have many essential functions that lead to the preservation and protection of many different species as well as ecological wellness and stability. A key factor that is linked to this productivity is the presence of chlorophyll in the sediments. Piston cores of the sediments at the low-tide mark at Waties Island, South Carolina, were collected across multiple years and subsequently analyzed for chlorophyll concentrations through fluorometry after acetone extraction and acidification. Each core profile was processed to calculate …

Neuronal Effects Of Cocaine In An Animal Model Of Social Stress: Analysis Of Neuronal Recordings, Eboni Eddins May 2023

Neuronal Effects Of Cocaine In An Animal Model Of Social Stress: Analysis Of Neuronal Recordings, Eboni Eddins

Honors Theses

Studies that use Intermittent (episodic) Social Defeat (ISD) in rats demonstrate that ISD increases cocaine-self administration several weeks after the end of the adverse experience and suggest that a history of social stress makes individuals more vulnerable to substance abuse in the long term. The medial prefrontal cortex (mPFC) plays a key role in regulating drug-seeking behavior. The present study investigates whether ISD enhances the response of mPFC neurons to cocaine. Male Long Evans rats (3-4 months) were implanted with electrode arrays in the mPFC (prelimbic area) and divided into two groups (Control, n= 4; Stress, n= 4). They were …

The Measurement And Treatment Of Overt Stuttering In Handwriting, Preston Dubberly May 2023

The Measurement And Treatment Of Overt Stuttering In Handwriting, Preston Dubberly

Honors Theses

While stuttering continues to be considered a speech disorder, other linguistic expressive modalities are reported as having stuttering-like disfluencies (SLD) as well. With research now defining the stuttering phenomenon as a medical disease, the modern stuttering research paradigm is now better equipped to account for SLDs in multiple expressive modalities, including handwriting. As our appreciation of stuttering crosses expressive modalities, the purpose of this manuscript is to document, describe, and define SLDs in handwriting, as well as to investigate fluency enhancement strategies for those who stutter in handwriting. The participant in this case study self-referred himself for an evaluation and …

Is Vdac1 A Novel Bcl2 Family Member That Binds Bax?, Claire Pearson May 2023

Is Vdac1 A Novel Bcl2 Family Member That Binds Bax?, Claire Pearson

Honors Theses

Apoptosis is a type of regulated cell death important for normal embryonic development and maintenance of adult tissues by removing excess or dysfunctional cells to ensure proper functioning of organs. The Bcl-2 family of proteins determines whether apoptosis remains suppressed or becomes activated through the balance of interactions among pro-survival and pro-death members. A defining feature of the Bcl-2 family is a BH3 domain that drives interactions between the family members. Isoform 1 of the voltage dependent anion channel (VDAC1) has an important role in metabolism, but was recently found to have high homology with known BH3 domains. This study …

Does Vdac2 Have A Bh3 Domain?, Lillian Ferkany May 2023

Does Vdac2 Have A Bh3 Domain?, Lillian Ferkany

Honors Theses

Mitochondrial outer membrane permeabilization (MOMP) by Bax oligomerization triggers apoptosis. BCl-2 family proteins, classified as BH3 only proteins, pro-survival proteins, or pro-apoptotic proteins, control apoptosis partly through their agonist or antagonistic effects on Bax, which are mediated by their conserved BH3 domains. All BH3 domains form an alpha helix containing 5-7 conserved hydrophobic residues, designated H0-H5, and one conserved aspartic acid that drive interaction with Bax and other ‘multi-domain’ BCl-2 members. BH3 agonists induce Bax oligomerization, while BH3 antagonists sequester Bax to prevent MOMP. We discovered that voltage dependent anion channels (VDACs) in the MOM contain a putative BH3-like domain …

Morphological And Functional Classification Of The Amblyomma Americanum (Lone Star Tick) Hemocytes, Julia Hanson May 2023

Morphological And Functional Classification Of The Amblyomma Americanum (Lone Star Tick) Hemocytes, Julia Hanson

Honors Theses

Ticks are a diverse group of obligate blood-feeding (hematophagous) arthropods that transmit a wide array of human and animal pathogens. These pathogens have caused an increase in tick-borne diseases that pose a significant threat to public health without preventative measures. These pathogens interact with the tick immune system upon uptake, which is primarily divided into humoral and cellular components. Cellular immunity in ticks encompasses several pathogen-eliminating mechanisms including phagocytosis. These mechanisms are mediated by the action of immune cells, commonly referred to as hemocytes, which are analogous to mammalian white blood cells. This study utilized a combination of morphological, functional, …

Play That Funky Music: A Study Of Bacterial Contamination Of Wind Instruments, Ashley Clark

Honors Theses

Should there be standards to routinely clean instruments and what methods are available for that? It has been found that the bacteria and other microorganisms inside of musical instruments may be the culprit of respiratory infections in musicians (King et al. 2016). This project was divided into two experiments. First, clarinet reeds were introduced to three different species of bacteria. Then mouthpieces of trombonists and clarinetists of Eastern Kentucky University were swabbed and analyzed using a Colony Forming Unit assay. Bacteria was found to live on unused reeds for up to 30 days. Mouthpieces house millions of bacteria, determined by …

Oxalic Acid Leaf Disk Soak Assay Is A New Possibility In Screening For Blight Resistance In Castanea Species, Kaitlyn Harden May 2023

Oxalic Acid Leaf Disk Soak Assay Is A New Possibility In Screening For Blight Resistance In Castanea Species, Kaitlyn Harden

Honors Theses

Chestnut blight is a disease caused by the ascomycete fungus Cryphonectria parasitica in the Castanea species. The fungus uses oxalic acid (OA) to attack the tree’s cells. Castanea dentata, the American chestnut, was wiped out by chestnut blight in the early to mid-20th century, but several East Asian Castanea species appear highly resistant to the fungus. To breed resistant American type trees, screening methods are used to enable selection of interspecific hybrids. The alternative small stem assay (aSSA) is a method of screening container-grown hybrid seedlings during their first growing season by directly infecting them with C. parasitica. Some plant …

Assessing The Utility Of High-Throughput Phenotyping For Ecological Applications, Annabree Corlew May 2023

Assessing The Utility Of High-Throughput Phenotyping For Ecological Applications, Annabree Corlew

Honors Theses

A phenotype describes the expressed traits of an organism. These traits can provide insight into interactions between an organism and its environment, making them a key focus of ecological research. But the process of phenotyping traits can be tedious to collect for large-scale experiments. High-throughput phenotyping software has the ability to speed up the phenotyping process by allowing multiple traits to be phenotyped at once. However, the utility of this relatively new and continually developing computational tool is still being assessed, especially beyond applications focused on agricultural crops and grasses. I investigated the accuracy and efficiency of the open-access high-throughput …

Species Diversity Of Coleoptera Populations In An Urban Garden In Chattanooga, Tennessee, Mallory Anderson May 2023

Species Diversity Of Coleoptera Populations In An Urban Garden In Chattanooga, Tennessee, Mallory Anderson

Honors Theses

Insects are an essential part of the function of ecosystems. The majority of entomological faunistic research has been conducted primarily in rural areas due to high diversity, space for bulky traps, and less outside influence from human activities. However, the need for time-series insect biodiversity data, in rural areas and urban areas alike, is paramount in the wake of our current sixth mass extinction event. Previous research has shown that the greater Chattanooga area is comprised of a diverse population of insects in the Coleoptera (beetles) order, and as shown in the University of Tennessee at Chattanooga’s (UTC) Insect Collection. …

Perceptions Of Stress: A Gendered Comparison Of Undergraduates In Stem, Molly Martin May 2023

Perceptions Of Stress: A Gendered Comparison Of Undergraduates In Stem, Molly Martin

Honors Theses

Students enrolled in undergraduate universities face numerous stressors every day. Outside of stress from their academic studies (i.e., homework, studying for exams, writing papers), they also encounter daily life stressors relating to self-sufficiency, career choices, families, housing, and many other areas. While every student deals with stress coming from multiple areas of their life, women tend to place more stress on themselves than men due to social pressure to perform well and the feeling of societal expectation to have a high stress load from which to derive personal value. Within the field of Science, Technology and Math (STEM), —a field …

Role Of Pseudomonas Aeruginosa Exoenzymes S, T, And Y In The Modulation Of Intrinsic Apoptosis In Pulmonary Microvascular Endothelial Cells, Andrea M. Vavrinek May 2023

Role Of Pseudomonas Aeruginosa Exoenzymes S, T, And Y In The Modulation Of Intrinsic Apoptosis In Pulmonary Microvascular Endothelial Cells, Andrea M. Vavrinek

Honors Theses

P. aeruginosa is the most common cause of ventilator-associated pneumonia in ICU patients. P. aeruginosa uses a type III secretion system to inject exoenzymes S, T, U, and Y into host cells, exhibiting unique effects. Pulmonary microvascular endothelial cells (PMVECs) are used to evaluate cellular changes caused by exoenzymes. ExoY infection of PMVECs leads to cell rounding but does not lead to cell death, unlike ExoS/T. The current study evaluated the exoenzyme-induced modulation of intrinsic apoptosis in endothelial cells. Strain PAK, producing ExoS, T, and Y, was compared to isogenic strains expressing either S, SY, T, TY, Y, or none …

Investigation Of The Vira Linker Domain To Characterize Its Phenol Interactions, Jessica Garofalo May 2023

Investigation Of The Vira Linker Domain To Characterize Its Phenol Interactions, Jessica Garofalo

Honors Theses

The soil bacterium Agrobacterium tumefaciens causes tumors in plants through interkingdom gene transfer. This transfer is initiated upon a wounding event that results in the release of plant signaling factors such as phenols and sugars that are recognized by protein machinery in the periplasm and inner membrane of A. tumefaciens. The histidine kinase protein VirA, in combination with the periplasmic protein ChvE, recognizes these signals and initiates induction of virulence genes via a signaling pathway, culminating with the insertion of tumor-inducing T-DNA into the wounded plant cells. While the interaction between the Periplasmic domain of VirA and sugar-bound ChvE …

Effect Of Temperature On The Microbiome Of A Laboratory-Reared Colony Of Haemaphysalis Longicornis Ticks, Brianna L. Mitchell May 2023

Effect Of Temperature On The Microbiome Of A Laboratory-Reared Colony Of Haemaphysalis Longicornis Ticks, Brianna L. Mitchell

Honors Theses

Haemaphysalis longicornis ticks are invasive to the United States with potential to transmit several tick-borne pathogens that are native to the United States. Based on existing locations of H. longicornis in its native regions in Asia, as well as its invasive populations that are established in the United States, several geographic range prediction models have been produced to help understand future range expansion and distribution of this invasive tick in North America. Unfortunately, these models do not all agree and there is uncertainty associated with the potential geographic range expansion of H. longicornis ticks in North America. Climate can affect …

Examining Ndufab1 Expression In Head And Neck Squamous Cell Carcinoma, Addison L. Stevens May 2023

Examining Ndufab1 Expression In Head And Neck Squamous Cell Carcinoma, Addison L. Stevens

Honors Theses

Head and neck squamous cell carcinoma (HNSCC) accounts for around 4% of all cancers in the USA. HNSCC includes cancers of the oral cavity, oropharynx, nasopharynx, hypopharynx, and larynx. Reprogramming of mitochondrial metabolism has been known to promote oncogenesis. NDUFAB1, a nuclear encoded subunit of respiratory complex I (RCI) in the inner mitochondrial membrane, is abundantly expressed at the mRNA level in HNSCC patients. Based on this finding, we hypothesize that NDUFAB1 protein expression is high in HNSCC and that NDUFAB1 expression predicts a poor prognosis in HNSCC patients. We determined NDUFAB1 expression in HNSCC using immunohistochemistry and pathology guided …

Pirna Expression In The Tube Feet Of Lytechinus Variegatus, Reagan Milliet May 2023

Pirna Expression In The Tube Feet Of Lytechinus Variegatus, Reagan Milliet

Honors Theses

Tissue regeneration is an area of research with implications for medicine and animal health. While nearly all living multi-cellular organisms are capable of regeneration, there are major differences in the animal kingdom. Some organisms are capable of regenerating virtually every cell in their bodies. In hopes of engineering tissue regeneration for medical applications, the mechanisms by which organisms regenerate are being widely investigated. To better understand regeneration, the role of P-element Induced WImpy testis (PIWI) proteins are being evaluated. In collaboration with PIWIinteracting RNAs (piRNAs), PIWI proteins have been proven instrumental to transposon silencing and maintenance of the genome in …

Dissolved Copper Distributions In The Western Mississippi Sound: Characterizing An Unrecognized Endmember, Megan Hansen May 2023

Dissolved Copper Distributions In The Western Mississippi Sound: Characterizing An Unrecognized Endmember, Megan Hansen

Honors Theses

This study examined the distribution of dissolved copper (Cu) in the western Mississippi Sound, as the element can be beneficial or harmful to organisms depending on its concentration and its chemical form. A mixing experiment eliminated flocculation as a control on Cu distributions during estuarine mixing in both dissolved and colloidal fractions. Field sampling of the western Mississippi Sound occurred during a low river discharge period, and distributions of Cu indicated conservative mixing between fresh ([Cu] ≈ 17 nM) and saline ([Cu] ≈ 2 nM) sources in mid-to-high salinities. Storm outfalls were sampled after rain events to evaluate the impact …

A Lost Reference Point: How Placing Our Identities In The State Has Facilitated Social Polarization Among Americans, Dylan Evans May 2023

A Lost Reference Point: How Placing Our Identities In The State Has Facilitated Social Polarization Among Americans, Dylan Evans

Honors Theses

We are living in a moment of societal breakdown, as America is increasingly plagued with fractious polarization along political and cultural lines. The potential causes of this are complex and exist within a broad spectrum of possibilities, with the potential solutions being even more contentious. However, it is my contention that identity is the central issue here. As people begin to place their identities in a religious devotion to the liberal state as opposed to a transcendent ideal, once simple, agreed-upon premises become harsh divides, and polarization ensues. To fully evaluate how this has happened, and thus how it may …

Developing A Continuous Cell Line Derived From The Gulf-Coast Tick (Amblyomma Maculatum) Embryos, Sabrina Walia May 2023

Developing A Continuous Cell Line Derived From The Gulf-Coast Tick (Amblyomma Maculatum) Embryos, Sabrina Walia

Honors Theses

Tick cell lines provide an affordable, reproducible, and efficient in vitro experimental system to study tick-pathogen interactions at the cellular level. Before the first tick cell lines were developed, there was no established method to perform in vitro studies with ticks. Tick research at the organismal level is full of difficulties and challenges. An increase in tick-borne infections and the availability of cutting-edge molecular tools have stimulated the research community's interest in using tick cell lines to answer important biological questions related to new and emerging tick-borne diseases. Unfortunately, not all medically relevant tick species have a cell line available. …

The Effects Of Social Media On Mental Health And Career Planning, Spencer A. Rowan May 2023

The Effects Of Social Media On Mental Health And Career Planning, Spencer A. Rowan

Honors Theses

Social media use is prevalent and necessary in society—nearly anything can be accomplished with a mobile device or smartphone. Among the US population, two thirds of American adults admit to using social media (Perrin, 2015) and in 2022, Georgiev (2023) found Americans spent an average of two and a half hours daily on social media. Furthermore, social media use is tied to mental well-being, work confidence levels, and feelings of being an imposter (Johnson et al., 2020; Uram & Skalski, 2022; Hernandez & Chalk, 2021; Myers, 2021; Ramm, 2019).

This project examined the role of social media use among college …
