Polychlorinated biphenyls are ubiquitous, persistent environmental contaminants that cause a variety of adverse health effects wildlife and humans. PCBs are the major contaminant in the Kalamazoo River Superfund Site, in which the Environmental Institute is funded to study. Projects at the Environmental Institute are focused on developing molecular biotechnology tools that will improve environmental risk assessment. Previous acute studies involved the exposure of developing tadpoles, Xenopus laevis to PCBs through aquarium water in order to measure gene expression. Diepoxybutane (DEB), a known industrial carcinogen, reacts with DNA primarily at the N7 position of deoxyguanosine residues and creates interstrand cross-links at the sequence 5'-GNC. Since N7-N7 cross-links cause DNA to fragment upon heating, quantative polymerase chain reaction (QPCR) is being used in this experiment to measure the amount of DEB damage (lesion frequency) with three different targets-mitochondrial (unpackaged), open chromatin region, and closed chromatin region. Initial measurements of DEB damage within these three targets were not consistent because the template DNA was not the limiting reagent in the PCR. Freshwater dinoflagellates, like their marine relatives, have the potential to reach large population sizes known as blooms. Though they are not toxic like some marine forms, freshwater blooms may have a large impact in their respective ecosystem. This study investigated the temporal and spatial changes in population density of four Peridinium species (P. deflandrei, P. volzii, P. wisconsinense, and P. limbatum) and one Peridiniopsis species (Peridiniopsis polonicium) in Boondoggle Lake, a shallow lake (4.5 m) in northern Mississippi (Lafayette Co.). The Soil Moisture Observatory (SMO) at the University of Mississippi (UM) is a 5 acre tract of a former agricultural field at the UM Biological Field Station. Preliminary investigations of this site included 60 continuous soil cores using the Geoprobe sampling technique. These soil cores were taken to a depth of 1.5 meters to correspond to the approximate depth of penetration for a Geonics EM38. This thesis examines age-related differences in religious practice and opinion in Spain and the influence of the declining status and increasing pluralism of the Catholic Church in the formation of these differences. It argues that there are three distinct age groups with respect to external religious manifestations and that the socialization of these cohorts coincides with three distinct time periods of differing degrees of power and unity of the Church. There are several classes of anticancer drugs, including the antimicrotubule drugs, which halt mitosis by interfering with the activity of microtubules, the cylindrical protein polymers that play a role in segregating chromosomes during mitosis. While most antimicrotubule drugs act by destabilizing the microtubule polymer and reducing microtubule assembly, paclitaxel, a drug first isolated from the bark of Taxus brevefolia, halts mitosis by stabilizing microtubules, thus interfering with the dynamics of polymerization and depolymerization that are crucial to their function. Wetlands are an interface between terrestrial and aquatic environments. As anthropogenic impacts on the environment increase it puts these sensitive areas at the forefront of change. Farming on William's Island has not occurred since 2001 but before that it was farmed to provide cost-free upkeep of the island. Simazine is a triazine herbicide, the most commonly used group of herbicides in modem agricultural practices. The purpose of this project was to identify all native and introduced vascular plant species near the Tennessee River Gardens. The field work resulted in 355 specimens that represent 81 families, 190 genera, and 268 species. In addition to collecting and identifying, I also compared my flora with that of Lookout Mountain Park to determine the degree of similarity between the two sites which are both south of the Tennessee River. Another comparison was completed contrasting my flora with the flora of Chickamauga and Chattanooga National Military Park. Diepoxybutane, diepoxyoctane, and mechlorethamine are cytotoxic DNA crosslinking agents that vary in their carcinogenic versus chemotherapeutic potentials. Interstrand cross-linking occurs between the N7 positions of deoxyguanosine residues on opposite strands of the DNA duplex. Each synthetic DNA oligomer used in this study contained four 5'-N\GN2CN3 sites (within a 32 base sequence) and we have systematically varied the bases in the N1 and N2 positions to determine the resulting cross-linking efficiencies of each cytotoxic agent. Each duplex was 5' -end labeled and incubated with cross-linking agent. Interstrand cross-links were purified through denaturing polyacrylamide gel electrophoresis and then subjected to piperidine cleavage. …
