Interference Of The Inflammasome Via Interferonβ, Madeline Sauer Apr 2018

Interference Of The Inflammasome Via Interferonβ, Madeline Sauer

Honors Theses

Metabolic disorders such as type two diabetes mellitus (T2DM) and obesity are known to have a chronic low grade inflammatory tissue environment as well as an increase in excess lipids. Current research suggests that a tightly regulated oligoprotein complex known as the NLRP3 inflammasome is highly activated in T2DM and obesity. However, it is not well understood the interplay between excess lipids, which was previously shown in our lab to cause lysosome damage, and inflammation. A key transcription factor that is known to have both an inflammatory effect as well as an effect on lipid metabolism is PPARγ. For this …

Do Normative And Pathological Personality Traits Overlap? Exploratory And Confirmatory Factor Analyses Of The Neo-Pi-3 And Pid-5, Lisa Eileen Stone Apr 2018

Do Normative And Pathological Personality Traits Overlap? Exploratory And Confirmatory Factor Analyses Of The Neo-Pi-3 And Pid-5, Lisa Eileen Stone

Honors Theses

Historically, personality disorders have been conceptualized as qualitatively distinct clinical syndromes, based on operational criteria. Consistent with this model, ten distinct set personality disorder criteria are outlined in the Diagnostic and Statistical Manual of Mental Disorders, Fifth Edition (American Psychiatric Association, 2013). However, debate persists about the clinical utility of this categorical model, with many (Krueger, et al.) researchers supporting a dimensional model that focuses on pathological levels of normative personality traits.

An exploratory factor analysis (De Fruyt et al., 2013) of the NEO Personality Inventory-3 (NEOPI-3; Costa & McCrae, 2010) and The Personality Inventory for DSM-5 (PID-5; Krueger, Derringer, …

“Eat, Sleep, Hydrate, Masturbate!” Sexuality Education, Digital Media And Creator Identity Implications, Emma Venetis Apr 2018

“Eat, Sleep, Hydrate, Masturbate!” Sexuality Education, Digital Media And Creator Identity Implications, Emma Venetis

Honors Theses

The introduction of sexuality information to young people has been a point of tension in our society for decades as adults argue over how, when, or if young people should learn such information. However, with the rise of digital technology, the ability of adults to regulate young people’s access to information about sexuality has minimized significantly. Yet the curriculum in sexuality education classrooms continues to be debated while little research has been done examining the easily-accessible information that lives on the Internet. This thesis analyzes two popular sexuality education channels on YouTube, sexplanations and lacigreen, with subscriber counts ranging from …

Caffeine Rush! Examining The Effects Of Caffeine On Spatial Working Memory, Alexander Lawriw Apr 2018

Caffeine Rush! Examining The Effects Of Caffeine On Spatial Working Memory, Alexander Lawriw

Honors Theses

Past research concerning caffeine and its effects on memory have yielded varying results. One study found that a 3-mg/kg dose of caffeine decreased reaction time during a selective attention task. However, this improvement was only found during a low display load, with no effect on reaction time occurring within a high display load (Lorist, Snell, Kok, & Mulder,1996). As such, the effects of caffeine may be dependent on a variety of other factors, such as the difficulty of the task at hand (Nehlig, 2004). This present study aimed to explore further the potential memory-enhancing qualities of caffeine with respect to …

If You Build It, They Will Come: A Habitat Mimicry-Based Assessment Tool For Estimating Larval Salamander Density In Temperate Forests Streams, Margaret E. Maloney Apr 2018

If You Build It, They Will Come: A Habitat Mimicry-Based Assessment Tool For Estimating Larval Salamander Density In Temperate Forests Streams, Margaret E. Maloney

Honors Theses

The biology of small, forested streams is critical to the ecology of larger waterways and broader watershed function. Because of their size, these “headwater streams” are strongly influenced by surrounding landscape conditions and their biology is intimately connected to the landscape through material and energy subsidies. In the United States, 50 to 80 percent of streams are primary headwater streams, making them a high priority for conservation. Often, stream salamanders are a useful indicator for biotic integrity of headwater streams due to their longevity, relatively stable populations, small home ranges, and abundance. Assessing stream salamanders is a challenging endeavor and …

Olfaction And Disgust As Predictors Of Elevated Perfectionism, Maia Mclin Apr 2018

Olfaction And Disgust As Predictors Of Elevated Perfectionism, Maia Mclin

Honors Theses

Odor detection and disgust sensitivity were once vital to survival by providing a means to assess if foods were safe for consumption. Along with odor detection and disgust sensitivity, obsessivecompulsive traits, such as checking, may have increased chance of survival by decreasing the likelihood of consuming contaminated foods leading to an evolutionary advantage (Rozin & Fallon, 1987). Current regulations that prevent the distribution of spoiled and contaminated foods in developed societies make these processes less necessary to survival today; as a result, obsessive compulsive traits that may have once been advantageous may now be pathological. Neural connections also suggest a …

Destruction, Reconstruction, And Remembrance: Exploring 'Memory' And 'Environment' Through Pennsylvania World War I Memorials In France, Amy Collins Jan 2018

Destruction, Reconstruction, And Remembrance: Exploring 'Memory' And 'Environment' Through Pennsylvania World War I Memorials In France, Amy Collins

Honors Theses

After examining the substantial efforts at land reclamation and environmental mitigation accompanying the State of Pennsylvania’s construction of memorials after World War I in France, I discovered a strong relationship between post-war memorialization and environmental mitigation in the areas in which the environmental consequences of WWI continue to affect humans and wildlife. My research illuminates how cultural impulses to build memorials that acknowledged the vast losses, acts of valor, and victories heavily influenced mitigation of France’s ecologically damaged Western Front. Many of France’s former battlefields, particularly in the devastated area known as the Red Zone, weren’t accessible to visitors before …

Investigating Phosphorylation Patterns And Their Effect On The Activity Of Transcription Factor Taabf1 In Imbibing Cereal Grains, Grace Uwase Jan 2018

Investigating Phosphorylation Patterns And Their Effect On The Activity Of Transcription Factor Taabf1 In Imbibing Cereal Grains, Grace Uwase

Honors Theses

The wheat transcription factor TaABF1 plays an important role in hormone-mediated regulation of seed dormancy and germination of cereal grains. Evidence shows that TaABF1 activity is regulated by phosphorylation, and previous work in our lab showed that when serine residues in its conserved regions; S36, S37, S113, S115 were altered to phosphomimetic aspartate, the 4xD TaABF1 mutant had increased activity as a transcription factor. However, when only S113 was altered, TaABF1’s activity was greatly reduced. The work presented here explored whether the S36D/S37D/S115D mutant would have stronger activity than the 4xD mutant. Using the particle bombardment technique to introduce the …

Presence And Potential Sources Of Pharmaceutical And Personal Care Product Chemicals In Messalonskee Lake, Alyssa T. Kullberg

Honors Theses

Pharmaceuticals and personal care product chemicals (PPCPs) represent a large group of contaminants that are mostly not regulated in surface or drinking water and whose presence and environmental and health impacts are poorly understood. We investigated the presence and potential sources of 18 PPCPs in Messalonskee Lake. We collected samples four times over the summer of 2017 at 13 sites around the lake. Samples were tested for PPCPs using liquid chromatography-tandem mass spectrometry. We detected five out of the 18 PPCPs we tested for at the ng/L level: caffeine, 1,7-dimethylxanthine, acetaminophen, sulfachloropyridazine, and amphetamine. PPCPs showed a significant positive correlation …

Haloferax Volcanii For Carotenoid Production, Erika Smith Jan 2018

Haloferax Volcanii For Carotenoid Production, Erika Smith

Honors Theses

Carotenoids are used as natural colorants and have been shown to have numerous health benefits, making these molecules highly desirable in the aquaculture, pharmaceutical, and nutraceutical markets. There is increasing consumer demand for the natural synthesis of carotenoids, but current methods are limited by the cost and difficulty of extraction from organisms. The halophilic archaeon Haloferax volcanii is amenable to extraction and has an endogenous carotenoid biosynthetic pathway that culminates in the accumulation of the carotenoid bacterioruberin; however, H. volcanii also synthesizes important precursors that can be converted into more highly-desired carotenoids, such as β-carotene and astaxanthin, in the presence …

From Forced To Voluntary Participation: The History Of Biomedical Human Experimentation In The United States After The Second World War, Haley L. Andonian

Honors Theses

For as long as medicine and medical practices have been around, so has the need for testing treatments in or procedures on the human body. Over the course of history, however, the nature, structure, and prominence of human biomedical experimentation has changed drastically both on an international and national level. My thesis focuses on revealing the driving forces behind these changes in administrative, legal and social factors related to human experimentation in an effort to connect the dots from the manipulative, forceful and unethical experimentation of early medical practitioners to the safe, voluntary and highly regulated experimentation characteristic of clinical …

The Identification And Application Of Generalizable Spatial Patterns Of Human-Wildlife Conflict, Vivian F. Hawkinson Jan 2018

The Identification And Application Of Generalizable Spatial Patterns Of Human-Wildlife Conflict, Vivian F. Hawkinson

Honors Theses

Many human-wildlife conflict studies focus on one location or one individual species or taxonomic group; fewer comparative studies analyze patterns of conflict across species and regions. As a result, numerous studies report similar conclusions across diverse cases of human-wildlife conflict. I found 133 scholarly articles published between 1975 and 2017 referencing distance from a protected area boundary as a variable associated with human-wildlife conflict. I identified three generalizable patterns of human-wildlife conflict that appear across taxonomic groups and geographic locations. The family Felidae had the highest maximum average conflict distance and furthest distance from a protected area that conflict was …

Disc1 Knockout Rats Reveal Sexually Dimorphic Patterns Of Impairment Across Development, Stephanie S. Desrochers Jan 2018

Disc1 Knockout Rats Reveal Sexually Dimorphic Patterns Of Impairment Across Development, Stephanie S. Desrochers

Honors Theses

Schizophrenia is a chronic disorder characterized by three symptom categories: positive (hallucinations, delusions), negative (anhedonia, anxiety), and cognitive (sensory processing and memory deficits). We worked with a biallelic deletion of the Disrupted-in-schizophrenia-1 (DISC1) gene in Sprague-Dawley rats to facilitate our understanding of the biological bases of schizophrenia. Mutations of DISC1 are associated with a higher prevalence of schizophrenia. Adult DISC1 knockout, compared to wildtype, rats display features of schizophrenia-like outcomes in rodent models. The present study investigated the progression of the cognitive symptom deficits in prepulse inhibition (PPI), assessing animals at postnatal days 17 (pre-weaning), 26 (post-weaning), 39 (adolescent), and …

Stable Bromine Isotope Signature Of Bromoform From Enzymatic And Abiotic Formation Pathways And Its Application In Identifying Sources Of Environmental Bromoform In The Damariscotta River, Chengyang Wang Jan 2018

Stable Bromine Isotope Signature Of Bromoform From Enzymatic And Abiotic Formation Pathways And Its Application In Identifying Sources Of Environmental Bromoform In The Damariscotta River, Chengyang Wang

Honors Theses

Bromoform is a major source of atmospheric bromine. Most bromoform is produced by marine organisms including macroalgae and phytoplankton, using the enzyme bromoperoxidase (BPO). Bromoform can also be a byproduct of industrial processes such as water disinfection. Identifying sources of environmental bromoform is still a challenge. A novel technique of using quadrupole mass spectrometry coupled to a gas chromatography (GCqMS) was developed and optimized for Br isotope analyses. The study shows that GCqMS in single ion monitoring (SIM) mode can measure 81Br with precision of around ±0.7‰ (60pmol bromoform injected). This study aims to investigate stable Br isotopes of bromoform …

The Effects Of Nutrient Limitation On Toxin Production For The Harmful Algal Bloom Species, Pseudo-Nitzschia, In The Gulf Of Maine, Julia M. Park Jan 2018

The Effects Of Nutrient Limitation On Toxin Production For The Harmful Algal Bloom Species, Pseudo-Nitzschia, In The Gulf Of Maine, Julia M. Park

Honors Theses

Pseudo-nitzschia is a pennate marine diatom that produces the neurotoxin domoic acid (DA). Domoic acid accumulates in the digestive tract of filter feeders and becomes concentrated at higher trophic levels. When DA concentrations reach critical levels, consumption of toxic shellfish can induce a condition known as Amnesic Shellfish Poisoning (ASP), which has been responsible for the deaths of marine mammals, seabirds and humans. This study worked to respond to a toxic Pseudo-nitzschia bloom in the Gulf of Maine during the fall of 2016, with the goal of quantifying and identifying Pseudo-nitzschia spp. in the bloom region and to determine the …

The Frequency Attenuations Of Foam Ear Plugs Affected By User Error Of College Students, Alyse J. Lemoine Jan 2018

The Frequency Attenuations Of Foam Ear Plugs Affected By User Error Of College Students, Alyse J. Lemoine

Honors Theses

This paper examines the relationship between the attenuating ability of foam ear plugs at low and high frequencies and the effects of incorrect ear plug fitting by college students. The National Institute for Occupational Safety and Health (1998) recommends the use of a hearing protection device (HPD) such as ear muffs or ear plugs to avoid noise induced hearing loss (NIHL). However, when not inserted properly, a HPD's effectiveness can be adversely affected by user error and present as a decrease in attenuation. Attenuation is measured and presented on packaging as the noise reduction rating (NRR). A high NRR affected …

Raman Spectroscopic And Computational Study Of Hydrogen Bond Interactions Between Guanidine Hydrochloride, Trimethylamine N-Oxide (Tmao), Urea, And Water, Andrew Kamischke

Honors Theses

Proteins are broken down in the body and produce water, CO2, and ammonia as by-products. Ammonia is toxic to cells, making it a priority to be excreted. Urea is the key route for the transportation of ammonia out of the body and is the main component of urine. Guanidine is another important nitrogen-containing molecule that is structural like urea and plays important biological roles in the stabilization of proteins. Guanidine hydrochloride and Urea are known to denature proteins and this functionality continues to be a topic of great interest. Urea and guanidine contain many hydrogen bonding sites and interactions with …

Purpose: Although hearing loss (HL) is prevalent in individuals with Down syndrome (DS), there has been little research investigating the effects of HL on language and literacy skills in this population. The purpose of this study is to document the frequency of HL in DS, as well as language and literacy outcomes, and to determine if there is a difference in language and literacy outcomes for children with DS with HL and those without HL. Parental knowledge of HL and language development was also measured. Method: A five-part survey was distributed to 77 members of DS-Connect, a National Institute of …

Cm-Dii And Mcf-7 Breast Cancer Cell Responses To Chemotherapeutic Agents, Ashten M.C. Anderson Jan 2018

Cm-Dii And Mcf-7 Breast Cancer Cell Responses To Chemotherapeutic Agents, Ashten M.C. Anderson

Honors Theses

CM-DiI is a lipophilic, red fluorescent dye used for staining and tracking the migration of cells. CM-DiI makes it possible to visualize cells in histological regions and can therefore be very useful for the tracking of cancer cell proliferation and metastasis in vivo. The ability to track and quantitate cancer cell proliferation in vivo is essential for cancer drug discovery. If CM-DiI labeled cancer cells respond to chemotherapy agents similar to unlabeled cancer cells, it facilitates screening of potential anti-cancer compounds using CM-DiI labeled cells in a xenotransplanted, transgenic zebrafish model (Danio rerio). To investigate whether CM-DiI labeling would affect …

Assembly Of A Protein-Based Hydrogel As A Vehicle For In Situ Drug Delivery, Austin Carroll Jan 2018

Assembly Of A Protein-Based Hydrogel As A Vehicle For In Situ Drug Delivery, Austin Carroll

Honors Theses

The goal of this work is to develop a self-assembling, protein-based gel to deliver drugs at the site of chronic inflammation. There are two engineered protomers that comprise the gel; calmodulin- and M13- are their critical components. Numerous enzymes have been shown to undergo calcium-dependent regulation by calmodulin, an intracellular protein highly resilient to its chemical environment. One of these enzymes, myosin light chain kinase, contains a remarkable 26 residue portion known as the M13 peptide that has been demonstrated to bind to calmodulin in the presence of calcium through hydrophobic interactions. Because the extracellular space in vivo constitutes an …

Effects Of Predation On The Biochemical Constituents Of The Bleached Soft Coral Sinularia Polydactyla, Carley Elizabeth Cole Jan 2018

Effects Of Predation On The Biochemical Constituents Of The Bleached Soft Coral Sinularia Polydactyla, Carley Elizabeth Cole

Honors Theses

Climate change can indirectly affect the level of predation on soft corals. As sea temperatures rise, more and more bleaching events are occurring, which can affect the production of chemical defenses and the concentrations of the biochemical constituents that make up the soft corals. These changes may make the soft corals more attractive to predators. This phenomenon is suggested by the environmental stress theory (EST), which states that under stressful conditions, more of an organism's energy is allocated to survival rather than to other factors, such as reproduction or predator deterrence. To test the effect of predation on already bleached …

Technique For The Synthesis Of Difluorinated Organic Molecules Using Magnesium Bases, Cassidy S. Baldwin Jan 2018

Technique For The Synthesis Of Difluorinated Organic Molecules Using Magnesium Bases, Cassidy S. Baldwin

Honors Theses

The presence of fluorine in organic molecules can confer biological advantages which are often exploited in drug development. For example, fluorine has been shown to limit drug metabolism and enhance drug distribution. However, the development of fluorinated pharmaceutics poses synthetic challenges. These challenges are especially pronounced when developing molecules with a difluoromethyl group. One difluoromethyl functional group of particular interest is the α,α-difluoro-β-amino carbonyl group, which has been seen in potential drug leads. The previous methods of accessing this group are inefficient and complex. The goal of this project was to develop a new technique to access this group by …

A Modernized Fairy Tale: Speculations On Technology, Labor, Politics, And Gender In The Oz Series, Zachary Hez Hollingsworth Jan 2018

A Modernized Fairy Tale: Speculations On Technology, Labor, Politics, And Gender In The Oz Series, Zachary Hez Hollingsworth

Honors Theses

On the surface, L. Frank Baum's Oz series would appear to merely be fourteen books of inventive children's fantasy, but in truth Baum communicates several personal progressive beliefs to his youthful audience through the use of his fantastical world upon closer examination. For my research, I reread every book in Baum's original Oz series and made note of any potentially relevant allegorical or metaphorical themes. Once I started to notice a trend of themes regarding technology, labor, politics, and gender, I settled on these themes to be the overall focus of my thesis's discussion. I read as many academic essays …

Characterizing Borrelia Burgdorferi In Virginia : Lyme Disease Prevalence As A Matter Of Bacterial Genetic Variation, Hannah Cornman

Honors Theses

yme Disease (LD) incidence and spatial distribution has grown dramatically in the past 20 years. In Virginia, the growth has been especially drastic, primarily in the Western part of the state. This research project investigates how genetic variability of the Lyme Disease causing bacteria, Borrelia burgdorferi, might be contributing to this increase in LD incidence in western Virginia. To do this, B. burgdorferi samples were isolated from ticks found across the state and were characterized by multilocus sequence typing (MLST). Phylogenetic and minimum spanning tree analyses were also performed. It was hypothesized that the increasing number of Lyme disease infections …
