The objective of this thesis was to optimize the hemolytic activity of two isolates of Clostridium septicum to evaluate if hemolytic titer of C. septicum antigen at time of formalin inactivation corresponded to increased serum antibody titer to the C. septicum alpha-toxin of turkeys immunized with an experimental formalin inactivated bacterin-toxoid vaccine. Chapter 1 provides a brief overview of the thesis. Chapter 2 consists of a literature review focused on clostridial dermatitis, specifically C. septicum-associated dermatitis (cellulitis) that affects commercial turkeys. Proper nitrogen (N) management in rice production can be difficult to achieve without the aid of tools such as soil and tissue tests. There are no tests currently available to rice producers that determine whether N has been managed properly throughout the season or that detect instances of over fertilization. Rice stalk samples were collected from N response trials within 3 days of harvest from the primary Arkansas rice growing region in 2016-2018. Stalk samples were analyzed for N concentration through a KCl extraction and spectrofluorometric quantification. Nitrogenase is the metalloenzyme only found in bacteria and archaea that is essential for biological nitrogen fixation (diazotrophy), but it can also serve as a catalyst in biofuel production. All diazotrophs contain a molybdenum (Mo) nitrogenase, while some species contain additional alternative nitrogenases where either vanadium (V) or iron (Fe) replace Mo in the active site cofactor. Nitrogen fixation by bacteria has been extensively studied. The limited investigation of nitrogen fixation in methanogenic archaea (methanogens) indicates production of nitrogenase is simpler than in bacteria and methanogen nitrogenase has different biochemical properties. In oncology clinical trials, when a treatment is administered to the patient population, a certain subset of patients may respond to the treatment while the other does not. The positive responders with long-term survival are considered “statistically cured” and can be referred to as cured patients or long-term survivors. When a proportion of patients achieve long-term survival, the hazard functions of two arms (control vs. treatment) are no longer proportional. Model compounds, 3,6,9-trithaiundecane-1,11-dicarboxylic acid (TTDPA), 2,5,8-trithianonane-1,9-dicarboxylic acid (TTDAA), and 1,11-diamide-3,6,9-trithiaundecane (TTDAce), closely related to the adducts formed by cysteine alkylation of the chemical weapon, sulfur mustard, were synthesized. It is shown that TTDPA forms complexes with key metal micronutrients: copper, nickel, cobalt, manganese, and zinc. Though the strength of binding to TTDPA varies, the complexes in many cases precipitate from solution. All metals produced a visible precipitate upon interaction with TTDPA under the conditions tested, however only Cu2+, Mn2+, and Zn2+ produced enough to be measured. Molecular dynamics (MD) simulations are routinely used to study the dynamics of proteins. However, conventional MD limited to the sampling of local conformational changes as the functionally important conformational transitions of proteins often extend beyond the timescales of the simulations employed, for example, membrane transport proteins. We have determined the combination of multiple MD based techniques that allows for a rigorous characterization of energetics and kinetics of large-scale conformational changes in membrane proteins. How communities respond to a natural hazard is influenced by how they perceive it. This dissertation evaluated the gap between intent and action regarding earthquake hazards in Muslim countries with a focus on Afghanistan and Pakistan. Cultural biases provided predictions of risk perceptions and risk-taking preferences that were often more powerful than measures of knowledge and experience. In Muslim communities, perception of risk was influenced by the teachings of Islam and related rituals, traditions, and culture.

This dissertation explores three aspects of conservation agriculture (CA) in the Sub-Saharan African region (SSA). The first article examines the demand side of CA and explores whether urban maize (Zea mays L.) consumers in the Democratic Republic of the Congo (DRC) would be willing to pay a premium for CA-produced maize flour. The second article estimates the effects CA provides to adopters and their farms in smallholder farming systems in the DRC, focusing on changes in soil properties and cowpea (Vigna unguiculata) yields. The western honey bee, Apis mellifera L., is a globally important pollinator plagued by several harmful stressors impacting colony health and survival. At least eight A. mellifera subspecies were imported and continue to be the genetic ancestors of U.S. honey bee populations today. Successive genetic bottle-neck events have led to reduced genetic diversity in U.S. honey bees. First, the subset of subspecies imported into the U.S. represents only a third of A. mellifera subspecies. Next, the parasitic varroa mite reduced managed and feral populations. Research shows that youth enrolled in formal mentoring programs often wait months before being matched with a mentor. This paper describes the development and pilot test of Caregiver-Initiated Mentoring (CG-IM), a program originally designed to equip caregivers to assist Big Brothers Big Sisters (BBBS) mentoring program staff in identifying and recruiting mentors from their social network. Using a mixed-methods design, the initial efficacy of the CG-IM program was evaluated via a small pilot test. Youth in foster care experience trauma exposure and subsequent trauma-related disorders at much higher rates than their peers, with prevalence rates comparable with veterans of war. Although youth in foster care appear to receive services at higher rates than their peers, little is known about what outpatient care consists of, particularly for trauma-related disorders. This dissertation describes transport experiments on quantum devices in layered Dirac nodal line topological semimetals and antiferromagnetic materials down to a few layers. We used gate-induced effects to alter the transport properties of these materials.

First, we introduced current annealing in topological semimetals to achieve high-quality devices. We demonstrate current annealing to substantially improve the electronic transport properties of 2D topological semimetal flakes. Contact resistance and resistivity were improved by factors up to 2,000,000 and 20,000, respectively, in devices based on exfoliated flakes of two topological semimetals, ZrSiSe and BaMnSb2. The aim of this dissertation is the investigation of thermoelectric and lattice dynamics properties of two-dimensional (2D) MX (M = Sn, Pb; X = S, Se, Te) and ZrS2 compounds based on the first-principles density functional theory. The dimensionality reduction (e.g., using 2D structure) of bulk materials is found to have enhanced thermoelectric efficiency. This enhancement is attributed to the increase of the Seebeck coefficient as a result of higher electronic density of states near the Fermi level in low-dimensional materials. Cancer cachexia (CC) is a devastating wasting syndrome characterized by marked weight loss including skeletal muscle atrophy that affects approximately 80% of cancer patients. Current therapeutic treatments including pharmacological and nutritional intervention are insufficient to prevent or reverse it. Prior studies demonstrated lower muscle mass, impaired muscle function, and mitochondrial health in the development of CC. Sustainable intensification of rice farming is crucial to meeting human food needs while reducing environmental impacts. Rice produces 8% of all anthropogenic CH4, which is a potent greenhouse gas. CH4 emissions can potentially be reduced by cultivation practices that minimize the number of days the fields are saturated, such as dry-seeding instead of water-seeding and irrigation using the alternate wetting and drying (AWD) technique instead of delayed, continuous flooding (DF). Survival under fluctuating environmental conditions, such as those increasing in frequency and magnitude under environmental change, requires a successful response to stress. Interspecific differences in stress responses may result in differential survival of species, even within a lineage. Psychopathy is a constellation of maladaptive interpersonal, affective, and behavioral features, including grandiosity, manipulativeness, emotional detachment, and impulsivity (Hare, 2003). Fearlessness, immunity to stress, self-assurance, and social dominance are considered to be adaptive features of psychopathy. Patrick and colleagues (2009) sought to reconcile differences between opposing conceptualizations of psychopathy by formulating a triarchic model of the condition. One core construct in this model, boldness, captures an ability to remain calm in the face of threat, an appetite for dangerous or risky activities, and an increased tolerance for uncertainty and danger. Healthcare-associated infections (HCAIs) affect 1.7 million hospitalized patients each year, resulting in over 98,000 deaths and anywhere from $28.4 to $45 billion in treatment costs. Furthermore, it has been established that more than 80% of these infections are caused by biofilms While HCAIs can be bacterial, viral, fungal, or parasitic, there are between 12 and 17 species cause 80 to 87% of all HCAIs. Nitrogen (N) biogeochemical cycling entails close networking between soil nutrients, plant species and resulting microbial communities, and abiotic factors such as temperature, moisture, and pH. To better understand transformations of N in soil microsites, which are known to be rich in N-cycling behavior, the microdialysis technique was employed to generate in situ data on temporal alterations of N during wetting-drying conditions in both the field and laboratory. This dissertation focused on the efficacy and validity of whole genome resequencing (WGR) for fine mapping genetic determinants of particular traits in a given organism. Previously, our research group used WGR to identify haplotype blocks of single nucleotide polymorphisms associated with ascites resistance with some as strong candidates for use in marker-assisted selection (MAS). Chapter 2 discusses the completion of a MAS project through evaluation of ascites incidence as well as production traits of economic value to poultry producers.
