Development Of A New Human Identity Testing Kit For The Simultaneous Amplification Of The 13 Codis Str Loci Plus Amelogenin Using Locus Specific Brackets (Lsbs), Lizmery Suarez Ferguson Jan 2012

Development Of A New Human Identity Testing Kit For The Simultaneous Amplification Of The 13 Codis Str Loci Plus Amelogenin Using Locus Specific Brackets (Lsbs), Lizmery Suarez Ferguson

Graduate Theses, Dissertations, and Problem Reports (ETD)

STR loci consist of short, repetitive sequence elements 2--7 base pairs in length. An STR genotype is typically determined by PCR amplification followed by fragment size estimation with electrophoresis. Commercial multiplex STR amplification kits are available for the 13 CORE loci used in the USA for human identity testing. The genotyping rationale with these multiplex STR amplification kits involves a two-fold calibration analysis: 1) An internal size standard (ISS) labeled with a separate dye establishes the size of the unknown DNA fragment and 2) an allelic ladder which contains most of the common alleles for each locus is also sized …

The Role Of Src-Homology 2 Domain-Containing Tyrosine Phosphatase 2 In Signaling And Tumorigenesis Of Basal Type/Triple Negative Breast Cancer, Fatimah Kh. Matalkah Jan 2012

The Role Of Src-Homology 2 Domain-Containing Tyrosine Phosphatase 2 In Signaling And Tumorigenesis Of Basal Type/Triple Negative Breast Cancer, Fatimah Kh. Matalkah

Graduate Theses, Dissertations, and Problem Reports (ETD)

Basal type/triple negative breast cancer (BTBC) represents a distinct tumor group that is characterized by an aggressive disease phenotype and poor clinical outcome. Patients with this subtype cannot benefit from the available anti-hormone and anti-HER2 targeted therapies (e.g. tamoxifen and herceptin). Therefore, characterizing potential therapeutic targets is urgently needed. We reasoned that the Src-homology 2 domain-containing tyrosine phosphatase 2 (SHP2) could serve as a therapeutic target in BTBC, because of its critical regulation of epidermal growth factor receptor (EGFR) which is overexpressed in BTBC. Here, we demonstrated that SHP2 knockdown in BTBC cell lines suppressed cell proliferation, migration, and transformation. …

Examination Of Novel Cardiac Mechanisms Influencing Mitochondrial Proteomes During Diabetes Mellitus, Walter Baseler

Graduate Theses, Dissertations, and Problem Reports (ETD)

Cardiac complications, including diabetic cardiomyopathy, are the leading cause of morbidity and mortality within diabetes mellitus. Mitochondrial dysfunction has been suggested as an underlying factor in the initiation and progression of the pathology. Cardiac mitochondria are characterized as two spatially distinct subpopulations within the myocyte including mitochondria located beneath the sarcolemma, termed subsarcolemmal mitochondria (SSM), and those located between myofibrils, termed interfibrillar mitochondria (IFM). Mitochondrial subpopulations have been shown to respond differently to physiological and pathological stimuli with the IFM being the most impacted in a type 1 diabetic setting. Proteomic evaluations within various models of diabetes have highlighted dynamic …

Development Of A Near-Field Magnetic Projectile Location System, Andrew D. Lowery

Graduate Theses, Dissertations, and Problem Reports (ETD)

Near-field magnetic principles and properties have been well studied and are used in a plethora of modern applications, ranging from medical applications to audio and video processing, and magnetic tracking. Current tracking applications are based in either AC or Pulsed-DC systems. Generally, AC systems have high resolution and accuracy, but perform very poorly in the presence of conducting magnetic materials. Pulsed-DC tracking has the benefit of not inducing large eddy currents in proximity to magnetic materials, thus increasing its overall accuracy. It has been suggested that pure DC systems are not feasible because they are unable to account for the …

Analog Signal Processing Elements For Energy-Constrained Platforms, Kyle Robert Mcmillan

Graduate Theses, Dissertations, and Problem Reports (ETD)

Energy constrained processing poses a number of challenges that have resulted in tremendous innovations over the past decade. Shrinking supply voltages and limited clock speeds have placed an emphasis on processing efficiency over the raw throughput of a processor. One of the approaches to increase processing efficiency is to use parallel processing with slower, lower resolution processing elements. By utilizing this parallel approach, power consumption can be decreased while maintaining data throughput relative to other more power-hungry architectures.;This low resolution / parallel architecture has direct application in the analog as well as the digital domain. Indeed, research shows that as …

Can Changes In Sr Protein Acetylation Affect Alternative Splicing?, Heather E. Knupp Dec 2011

Can Changes In Sr Protein Acetylation Affect Alternative Splicing?, Heather E. Knupp

Graduate Theses, Dissertations, and Problem Reports (ETD)

Cellular phenotype and function is determined by the cellular proteome. Within the cellular proteome there are processes that have a potential to significantly alter the composition of the proteome. One such process is called alternative splicing. Splicing is the removal of introns from a pre-mRNA sequence and the remaining pre-mRNA sequences, called exons, are combined. Alternative splicing modifies the intron-exon combination, and can form novel products from the different arrangement of the coding sequences. This process, due to the high potential for new, possibly unwanted products, is highly regulated and can occur in a tissue and developmental-specific fashion. The misreguation …
