Early embryonic development is a complex process that involves timely and quantitative control of gene transcription. Nuclear proteins, including transcription factors and chromatin remodeling proteins, are required for initiation of transcription in early embryos prior to embryonic genome activation. The nuclear transport of these proteins is mediated by transport factors such as importins. Through analysis of EST sequences from a bovine oocyte cDNA library and the database, we identified a novel transcript which shows sequence similarity to the importin α gene. Research suggests that physical activity is positively associated with mental and physical health outcomes for adults of all ages. However, despite concerted effort to increase the activity levels of adults, the rates of adults who engage in regular physical activity remain low. This study investigated metabolic activation by benzopyrenes (BP and B[e]P) in human mammary cells and modulation by chlorophyllin (CHL, a chemopreventive agent). Among 6 NHMEC strains monitored using microarrays, 54 genes were up-regulated and 11 down-regulated by signal log ratio (SLR) ≥ 1.5 on treatment with BP alone. Pre CHL + post BP+CHL treatment up-regulated the expression of 129 genes and down-regulated those of 35 genes by SLR ≥ 1.5. Studies on CYP1 gene induction and BP-DNA adduct formation among 20 NHMECs revealed wide inter-individual variations both in the induction (3-96-fold for CYP1A1 and 4-43-fold for CYP1B1, respectively) and modulation …

Producers require information about incorporating annual forages into grazing systems during months of low productivity of the cool-season species. COPD is ranked as the fourth leading cause of death with an estimated cost of {dollar}37.2 billion in the U.S. primarily due to exacerbations. Therefore, the Global Initiative for Obstructive Lung Disease (GOLD) committee has identified a need for assessing the effect of drug therapies on outcomes of exacerbations, costs, and mortality. This study used Medicaid data from West Virginia and Kentucky, and involved three phases. Phase I assessed the costs incurred due to COPD, and the additional cost burden imposed by a concomitant diagnosis of asthma. The expression of most lipogenic enzymes is regulated through the effects of dietary polyunsaturated fatty acids on transcription. However, we have observed that the regulation of Glucose-6-phosphate dehydrogenase (G6PD) by polyunsaturated fatty acids occurs through a novel posttranscriptional mechanism. RNA reporter assays were used to map the cis-acting element by measuring the regulation of each RNA reporter construct transfected into primary rat hepatocytes. All RNA reporter constructs were regulated by polyunsaturated fatty acids with the exception of RNA reporter constructs lacking exon 12. Ovine reproduction is characterized by a breeding season and an anestrous season. Seasonal anestrus is induced by increased responsiveness of the GnRH neurosecretory system to estrogen (E) negative feedback. Previous studies have shown that inhibitory A15 dopamine neurons mediate this increased sensitivity to E, however, these neurons lack E receptors. In this thesis we performed three experiments. For experiment 1 we looked at the specific sites of actions that thyroid hormones act to cause neuronal plasticity of A15 DA neurons, utilizing dual-labeled immunocytochemistry. This dissertation integrates both theoretical and empirical insights to facilitate understanding of the current obesity epidemic in WV given heterogeneity in socioeconomic, demographic and built environment characteristics of the state. In meeting this objective, county-level and individual-level health demand analyses using secondary data sources were conducted. County-level obesity differences were studied using spatial and non-spatial random and fixed effects frameworks under a panel data structure. The Effect Of A Diet Supplemented With Flaxseed Oil On The Lipid Content And Fatty Acid Profile Of Rainbow Trout (Oncorhynchus Mykiss) Muscle Tissue, Jason P. Nguyen

This experiment examined the effect of nine diets supplemented with varied combinations of flaxseed oil and vitamin E on the total lipid content and fatty acid profile of rainbow trout over four months. Mean values were tested for significant differences between groups using one-way analyses of variance.;The most flaxseed oil supplementation, 23.5%, resulted in the highest concentration of total omega-3 fat in fillets (P<0.05). This was due to the increased (P<0.05) concentration of alpha-linolenic acid (ALA). The 23.5% flaxseed oil trout had the lowest (P<0.05) proportion of saturated fatty acids and the highest amount of total unsaturated fatty acid. Unexpectedly, eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) contents significantly decreased as the experiment progressed. No significant differences were seen among total fat contents. The results indicate that feeding a diet supplemented with flaxseed oil and vitamin E produces omega-3 enhanced trout with higher ALA but lower EPA and DHA.

Agonists of the liver X receptor (LXR) prevent and decrease arterial plaque formation in experimental models of atherosclerosis. The anti-atherosclerotic effects of LXR agonists are mediated by an increase in expression of genes involved in cholesterol export. Currently, the therapeutic utility of LXR agonists is limited by the fact that these agents increase triglyceride levels in the blood and liver. These undesired effects are caused by an increase in transcription of genes controlling triglyceride synthesis, such as acetyl CoA carboxylasealpha (ACCalpha). Fee-fishing businesses in West Virginia are often characterized as small businesses, and they could benefit from connecting with larger travel packages that are more likely to attract out-of-state anglers. The objectives are: (1) identify mini-market segments based on fee-fishing experiences; (2) examine how fee-fishing mini-markets can better connect with the larger outdoor recreation markets; and (3) to use this information to identify gaps in recreational offerings and develop tourism packages in a West Virginia test market. Aluminum (Al) in acidic soils is toxic to plants, affecting growth, water uptake, and nutrient assimilation. Aluminum resistance of plants has been ascribed to organic acid exudation from roots and changes in plant physiology resulting from the arbuscular mycorrhizal (AM) fungal symbiosis. This dissertation describes four studies that address questions regarding Al resistance of mycorrhizal plants.;In chapter 2, Liriodendron tulipifera, a common tree species in northeastern U.S. forests, was inoculated with several species of AM fungi and exposed to Al. The purpose of this study was to determine the knowledge and attitudes on meat goats and the meat goat industry in West Virginia by West Virginia Extension agents and high school agricultural teachers. A descriptive research design was selected for the study. A mailed questionnaire was sent to 51 Extension agents and 88 agriculture teachers who were the accessible population. Of the 139 agriculture educators 99 responded to the survey, for a response rate of 71.2%. A large portion of the childhood population in the US is overweight, and beverage consumption may be a contributing factor. A total of 125 students participated in both Healthy Hearts and CARDIAC over two school years. Beverage consumption data from Healthy Hearts and cardiovascular risk factors data (BMI, total cholesterol, HDL, triglycerides, and acanthosis nigricans) from CARDIAC were analyzed. One percent milk was consumed the least and 100% fruit juice was consumed the most. When separated into BMI categories, 69 students were at a healthy weight (BMI85th percentile). Recently, there has been a rise in overweight in children. Overweight in children is associated with chronic diseases in childhood and adulthood. This study aimed to analyze factors that may influence weight of children. Specifically, average soft drink consumption, average hours of television/video viewing/video game playing, and mothers' BMI were compared among overweight and normal weight preschool-aged children in rural West Virginia.;Children, mean age 52 months, residing in McDowell or Mercer counties in West Virginia. Objective. The purpose of this study is to determine the nutritional knowledge of high school football players and to determine the application of this knowledge through their food choices. A total of 56.1% participants (n=32) answered 17 or more questions correctly on the nutritional knowledge questionnaire. There was a significant difference between the number correct on the nutritional knowledge questionnaire and year in school between freshman and sophomores. A significant difference was found between year in school and responses for question number seven on the diet recall between freshman and seniors, and juniors and seniors. Pipefishes and seahorses of the family Syngnathidae are important inhabitants of diminishing grass beds threatened by excessive nutrient loads, sedimentation, and chemical contaminants. This research examined the influence of these environmental factors on two sympatric pipefishes from the Chincoteague Bay, Virginia. Syngnathids are characterized by a unique mode of reproduction in which the male carries developing embryos in a placenta-like ventral brood pouch until the fry are free-swimming. Interspecific comparisons revealed differences in parental nutrient allocation to embryos. In Syngnathus fuscus, females produce nutritionally poor eggs and males implant developing embryos adjacent to blood vessels. The loose connection between eggs …

Glucose-6-phosphate dehydrogenase (G6PD) is the rate-limiting enzyme of pentose phosphate pathway and provides NADPH for de novo fatty acid biosynthesis. For the later reason, G6PD is known as a lipogenic enzyme. Insulin induces expression of G6PD mRNA via activating the PI3 kinase pathway in primary rat hepatocytes. Addition of arachidonic acid, a polyunsaturated fatty acid, in the medium interferes with the PI3 kinase pathway, and thus inhibits the insulin-mediated induction of G6PD. Arachidonic acid in the presence of insulin induces p38 MAP kinase pathway over insulin treatment alone. Activation of p38 MAP kinase by arachidonic acid interferes with the insulin …

Although the organic market has experienced rapid growth in recent decades, growth in production has not been distributed evenly across the country, instead concentrating in certain regions. Employing measures of spatial concentration and association, we identify those counties in which organic production is clustered or represents a proportion of the agricultural economy greater than would be expected by national trends. Results show that spatial clustering of organic agriculture does exist. Large location quotients for organic production were most often found for the western U.S., the Great Plains states, and New England. Clusters measured by the local Moran’s I statistic followed …

The radiographs acquired after a person is deceased are called the Post-mortem (PM) radiographs, and the radiographs acquired while the person is alive are called the Ante-mortem (AM) radiographs. This study examines relations between adolescent reports of conformity to their parents and religiosity and if these are moderated by the sex of the adolescent and parent. Self-report data was collected from 121 ninth and tenth graders enrolled in three non-metropolitan high schools. Religiosity was measured by Schumm et al.'s (1991) modified version of Gorsuch and Venable's (1983) scales. Conformity to Parents was assessed using Peterson's (Peterson, Rollins, & Thomas, 1985) Conformity to Parents scales. Four regressions were used. Poverty and food insecurity are challenging socio-economic problems that policy makers are trying to address for a long time. The intent of this study was to examine the attitudes of early childhood personnel on preschool inclusion before and after participating in a preschool inclusion class that was specifically designed for preschool teachers. The attitudes of teachers were measured using a 7 point Likert type pre and post-assessment scale. The items on the scale dealt with the inclusion of children with disabilities in a typically developing classroom. The participants in the study included 96 early childhood personnel working in West Virginia. Because lay vision screeners rarely receive formal training in how to screen vision and because older children were seen in clinic with vision disorders that should have been detected earlier through vision screening, a pediatric ophthalmologist and the researcher created the Vision Initiative for Children (VIC) in 2001 as a statewide program of the West Virginia University Eye Institute. VIC has trained, equipped, and provided ongoing support to lay individuals who are mandated or want to screen preschoolers' vision. Implementation of modern fishing techniques and hatchery technologies necessitates use of genetics in management of fishery stocks. Genetic analysis of rainbow trout, Oncorhynchus mykiss, will assist development of superior strains of a valuable aquacultured species. This study was to (1) develop polymorphic microsatellite markers to examine for variation among rainbow trout strains and develop an identification panel of microsatellites to distinguish individual strains, and (2) to identify novel genes and estimate relative gene expression profiles (expressed sequence tags from rainbow trout liver, intestine, kidney, and ovary non-normalized libraries).
