Brook trout (Salvelinus fontinalis) populations have experienced dramatic declines throughout their native range, in part, due to anthropogenic land use and habitat fragmentation. In the mid-Atlantic region, brook trout populations often occupy small, headwater habitat fragments in demographic and genetic isolation, making them vulnerable to inbreeding and genetic drift. My dissertation evaluates different methods for genetic assessment, monitoring, and management of small, isolated brook trout populations. Cells are constantly monitoring their extracellular environment for danger signals that warn of imminent threats. When cells are killed by physical or other non-infectious injuries, they release concentrated intracellular molecules into the surrounding area. These danger-associated molecules alert other cells that danger is nearby. When cells are killed by pathogens, they release both damage-associated and pathogen-associated signals. Both eukaryotic and bacterial cells sense these molecules and make threat assessments of cellular injury. Little is known about the energetic costs to insects of raising young. Honey bees collectively raise young, or brood, through a series of complex behaviors that appear to accelerate and synchronize the timing of brood maturation. These include maintaining the brood nest at warmer and consistent temperatures and the exceptional activity of "heater bees." The temperature at which juvenile insects are raised can profoundly affect their development. Monies from the sale of hunting licenses, as well as excise taxes on firearms, ammunition, and archery tackle through the Pittman-Robertson Federal Aid in Wildlife Restoration Act (PR), currently fuel a complex system of wildlife conservation via multiple levels of government. Previous studies have used the doubly labelled water (DLW) method to evaluate the total energy expenditure (TEE) during Ironman, ultra-marathon trail runs and competitive road cycling. However, the technique has not been applied to a 24-hour cross-country mountain bike event. This case study examined the TEE, cycling metrics and ad libitum nutrient/fluid intake in a trained male cyclist during a 24-hour cross-country mountain bike race. A trained male cyclist (41 y, 74.1 kg, 172.4 cm) received an oral dose of DLW prior to the 24-hour event for the calculations of TEE and water turnover (rH2O). Nude body …

Poly-N-acetylglucosamine (PNAG) is an extracellular polysaccharide consisting of β-1,6 N-acetylglucosamine monomers. PNAG is a key component of biofilms produced by diverse bacteria including Escherichia coli, Staphylococcus aureus, Yersinia pestis, Bacillus subtilis¸and many more. The structure of PNAG is highly conserved across the microbial world. Here, we demonstrate that PNAG is produced by two Borrelia species, Borrelia (Borreliella) burgdorferi and Borrelia hermsii. PNAG production in B. burgdorferi, the Lyme disease spirochete, depends on pngA (bb0620), a gene annotated to express a glycosyl hydrolase family 3 (GH3) enzyme, which is a novel pathway. Studies have been conducted on DNA cross-contamination throughout the criminal investigation process in order to evaluation the possibility that DNA from one sample could potentially influence the outcome of another. However, no published studies have examined the potential for contamination during the storage of genetic evidence after samples have been taken from crime scenes or suspects. Thermophilic microorganisms have been scientifically observed since the early nineteenth century and have spurred many questions about the limits of life and the capacity of organisms to survive extreme conditions. Decades of research on thermophile proteins and genomes have yielded several proposed correlates of temperature that may contribute to adaptation of bacteria and archaea to high temperature. The cough reflex is complex and requires integration of sensorimotor information from the central and peripheral nervous system. Cough motor output can be volitionally modified even when a strong urge-to-cough (UTC) is felt which reinforces the idea of cortical involvement in cough and opens the door to consider how manipulating attention or changing cognitive load may impact cough reflex sensitivity. Two prior studies have shown healthy individuals have reduced cough reflex sensitivity to inhaled tussigenic stimulants when completing a distracting task, presumably due to influence from increased cognitive load. Guanine nucleotide-binding proteins, or G-proteins, are crucial to cell signaling by acting as molecular switches through interactions with membrane-bound receptors. G-proteins have become of greater interest due to the possibility of novel drug targets. The G-protein alpha subunit, Gα, binds GTP before localizing to the membrane to interact with other G-protein subunits and G-protein-coupled receptors (GPCRs). Gi lacks a hydrophobic region to promote localization to the membrane, but a co-translational lipidation of the N-terminal residue stabilizes this interaction.

Patterns of cultural mountain adaptations are recorded throughout the Western Cordillera of North America. They demonstrate prolonged and complex uses of these ecosystems over the Holocene. What behaviors or circumstances in the Early Holocene transitioned mountains from inhospitable to reliable territory? This research addresses this question in the Beartooth Mountains of the Greater Yellowstone Ecosystem (GYE).

N-methyl-D-aspartate (NMDA) receptors are a member of ionotropic glutamate receptors that mediate excitatory neurotransmission across the mammalian central nervous system. NMDA receptors are organized into heterotetrameric assemblies containing two obligatory GluN1 subunits and two GluN2A-D, or GluN3A-B subunits. GluN1 and GluN3A-B bind glycine or D-serine, whereas GluN2A-D bind glutamate. The structural, functional, and pharmacological investigation of GluN1/3 NMDA receptors has been greatly hindered by their unconventional properties such as impermeability to Ca2+, insensitivity to the voltage-dependent block by Mg2+, and robust desensitization upon binding their endogenous agonist glycine. Heat production relative to dissipation rates have constrained reproductive effort and reduced fitness in short-lived, temperate birds. To determine whether heat constrains reproductive effort in long-lived taxa with low reproductive effort, we experimentally clipped plumage from tropical Gray-throated babblers (Stachyris nigriceps) to increase heat dissipation rates. Contrary to findings in short-lived species, we found no strong evidence of heat dissipation constraints on reproductive effort in our mid-elevation study. Clipped adults did not increase feeding rates compared to controls, but clipped females did spend more time incubating and brooding eggs and young. In this paper I argue that many of the philosophical problems with the concept of wilderness can be mitigated by thinking of wilderness as an aesthetic concept rather than as a real feature of the world or a special metaphysical category. In Section 1, I respond to arguments for wilderness no longer being extant in the world as well as being conceptually contradictory by examining the ways in which people continue to experience wilderness in spite of these challenges. Wildfires in the western United States have been increasing in size and severity over the last several decades, and climate forecasts suggest a continued increase in wildfire activity in the future. Large-scale wildfires often burn in a mosaic-like pattern of varying fire severity creating a heterogenous landscape with concomitant effects on vegetation communities. Research in motivation suggests that individuals are highly attuned to perceived risk and danger and tend to form groups (physical and ideological) on the basis of the threats they collectively share with like-minded others. These initial stages of threat detection and evaluation are often found to occur through subtle environmental cues. This study examined the relationship between feelings of uneasiness and political ideology through the framing of temporal cues (e.g., past- vs. future-tense).
