An increasing number of spontaneous mutations in the GABA transporter gene SLC6A1 (GAT-1) have been associated with pediatric epilepsy and neurodevelopmental disorders. The molecular mechanisms and impact of mutations on GABA homeostasis and neuronal circuit function are poorly understood. Furthermore, there are currently no validated therapeutic approaches to treat patients. A de novo mutation (S295L) in the GABA transporter gene SLC6A1 that was identified in a pediatric patient was introduced into a human cDNA construct, and mutant and wild-type transporters were expressed and studied in Xenopus oocytes by voltage clamp and radiolabeled GABA flux measurement. Prospective memory necessitates the deliberate formation and timely execution of preplanned intentions. The degree of cognitive demand required is thought to be lower for focal compared to non-focal prospective memory cues. While negatively evocative focal cues have been shown to improve prospective memory performance, it is unclear if and to what extent the cognitive reappraisal of negative emotion impacts prospective memory performance. Cognitive reappraisal is often utilized to help individuals effortfully attenuate negative emotions and reduces the chronic burden on cognitive resources. In the present study, we hypothesized that more effective cognitive reappraisal is associated with better prospective memory performance. …

This thesis investigates the impact of an AI-powered educational tool on high school student motivation in asynchronous online courses using Self-Determination Theory (SDT) to examine perceived competence, autonomy, and relatedness. A pilot study involving 129 students utilized an AI-powered automated feedback (AF) writing review tool. Data was collected using the Autonomy and Competence in Technology Adoption (ACTA) Questionnaire and teacher communications. Findings indicate that AI tools can enhance perceived competence through immediate, personalized feedback. Pre-mRNA intron splicing is essential to the production of many eukaryotic proteins, but it is relatively uncommon for sequence analysis tools to account for this process in their methods. In this dissertation I present three tools for biological sequence analysis, demonstrating the benefits that they derive from their explicit awareness of intron splicing. First, I present Mirage2, an updated implementation of the Mirage multiple sequence alignment program, which produces alignments of protein isoforms according to spliced genome mapping coordinates. Background: This study explored the concept of specialized substance use treatment services among a sample of Lesbian, Gay, Bisexual, Transgender, Queer and/or Questioning, Intersex, Asexual, and Two-Spirit (LGBTQIA2+) individuals: their definition, perceived barriers to receiving these services, and ways of improving such services. Having direct input from past and potential treatment-seeking LGBTQIA2+ individuals could help to deepen our understanding of how to meet the unique treatment needs of these populations.

The information included are a complete biological profile, any taphonomic features or pathological conditions present, and a literature review on forensic odontology due to the severe nature of the teeth. This individual is consistent with a 5'2"-5'6" adult male ranging between the ages of 35-50 of ambiguous population affinity. Evidence of osteoarthritis and bone spurs are present along the vertebral spine, clavicle, and articulation points throughout the post-cranial skeleton. Fort Missoula is a rare site of history, memory, and narrative. This project seeks to trace the complex web of identities and relationalities at the Fort through a narrative map: a method that combines site-based ethnography and qualitative storytelling relying on first-person perspective. This methodological framework reveals that Fort Missoula’s distinct character lies in its role as a living archive of cultural memory, accessible through the past, present, and future narratives of its places and spaces. Mountain aquifer groundwater systems remain one of the least understood hydrologic systems in upland hydrology. In this study, we aim to characterize bedrock groundwater system dynamics and bedrock-saprolite interactions across a variety of landscape positions and lithology. We investigate groundwater hydraulics, recharge temperature and elevation, residence times, and bedrock recharge rates across two mountainous headwater catchments in west central Montana. We used a suite of environmental tracers (3H, CFCs, SF6, and 4He) and residence time distribution models to estimate groundwater mean residence times over a range of timescales from decades to millennia. River recreation in the U.S. increased dramatically in the 1970s as boating technology and recreational skill level shifted, thus a number of federally- and state-managed river areas instituted use limits over the years to prevent the environmental and social impacts perceived to have stemmed from increased use. These river managers were then faced with the task of allocating this use between private and commercial users (allotment) and within these groups (rationing), complicated by the fact that members of the public perceive equity differently. This report and plan will: 1) review bison ecology and the history of reintroduction efforts on the reservation; 2) identify potential issues and barriers to successful reintroduction; 3) present a summary of current land policies and patterns of land use on the reservation; and 4) provide recommendations for dealing with those potential issues. In this work, we present a solution to William Thurston's edge gluing equations for Dehn fillings of hyperbolic 3-manifolds. This is done for triangulations that involve the layered solid torus. Our approach uses Farey recursive functions, and we present a Farey recursive function that provides a solution to the gluing equations for any hyperbolic Dehn filling admitting a triangulation by the layered solid torus. We provide examples that demonstrate our solution for multiple 3-manifolds, and study the roots of the corresponding Farey recursive polynomials. This is a full memoir manuscript that I wrote for my MFA thesis, which is tentatively titled Foreign Expert. In this manuscript, we follow myself as narrator as I struggle to adapt to life in Beijing (prior to the pandemic) while also recovering from a recent divorce. The goal of this project is to better understand and test theories about consumer behavior and meat consumption through faunal remains using orphaned and salvaged historic archaeological collections from three sites across Missoula, Montana, from the 1860s through the 1920s. Previous research underscored the need for properly defined units of analysis that represent what was being purchased and consumed. Presented in this study is a detailed investigation of the paragenesis and diagenetic processes of the Austin Chalk Formation in southwest Texas, utilizing a combination of core-based, petrographic, mineralogic, SEM, and geochemical based analyses. This study is conducted on a single whole rock core from an oil and gas producing well in Webb County, Texas. At this distal shelf location within the Maverick Basin, the Austin Chalk Formation consists of foraminiferal wackestones, siliceous wackestones, foraminiferal mudstones, bentonites, and foraminiferal packstone facies. Most current ecohydrologic models simplify the hydraulics that are at play in plant-water relationships by assuming that the water content within the plant is static. In reality, plants have a relative water content that varies with time in response to environmental stresses. To some extent, variations in plant relative water content are regulated by changes in the osmotic potential of water within plant cells, which contributes to the resilience of plants during periods of water shortage. Organisms living in challenging conditions, such as high elevation, must evolve a suite of physiological and genetic adaptations to survive and thrive. In this work, we present results employing the flower base method. We work in the field of extremal set theory, focusing our attention on r-uniform intersecting families. The classic Erd˝os-Ko-Rado Theorem states that, for n large enough, the largest runiform intersecting family, a so-called maximum star, consists of taking all r-sets containing a fixed element. Lemons and Palmer [31] investigated the diversity function, showing that the largest intersecting family that is the most “un-star-like” consists of taking all r-sets containing at least two members of a set of size three. brat olympics is an exploration of feminine disgust, apprehension, and disorientation contextualized through interpersonal relationships and societal shapeshifting. It pays particular attention to gender performance, surburban queerness, and strangerness. The manuscript plays on the history of the bildungsroman, but instead of one consistent singular timeline, it provides three separate attempts at a characterization of a life.
