A Derived Honey Bee Stock Confers Resistance To Varroa Destructor And Associated Viral Transmission, Thomas A. O’Shea-Wheller, Frank D. Rinkevich, Robert G. Danka, Michael Simone-Finstrom, Philip G. Tokarz, Kristen B. Healy

Faculty Publications

The ectoparasite Varroa destructor is the greatest threat to managed honey bee (Apis mellifera) colonies globally. Despite significant efforts, novel treatments to control the mite and its vectored pathogens have shown limited efficacy, as the host remains naïve. A prospective solution lies in the development of Varroa-resistant honey bee stocks, but a paucity of rigorous selection data restricts widespread adoption. Here, we characterise the parasite and viral dynamics of a Varroa-resistant honey bee stock, designated ‘Pol-line’, using a large-scale longitudinal study. Results demonstrate markedly reduced Varroa levels in this stock, diminished titres of three major viruses (DWV-A, DWV-B, and CBPV), …

Background: Automated and data-driven methods for screening using natural language processing (NLP) and machine learning may replace resource-intensive manual approaches in the usual care of patients hospitalized with conditions related to unhealthy substance use. The rigorous evaluation of tools that use artificial intelligence (AI) is necessary to demonstrate effectiveness before system-wide implementation. An NLP tool to use routinely collected data in the electronic health record was previously validated for diagnostic accuracy in a retrospective study for screening unhealthy substance use. Our next step is a noninferiority design incorporated into a research protocol for clinical implementation with prospective evaluation of clinical …

There is conflicting evidence on the role of lipid biomarkers in breast cancer (BC), and no study to our knowledge has examined this association among African women. We estimated odds ratios (ORs) and 95% confidence intervals (95% CI) for the association of lipid biomarkers—total cholesterol, high-density lipoprotein (HDL), low-density lipoprotein (LDL), and triglycerides—with odds of BC overall and by subtype (Luminal A, Luminal B, HER2-enriched and triple-negative or TNBC) for 296 newly diagnosed BC cases and 116 healthy controls in Nigeria. Each unit standard deviation (SD) increase in triglycerides was associated with 39% increased odds of BC in fully adjusted …

Background: Domestication usually has negative effects on insect performance, especially when they are reared continuously for many generations. Rejuvenation can reduce the negative effects of domestication in the parasitoid, Trichogramma brassicae Bezdenko (Hymenoptera: Trichogrammatidae) reared for 30 generations (G) on Sitotroga cerealella (Olivier) (Lepidoptera: Gelechiidae). Life table and parasitism parameters in the primary colony (G31–G45) were compared with the respective colony rejuvenated with field-collected individuals. Results: There were significant differences in life history traits between domesticated and rejuvenated populations. The highest (38.4 eggs/female) and lowest (13.3 eggs/female) fecundity was observed in rejuvenated G31 and domesticated G45, respectively. The highest values …

Spodoptera frugiperda (fall armyworm, FAW) is one of the most devastating insect pests to corn and soybean production in the Americas and is rapidly expanding its range worldwide. It is known to be hard to control either by chemical insecticide applications or by GM. Although the use of GM traits can be an effective way to control this pest, it is very rare to find native insecticidal proteins that provide the necessary level of FAW control in crop fields where FAW pressure and damage are high. Insecticidal Cry proteins sourced from Bacillus thuringiensis have been heavily utilized in the development …

Faculty Publications

The fall armyworm (Spodoptera frugiperda) is a highly polyphagous lepidopteran pest of relevant food and fiber staple crops. In the Americas, transgenic corn and cotton producing insecticidal proteins from the bacterium Bacillus thuringiensis (Bt) have controlled and reduced the damage caused by S. frugiperda. However, cases of field-evolved S. frugiperda resistance to Bt corn producing the Cry1F insecticidal protein have been documented in North and South America. When characterized, field resistance to Cry1F is linked to insertions and mutations resulting in a modified or truncated ABC transporter subfamily C2 (SfABCC2) protein that serves as Cry1F receptor in susceptible S. frugiperda. …

Spatial And Temporal Changes Of Sand Mining In The Yangtze River Basin Since The Establishment Of The Three Gorges Dam, Yugai Ma, Yingying Chai, Y. Jun Xu, Zijun Li, Shuwei Zheng

Faculty Publications

The global demand for sand and gravel is at 50 billion tons per year, far exceeding global resource capacities. It reached 7.6 billion tons in 2021 in the Yangtze River Basin (YRB), China. However, production is severely limited in the YRB. Therefore, the incongruity between the supply and demand of river sand is prominent. Wise management of decreasing sand resources in the YRB has become critical since the Three Gorges Dam became operational in 2003. This study synthesized spatial and temporal changes in sand mining activities and quantities along the Yangtze River and its major tributaries from 2004 to 2020. …

Deltaic channels are significant landforms at the interface of sediment transfer from land to oceanic realms. Understanding the dynamics of these channels is urgent because delta processes are sensitive to climate change and adjustments in human activity. To obtain a better understanding of the morphological processes of large deltaic channels, this study assessed the evolution and response mechanism of the South Channel and South Passage (SCSP) in the Yangtze Estuary between 1983 to 2018 using hydrology, multibeam echo sounding and historical bathymetry datasets. Decadal changes in riverbed volume and erosion/deposition patterns in the SCSP were assessed. The results showed that …

Water Quality And The Co2-Carbonate System During The Preconditioning Of Pacific Oyster (Crassostrea Gigas) In A Recirculating Aquaculture System, Salvador Villasuso-Palomares, María T. Gutiérrez-Wing, Carmen G. Paniagua-Chávez Dec 2022

Water Quality And The Co2-Carbonate System During The Preconditioning Of Pacific Oyster (Crassostrea Gigas) In A Recirculating Aquaculture System, Salvador Villasuso-Palomares, María T. Gutiérrez-Wing, Carmen G. Paniagua-Chávez

Faculty Publications

The continued increase of the demand for seed of the Pacific oyster (Crassostrea gigas) has driven the aquaculture industry to produce land-based hatcheries using broodstock conditioning. This has led to the need to create closed systems to control the main factors involved in reproduction (temperature and food). Additionally, reproductive synchronization of broodstocks may be considered to ensure homogeneous maturation and spawning among the organisms. In this work, we synchronized the broodstock reproductive stage of Pacific oysters in a recirculating aquaculture system (RAS) using a “preconditioning” process and evaluated the effect of the water quality and the CO2-carbonate system on preconditioned …

Evaluation Of Light-Logging Geolocators To Study Mottled Duck Nesting Ecology, Kevin M. Ringelman, Michael G. Brasher, Joseph R. Marty, David A. Butler, Stephen Mcdowell, Richard Temple Dec 2022

Evaluation Of Light-Logging Geolocators To Study Mottled Duck Nesting Ecology, Kevin M. Ringelman, Michael G. Brasher, Joseph R. Marty, David A. Butler, Stephen Mcdowell, Richard Temple

Faculty Publications

Geolocators are small devices that record and store time-stamped light levels that researchers typically use to approximate the latitude and longitude of small birds across the annual cycle. However, when geolocators are affixed to leg bands of larger-bodied birds, nest incubation by females interrupts the daily pattern of light and darkness. Thus, geolocators can provide information on nesting propensity, nest success, and renesting intensity; these demographic parameters are both difficult to measure unobtrusively and are critically important in determining population dynamics of birds, especially ducks. Here, we deployed 240 geolocators on mottled ducks Anas fulvigula in Louisiana and Texas in …

Considering the global intensification of aridity in tropical biomes due to climate change, we need to understand what shapes the distribution of drought sensitivity in tropical plants. We conducted a pantropical data synthesis representing 1117 species to test whether xylem-specific hydraulic conductivity (KS), water potential at leaf turgor loss (ΨTLP) and water potential at 50% loss of KS (ΨP50) varied along climate gradients. The ΨTLP and ΨP50 increased with climatic moisture only for evergreen species, but KS did not. Species with high ΨTLP and ΨP50 values were associated with both dry and wet environments. However, drought-deciduous species showed high ΨTLP …

Social Inequalities In Climate Change-Attributed Impacts Of Hurricane Harvey, Kevin T. Smiley, Ilan Noy, Michael F. Wehner, Dave Frame, Christopher C. Sampson, Oliver E.J. Wing

Faculty Publications

Climate change is already increasing the severity of extreme weather events such as with rainfall during hurricanes. But little research to date investigates if, and to what extent, there are social inequalities in climate change-attributed extreme weather event impacts. Here, we use climate change attribution science paired with hydrological flood models to estimate climate change-attributed flood depths and damages during Hurricane Harvey in Harris County, Texas. Using detailed land-parcel and census tract socio-economic data, we then describe the socio-spatial characteristics associated with these climate change-induced impacts. We show that 30 to 50% of the flooded properties would not have flooded …

Goal-Oriented A-Posteriori Estimation Of Model Error As An Aid To Parameter Estimation, Prashant K. Jha, J. Tinsley Oden Dec 2022

Goal-Oriented A-Posteriori Estimation Of Model Error As An Aid To Parameter Estimation, Prashant K. Jha, J. Tinsley Oden

Faculty Publications

In this work, a Bayesian model calibration framework is presented that utilizes goal-oriented a-posterior error estimates in quantities of interest (QoIs) for classes of high-fidelity models characterized by PDEs. It is shown that for a large class of computational models, it is possible to develop a computationally inexpensive procedure for calibrating parameters of high-fidelity models of physical events when the parameters of low-fidelity (surrogate) models are known with acceptable accuracy. The main ingredients in the proposed model calibration scheme are goal-oriented a-posteriori estimates of error in QoIs computed using a so-called lower fidelity model compared to those of an uncalibrated …

Although both salinity and alkalinity result from accumulation of soluble salts in soil, high pH and ionic imbalance make alkaline stress more harmful to plants. This study aimed to provide molecular insights into the alkalinity tolerance using a recombinant inbred line (RIL) population developed from a cross between Cocodrie and Dular with contrasting response to alkalinity stress. Forty-six additive QTLs for nine morpho-physiological traits were mapped on to a linkage map of 4679 SNPs under alkalinity stress at the seedling stage and seven major-effect QTLs were for alkalinity tolerance scoring, Na+ and K+ concentrations and Na+:K+ ratio. The candidate genes …

Uterine Lumen Fluid Is Metabolically Semi-Autonomous, Constantine A. Simintiras, Jessica N. Drum, Hongyu Liu, M. Sofia Ortega, Thomas E. Spencer

Faculty Publications

Uterine lumen fluid (ULF) is central to successful pregnancy establishment and maintenance, and impacts offspring wellbeing into adulthood. The current dogma is that ULF composition is primarily governed by endometrial glandular epithelial cell secretions and influenced by progesterone. To investigate the hypothesis that ULF is metabolically semi-autonomous, ULF was obtained from cyclic heifers, and aliquots incubated for various durations prior to analysis by untargeted semi-quantitative metabolomic profiling. Metabolite flux was observed in these ULF isolates, supporting the idea that the biochemical makeup of ULF is semi-autonomously dynamic due to enzyme activities. Subsequent integrative analyses of these, and existing, data predict …

Characterization Of A Stress-Enhanced Promoter From The Grass Halophyte, Spartina Alterniflora L., Sonali Sengupta, Necla Pehlivan, Venkata Mangu, Kanniah Rajasekaran, Niranjan Baisakh

Faculty Publications

Stress-inducible promoters are vital for the desirable expression of genes, especially transcription factors, which could otherwise compromise growth and development when constitutively overexpressed in plants. Here, we report on the characterization of the promoter region of a stress-responsive gene SaAsr1 from monocot halophyte cordgrass (Spartina alterniflora). Several cis-acting elements, such as ABRE (ABA-responsive element), DRE-CRT (dehydration responsive-element/C-Repeat), LTRE (low temperature-responsive element), ERE (ethylene-responsive element), LRE (light-responsive element), etc. contributed at varying degrees to salt-, drought- and ABA-enhanced expression of gusA reporter gene in Arabidopsis thaliana under the full-length promoter, pAsr11875 and its deletion derivatives with an assortment of cis-regulatory motifs. …

Angles Between Subspaces And Nearly Optimal Approximation In Gfem, Robert Lipton, Paul Sinz, Michael Stuebner

Faculty Publications

The Generalized Finite Element Methods (GFEM) are known for accurately resolving local features in heterogeneous media. Recent numerical experiments have shown that the use of local bases made from A-harmonic extensions of well chosen boundary data have nearly optimal approximation properties. In this paper we show that the explanation is geometric and given by the angle between any finite dimensional approximation space and the optimal finite dimensional approximation space introduced in (Babǔska and Lipton, 2011). The effect of the angle is quantified by an upper bound on the convergence rate. This rate is seen in the numerical simulations where local …

Smart Quantum Statistical Imaging Beyond The Abbe-Rayleigh Criterion, Narayan Bhusal, Mingyuan Hong, Ashe Miller, Mario A. Quiroz-Juárez, Roberto De J. León-Montiel, Chenglong You, Omar S. Magaña-Loaiza

Faculty Publications

The wave nature of light imposes limits on the resolution of optical imaging systems. For over a century, the Abbe-Rayleigh criterion has been utilized to assess the spatial resolution limits of imaging instruments. Recently, there has been interest in using spatial projective measurements to enhance the resolution of imaging systems. Unfortunately, these schemes require a priori information regarding the coherence properties of “unknown” light beams and impose stringent alignment conditions. Here, we introduce a smart quantum camera for superresolving imaging that exploits the self-learning features of artificial intelligence to identify the statistical fluctuations of unknown mixtures of light sources at …

The Rotor-Vibrator Plus Multi-Particle-Hole Description Of 154gd, Yingxin Wu, Aoxue Li, Feng Pan, Lianrong Dai, Jerry P. Draayer

Faculty Publications

Based on the well-known rotor-vibrator model and the particle-plus-rotor model, multi-particle-hole excitations from a collective even-even core described by the rotor-vibrator is considered to describe well-deformed even-even nuclei. Like the particle-plus-rotor model, the intrinsic Vierergruppe (D2) symmetry is still preserved in the rotor-vibrator plus multi-particle-hole description. It is shown that a series of experimentally observed 0+ states in these nuclei may be interpreted as the multi-particle-hole excitations in a complementary manner to the beta and gamma vibrations described by the rotor-vibrator model. As a typical example of the model application, low-lying positive parity level energies below 1.990 MeV in …

Niobium is one of the most studied superconductors, both theoretically and experimentally. It is tremendously important for applications, and it has the highest superconducting transition temperature, Tc=9.32 K, of all pure metals. In addition to power applications in alloys, pure niobium is used for sensitive magnetosensing, radio-frequency cavities, and, more recently, as circuit metallization layers in superconducting qubits. A detailed understanding of its electronic and superconducting structure, especially its normal and superconducting state anisotropies, is crucial for mitigating the loss of quantum coherence in such devices. Recently, a microscopic theory of the anisotropic properties of niobium with the disorder was …

Machine learning has been applied to a wide variety of models, from classical statistical mechanics to quantum strongly correlated systems, for classifying phase transitions. The recently proposed quantum convolutional neural network (QCNN) provides a new framework for using quantum circuits instead of classical neural networks as the backbone of classification methods. We present the results from training the QCNN by the wavefunctions of the variational quantum eigensolver for the one-dimensional transverse field Ising model (TFIM). We demonstrate that the QCNN identifies wavefunctions corresponding to the paramagnetic and ferromagnetic phases of the TFIM with reasonable accuracy. The QCNN can be trained …

By the end of May 2020, all states in the US have eased their COVID-19 mitigation measures. Different states adopted markedly different policies and timing for reopening. An important question remains in how the relaxation of mitigation measures is related to the number of casualties. To address this question, we compare the actual data to a hypothetical case in which the mitigation measures are left intact using a projection of the data from before mitigation measures were eased. We find that different states have shown significant differences between the actual number of deaths and the projected figures within the present …

Exploring the responses of microbial communities and their functions to marine frontal systems are the foci of marine ecologists. However, most of the existing studies have focused on only microbial communities or their functions. The relationships between microbial communities and their functions across coastal fronts therefore remain unclear. Here we studied a coastal front in the Taiwan Strait during late spring and examined the bacterial community structure, extracellular enzymatic activity (EEA), and their linkages. The results showed that the coastal front strongly delineated zones of bacterial community composition and diversity and functioned as a transitional zone rather than a hotspot …

There is considerable uncertainty regarding which ecosystems are most vulnerable to warming. Current understanding of organismal sensitivity is largely centred on species-level assessments that do not consider variation across populations. Here we used meta-analysis to quantify upper thermal tolerance variation across 305 populations from 61 terrestrial, freshwater, marine and intertidal taxa. We found strong differentiation in heat tolerance across populations in marine and intertidal taxa but not terrestrial or freshwater taxa. This is counter to the expectation that increased connectivity in the ocean should reduce intraspecific variation. Such adaptive differentiation in the ocean suggests there may be standing genetic variation …
