Increasing evidence suggests that mitoNEET, a target of the type II diabetes drug pioglitazone, is a key regulator of energy metabolism in mitochondria. MitoNEET is anchored to the mitochondrial outer membrane via its N-terminal α helix domain and hosts a redox-active [2Fe-2S] cluster in its C-terminal cytosolic region. The mechanism by which mitoNEET regulates energy metabolism in mitochondria, however, is not fully understood. Previous studies have shown that mitoNEET specifically interacts with the reduced flavin mononucleotide (FMNH2) and that FMNH2 can quickly reduce the mitoNEET [2Fe-2S] clusters. Here …

Associations of hepatitis C virus infection with Alzheimer’s disease have not been studied among higher risk, bipolar disorder patients. This population-based case-control study investigated the risks of hepatitis C virus infection among Alzheimer’s disease patients with bipolar disorder in the years preceding their Alzheimer’s disease diagnosis. We used 2000–2013 data from the Longitudinal Health Insurance Database in Taiwan. Among patients with bipolar disorder, 73 were diagnosed with Alzheimer’s disease (cases), who were compared with 365 individuals with bipolar disorder but without Alzheimer’s disease (randomly selected controls matched on sex, age, and index year with cases). The phosphatidylinositol 3-kinase (PI3K)/protein kinase B (PKB/Akt) pathway plays an important role in regulating cell proliferation, metabolism, and survival. However, the distinct roles of Akt isoforms (Akt1, Akt2, and Akt3) in pluripotent stem cell maintenance are not fully defined. Using mouse embryonic stem cells (ESCs), we show that direct inhibition of Akt activity leads to ESC apoptosis. The Akt3, but not Akt1 or Akt2, activity specifically regulates this effect. Inhibiting Akt3 also leads to a cell cycle arrest at G1 phase. These regulatory roles of Akt3 are dependent on its kinase …

Phenology reflects the interplay of climate and biological development. Early spring phenological phenomena are particularly important because the end of diapause or dormancy is related not only to heat accumulation in the early spring but also probably to winter low temperatures. Although a warmer winter can reduce overwintering mortality in many insects and plants, it also reduces the accumulation of chilling time that often triggers the end of diapause or dormancy. Position Statement: The International Society of Sports Nutrition (ISSN) bases the following position stand on a critical analysis of the literature regarding the effects of diet types (macronutrient composition; eating styles) and their influence on body composition. Throughout biology, amyloids are key structures in both functional proteins and the end product of pathologic protein misfolding. Amyloids might also represent an early precursor in the evolution of life because of their small molecular size and their ability to selfpurify and catalyze chemical reactions. They also provide attractive backbones for advanced materials. When β-strands of an amyloid are arranged parallel and in register, side chains from the same position of each chain align, facilitating metal chelation when the residues are good ligands such as histidine. An implicit assumption of speciation biology is that population differentiation is an important stage of evolutionary diversification, but its significance as a rate-limiting control on phylogenetic speciation dynamics remains largely untested. If population differentiation within a species is related to its speciation rate over evolutionary time, the causes of differentiation could also be driving dynamics of organismal diversity across time and space. Alternatively, geographic variants might be short-lived entities with rates of formation that are unlinked to speciation rates, in which case the causes of differentiation would have only ephemeral impacts. Toxoplasma gondii (T. gondii) is a microscopic, apicomplexan parasite that can infect muscle or neural tissue, including the brain, in humans. While T. gondii infection has been associated with changes in mood, behavior, and cognition, the mechanism remains unclear. Recent evidence suggests that T. gondii may harvest folate from host neural cells. Reduced folate availability is associated with an increased risk of neurodevelopmental disorders, neurodegenerative diseases, and cognitive decline. We hypothesized that impairment in cognitive functioning in subjects seropositive for T. gondii might be associated with a reduction of folate availability in neural cells. The Karamojong people of Uganda are marginalized and likely to have difficult lives. Research is needed to understand Karamojong children’s challenges, adjustment, and resiliency to help guide interventions and policies to improve their lives. Thus, 18 Karamojong 10–16-year-olds (10 girls; M = 13.33 years; SD = 1.81) were recruited from a nongovernmental organization in Tororo District, Uganda, and interviewed about their life events, coping strategies, social support, and hope. Adolescents also were verbally administered questionnaires about their life events and adjustment problems. Participants reported many negative life events (M = 9.28 of 16). Integrating spatial heterogeneity into assessments of salt marsh biogeochemistry is becoming increasingly important because disturbances that reduce plant productivity and soil drainage may contribute to an expansion of shallow ponds. These permanently inundated and sometimes prominent landscape features can exist for decades, yet little is known about pond biogeochemistry or their role in marsh ecosystem functioning. © The Royal Society of Chemistry 2017. Iron is an essential nutrient for bacteria but the reactivity of Fe2+ and the insolubility of Fe3+ present significant challenges to bacterial cells. Iron storage proteins contribute to ameliorating these challenges by oxidizing Fe2+ using O2 and H2O2 as electron acceptors, and by compartmentalizing Fe3+. Two types of iron-storage proteins coexist in bacteria, the ferritins (Ftn) and the heme-containing bacterioferritins (Bfr), but the reasons for their coexistence are largely unknown. Vaccination remains the best option to combat equine herpesvirus 1 (EHV-1) infection, and several different strategies of vaccination have been investigated and developed over the past few decades. The supply of inorganic carbon (Ci) at the site of fixation by Rubisco is a key parameter for efficient CO2 fixation in aquatic organisms including the green alga, Chlamydomonas reinhardtii. Chlamydomonas reinhardtii cells, when grown on limiting CO2, have a CO2-concentrating mechanism (CCM) that functions to concentrate CO2 at the site of Rubisco. Pseudomonas fluorescens strain 3JW1, which has a broad-spectrum antimicrobial activity, was studied to investigate whether it affects the amounts of aflatoxin B1 (AFB1) produced by Aspergillus flavus. It was found that the bacterium reduced the amounts of AFB1 in potato dextrose broth (PDB) and peanut medium by 97.8% and 99.4%, respectively. It also reduced AFB1 by ~183 μg/kg (55.8%) when applied onto peanut kernels. This strain reduced AFB1 via three mechanisms. Field experiments were conducted in a mango orchard in Croix-des-Bouquets, Haiti, to compare the effectiveness of 2 inexpensive artisanal trap types and the standard McPhail trap (Great Lakes IPM, Inc., Vestaburg, Michigan) for capturing fruit flies Anastrepha obliqua (Macquart) and Anastrepha suspensa (Loew) (Diptera: Tephitidae) and to ascertain the optimal trap density for mass trapping these flies. Two artisanal trap designs were constructed from inexpensive plastic soda bottles. Background--Plasminogen activator inhibitor type 1 (PAI-1) plays an essential role in the fibrinolysis system and thrombosis. Population studies have reported that blood PAI-1 levels are associated with increased risk of coronary heart disease (CHD). However, it is unclear whether the association reflects a causal influence of PAI-1 on CHD risk. Methods and Results--To evaluate the association between PAI-1 and CHD, we applied a 3-step strategy. Geographic origin and phenotypic variants such as coat color and body size have commonly been used to identify different species. Here, we investigate the role of song as a behavioural and genetic barrier in a contact zone between two subspecies of white-crowned sparrows (Zonotrichia leucophrys). We employed a reduced genomic data set to assess population structure and infer the history underlying divergence, gene flow and hybridization. Background: Over the last decade, several theoretical tumor-models have been developed to describe tumor growth. Oncology imaging is performed using various modalities including computed tomography (CT), magnetic resonance imaging (MRI), single-photon emission computed tomography (SPECT) and fluorodeoxyglucose-positron emission tomography (FDG-PET). Our goal is to extract useful, otherwise hidden, quantitative biophysical parameters (such as growth-rate, tumor-necrotic-factor, etc.) from these serial images of tumors by fitting mathematical models to images. These biophysical features are intrinsic to the tumor types and specific to the study-subject, and expected to add valuable information on the tumor containment or spread and help treatment plans. © 2017 World Scientific Publishing Company. Syntheses of three new chlorin e6 conjugates for PDT of tumors are reported. One of the new compounds 17 is conjugated with lysine at the 131-position, but the others are mono-conjugated 14 or diconjugated 15 with the non-amino acid species ethanolamine. Cellular experiments with the three new compounds and previously synthesized non-amino acid 152-conjugates (7-10), 131-monoconjugates 14, 16, and a 131,152-diconjugate 12 are reported. In vitro cytotoxicity experiments show that the 131-conjugates are more toxic than the 152-conjugates, and the most toxic derivative (dark- and photo-toxicity) is the 131-ethylenediamine conjugate 11. Novel infection control methods are needed to prevent further increases in the morbidity and mortality associated with hospital acquired infections. One method is to minimize bacterial transmission is to prevent bacterial growth on hospital surfaces. Copper has anti-microbial properties but is used sparingly in health care settings due to the cost associated with retrofitting surfaces with copper sheet metal or less durable foil. However, copper nanoparticles (CNPs) embedded in a polymer matrix may be similarly anti-microbial, which would offer a viable alternative to copper metal. The management of wintering North American waterfowl is based on the premise that the amount of foraging habitat can limit populations. To estimate carrying capacity of winter habitats, managers use bioenergetic models to quantify energy (food) availability and energy demand, and use results as planning tools to meet regional conservation objectives. Regional models provide only coarse estimates of carrying capacity because habitat area, habitat energy values, and temporal trends in population-level demand are difficult to quantify precisely at large scales. Insects are important components in coastal marsh ecosystems and can be used as an indicator of marsh health. Collections of insects in coastal marshes are usually made using either sweep nets or insect vacuums. Differences in these 2 methods have not been compared within the marsh ecosystem. Therefore, we compared collections made using these methods in oiled and non-oiled marshes along multiple transects on the Louisiana coast. We found that sweep net collections accounted for 5.9 times more individual arthropods and more total insect taxa when compared with the insect vacuum collections. Analytic philosophy is rediscovering Hegel. This essay examines a particularly strong thread of new analytic Hegelianism, sometimes called ‘Pittsburgh Hegelianism’, which began with the work of Wilfrid Sellars. In trying to bring Anglo-American philosophy from its empiricist phase into a more sophisticated, corrected Kantianism, Sellars moved in substantially Hegelian directions. Sellars’ work has been extended, and revised by his Pittsburgh colleagues John McDowell and Robert B. Brandom. Substance use during adolescence can lead to the development of substance use disorders and other psychosocial problems. These negative outcomes are especially likely for individuals who use substances at earlier ages and those who engage in heavier use during adolescence, behaviors which are both more common among youth at higher risk for developing a substance use disorder, such as those with a family history of substance use disorders (FH +). In the recent years, engineered nanoparticles (NPs) such as PLGA or poly-lactic-co-glycolic acid, have raised a substantial interest due to their possible medical applications in vaccination, diagnostic imaging procedures, cancer therapy or sustained delivery of drugs. The main aim of the present work is to evaluate key oxidative stress parameters in several organs following NPs administration in an animal model. Our data shows that acute oral administration of PLGA NPs induces a change in the antioxidant status in both rat liver and spleen, but may not induce oxidative stress damage to cell structures such as lipid or protein oxidation.

Evolution typically occurs in response to a suite of selective pressures. Yet, many studies of natural selection in the wild only investigate a single selective agent at a time. This can be problematic when selective agents act in non-additive ways. Here we evaluate the interactive effects of diet and predation on the evolution of body shape in the cyprinid fish Utah chub (Gila atraria). We found that both factors and the interaction between them are significant predictors of body shape. Despite their potential benefits, antibody microarrays have fallen short of performing reliably and have not found widespread use outside of the research setting. Experimental techniques have been unable to determine what is occurring on the surface of an atomic level, so molecular simulation has emerged as the primary method of investigating protein/surface interactions. Simulations of small proteins have indicated that the stability of the protein is a function of the residue on the protein where a tether is placed.
