How Do Harpacticoid Grazing Rates Differ Over A Tidal Cycle? Field Verification Using Chlorophyll-Pigment Analyses, Alan W. Decho
How Do Harpacticoid Grazing Rates Differ Over A Tidal Cycle? Field Verification Using Chlorophyll-Pigment Analyses, Alan W. Decho
Faculty Publications
Four species of meiobenthic copepods were examined for diatom feeding. Microscopic analysis of gut-pellet contents from field-collected individuals indicated frequent ingestion of diatoms by 3 harpacticoids (Scottolana canadensis, Microarthridion littorale, Cletocamptus deitersi) while occasional diatom ingestion occurred Paronychocamptus huntsmani. Frustules were usually empty and broken, indicating that contents were digested. Laboratory experiments using 14C-labeling showed assimilation of diatoms by the 3 species examined. Field grazing rate studies were conducted over different portions of a tidal cycle using fluorescent chlorophyll-pigment analysis of gut-contents. Highest diatom consumption (p<0.05) occurred just after the mudflat became exposed (i.e. Early Low Water level, ELW) for S. canadensis, while consumption at Late Low Water …0.05)>

Immunocytochemical localization and in situ hybridization techniques were used to investigate the presence of spermatid nuclear transition protein 1 (TP1) and its mRNA during the various stages of spermatogenesis in the rat. A specific antiserum to TP1 was raised in a rabbit and used to show that TP1 is immunologically crossreactive among many mammals including humans. During spermatogenesis the protein appears in spermatids as they progress from step 12 to step 13, a period in which nuclear condensation is underway. The protein is lost during step 15. An asymmetric RNA probe generated from a TP1 cDNA clone identified TP1 mRNA …

The relationship between doctors and patients in the medical and health field is a topic of recurrent discussion today. The topic is of immense interest because of the questions raised about value received for services purchased and because of the large amount of economic resource consumed. The size of the stakes are such that the debate will grow in the near future until some solution is found to remedy the value and cost for service dilemma.

Southern Pine Beetle And Fire In Wilderness Areas: The Kisatchie Hills Wilderness, Kisatchie National Forest, David Kulhavy, William G. Ross
Faculty Publications
Southern pine beetle infestations affect wilderness areas in the southeastern United States. In the Kisatchie Hills Wilderness area in Louisiana, a southern pine 'beetle outbreak resulted in widespread destruction of longleaf pine. Nest trees of the red-cockaded woodpecker, an endangered species, also were killed. Following the epidemic, a fire fueled by beetle-killed pines went through the wilderness. Forest structure, species composition, successional processes and general ecosystem function were substantially altered as a result of these two related disturbances. Most wilderness areas containing southern pines were managed for pine timber before being designated as wilderness. Bark beetle outbreaks are a predictable …

Interpreting and preaching on these Markan texts in the Epiphany Season requires attention to a series of contexts, including the narrative setting provided for these separate units and the congregational setting within which they will be interpreted. The interpretive task would seem to be to get at the promise in these epiphany texts while taking seriously the particularity of the good news and the specific setting in which the lives of hearers are planted.

Platelet-Derived Growth Factor Mrna Detection In Human Atherosclerotic Plaques By In Situ Hybridization, J. N. Wilcox, K. M. Smith, L. T. Williams, S. M. Schwartz, D. Gordon
Faculty Publications
Platelet-derived growth factor (PDGF) mRNA, and mRNA for its receptor, have been localized to specific cell types within the human atherosclerotic plaque, using in situ hybridization. The predominant cell types found to express PDGF A and B chain mRNA are mesenchymal-appearing intimal cells and endothelial cells, respectively, with little or no expression detected in macrophages. The distribution of PDGF receptor mRNA containing cells was also examined and found to be localized predominantly in the plaque intima.

Purification, Partial Characterization, And Seroreactivity Of A Genuswide 60-Kilodalton Legionella Protein Antigen, Chou-Pong Pau, B. B. Plikaytis, G. M. Carlone, I. M. Warner
Faculty Publications
A genuswide protein antigen extracted from Legionella pneumophila serogroup 1 (strain Philadelphia 1) cells was enriched by differential pelleting and ammonium sulfate precipitation and subsequently purified with a combination of high-performance size-exclusion and ion-exchange chromatography. The protein has an apparent molecular weight of 650,000 before and 63,000 after urea (5 M) treatment, as determined by size-exclusion chromatography. These proteins resolved to a single band of 60,000 after sodium dodecyl sulfate-polyacrylamide gel electrophoresis. The urea-treated protein had an isoelectric point of 5.8. This purified 60-kilodalton protein reacted with a convalescent-phase serum sample from a patient with legionellosis and rabbit immune sera …

We have investigated the mechanisms whereby equimolar quantities of ribosomal proteins accumulate and assemble into ribosomes of the yeast Saccharomyces cerevisiae. Extra copies of the cry1 or RPL16 genes encoding ribosomal proteins rp59 or L16 were introduced into yeast by transformation. Excess cry1 or RPL16 mRNA accumulated in polyribosomes in these cells and was translated at wild-type rates into rp59 or L16 proteins. These excess proteins were degraded until their levels reached those of other ribosomal proteins. Identical results were obtained when the transcription of RPL16A was rapidly induced using GAL1-RPL16A promoter fusions, including a construct in which the entire …

Dehalogenation In Marine Sediments Containing Natural Sources Of Halophenols., G. M. King
Faculty Publications
Halophenols such as 2,4-dibromophenol (DBP) occur naturally in some marine sediments, as a consequence of various animal and algal activities. In an earlier study, DBP was observed in the burrow microenvironment of the hemichordate Saccoglossus kowalewskii. At the concentrations found in the burrow lining, aerobic respiration appeared to be inhibited significantly relative to anaerobic catabolism. This effect, as well as factors contributing to the degradation of DBP, has been documented further here. Results from the addition of radiolabeled DBP to oxic and anoxic sediment slurries and growth experiments with aerobic and anaerobic enrichments suggested that aerobes did not significantly metabolize …

Determination Of The Range In Binding-Site Densities Of Rat Skin Heparin Chains With High Binding Affinities For Antithrombin., A. A. Horner, M. Kusche, U. Lindahl, C. B. Peterson
Faculty Publications
Rat skin heparin proteoglycans vary markedly in the proportions of their constituent polysaccharide chains that have high binding affinity for antithrombin. As the proportion of such chains in a proteoglycan rises, their degree of affinity for antithrombin also increases [Horner (1987) Biochem. J. 244, 693-698]. The antithrombin-binding-site densities of such chains have now been determined, by measuring heparin-induced enhancement of the intrinsic fluorescence of antithrombin and by chemical analysis for the disaccharide sequence glucuronosyl-N-sulphoglucosaminyl (3,6-di-O-sulphate), which is unique to this site in heparin [Lindahl, Bäckström, Thunberg & Leder (1980) Proc. Natl. Acad. Sci. U.S.A. 77, 6551-6555]. Antithrombin-binding-site density ranged from …

Since 1976 there has been a leveling off or slowdown in the rate of decline in coronary heart disease (CHD) mortality. The age-adjusted absolute annual rate of decline in CHD mortality rates during 1968-75 (Δ rate/100,000 population/year) was virtually identical for White males (-7.54), Black males (-7.85), and Black females (-7.20), and somewhat lower for White females (-4.25). During 1976-85, however, the secular trends diverged considerably. Age-adjusted rates continued to decline at the same annual rate for White males, while the decline was approximately half as steep for the other three race-sex groups. During 1976-85 there was also a leveling …

Explaining Policy Failure: Japan And The International Economy, 1969-1971, Robert C. Angel
Faculty Publications
This paper examines the determinants of Japan's most serious postwar blunder: failure to define and implement effective and timely countermeasures to deal with its change from deficit to surplus international monetary status during the 1969-1971 period. It concludes that intense bureaucratic compartmentalization and a lack of supra-ministerial leadership of national policy were key determinants of this failure, leaving Japan's political system dependent upon irresistible external pressure (gai-atsu), in this case from the United States, to define and force implementation of necessary policy changes. This critical but largely ignored episode illustrates a negative aspect of the traditional insulation of Japan's national …

The Purposes And Limits Of Independent Agencies, Paul R. Verkuil
The Purposes And Limits Of Independent Agencies, Paul R. Verkuil
Faculty Publications
The independent agency has been around for 100 years now, but we are still trying to understand how it best relates to the administration of government. Its popularity as an organizational mechanism is more a function of competing political forces within the legislative and executive branches than of any systematic analysis of its effectiveness. Yet one can discern reasons why independent agencies might be superior mechanisms for administering government programs if their structure and purpose are analyzed functionally. This essay proposes to do that and, in the process, reach some conclusions about both the potential and the limits of the …
Role Of The Criminal Defense Lawyer In Representing The Mentally Impaired Defendant: Zealous Advocate Or Officer Of The Court, Rodney J. Uphoff
Role Of The Criminal Defense Lawyer In Representing The Mentally Impaired Defendant: Zealous Advocate Or Officer Of The Court, Rodney J. Uphoff
Faculty Publications
This article examines a difficult question in the representation of mentally impaired criminal defendants: should counsel be obligated to inform the court of doubts about a client's competency to stand trial even though doing so may be contrary to the client's wishes or best interests? Professor Rodney J. Uphoff analyzes authorities that impose such an obligation on defense lawyers, including an American Bar Association Criminal Justice Standard and a recent decision of the Wisconsin Supreme Court, State v. Johnson. Uphoff concludes that these authorities needlessly undercut the mentally impaired defendant's right to zealous representation. He proposes an alternative ethical model …

The glycation (nonenzymatic glycosylation) of several proteins was studied in various buffiner os rder to assess the effects of buffering ions on the kinetics and specificity of glycation of protein. Incubation of RNase with glucose in phosphate buffer resulted in inactivation of the enzyme because of preferential modification of lysine residues ino r near the activsei te. In contrast, in the cationic buffers, 3-(N-morpholino)propanesulfonic acid and 3-(N-tris(hydroxymethyl)rnethylamino)- 2-hydroxypropanesulfonica cid, the kineticso f glycation of RNase were decreased 2- to 3-fold, there was a decrease in glycation of active site versus peripheral lysines, and the enzyme was resistant to inactivation by …
