The differentiation of adipocytes is regulated by the activity of a variety of transcription factors, including peroxidase proliferator-activated receptor (PPAR)-γ and C/EBPα. Our current study demonstrates that ectopic expression of STAT5A, such as that of PPAR-γ and C/EBPα, promotes adipogenesis in two nonprecursor fibroblast cell lines. Using morphologic and biochemical criteria, we have demonstrated that STAT5A and the combination of STAT5A and STAT5B are sufficient to induce the expression of early and late adipogenic markers in BALB/c and NIH-3T3 cells. Yet, the ectopic expression of STAT5B alone does not induce the expression of adipocyte genes, but enhances the induction of …

Methylation-Sensitive Binding Of Transcription Factor Yy1 To An Insulator Sequence Within The Paternally Expressed Imprinted Gene, Peg3, Joomyeong Kim, Angela Kollhoff, Anne Bergmann, Lisa Stubbs

The 5�-ends of two paternally expressed mouse genes, Peg3 and Usp29, are jointly associated with a CpG island that exhibits allele-specific methylation. Sequence comparison of the regions derived from human, mouse and cow revealed the presence of two evolutionarily conserved sequence motifs including one that is repeated multiple times within the first intron of Peg3 in all three mammals. DNA mobility shift and chromatin immunoprecipitation (ChlP) assays clearly demonstrated that this motif is an in vivo binding site for the Gli-type transcription factor YY1. The YY1-binding site contains one CpG dinucleotide, and methylation of this CpG site abolishes the binding …

Transcriptional Initiation Under Conditions Of Anoxia-Induced Quiescence In Mitochondria From Artemia Franciscana Embryos, Brian D. Eads, Steven C. Hand

In response to anoxia, embryos of the brine shrimp Artemia franciscana are able coordinately to downregulate metabolism to levels low enough to permit survival for several years at room temperature. In addition to dramatic decreases in free ATP levels and heat production, intracellular pH drops from 7.8 to 6.3 overnight. Use of isolated mitochondria to study transcriptional responses to anoxia offers several advantages: (1) the localized nature of transcript initiation, processing and degradation, all of which may be followed in organello; (2) the relatively simple cis- and trans-machinery involved and (3) the ability to provide relevant physiological treatments in vitro. …

Localization And Characterization Of Phenamil-Sensitive Na+ Influx In Isolated Rainbow Trout Gill Epithelial Cells, Scott D. Reid, G. S. Hawkings, F. Galvez, G. G. Goss

Percoll density-gradient separation, combined with peanut lectin agglutinin (PNA) binding and magnetic bead separation, was used to separate dispersed fish gill cells into sub-populations. Functional characterization of each of the sub-populations was performed to determine which displayed acid-activated phenamil- and bafilomycin-sensitive Na+ uptake. Analysis of the mechanism(s) of 22Na+ influx was performed in control and acid-activated (addition of 10 mmoll-1 proprionic acid) cells using a variety of Na+ transport inhibitors (ouabain, phenamil, HOE-694 and bumetanide) and a V-type ATPase inhibitor (bafilomycin). Background: Four multicenter Networks (GenNet, GENOA, HyperGEN, SAPPHIRe) form the National Heart, Lung and Blood Institute Family Blood Pressure Program (FBPP), to search for hypertension/blood pressure (BP) genes. The networks used different family designs and targeted multiple ethnic groups, using standardized protocols and definitions. Linkage genome scans were done on samples within each network (N = 6245 relatives). Methods: The evidence was synthesized using meta-analysis. Results: Combining ethnic groups, no region reached LOD >2, but several small peaks were identified, including chromosome 2p where two other recent reports find hypertension linkage. Like many deposit-feeding organisms, the burrowing brittlestar Amphipholis gracillima feeds on particulate organic matter in surface sediments. Microbial exopolymeric secretions (EPS) are carbohydrate-enriched polymers produced by microalgae and bacteria that bind aggregates and form dense biofilms near the sediment-water interface. EPS are assimilable by some benthic infauna and may be utilized as a significant carbon source. EPS are absorbed by some deposit-feeders, including a holothurian, and may be supplemental sources of nutrition. The burrowing brittlestar A. gracillima is a deposit-feeder that was used in a mass balance approach to model the incorporation of radiolabeled EPS by bottom feeders. Brittlestars were …

Dry ravel is a general term that describes the rolling, bouncing, and sliding of individual particles down a slope and is a dominant hillslope sediment transport process in steep arid and semiarid landscapes. During fires, particles can be mobilized by the collapse of sediment wedges that have accumulated behind vegetation. On a daily basis, particles may be mobilized by bioturbation and by small landslides. Experiments on a dry ravel flume indicate that a basic expression of the momentum equation predicts the distance traveled by particles propelled down a rough surface. The fault restoration technique of De Chabalier and Avouac [1994] is applied to an ultra-highresolution bathymetry data set from the East Pacific Rise (EPR) at 18140S. Fault offsets are calculated and subtracted from the original seafloor bathymetry to examine the morphology of the unfaulted seafloor surface within an area encompassing the ridge axis 400 [1] 1600 m in dimension. The restored topography reveals a gently sloping seafloor 200 m wide, which slopes 5 inward toward the spreading axis. We attribute this inward sloping seafloor to subsidence within the axial trough due to subsurface magmatic deflation. The vertical deformation field extracted …

Environmental Sovereignty Discourse Of The Brazilian Amazon: National Politics And The Globalization Of Indigenous Resistance, Anne Marie Todd

This analysis explores the connection between globalization and national identity in cultural expressions of environmental sovereignty. Competing claims to resources in the Brazilian Amazon reflect changing notions of state authority and the role of indigenous citizens in protection of biodiversity. The debate between Brazilian state governments and the Yanomami Indians and seringueiros (Brazilian Rubber Tapper communities) illustrates the complex interaction of national identity and expressions of local culture in a global ecological context.

Phenotypic plasticity, such as morphological and behavioral changes in response to predators, is common in larval anurans. Less is known about inducible defenses in the embryonic stages of development. We investigated the predation risk imposed by crayfish (Procambarus nigrocinctus) on southern leopard frog (Rana sphenocephala) eggs aud whether crayfish presence induces a change in the timing of hatching of R. sphenocephala eggs. We found that crayfish significantly reduce the hatching success of R. sphenocephala eggs by eating them and that eggs hatch significantly faster in the presence of crayfish than when crayfish are not present. We tested the hypotheses that potential egg predators, crayfish Procambarus nigrocinctus and dytiscid Cybister sp. larvae, would accelerate the timing of hatching and that a larval predator, dragonfly naiad Anax junius, would delay hatching in the southern leopard frog (Rana sphenocephala). We also tested the hypothesis that differences in response would be proportional to predator lethality. Our results indicate that our hypotheses were partially supported. The presence of an efficient egg predator (crayfish) induces hatching faster than a less efficient predator (dytiscid larvae). However, the presence of a larval predator (naiads) did not delay hatching. Eggs that …

A proper dietary electrolyte balance (DEB) is essential to ensure an optimum acid-base equilibrium and broiler performance. In low-CP diets, this balance can be affected by reduction of soybean meal and inclusion of high levels of synthetic amino acids. Although, some studies have related low-protein diets supplemented with amino acids and DEB, these relations are not well explained, because some research demonstrates confusion about the deficiency and balance of nutrients. Sodium and Cl are low-cost nutrients with great influence on feed conversion ratio (FCR), eggshell quality, and excreta moisture. Actual values of dietary requirements of these minerals for commercial laying hens are not well defined. These requirements were reevaluated in a factorial experiment using corn-soybean meal basal diets. No significant influence of Na and Cl levels was observed on egg production (%), egg weight (g), or feed intake (g/d), but levels of these minerals had variable effects on FCR, eggshell quality, and excreta moisture. The optimum requirement of Na changed according to the variable evaluated and level of Cl used.

During the past two decades, the United States has experienced dramatic developments in the area of biomedical research. Expanding budgets, augmented computer capabilities, and the Human Genome Project have all significantly enhanced research capabilities. Understanding the responses of white-tailed deer (Odocoileus virginianus) to controlled dog hunting can aid in the effective implementation of canine-assisted population management strategies. We examined the 24-h diel movements of 13 radio-collared female deer exposed to dog hunting on the Savannah River Site (SRS) near Aiken, South Carolina, where regulated dog hunting has occurred since 1965. We compared diel home range size, rate of travel, and distance between extreme diel locations before, during, and after hunts from 14 September-14 December 2002. Senate Bill 297, which was enacted by the Kansas legislature in 2002 and which became effective on January 1, 2003, is a substantial adoption of the Uniform Trust Code (2000) (“UTC”). The Kansas UTC is codified in a new chapter, Kansas Statutes Annotated chapter 58a. The UTC is the Uniform Law Commissioners' first effort to provide the states with a comprehensive model for codifying their law on trusts.

We counted the numbers of seals hauled out at low tide on two near-shore ledges in a protected cove in Casco Bay, Gulf of Maine a minimum of 12 times per month, for four years starting in August 1997. The highest mean monthly counts were in August (molting season) and the lowest in either January or February. Counts during pupping season (May and June) were lower than during April or July. As no mother-pup pairs were observed, these ledges are molting but not pupping ledges. Time-series analysis revealed no overall trend in the number of seals present. but did show …

Two federal circuits, borrowing from Title VII jurisprudence, recently recognized a cause of action for a disability-based hostile environment under the Americans with Disabilities Act (ADA). Neither opinion, however, considered how the analysis of a disability-based hostile environment claim under the ADA might differ from that of a race- or sex-based hostile environment claim under Title VII. This Article examines the differing theories of equality underlying the two statutes and argues that, because the statutes prohibit discrimination in fundamentally different ways, courts must resist the temptation to copy and paste Title VII doctrine into ADA hostile environment opinions. In this study, I experimentally examined how the landscape matrix influenced the movement, oviposition behavior, and spatial distribution of Anagrus columbi, a common egg parasitoid of the planthopper Prokelisia crocea. Both species exist among discrete patches of prairie cordgrass (Spartina pectinata), the sole host plant of P. crocea. Based on out-planted cordgrass pots bearing host eggs (to assess parasitism) or sticky leaves (traps for adult A. columbi), I found that the distribution of adult female A. columbi and pattern of ovipositions within a cordgrass patch were strongly matrix dependent. Female densities were 59% lower on the edge than interior of …

To date, there is a lack of well-controlled field experiments that disentangle the effects of the intervening matrix from other landscape variables (e.g., patch geography or quality) that might influence animal dispersal among patches. We performed a field experiment to investigate how the movement of a delphacid planthopper (Prokelisia crocea) among discrete patches of prairie cordgrass (Spartina pectinata) is affected by the composition of the matrix (mudflat, native nonhost grasses, and the introduced grass smooth brome [Bromus inermis]). AGAMOUS, a key player in floral morphogenesis, specifies reproductive organ identities and regulates the timely termination of stem cell fates in the floral meristem. Here, we report that strains carrying mutations in three genes, HUA1, HUA2, and HUA ENHANCER4 (HEN4), exhibit floral defects similar to those in agamous mutants: reproductive-to-perianth organ transformation and loss of floral determinacy. HEN4 codes for a K homology (KH) domain-containing, putative RNA binding protein that interacts with HUA1, a CCCH zinc finger RNA binding protein in the nucleus.
