The current investigation was an examination of the kinetic and kinematic characteristics of the back squat using accentuated eccentric loading (AEL) and cluster set programming strategies. Trained male subjects (age = 26.1 ± 4.1 years, height = 183.5 ± 4.3 cm, body mass = 92.5 ± 10.5 kg, back squat to body mass ratio = 1.8 ± 0.3) volunteered to complete four different load condition sessions involving traditionally loaded straight sets (TL), traditionally loaded cluster sets (TLC), AEL cluster sets (AEC), and AEL straight sets where only the first repetition of each set used eccentric overload (AEL1). This research was designed to determine the conditions necessary to remove c-myc and 6x-His tags from a flavonol specific glucosyltransferase found in grapefruit (CP3GT) using thrombin in preparation for crystallization. X-ray crystallography of CP3GT crystals may elucidate structural features that account for flavonol specificity in some glucosyltransferase enzymes. A thrombin cleavage site was inserted into WT CP3GT and one mutant. Recombinant CP3GT was expressed in yeast and purified. Optimal conditions for thrombin digestion were explored. Digestion with 100U of thrombin for 2 hours at 4o C was optimal for removing tags from CP3GT. Storage at 4o C for …

This study explores the rationale, efficacy, and social validity of a professional development model designed to move elementary school science activities closer to the practices of working scientists as required by the United States’ “Next Generation Science Standards.” The model is culturally sensitive and aims to create experiences with high subjective task value. In 1965, Duke Power announced the construction of a series of dams along the Keowee and Little rivers in Oconee County, South Carolina. The dams would create water reservoirs for one of the largest hydro-electric and nuclear power facilities in the nation. The dominant narrative focuses on the recreational activities, power generation, and economic development facilitated by the creation of lakes Jocassee and Keowee. However, residents of the Keowee and Jocassee valleys had to be removed, a process that started years earlier when Duke Power began serious land purchasing efforts. The purpose of this qualitative study was to determine how a university aligns the budget allocation process to the strategic plan process. This case study was conducted at a four-year, liberal arts university in the southeastern part of the United States. The qualitative data was collected through personal individual interviews from a purposeful sample of administrators at the university who were knowledgeable about the budget allocation process and the strategic plan. Data were also collected through a document review from items publically accessible online and information received from the interviews.

Hepatitis C virus (HCV) or human immunodeficiency virus (HIV) infection leads to a phenomenon of inflammaging, in which chronic infection or inflammation induces an immune aged phenotype with T cell dysfunction. Thus, HCV or HIV infection has been deemed as a model to study the mechanisms of T cell infammaging and viral persistence in humans. In this dissertation, T cell homeostasis, DNA damage and repair machineries were investigated in patients with chronic HCV or HIV infection at risk for inflammaging. Tennessee has 2 programs: the Tennessee Promise and the Tennessee Reconnect programs that offer students tuition-free access to the state’s community college system. Childhood adversity is associated with numerous negative outcomes across multiple domains, including mental and physical health, interrelationships, and social functioning. Notably, research suggests that childhood adversity has a dose-response relationship with these outcomes; that is, greater numbers of adverse experiences in childhood are associated with worse outcomes. These outcomes overlap with many risk factors of homelessness. This study sought to address two questions: 1) Does a dose-response relationship exist between childhood adversity and chronic homelessness? 2) Does childhood adversity negatively predict the age at which homelessness first occurs? Suicide is a national public health concern, and college students may be at increased risk. Symptoms of psychopathology (i.e., stress, anxiety, and depression) may contribute to risk, whereas religiosity (i.e., religious attendance, surrendering to God, and feeling forgiven by God) may reduce risk. Students from a rural southeastern university (N=249) completed self-report measures. Serial mediation analyses indicate that attendance and surrender to God are inversely- predictive of suicide risk, both directly and through the indirect pathways of feeling forgiven by God (1st order mediator) and psychopathology (2nd order mediators). In all models, specific indirect effects occurred through feeling forgiven by …

Using the lives of impaired individuals catalogued in the Íslendingasögur as a narrative framework, this study examines medieval Scandinavian social views regarding impairment from the ninth to the thirteenth century. Beginning with the myths and legends of the eddic poetry and prose of Iceland, it investigates impairment in Norse pre-Christian belief; demonstrating how myth and memory informed medieval conceptualizations of the body. This thesis counters scholarly assumptions that the impaired were universally marginalized across medieval Europe. It argues that bodily difference, in the Norse world, was only viewed as a limitation when it prevented an individual from fulfilling roles that …

The purpose of this quantitative study was to investigate educator factors that have an impact on student achievement and overall school performance as indicated in the American College Test (ACT) scores from the district and the individual schools. Educators from a moderate-sized public school district participated in an anonymous online survey. According to the Tennessee Department of Education (TDOE) Report Card, the district ACT composite is a 20.1 (TDOE, 2018e). A qualitative investigation was conducted to explore the characteristics of high quality teachers. This study used a phenomenological methodology, enabling the researcher to gather information from a purposeful selection of people directly identified as high quality teachers by their principals. This study employed processes of data collection commonly used in observational analysis and included the use of interviews found in qualitative design.

The purpose of this study was to identify existing relationships between the perceptions of novice teachers regarding their teacher preparation programs, the support given to teachers from their schools, and the purpose of school. The study took place in Northeast Tennessee. Seventeen teachers from three different school systems participated in individual qualitative interviews, which lasted about one hour each.
The finding corroborated the theory analyzed for this study and revealed areas for improvement in all levels of the education system. Teachers suggested meaningful changes to teacher preparation programs, including changes to general prerequisites and reorganizing education programs around meaningful field …

Self-control has been measured using a variety of methods including self-report measures, cognitive inhibition tasks, delay discounting and delay of gratification tasks, and persistence and willpower tasks. Although these are all theoretically linked to processes involved in self-control, recent evidence has shown that these diverse measurement techniques relate only minimally to one another. Assuming that self-control is a reflective construct, this would indicate that many of these tasks are poor indicators of self-control. The present research challenges the common assumption that self-control is a reflective construct and instead proposes that self-control is a formative construct. The purpose of this study was to assess technology use, on-the-job technology training, education levels, and salary ranges of employees in low-, middle-, and high- skill jobs in a rural county in East Tennessee to create an example of a small town workforce. For this study, technology included computers, robotics, and mobile technology. A survey determined the technology used, training provided, salaries, and job skill levels based on education obtained and required by major employers. The researcher used the framework of This We Believe and the School To Watch application process to determine schools worthy of the study. The purpose of this study is to explore the perceptions of soring in the Tennessee Walking Horse industry. Although a limited amount of research has focused on the practice, this is the first known study to utilize a criminological lens to better understand the perceptions and motivations of its use. The study relies upon data collected from Tennessee Walking Horse trainers, event attendees, and Designated Qualified Persons (DQPs), which are analyzed via a mixed-methods approach. In this thesis, we investigated T cell homeostasis and DNA damage repair machineries in HIV infection. We found that the frequencies of CD4T cells were low, which is associated with cell apoptosis in HIV patients compared to healthy subjects. Importantly, these events were closely correlated to the increase in T cell exhaustion, senescence, DNA damage, and telomere attrition. Mechanistically, while the DNA damage sensors Mer11, Rad50, and NBS1 (MRN) complexes remained intact, the ataxia-telangiectasia mutated (ATM) kinase and its downstream checkpoint kinase 2 (CHK2) were significantly inhibited during HIV infection. Additionally, telomeric repeat-binding factor 2 (TRF2) that functions to protect …

Using part of James Cook’s first voyage of discovery in which he explored the Australian coast, and Joseph Banks’s 1772 voyage to Iceland as case studies, this thesis argues that late eighteenth-century travelers used scientific voyages to present audiences at home with a new understanding and scientific language in which to interpret foreign places and peoples. As a result, scientific travelers were directly influential not only in the creation of new forms of knowledge and intellectual frameworks, but they helped direct the shape and formation of the Empire. Current practices support the use of probiotic and prebiotic supplementation to improve chronic gastrointestinal distress (GID). The aim of this study was to determine the tolerance and benefits of GlutenShield (GS), a prebiotic, probiotic, and enzyme supplement, on adults with GID. Subjects (n=20) took either GS or the placebo for 30 days and completed a pre-treatment FFQ as well as a pre- and post-treatment GID questionnaire, blood draw, and stool sample. Participants consumed more total and saturated fat, and less fiber and whole grains compared to the recommended intake. A significant reduction in IgG2 was observed in the GS group …

Peptide identification using tandem mass spectrometry depends on matching the observed spectrum with the theoretical spectrum. The raw data from tandem mass spectrometry, however, is often not optimal because it may contain noise or measurement errors. Denoising this data can improve alignment between observed and theoretical spectra and reduce the number of peaks. The method used by Lewis et. al (2018) uses a combined constant and moving threshold to denoise spectra.
