15 households in the Louisville Metro area were sampled, 7 had a patient with cystic fibrosis (CF), and 8 did not. Houses were sampled between 3 and 8 times each, and samples were collected from 123 different sites within and around the homes. These sites were categorized into the variable environment type based on ecological similarity of sites. Between 75 and 168 samples were collected from each home per visit. Recovery was examined for biogeographical patterns by environment type and season at multiple taxonomic levels. Approximately 10% of samples taken yielded Pseudomonas, and of these 61% were P. putida group, …

Spontaneous recovery in rats after spinal cord injury (SCI), and interventions that improve or impede spontaneous recovery are not well understood. During a study to characterize recovery landmarks after SCI using telemetry EMG, we discovered loss of function in telemetry implanted animals. To investigate this difference further, we implanted animals with leads in various hindlimb muscles to look for behavioral and kinematic differences in recovery. We found significant differences in both open field locomotor testing (BBB) and kinematics between implant and non-implant groups where implanted groups showed loss of coordination, as well as trends for animals implanted below the knee …

Altered metabolism has long been recognized as a defining characteristic of tumor cells. The increased glycolytic phenotype, an observation credited to Otto Warburg in 1956, is almost universal for cancer cells, making metabolic enzymes attractive targets for cancer therapy. However, anti-metabolic drugs, thus far, have not lived up to expectations as stand-alone agents to treat this disease. Unlike glycolytic enzymes that directly catabolize glucose to pyruvate, the family of 6-phosphofructo-2-kinase/fructose-2,6-bisphosphatases (PFKFBs) control the conversion of fructose-6-phosphate to and from fructose-2,6-bisphosphate, a key regulator of the glycolytic enzyme phosphofructokinase-1 (PFK-1). Autism spectrum condition (ASC) consists of a set of pervasive developmental problems marked by measurable deficits in social interaction and communication, often coupled with specific and repetitive patterns of behavior. Featured restrictions in the capability to communicate and remain attentive can directly relate to the individual’s ability to interact with others within societal norms. The biologically active derivatives of vitamin B12 (cobalamin) possess the only examples containing organometallic Co-C bonds in living systems where the cleavage of Co-C bond initiates the catalytic cycle of the enzymatic reactions. In this dissertation, the electronic structure of various intermediates and the mechanistic details of the methyl transfer reactions involved in the catalytic cycle of the methionine synthase (MetH) have been computationally investigated using hybrid quantum mechanics/molecular mechanics (QM/MM) simulations, density functional theory (DFT), and complete active space self-consistent field with second order perturbation theory (CASSCF/QDPT2) computational methodologies. Griffithsin (GRFT) is a protein derived from the red alga Griffithsia sp. off the coast of New Zealand. It binds and inactivates number of enveloped viruses including HIV-1. For a product to be developed as a microbicide, it must be safe and effective after repeated use. For initial in vivo safety testing we exposed mice repeatedly to various vaginal products and found that increased concentration of mouse serum albumin (MSA) in the vaginal lumen is predictive of epithelial damage. Repeated intravaginal administration of GRFT did not result in increased amounts of MSA, but the known epithelial disrupters nonoxynol-9 and benzylkonium …

An application of a mixture of Poisson distributions is proposed to model the discrete changes in stock price based on the minimum price movement known as `tick-size'. The parameters are estimated using the Expectation-Maximization (EM) algorithm with a constant mixing probability as well as mixing probabilities which depend on order size. The model is evaluated using simulations and real data. Both the simulated and real data show reasonable estimates. A population of the rare Southern Appalachian endemic species Lilium grayi, (Gray’s lily) Roan Mountain, TN/NC was monitored for 2 years to determine the cause and impact of an early season collapse. High concentrations of the Lilium spp. host-specific fungal phytopathogen, Pseudocercosporella inconspicua (G. Winter) U. Braun were associated with 19/20 symptomatic and 0/30 asymptomatic plants. Strength of the association between pathogen and disease and the replication of disease symptoms in 4/4 healthy hosts showed that P. inconspicua was the causal agent of the disease referred to as lily leaf spot. This study focuses on an evolving, interdisciplinary area of research involving Exercise Science and Clinical Psychology. It investigated the relationship between the perception of present-moment exertion or effort during exercise and a concept called mindfulness. Exertion is commonly measured more objectively using physiological measures (e.g., heart rate) or more subjectively using self-rated Ratings of Perceived Exertion (RPE). Mindfulness is characterized as “present-moment, non-judgmental awareness,” or “living in the present.” Despite the acknowledged benefits of physical activity, many people find it burdensome, stressful, and emotionally taxing, especially when first starting an exercise program. The prevalence of child anxiety is high and anxiety disorders are often left untreated throughout childhood and adolescence. Many studies have focused on the role of parenting in understanding the development of anxiety during childhood. Given the importance of the parent-child relationship in a child's development of emotional competence, the role of the parent in facilitating their child's emotional development was proposed here to provide a more specific method of assessing the impact of parenting on risk for developing anxiety. Lonicera maackii (amur honeysuckle) is an aggressive alien shrub that invades many habitats in the Eastern United States, including along streambanks. This study investigated the direct and indirect effects of L. maackii invasion on leaf litter decomposition in an urban stream by placing leaf litter packs of L. maackii and the native Acer saccharum (sugar maple) in stream segments invaded by or managed for L. maackii. We found L. maackii litter decomposed two times faster than native A. saccharum, and A. saccharum leaf litter supported a higher abundance of macroinvertebrates than L. maackii. The current study examined the mediating role of possible food preferences on the temperamentweight relationship among 18-month-old toddlers. Parents of 37 typically developing toddlers completed the Early Childhood Behavior Questionnaire (ECBQ). During a lab visit toddlers’ weight and recumbent length were measured and recorded. Toddlers also completed a sequential touching task to examine their ability to categorize a healthy group of foods and an unhealthy group of foods. The only temperament measure found to associate with both child weight status and food categorization was inhibitory control. The purpose of this modified replication study is to examine the relationship between the stress levels and the wellness practices of U.S. radiologic technology program directors and to determine if correlations exist between wellness practices and stress levels of the group. Additionally, relaxation activities and the feelings, characteristics, and symptoms associated with stress were considered. The design of the study was ex post facto correlational. The population of the study was U.S. radiologic technology program directors working in JRCERT accredited programs. Purpose: Currently, heart transplantation and mechanical circulatory support (MCS) are the preferred treatments for end stage heart failure. However, these therapies are associated with a shortage of donor organs, a 5-yr survival rate of 80%, and adverse events associated with long-term MCS usage. To overcome these limitations, new clinical paradigm is proposed using a combination therapy of MCS and extracellular matrix (ECM) injection therapy to promote myocardial recovery. Older adults entering a nursing home face many physical and psychosocial challenges, all at a point in life when their bodies may be least equipped to handle such adversity. Data for the present study were collected for 186 incoming residents from 6 nursing homes over the course of 1 year. Fifty-six participants were interviewed at the earliest possible time point following admission and rest-activity data were collected via actigraphy for 1 week, after which participants were interviewed a second time. One of the numerous complications of obesity is infertility. Leptin has been shown to reverse infertility; however, exact mechanism is poorly understood. Recent evidence indicates Ecadherin/ β-catenin complex, which is a structural constituent of adherens junction, is expressed in the rat ovary during folliculogenesis. We hypothesized that systemic leptin modulates the expression of E-cadherin and β-catenin in dietary-induced obese infertile rats to reverse infertility. Female Sprague-Dawley rats were fed either regular chow diet (RCD) (n=6) or high fat diet (HFD) (n=14). Glyptosaurinae is an extinct subfamily of lizards of the family Anguidae. Glyptosaurine lizards are known exclusively from the Paleogene of North America and Eurasia, reaching their peak of diversity and distribution in the Eocene. In North America these lizards are largely restricted to the intermontane basins along the Rocky Mountain range, with only sparse, indeterminately-identified skeletal elements known from outside of this region. Glyptosaurine lizards are split into two tribes: the monophyletic Glyptosaurini and paraphyletic “Melanosaurini”. Within Glyptosaurini, the most common and widespread genus is Glyptosaurus. The purpose of this quantitative study was to investigate the perceptions of K-12 teachers as they relate to the implementation of the Tennessee Educator Acceleration (TEAM) evaluation framework. Survey links were sent to 1,115 K-12 teachers from 4 Northeast Tennessee school districts. The survey achieved a 24% return rate for a total of 270 participants. The research evaluated K-12 teachers’ overall perceptions of the TEAM evaluation framework, their perceptions of changes to their lesson planning processes, their perceptions of changes in the use of instructional strategies in their classrooms, and their perceptions of changes in the amount of time needed …

Microsporidia infect invertebrate and vertebrate animals. Human pathogenic microsporidia are associated with severe disease in immunocompromised individuals, and mostly asymptomatic infection in the immunocompetent. Treatment options for microsporidiosis are limited, incompletely effective, and associated with toxicity. Furthermore, microsporidia infection of healthy individuals is poorly understood, and the consequences of asymptomatic infection have not been determined. This study was an investigation of the association between executive functioning (EF) ability and language development in the latter half of the second year. Fifty-five typically developing 18- month-olds were brought into the lab. The elicited imitation with inappropriate object substitution (EIIOS) task was used as a developmentally sensitive measure of EF. Language acquisition was assessed using a real-time word learning task as well as a parent report measure of vocabulary size (MacArthur-Bates Communicative Development Inventory: Words and Sentences Version). The yeasts Pichia pastoris and Kluyveromyces lactis were used to express several recombinant human proteins for further biochemical characterization. Two substitution variants of recombinant human enteropeptidase light chain (rhEPL) were engineered to modify the extended substrate specificity of this serine protease. Both were secreted as active enzymes in excess of 1.7 mg/L in P. pastoris fermentation broth. The substitution variant rhEPL R96Q showed significantly reduced specificities for the preferred substrate sequences DDDDK and DDDDR; however, the rhEPL Y174R variant displayed improved specificities for these substrate sequences relative to all other reported variants of this enzyme. The Late Hemphillian (latest Miocene or earliest Pliocene, 7-4.5 Ma) Gray Fossil Site in northeastern Tennessee is interpreted to represent a lacustrine paleokarst fed by a river or stream. This research focuses on the morphological and systematic relations of Nyssa endocarps (fruit pits) from the fossil site to extinct and extant Nyssa species. A combination of metric and nonmetric traits allows recognition of a new species: Nyssa grayensis sp. nov. This fossil species shares the most similarities with the extant Nyssa ogeche Bartram ex Marshall from southeast North America and the Eocene fossil Nyssa eolignitica Berry from western Tennessee. The interest in hoarding has peaked since its first clinical definition in 1996 and is evident by six television shows centered on the topic. This thesis reports the results a content analysis of two seasons (21 episodes) of the popular T.V. series A&E’s Hoarders. People rationalize hoarding in ways that both differ and overlap. Doctors, professional organizers, hoarders and their loved ones collectively frame hoarding as a medical and mental health problem. The results suggest that Americans’ perceptions of hoarding behavior has shifted from one that is deviant behavior to one that is medicalized.

This thesis examines the sit-in demonstrations that used direct action and civil disobedience to target segregation at store lunch counters. The Nashville demonstrations were the last sit-in protests to occur that are discussed in this thesis, which also examines the protests in Wichita and Greensboro. Historians argue that the Wichita and Greensboro sit-ins were the most important demonstrations of their kind. The movement in Wichita was the first protest to end segregation policies at targeted stores, and the Greensboro protests led to a direct action movement in over fifty other cities targeting lunch counters. Pseudomonas aeruginosa, a Gram-negative bacillus, commonly infects immunocompromised individuals and uses a variety of virulence factors to persist in these hosts. The posttranscriptional regulator, RsmA, plays a role in the expression of many virulence factors in P. aeruginosa. RsmA up regulates virulence factors used in colonizing hosts. However, regulation of rsmA is not well elucidated. Transposon mutagenesis was performed on P. aeruginosa containing a transcriptional rsmA-lacZ fusion to answer this question. Mutants were screened via β-galactosidase assay and transposon insertions identified via arbitrary PCR. A probable MFS transporter, we named mtpX, was one significant transposon mutant identified. …

Resveratrol is a naturally occurring phytoalexin of the stilbene family produced by various plants in response to stress, UV radiation, and fungal attack. It is primarily found in peanuts, berries, grape skin, and red wine. Resveratrol has been found to exhibit anti-cancer, anti-inflammatory, anti-aging, and anti-oxidant properties. Research indicates that diets enriched with resveratrol containing substances result in less incidence of cancer. Unfortunately, the low bioavailability and solubility has been a huge setback for its potential prospects. Human replication protein A (RPA) is an eukaryotic single-stranded DNA binding protein directly involved in a variety of DNA metabolic pathways including replication, recombination, DNA damage checkpoints and signaling, as well as all DNA repair pathways. This project presents 2 novel roles of RPA in the cellular response to DNA damage. The first elucidates the regulation of RPA and p53 interaction by DNA-dependent protein kinase (DNA-PK), ataxia telangiectasia mutated (ATM) and ATM- and Rad3-related (ATR) in homologous recombination (HR).
