Towards Structural Determination Of Human Α1-Glycine Receptor Allostery, Rathna Jyothi Veeramachaneni Jan 2016

Towards Structural Determination Of Human Α1-Glycine Receptor Allostery, Rathna Jyothi Veeramachaneni

Electronic Theses and Dissertations

Recent advances in technology have led to the determination of numerous notable structures of membrane proteins. While they provide valuable information about the structure of membrane proteins these studies often provide static images with potentially limited dynamics, and structural determination often requires truncation of flexible regions, and often utilizes bacterial homologs given the need for stable, heterologous overexpression. In order to better understand allostery at a molecular level, state-dependent crosslinking studies coupled with multidimensional mass spectrometry (MS) were conducted on glycine receptor (GlyR) stabilized in different allosteric states. Predominant allosteric states were stabilized using wild type or mutated receptor in …

The Ecology And Evolution Of Rare, Soil Specialist Astragalus Plants In The Arid Western U.S., Joseph M. Statwick Jan 2016

The Ecology And Evolution Of Rare, Soil Specialist Astragalus Plants In The Arid Western U.S., Joseph M. Statwick

Electronic Theses and Dissertations

Organisms that specialize in uncommon habitats are, by their very nature, inherently uncommon. Specialization has its advantages, namely reduced competition and predation, but it also incurs costs. Specialists often have small population sizes, narrow ranges, and fragmented habitat, all of which engender negative consequences on an evolutionary timescale. Herein, I examine benefits and costs of specialization in selenium-hyperaccumulating plants in the genus Astragalus (Fabaceae). These plants are disproportionately likely to be rare and of conservation concern. Thus, I optimized germination pretreatments for Astragalus species such that seed loss can be minimized during ex situ cultivation, and found that physical scarification …

Moving From The Shadows: Shedding Light On Mixed-Citizenship Status Latino Families And Emotional Well-Being—A Mixed Methods Study, Ashley-Marie Vollmer Hanna

Electronic Theses and Dissertations

Throughout history and in contemporary U.S. society, immigration policies and practices have been laced with racist nativism, benefiting the dominant (white, male elite) society at the expense of Immigrants of Color (Chavez, 2008; Hanna & Ortega, 2016; Huag, n.d.; Ortega, Hanna, & Haffejee, 2014; Pérez-Huber, 2008). Today, Latinos are particularly vulnerable as they are the target of racist nativist immigration policy and practices and anti-immigrant rhetoric.

This dissertation examines the emotional well-being of Latino youths and young adults in mixed-citizenship status families using mixed methods research methodology. The study is informed by Critical Race Theory (CRT) and Latina/o Critical Race …

Living Divided No More: An Exploration Of Authenticity In Philosophically Inspired Schools, Melanie Reiser Jan 2016

Living Divided No More: An Exploration Of Authenticity In Philosophically Inspired Schools, Melanie Reiser

Electronic Theses and Dissertations

This study explores school authenticity (SA) by conducting research at three philosophically inspired schools. The study investigates consciousness to identity, integrity to that identity, and ascertains characteristics of authenticity across settings through an investigation of the school's congruence between the philosophy and the school ecology. Two frameworks are utilized to explore school congruence. The first framework addresses establishment of identity--the school's attention to and congruence between the philosophy and their identified core educational principles. The second investigates integrity by exploring adherence to identity--in other words, the school's attention to and congruence between educational principles and the school ecology. I explore …

Pathways From Childhood Abuse To Positive Adapation: The Moderating Roles Of Social Support And Coping Style, Sarah Elizabeth Cleary Jan 2016

Pathways From Childhood Abuse To Positive Adapation: The Moderating Roles Of Social Support And Coping Style, Sarah Elizabeth Cleary

Electronic Theses and Dissertations

Studies show that up to 50% of children worldwide are affected by physical, emotional, sexual abuse and/or neglect. While these traumatic events can have profound consequences on development across the lifespan, it is important to note that approximately 20-30% of childhood abuse survivors do not report negative impacts. One explanation for this difference in outcomes is the concept of resilience, defined as successful adaptation in spite of the experience of high-risk trauma. Many studies have been conducted to delineate factors fostering resilience. Some researchers argue that individuals' ability to achieve resilience is a direct result of the environment, while others …

Missed Appropriations: Uncovering Heidegger's Debt To Kierkegaard In Being And Time, Kenneth David Geter Jan 2016

Missed Appropriations: Uncovering Heidegger's Debt To Kierkegaard In Being And Time, Kenneth David Geter

Electronic Theses and Dissertations

It is widely held that Martin Heidegger appropriated several existential concepts from Søren Kierkegaard in his 1927 work, Being and Time. Most scholars agree that Heidegger did not sufficiently credit Kierkegaard. What was the extent of the appropriation, and why did Heidegger not duly cite Kierkegaard? This work will focus on the concept of anxiety which appears throughout Being and Time and which was influenced by the concept of the same name presented in Kierkegaard's 1844 work The Concept of Anxiety. It will also be seen how the structure of Being and Time closely resembles that of Concept …

Electronic Theses and Dissertations

This research set out to continue the exploration and diversification of excited state o-azaxylylene cycloadditions, utilizing both pre-photochemical and post-photochemical modifications.

Pre-photochemical modifications came through the addition of heteroatoms, namely nitrogen and oxygen, to the tether that links the unsaturated photopendant to the photogenerated o-azaxylylene. Photochemistry resulted in the formation of new, interesting N, O, S polyheterocycles.

Post-photochemical modifications took place through several different cycloaddition reactions, including a [4+2] hetero-Diels Alder reaction, a [4+2] Povarov cyclization, a [3+2] nitrile oxide addition, and a [3+2] nitrone cyclization. These reactions were applied first to a model system, and then to …

The Online Dating Preferences Of Chinese American Older Adults, Lin Jiang Jan 2016

The Online Dating Preferences Of Chinese American Older Adults, Lin Jiang

Electronic Theses and Dissertations

In recently years, there has been a dramatic increase in the use of online dating websites among adults over age 50. Nevertheless, samples collected in existing literature are mainly centered on White/Caucasian Americans. Furthermore, the only theoretical guide has been sexual strategies theory. This study fills the gap by using sexual strategies theory and acculturation theory to test Chinese American older adults dating preferences.

The sexual strategies theory discusses how males and females use different mating strategies in short-term and long-term contexts. Acculturation theory describes changes in which individuals or groups adopt another culture when it makes contact with their …

The Limits Of Human Flourishing: Reconstructing Theological Anthropology In Light Of Cognitive Disability, David N. Scott Jan 2016

The Limits Of Human Flourishing: Reconstructing Theological Anthropology In Light Of Cognitive Disability, David N. Scott

Electronic Theses and Dissertations

The concept of human flourishing currently holds a position of prominence within Christian theology. Numerous theologies assert that a person enjoys the fullness of humanity, not through conformity to an essential nature, but through being a living demonstration of what human being can become. This approach to theological anthropology has proven especially useful for advocacy on behalf of marginalized groups. Nevertheless, because this approach identifies human being with the capacity for purposive agency, it remains incapable of affirming the full humanity of persons with profound cognitive disabilities. These persons lack abilities that purposive agency presupposes, such as self-representation, language, and …

The Spatial Distribution Of Terrestrial Stable Carbon Isotopes In North America, And The Impacts Of Spatial And Temporal Resolution On Static Ecological Models, Sydney M. Firmin Jan 2016

The Spatial Distribution Of Terrestrial Stable Carbon Isotopes In North America, And The Impacts Of Spatial And Temporal Resolution On Static Ecological Models, Sydney M. Firmin

Electronic Theses and Dissertations

Due to the unique spatial and temporal characteristics of ecological phenomena, the extent and grain size of spatial data sets essentially filter the observations. This thesis examines the impacts of temporal and spatial resolution on the modeling of terrestrial stable carbon isotopic landscapes (isoscapes). I model the distribution of leaf stable carbon isotope composition (delta13C) for the continent of North America at multiple temporal and spatial resolutions. I generate each delta13C isoscape variation by first predicting the relative abundance of C3/C4 vegetation cover using monthly climate grids, crop distribution/type grids, and remote sensing data of plant …

Fluorine Dating Of Human Bone At The Pre-Columbian Maya Cemetery Of Caves Branch Rockshelter, Belize, Morgan Isaacs

Electronic Theses and Dissertations

This thesis tests the ion-selective electrode fluorine dating method on the remains from the Maya mortuary site Caves Branch Rockshelter (CBR) in Belize. CBR is a cemetery containing at least 400 burials from the late Preclassic to the Postclassic periods. The intensive use and reuse of the site has disturbed the burial matrices, making it difficult to seriate the burials. Fluorine dating analyzes the amount of fluorine that has accumulated in bone over time. In principle, an older burial will contain more fluorine from groundwater than a more recently buried bone; however, this principle must be tested at each site …

Using Fluorescence To Characterize Four Day Simulated Distribution System Trihalomethane Content In Florida Groundwaters, Jonathan Ousley

Electronic Theses and Dissertations

The United States Environmental Protection Agency (USEPA) regulates public water systems and has established limits for certain disinfection by products (DBPs) that have been linked to health effects, such as bladder cancer. The regulation of DBPs, specifically total trihalomethanes (TTHMs) and haloacetic acids (HAAs), have encouraged water treatment professionals to assess the type and amount of organic precursors in their supplies. Three of the more common water quality parameters that are monitored as DBP surrogates include dissolved organic carbon (DOC), ultraviolet absorbance (UV254), and specific ultraviolet absorbance (SUVA). Although DOC, UV254, and SUVA have been effectively correlated to DBP formation, …

Vo-Ohpic Treatment Reduces Cardiac Remodeling In Doxorubicin-Induced Cardiomyopathy, Taylor Johnson

Electronic Theses and Dissertations

Doxorubicin (Doxo) is one of multiple anthracycline drugs used to effectively treat various forms of cancer. Unfortunately, Doxo treatment, as a side effect, induces cardiomyopathy and subsequent heart failure. We have previously demonstrated that transplanted embryonic stem (ES) cells and their conditioned medium (CM) modulate the PTEN pathway and reduce apoptosis, fibrosis and hypertrophy in a Doxo induced cardiomyopathy (DIC) model. However, mechanisms of inhibited apoptosis mediated through PTEN pathway are completely unknown. Therefore, we used VO-OHpic (VO), a potent PTEN inhibitor to understand the mechanism of apoptosis as well as its effect on cardiac remodeling in DIC. Animals were …

Structural And Functional Characterization Of Enzymes Of A Novel Group Of Tryptophylquinone Cofactor Containing Oxidases, Esha Sehanobish

Electronic Theses and Dissertations

Protein-derived cofactors are redox and catalytic centers that are generally formed by the posttranslational modifications of one or more amino acids. An important class of these cofactors are the quinones derived from tyrosine and tryptophan. Amongst redox proteins, it has been known till now that oxidases either contain a flavin or a tyrosylquinone cofactor, whereas tryptophylquinone ones are present within the dehydrogenases. In recent times, oxidases from a marine bacterium, Marinomonas mediterranea, have been shown to possess the latter. This study involves the characterization of two such proteins, lysine-?-oxidase (LodA) and glycine oxidase (GoxA). They have been reported to contain …

Probabilistic-Based Computing Transformation With Reconfigurable Logic Fabrics, Mohammed Alawad Jan 2016

Probabilistic-Based Computing Transformation With Reconfigurable Logic Fabrics, Mohammed Alawad

Electronic Theses and Dissertations

Effectively tackling the upcoming "zettabytes" data explosion requires a huge quantum leap in our computing power and energy efficiency. However, with the Moore's law dwindling quickly, the physical limits of CMOS technology make it almost intractable to achieve high energy efficiency if the traditional "deterministic and precise" computing model still dominates. Worse, the upcoming data explosion mostly comprises statistics gleaned from uncertain, imperfect real-world environment. As such, the traditional computing means of first-principle modeling or explicit statistical modeling will very likely be ineffective to achieve flexibility, autonomy, and human interaction. The bottom line is clear: given where we are headed, …

Passive, Wireless Saw Ofc Strain Sensor And Software Defined Radio Interrogator, James Humphries Jan 2016

Passive, Wireless Saw Ofc Strain Sensor And Software Defined Radio Interrogator, James Humphries

Electronic Theses and Dissertations

Surface acoustic wave (SAW) devices have exhibited unique capabilities to meet the demands for many applications due to the inherent properties of SAW devices and piezoelectric materials. In particular, SAW devices have been adapted as sensors that can be configured to operate both passively and wirelessly. SAW sensors can be operated in harsh environmental extremes where typical sensor technologies are not able to operate. Because the sensors are passive, a radio transceiver is required to interrogate the sensor and receive the reflected response that has been modulated by the SAW device. This dissertation presents the design of a passive, wireless …

Integrating Spray Aeration And Granular Activated Carbon For Disinfection By-Product Control In A Potable Water System, Angela Rodriguez

Electronic Theses and Dissertations

Public water systems add disinfectants in water treatment to inactivate microbial pathogens. Chlorine, when used as a disinfectant, reacts with natural organic matter in the water to form trihalomethane (THM) and haloacetic acid (HAA5) disinfection by-products (DBPs), which are suspected carcinogens. The Safe Drinking Water Act's Disinfectant and Disinfection By-Product (D/DBP) Rules were promulgated by the U.S. Environmental Protection Agency to regulate the amount of DBPs in water systems. Regulatory compliance is based on maximum contaminant levels (MCL), measured as a locational running annual average (LRAA), for total THM (TTHM) and HAA5 of 80 µg/L and 60 µg/L, respectively. Regulated …
