Does Thermotolerance In Daphnia Depend On The Mitochondrial Function?, Rajib Hasan Aug 2019

Does Thermotolerance In Daphnia Depend On The Mitochondrial Function?, Rajib Hasan

Electronic Theses and Dissertations

Thermotolerance limit in aquatic organism is set by the ability to sustain aerobic scope to sudden temperature shifts. This study tested the genetic and plastic differences in thermotolerance of Daphnia that can be explained by the differences in the ability to retain mitochondrial integrity at high temperatures. Five genotypes with different biogeographic origins were acclimated to 18ᵒC and 25ᵒC. We developed a rhodamine 123 in-vivo assay to measure mitochondrial membrane potential and observed higher fluorescent in heat damaged tissues as the disruption of the mitochondrial membrane potential. Significant effects on temperature tolerance were observed with CCCP …

Purification Of Biomass Hydrolyzates For The Promotion Of Xylose Diester Formation, Laura Fernandez Aug 2019

Purification Of Biomass Hydrolyzates For The Promotion Of Xylose Diester Formation, Laura Fernandez

Electronic Theses and Dissertations

The application of a novel xylose isolation procedure using phenyl boronic acid esterification has been limited to the hydrolyzates of low extractive biomasses like hardwoods and agricultural resides. Hydrolyzates with lower sugar and higher extractive concentrations, like softwoods, have not yielded xylose, or even xylose diester (XDE), the first intermediate product in the new process, under the same conditions. This research applied evaporation concentration, adsorbent separation and membrane filtration to these lower performing biomass hydrolyzates to maximize the yield of the XDE formation step of the new xylose extraction process. Lab-scale tests of these techniques were applied to pine hydrolyzate …

Fluid Flow Analysis Of Diluted Evaporating American Whiskey Droplets., Martin J. Brown Vi Aug 2019

Fluid Flow Analysis Of Diluted Evaporating American Whiskey Droplets., Martin J. Brown Vi

Electronic Theses and Dissertations

The recent discovery of the unique structures, whiskey webs, formed when the evaporation of diluted American whiskey, has raised many questions as to the nature of the structures. Their formation process follows as such: (1) dilution of the whiskey to form nanoaggregates, (2) formation of a monolayer at the air-liquid interface, (3) chaotic ethanol evaporation caused monolayer collapse (via dynamic pressure), (4) bulk fluid evaporation caused monolayer collapse (via reduction of surface area), where the web-like structures reside on the surface, and finally, (5) web-like structures deposit on the substrate. The webs imaged via SEM had a striking resemblance to …

Comparative Spending Of Medicaid Dollars On Child Participants Of Kentucky’S Sobriety Treatment And Recovery Teams Program Versus A Matched Comparison Group., Matthew Thomas Walton

Electronic Theses and Dissertations

Child protective services agencies have long observed the complicating role that parental substance use and addiction plays in cases of child maltreatment. Families who struggle with these problems present unique challenges for child welfare professionals. These families are typically more difficult to engage, more likely to have children removed from the home, and have poorer outcomes when compared to other families. These poorer outcomes often include health problems. Addiction has well-known effects on health, and the specific manifestations of these problems for parents have been documented for years in child protection casework. However, what has been less investigated are the …

An Integrated Model Of Eating Disorder Risk And Protective Factors: Implications For Research And Treatment, Leslie Alison Davis-Waddle Aug 2019

An Integrated Model Of Eating Disorder Risk And Protective Factors: Implications For Research And Treatment, Leslie Alison Davis-Waddle

Electronic Theses and Dissertations

Body dissatisfaction and disordered eating behaviors are significant public health concerns, especially among college-age women. Despite extensive research, the complex nature of disordered eating and its etiology has prevented a satisfactory predictive model from being developed and validated and has therefore hindered the development of effective prevention and intervention strategies. This study aimed to integrate four of the most common etiological theories into a model to predict disordered eating behaviors. Bivariate correlations and moderation analyses were conducted to determine the strength of relationships between variables and to determine whether a protective factor (i.e., self-compassion) moderates the effects of risk factors …

Peroxide Sensing Using Nitrogen-Doped And Platinum Nanoparticle-Modified Screen-Printed Carbon Electrodes, Chidiebere Ogbu

Electronic Theses and Dissertations

Nitrogen-doped carbon materials have garnered much interest due to their abilities to behave as electrocatalysts for reactions important in energy production (oxygen reduction) and biosensing (hydrogen peroxide reduction). Here, we demonstrate fabrication methods and determine electrocatalytic properties of nitrogen-doped screen-printed carbon (N-SPCE) electrodes. Nitrogen doping of graphite was achieved through a simple soft-nitriding technique which was then used in lab-formulated screen-printing inks to prepare N-SPCEs. N-SPCEs displayed good electrocatalytic activity, reproducibility and long term stability towards the electrochemical reduction of hydrogen peroxide. N-SPCEs exhibited a wide linear range (20 µM to 5.3 mM), reasonable limit of detection of 2.5 µM, …

The Effects Of Urbanization On Avian Seed Dispersal Success Of Toxicodendron Radicans (Anacardiaceae), Aiden M. Stanley Aug 2019

The Effects Of Urbanization On Avian Seed Dispersal Success Of Toxicodendron Radicans (Anacardiaceae), Aiden M. Stanley

Electronic Theses and Dissertations

Urbanization is increasing at a dramatic rate as the human population increases. While it is well-known that urbanization tends to decrease species diversity (i.e., biotic homogenization), it is not known how urbanization affects the frequency and efficiency of species interactions. Seed dispersal is a plant-animal interaction that depends on disperser feeding rate, disperser diversity, probability of seed dispersal and germination. How these factors are affected by urbanization however is unknown. In this study, we evaluate how urbanization alters these factors. Urban sites had 2x higher feeding rate and 3x higher number of disperser species. The probability of seed dispersal however …

The Distinct Expressions Of Integrins Αdβ2 And Αmβ2 Differently Regulate Macrophage Migration In 3d Matrix In Vitro And In Tissue During Inflammation, Kui Cui Aug 2019

The Distinct Expressions Of Integrins Αdβ2 And Αmβ2 Differently Regulate Macrophage Migration In 3d Matrix In Vitro And In Tissue During Inflammation, Kui Cui

Electronic Theses and Dissertations

Chronic inflammation is an essential mechanism during the development of cardiovascular and metabolic diseases. The outcome of diseases depends on the balance between the migration and accumulation of macrophages in damaged tissues. Macrophage motility is highly regulated by adhesive receptors, integrins. Namely, intermediate expression of integrin supports macrophage migration, while a high integrin density inhibits it. Our studies are focused on evaluation of the contribution of related integrins αDβ2 and αMβ2 to macrophage migration and development of chronic inflammation.

We found that integrin αDβ2 is upregulated on M1-macrophages in vitro and …

Exploring The Moderating Effect Of Maternal Scaffolding On The Temperament - Language Development Relationship, Chelsea L. Robertson

Electronic Theses and Dissertations

Many studies have examined the relationship between a child’s temperament and its effect on his or her early language development. However, few studies have investigated the detrimental effects a child’s negative affectivity may have on their language development and potential ways these effects may be limited through parental behaviors. The current study aimed to investigate if physical or verbal maternal scaffolding behaviors moderated the effect negative affect has on language development. Although it was expected that maternal encouragement of physical activity would play a moderating role in the relationship between temperament and language development, no such relationship was found. One …

Self-Compassion And Suicide Risk In Veterans: Serial Effects Of Shame, Guilt, And Ptsd, Jessica Mckinney Aug 2019

Self-Compassion And Suicide Risk In Veterans: Serial Effects Of Shame, Guilt, And Ptsd, Jessica Mckinney

Electronic Theses and Dissertations

Suicide is a significant public health concern and ranks as the 10th leading cause of death in the U.S. Veterans are at a disproportionately higher risk for suicide, due to risk factors such as exposure to trauma and its negative cognitive-emotional sequalae, such as PTSD, shame, and guilt. However, not all veterans exposed to traumatic events, or who experience shame and guilt, die by suicide, perhaps as a result of the presence of individual-level protective factors such as self-compassion. Conceptualized as self-kindness, mindfulness and common humanity, self-compassion is beneficially associated with mental and physical health, including reduced suicide risk. …

Evaluating Fluid Fluxes From Deep-Sea Seepage Habitats, Leigha E. Peterson Jul 2019

Evaluating Fluid Fluxes From Deep-Sea Seepage Habitats, Leigha E. Peterson

Electronic Theses and Dissertations

The permanently dark deep-sea, located at oceanic water depths greater than 200 m, represents the largest potential habitat space on Earth. The physicochemical conditions of the planet’s largest biome are tightly coupled to the exchange of matter and energy from terrestrial and sea-floor end-members. In fact, global ocean and climate systems are significantly impacted by deep-sea processes. Seafloor vents and seeps appear to act as geologic exchange conduits, returning recycled materials to the hydrosphere to sustain another generation of life. Despite submarine seepage having control on global elemental cycling, it is estimated that less than 1% of the deep-sea has …

Simulation Of A Continuum Tumor Model Using Distributed Computing., Dylan A Goodin Jul 2019

Simulation Of A Continuum Tumor Model Using Distributed Computing., Dylan A Goodin

Electronic Theses and Dissertations

Mathematical modeling aims to provide a theoretical framework for understanding tissue dynamics and for establishing treatment response for diseased tissues, such as tumors. Previously published continuum models have successfully represented idealized two-dimensional and three-dimensional tissue for short periods of time. A recently published continuum model of cancer increases model complexity and describes three-dimensional tissue that, due to the required complexity of the geometric multigrid solver, can only be feasibly applied to millimeter-scale simulations. Furthermore, the computational cost for such models has hindered their application in the laboratory and in the clinic. With computational demands greatly outpacing current openMP-based approaches on …
